Microvascular permeability is a pharmacologic indicator of tumor response to therapy, and it is expected that this biomarker
will evolve into a clinical surrogate endpoint and be integrated into protocols for determining patient response to antiangiogenic
or antivascular therapies. This review discusses the physiological context of vessel permeability in an imaging setting, how
it is affected by active and passive transport mechanisms, and how it is described mathematically for both theoretical and
complex dynamic microvessel membranes. Many research groups have established dynamic-enhanced imaging protocols for estimating
this important parameter. This review discusses those imaging modalities, the advantages and disadvantages of each, and how
they compare in terms of their ability to deliver information about therapy-associated changes in microvessel permeability
in humans. Finally, this review discusses future directions and improvements needed in these areas. 相似文献

Imaging of the liver is undertaken for the detection and characterization of suspected primary or secondary neoplasms, prior to planning a surgery or chemotherapy pump placement, for assessing treatment response, for evaluating biliary pathology, and for screening for liver neoplasms in high-risk groups. In this article, we review the advantages and disadvantages of various imaging modalities in the evaluation of the liver and formulate guidelines for the imaging of common clinical indications. Imaging data have the potential to provide information on disease profiling pertaining to diagnosis, prognosis, selection of therapy, monitoring of response to therapy and pharmacokinetic information of drugs. Selection of the most appropriate imaging modality for a specific task will be vital for diagnosis, stratification, treatment response or treatment efficacy, toxicity assessment, and treatment outcome measures (progression-free survival). The EORTC Imaging Group was formed to establish and maintain the scientific and clinical value of advanced imaging in EORTC clinical trials. The group focuses on the development of specific analytical and review procedures as well as quality control procedures, in the context of clinical trials conducted by the EORTC groups. 相似文献
