Lymphocytes were collected from the brains of guinea pigs immunized with syngeneic spinal cord emulsified in complete Freund's adjuvant. Sixteen to 32 X 10(6) lymphocytes were inoculated into the vein of each inbred guinea pig (NIH 13). Recipient animals without clinical signs of experimental allergic encephalomyelitis (EAE) were sacrificed 20 days later. Some of the recipients had perivascular infiltrates of a large number of mononuclear cells mostly in choroid plexus and meninges of the brain and spinal cord. Demyelination, which was not so intense, was also observed in the vicinity of the perivascular infiltrates. Thus, the lymphocytes from the brain as well as lymph node and spleen were clear to have the ability to transfer EAE.  相似文献

在建立外周血液淋巴细胞β受体测定法的基础上,动态地观察了实验性过敏性哮喘豚鼠外周血液淋巴细胞中β受体最大结合容量(Bmax)、cAMP含量,以及血浆中儿茶酚胺(CA)含量的变化,发现它们都呈明显的时相性改变。通过分析它们发生的时间过程和相互关系,认为:在过敏性哮喘时所观察到的淋巴细胞β受体Bmax值的降低,乃是高水平内源性儿茶酚胺所致受体失敏的结果;并对所观察到的变化的意义作了初步探讨。  相似文献

In 18 immature guinea pigs both sides of the vasa efferentia were ligated; 18 control animals were not altered. The ligation did not prevent the formation in mature animals of active spermatogenesis in the majority of seminiferous tubules, but did cause a reduction in the number of layers of seminiferous epithelium in some, and atrophy of the epididymis due to the absence of germ cells. Immunization, in the adult stage, with homogenate of homologous testis mixed with complete Freund adjuvant created changes in the permeability of the blood-testis barrier in relation to endogenous globulins and rivanol and the development of autoimmune orchitis of the same intensity and terms as in the control animals. In contrast to the control group, destructive changes in the testis did not follow with inflammatory processes in the epididymis.  相似文献

目的探讨哮喘豚鼠白介素-5(IL-5)、粒细胞-巨噬细胞集落刺激因子(GM-CSF)及CD4+和CD8+T细胞在哮喘发病中的作用及雷公藤的干预作用.方法实验分为哮喘组、雷公藤治疗组(处理组)和对照组,每组各10只豚鼠,采用原位杂交方法检测豚鼠外周血淋巴细胞的IL-5、GM-CSFmRNA表达;应用免疫细胞化学方法检测外周血淋巴细胞CD4+和CD8+的表达.结果哮喘组外周血淋巴细胞IL-5、GM-CSF-mRNA表达均明显高于处理组和对照组(P<0.01),而处理组和对照组无显著差异.哮喘组外周血淋巴细胞CD4+表达明显高于对照组和处理组(P<0.01),CD8+表达低于对照组和处理组(P<0.05).结论哮喘豚鼠外周血淋巴细胞IL-5、GM-CSFmRNA表达增高、CD4+表达增高,CD8+表达降低,而雷公藤甲素可降低外周血淋巴细胞IL-5、GM-CSFmRNA表达,并可提高CD8+表达,降低CD4+表达.表明雷公藤可能在抗哮喘气道炎症中发挥一定作用.  相似文献

The anti-allergic activity and mechanism of cinnarizine was investigated in guinea pigs. Nifedipine, a calcium antagonist, and tranilast, a potent, orally active anti-allergic agent, were used as comparative drugs. Cinnarizine protected against fatal systemic anaphylactic shock in guinea pigs passively sensitized with IgE antibody. Cinnarizine reduced many of the features of severe respiratory disorders. Nifedipine and tranilast showed similar effects. Cinnarizine and nifedipine inhibited the contractile response to antigen of sensitized tracheal smooth muscle when the challenge was carried out at low antigen concentrations. Tranilast showed a tendency to inhibit the antigen-induced contraction of tracheal smooth muscle. Cinnarizine and nifedipine inhibited Ca-induced contraction in potassium-depolarized tracheal smooth muscle, tranilast had no effect. Cinnarizine showed antagonistic action to the contraction by histamine or leukotriene D4 (LTD4) of tracheal muscle. Nifedipine showed similar antagonistic action, although its potency is lower than cinnarizine. Tranilast showed slight antagonistic action to LTD4. Antigen-induced release of histamine and slow reacting substance of anaphylaxis (SRS-A) from sensitized lung tissues was inhibited by nifedipine and tranilast but not by cinnarizine. The release of histamine and SRS-A from lung tissues by calcium ionophore A23187 was inhibited by nifedipine and tranilast but not by cinnarizine. These results suggest that the anti-allergic action of cinnarizine is mainly due to the antagonistic action to allergic mediators and not by interfering with the release of mediators. Cinnarizine's mechanism seems to be related to its antagonistic action to Ca in smooth muscle, but not to the transport of Ca in releasing the anaphylactic chemical mediators in mast cells and other target cells.  相似文献

