Objective: To investigate the expression of MMP-13 in non-small cell lung cancer (NSCLC), so as to analyze its correlation with prognosis of NSCLC. Methods: MMP-13 expression was detected in 99 NSCLC tissues and 32 normal lung tissues by immunohistochemical method. Results: (1) Expression of MMP-13 (51.5%, 51/99) in cancer tissues was significantly higher than that in normal tissues 0% (P < 0.05). Expression level of MMP-13 was significantly related to lymph node metastasis and clinical stage (P<0.01). (2) Kaplan-Meier analysis showed that expression level of MMP-13 was closely correlate with the prognosis of NSCLC. Multivariate Cox model analysis suggested that the survival time was significantly related to clinical stage and the expression of MMP-13. Conclusion: MMP-13 is an independent factor that affect prognosis.  相似文献

The Rho-specific guanine nucleotide exchanging factor p114RhoGEF is involved in RhoA activation and cell motility. Previous studies suggest that altered expression of p114RhoGEF could contribute to cancer progression. We investigated an association of p114RhoGEF expression with progression and prognosis of non-small cell lung cancer (NSCLC). Immunohistochemistry was performed to detect p114RhoGEF expression in 105 NSCLC (34 adenocarcinoma and 71 squamous-cell carcinoma) and 32 normal lung tissues. We found that p114RhoGEF expression was upregulated in squamous-cell lung carcinoma and that p114RhoGEF expression was significantly higher in squamous-cell carcinoma than in adenocarcinoma or normal tissues (P?<?0.05, both). Expression of p114RhoGEF protein was significantly associated with lymph node metastasis of lung cancer (P?<?0.05), but not with patients’ age, gender, tumor size, differentiation, or stage. Expression of p114RhoGEF protein was also associated with poor overall and event-free survival of squamous-cell lung carcinoma patients (P?<?0.05). Taken together, p114RhoGEF expression may be useful in predicting progression and survival of squamous-cell lung carcinoma patients. A future study will investigate whether p114RhoGEF can serve as a novel therapeutic target in squamous-cell lung cancer.  相似文献

目的探讨基质金属蛋白酶(matrixmetalloproteinases,MMPs)MMP-2和MMP-3在肺癌组织中的表达及与患者临床病理特征的关系。方法用免疫组化S-P染色技术对45例非小细胞肺癌标本及20例癌旁正常肺组织标本中MMP-2和MMP-3的表达水平进行检测,并结合患者的临床病理资料对其结果进行对比分析。结果MMP-2和MMP-3在肺癌组织中的阳性表达率明显高于癌旁正常肺组织(P<0.05),MMP-2的表达与肺癌临床分期、组织分化程度、淋巴结转移有密切关系(P<0.05〉,而MMP-3的表达与上述临床病理特征无明显关系(P>0.05)。结论MMP-2、MMP-3均是良好的肺瘤标记物,MMP-2与肺癌侵袭转移关系密切,MMP-3可能在肺癌侵袭转移中作用不显著。  相似文献

Objectlve:To investigate the expression characteristics of Livin and second mitochondrial activator of Caspase(Smac) proteins in non-small cell lung cancer(NSCLC),and analyze their effect on patients'prognosis.Methods:The expressions of Livin and Smac proteins were detected in 89 NSCLC tissue samples and 25 normal lung tissue samples by immunohistochemical technique.Results:The positive expression rates of Livin and Smac proteins in NSCLC tissues were 53.9%.and 58.4%respectively,higher than that in normal lung tissues(P<0.01).Livin protein expression correlated with Smac protein significantly(χ2=18.451,P=0.000,r=0.455).The expression level of Livin protein was closely related to lymph node metastasis,TNM stage and histological type(P<0.05),but not to sex,age,differentiation grade(P>0.05).The expression level of Smac protein was closely related to lymph node metastasis,TNM stage(P<0.01),but not to sex,age,histological types(P>0.05).Kaplan-Meier analysis revealed a significant impact on survival by Livin protein in NSCLC (P<0.01),but not by Smac protein(P>0.05).Conclusion:Overexpression of Livin protein may play a promoting role in the occurrence and progression of NSCLC.Moreover,it may bring an adverse effect on patients'prognosis.Although overexpression of Smac protein affects the occurrence and progression of NSCLC,it has no relationship with the prognosis.Livin protein may be helpful to evaluate the progression of NSCLC,and to predict the prognosis.  相似文献