Lymphocytes from the cervical lymph nodes of guinea pigs were incubated in medium No. 199 for 24 h in the presence of cortisol in a concentration of 20 or 100 g%. The survival rate of the lymphocytes and their cortisol metabolism were determined and the nucleic acid content estimated cytophotometrically. A considerable decrease was found in cortisol metabolism by the lymphocytes from the 6th day after addition of an encephalitogenic mixture and there was a marked increase in the cortisol-resistant population of lymphocytes in guinea pigs with encephalomyelitis on the 17th–30th days after injection of the complete adjuvant. Cortisol in a concentration of 100 g% lowered the nucleic acid content of the lymphocytes of the intact animals but had no effect on lymphocytes of guinea pigs of the two experimental groups during the period of a considerable increase in the cortisol-resistant population. Progesterone depressed the lympholytic action of cortisol and the metabolism of this hormone by lymphocytes of intact guinea pigs. The ability of progesterone to reduce the lympholytic action of cortisol was weakened in guinea pigs with encephalomyelitis and guinea pigs receiving the adjuvant.Department of General Pathology, N. The effect of amlexanox given orally for 3 weeks was studied on the IgE-mediated experimental allergic rhinitis in the actively sensitized guinea pigs. The intranasal instillation of antigen (egg albumin) induced the increase of nasal vascular permeability (dye leakage), histamine content in nasal perfusate and nasal resistance in sensitized guinea pig. Amlexanox, 20 and 60 mg/kg/day given orally for 3 weeks significantly inhibited the increase of dye leakage into the nasal cavity, histamine content and nasal resistance in a dose-dependent manner. These results suggest that amlexanox given orally may be useful therapeutic agent for human allergic rhinitis.  相似文献

Experimental allergic encephalomyelitis (EAE) was induced in guinea pigs by injection of myelin from homologous brain in Freund's complete adjuvant. In the incubation period of EAE some animals showed a marked increase in the spontaneous proliferative activity of the blood lymphocytes, assessed in cultures using thymidine-H3. With the development of clinical manifestations of the disease, the spontaneous activity of the lymphocytes reached a maximum and was exhibited in the overwhelming majority of animals. When the clinical picture of EAE was well marked, the response of the lymphocytes to stimulation by myelin preparations was observed mainly if the spontaneous mitotic activity of the cells was low or only moderately increased. Specific antigenic stimulation in vivo connected with the immunopathological process could be the cause of the increasing spontaneous activity of the lymphocytes. The state of nonspecific reactivity of the lymphocytes to phytohemagglutinin in EAE was indistinguishable from that in the control animals.  相似文献

Ribonucleic acid (RNA) extracts were obtained from lymph nodes of guinea-pigs that had previously been immunized with a purified testicular antigen emulsified in Freund's complete adjuvant. The RNA extracts were incubated with normal guinea-pig peritoneal exudate cells (NGP-PEC). After treatment, the NGP-PEC cells showed specific inhibition of migration when tested with the specific antigen. No inhibition of migration was observed when cells were tested with an unrelated antigen or when the cells were incubated with RNA obtained from animals immunized with adjuvant alone. Failure of inhibition of migration was also observed when the 'immune' RNA was degraded with RNAse. The appearance of this I-RNA in the immunized guinea-pigs correlates with the appearance of delayed hypersensitivity in vivo.  相似文献