Objective: To study the expressive characteristics of iNOS to the prognosis NSCLC patients. Methods: The expression in non-small cell lung carcinoma (NSCLC) and it's affection of iNOS was detected in 50 NSCLC tissues by Immunohistochemistry technology. Results: There was positive expression in NSCLC (positive rate 38.00%). The expression of iNOS had a correlation with lymph node metastasis (P〈0.05). There was no significant difference between positive expression of iNOS and lung cancer histological types, TNM stages, T stages (P〉0.05). But the data suggested that there was a direct correlation between expression of iNOS and clinical prognosis (P〈0.05). Conclusion: Positive expression of iNOS was observed in NSCLC, which correlated with postoperative survival time and lymph node metastasis. Although iNOS level had no significant relation with T stage, we find the positive rate in T2 stage was much higher than that in TI stage. The result suggested that positive expression of iNOS was related to bad clinical prognosis because of promoting tumor growth and metastasis.  相似文献

背景与目的:基质金属蛋白酶26(matrix metalloproteinase-26,MMP-26)与多种恶性肿瘤的发生、发展及转移相关。本实验通过检测MMP-26蛋白在浸润性非小细胞肺癌(non-small cell lung cancer,NSCLC)、浸润前肺癌和正常肺组织中的表达.探讨MMP-26在NSCLC发生发展中的作用及与预后的关系。方法:采用免疫组织化学(SP)法,分别检测72例NSCLC、14例非典型增生和10例正常肺组织中MMP-26蛋白的表达。结果:MMP-26蛋白高表达率在正常肺组织中为0(0/10),在非典型增生巾为14.3%(2/14),在NSCLC中为59.7%(43/72)。MMP-26蛋白在NSCLC中的表达高于在非典型增生及正常肺组织中(P〈0.01),在非典型增生中高于正常肺组织中,但二者差异无统计学意义(P〉0.05)。MMP-26蛋白表达与NSCLC分期(P〈0.05)和淋巴结转移有关(P〈0.05),而与患者年龄、性别以及肿瘤大小、分化无关(P〉0.05)。Cox比例风险模型进行多因素生存分析显示:MMP-26表达和临床分期是有意义的NSCLC预后指标(P〈0.05)。MMP-26蛋白高表达的NSCLC患者无复发生存期和总生存期低于阴性表达的患者(分别为log-rank=19.34、23.2,P〈0.001、0.001)。结论:MMP-26蛋白高表达与NSCLC发生、淋巴结转移、临床分期及预后相关,有可能作为判断NSCLC进展和预测预后的一项指标。  相似文献

目的:探讨MMP-2、MMP-9、TI MP-1和TI MP-2在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达及临床意义。方法:用免疫组化法测定124例NSCLC组织、124例癌旁组织和10例良性肺病组织(错构瘤)中MMP-2、MMP-9、TI MP-1和TI MP-2的表达。结果:在NSCLC组织中,MMP-2、MMP-9、TI MP-1和TI MP-2表达水平显著高于癌旁组织和良性肿瘤(P均〈0.05)。MMP-2在鳞癌组织中的表达水平显著高于腺癌和腺鳞癌(P〈0.05);TI MP-2在鳞癌中的表达水平显著高于腺癌(P〈0.05);MMP-9和TI MP-1在腺癌组织中的表达水平显著高于鳞癌(P〈0.05)。在NSCLC组织中,随着TNM分期增加,MMP-2、MMP-9、TI MP-1和TI MP-2的表达水平增加(P均〈0.05)。MMP-2、MMP-9、TI MP-1和TI MP-2在淋巴结转移组中的表达水平显著高于无淋巴结转移组(P均〈0.05)。MMP-2、MMP-9、TI MP-1和TI MP-2在低分化组中的表达水平显著高于高分化组(P均〈0.05)。MMP-9在女性组中的表达水平显著高于男性组(P〈0.05)。MMP-2、MMP-9、TI MP-1和TI MP-2之间的表达水平呈显著性正相关(P均〈0.05)。结论:MMP-2、MMP-9、TI MP-1和TI MP-2均参与NSCLC的形成且与NSCLC临床病理特征关系密切,同时可能是判断预后的较好评价指标。  相似文献

目的:检测MMP-1、MMP-3、MMP-7和尿激酶纤溶酶原激活物受体(uPAR)在非小细胞肺癌(NSCLC)的表达水平,分析其表达水平与肺癌侵袭转移以及相关临床病理特征之间的关系。方法:免疫组织化学方法检测66例NSCLC和10例正常肺组织中MMP-1、MMP-3、MMP-7和uPAR的蛋白表达水平。结果:免疫组织化学检测显示NSCLC组织MMP-1和MMP-3表达率较低,而MMP-7和uPAR表达率较高,分别达到55%和61%,明显高于正常肺组织。MMP-7的表达与NSCLC淋巴结转移、分化程度以及预后密切相关;uPAR的表达与NSCLC预后密切相关,与相关临床病理因素无关;MMP-7和uPAR的表达呈正相关。结论:MMP-7和uPAR的表达可作为NSCLC患者预后评价的指标,可能成为抗肿瘤治疗的靶点。  相似文献

目的:探讨组织蛋白酶D(CD)、表皮生长因子受体(EGFR)、转移抑制基因nm23、抑癌基因p16在非小细胞肺癌(NSCLC)中表达的意义。方法:采用快速免疫组化法检测64例肺癌标本四种蛋白的表达。结果:肺癌组织CD、EGFR、nm23和p16表达的阳性率分别为65.6%、67.2%、60.9%和51.6%,p16蛋白表达缺失率达48.4%,CD和EGFR表达与肺癌患者临床分期(P<0.05,P<0.01)、淋巴结转移(P<0.01,P<0.05)密切相关。COX比例风险模型分析和Kaplan-Meier生存曲线分析显示CD和EGFR是影响肺癌患者预后的重要因素(P<0.05)。结论:在NSCLC中nm23、p16、CD和EGFR异常改变的频率很高,在NSCLC的发展中可能起促进作用。CD和EGFR可作为评价NSCLC患者的预后指标。  相似文献

目的:分析肺腺癌患者癌组织中微管关联蛋白4(MAP4)的表达水平及其与病理参数、预后的关系。方法:选取我院2014年1月至2015年1月82例非小细胞肺癌(NSCLC)患者,利用免疫组织化学染色法对手术切除的癌组织及其癌旁正常组织的MAP4蛋白表达水平进行检测,并分析MAP4蛋白表达与患者临床病理参数和预后的关系。结果:MAP4蛋白在肺腺癌组织中的高表达率和表达水平均明显高于癌旁正常组织(P<0.05);其表达水平与肺腺癌患者的肿瘤浸润分期、淋巴结转移、分化等级相关(P<0.05)。3年随访显示MAP4蛋白高表达组患者的中位生存时间、无病生存率、总生存率明显低于低表达组(P<0.05);Logistic回归分析MAP4表达水平、肿瘤浸润分期、淋巴结转移、TNM分期、分化等级均是肺腺癌患者预后不良的危险因素。结论:肺腺癌患者肿瘤组织中MAP4蛋白水平高表达可能会促进患者的病程发展,且是预测患者不良预后的危险因素。  相似文献

目的检测白细胞介素-17(IL-17)和基质金属蛋白酶-9(MMP-9)在汉族、维吾尔族NSCLC患者中的表达,分析其表达在两民族中是否有差异,探讨其临床意义。方法随诊资料完整的术后NSCLC患者组织标本60例,其中汉族、维吾尔族各30例,采用PV9000二步免疫组织化学法检测IL-17和MMP-9在癌组织及癌旁组织中的表达。 结果IL-17、MMP-9在NSCLC中表达的阳性率为78.3%、80.0%,在癌旁组织中分别为48.3%、35.0%,癌组织中IL-17与MMP-9的表达高于癌旁组织(P<0.05);IL-17的表达与肿瘤的肿块大小、淋巴结转移和TNM分期呈正相关(P<0.05),MMP-9与肿瘤大小、TNM分期呈正相关,差异有统计学意义(P<0.05);IL-17和MMP-9的表达与患者的民族、年龄、性别、病理类型及分化程度差异无统计学意义(P>0.05);汉族与维吾尔族的术后生存差异无统计学意义(P<0.05),IL-7和MMP-9的阴性表达组的3年生存率较阳性表达组高,差异有统计学意义(P<0.05)。结论IL-17和MMP-9在癌组织中的表达不是汉族与维吾尔族患者在本院构成比差异的主要原因;IL-17和MMP-9的高表达对NSCLC的侵袭和转移起着重要作用;IL-17和MMP-9的高表达与肺癌的不良预后有关。  相似文献

背景与目的非小细胞肺癌(non-small cell lung cancer,NSCLC)的预后与众多因素有关,凋亡相关基因在肿瘤的发生发展过程中发挥重要的作用,但其在NSCLC中作为生物预后因素的意义还不明确。本研究的目的是探讨凋亡相关基因caspase-3、survivin、k-ras在NSCLC中的表达与不同临床病理特征和预后的关系,并探讨三者在NSCLC发生过程中可能存在的相关性。方法采用免疫组化SP二步法检测91例石蜡标本癌组织和癌旁正常组织中caspase-3、survivin、k-ras蛋白的阳性表达。结果Caspase-3在癌组织中阳性表达率低于癌旁正常组织,survivin、k-ras在癌组织阳性表达高于癌旁正常组织,均具有统计学差异(P<0.05)。Caspase-3阳性表达与年龄、病理类型、TNM分期、淋巴结状态、分化程度有关(P<0.05),而与病灶大小、有无吸烟史等无关(P>0.05);survivin阳性表达与年龄、TNM分期、分化程度、淋巴结状态有关(P<0.05),而与病理类型、病灶大小、有无吸烟史等无关(P>0.05);k-ras阳性表达与病理类型、吸烟有关(P<0.05),与年龄、TNM分期、淋巴结状态、分化程度、病灶大小等均无关(P>0.05)。Caspase-3与survivin的阳性表达呈负相关(r=-0.662,P<0.001)。Cox多因素分析survivin,caspase-3,TNM分期、病理类型、分化程度5个因素具有独立预后价值(P<0.05)。结论Survivin和k-ras在NSCLC中高表达,而caspase-3在NSCLC中低表达,可能在肿瘤细胞凋亡过程中起着重要的作用;Caspase-3低表达、survivin高表达可作为术后NSCLC预后不良的独立性因素;K-ras与NSCLC预后的关系需要进一步研究。  相似文献

目的:检测Syntenin在非小细胞肺癌(NSCLC)组织中的蛋白表达水平并探讨其临床意义。方法:采用组织芯片和免疫组织化学染色法,检测Syntenin在147例NSCLC及相应癌旁组织中的表达水平,并分析Syntenin的表达与NSCLC患者临床病理指标及预后的关系。结果:Syntenin在NSCLC组织中的阳性表达率(49.7%)显著高于癌旁组织(4.1%)(P < 0.01)。Syntenin的表达与NSCLC的淋巴结转移、临床分期和病理类型显著相关(P值分别为0.039、0.038、0.024),与患者的性别和年龄无显著相关(P > 0.05)。Kaplan-Meier分析表明,Syntenin的表达与NSCLC患者的总生存期显著相关(P=0.028)。结论:Syntenin在NSCLC组织中高表达,并且其高表达预示了患者的不良预后。  相似文献

BACKGROUND: Matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) play a role in the processes of extracellular matrix degradation. Changes in their expression levels have been observed in various tumor types, including lung carcinoma. However, their clinical significance and their prognostic importance in the progression of nonsmall cell lung carcinoma (NSCLC) remain to be specified. In this study, mRNA expression levels of MMP-1, MMP-9, TIMP-1, and TIMP-2 were evaluated in patients with resected NSCLC, and their associations with disease progression and prognosis were determined. METHODS: Between June 1996 and December 1999, 116 patients underwent resection for NSCLC. Expression levels of MMPs and TIMPs were evaluated using Northern blot analysis in these NSCLC tissue samples and in 39 matched samples of normal lung tissue. RESULTS: MMP-1, MMP-9, and TIMP-1 expression levels were increased in tumor samples compared with matched, corresponding normal tissues. In contrast, TIMP-2 expression was decreased in tumor samples. MMP-1 tumor expression was correlated significantly with the evolution of lymph node status and tumor-lymph node-metastasis (TNM) stage. In contrast, MMP-9 tumor expression was correlated significantly with increased T stage. TIMP-1 overexpression was an independent predictor of worse survival in patients with NSCLC that was not associated with other prognosis factors, such as TNM stage. CONCLUSIONS: The overexpression of TIMP-1 was an independent prognostic marker in patients with NSCLC, and evaluating TIMP-1 may be important for identifying patients who are at greater risk of disease recurrence.  相似文献

Objective: This study was designed to detect the protein expression of VEGF, EGFR and MMP-9, to investigate the potential roles they might play in the pathogenesis of NSCLC and to discuss their relationship and their clinical significance. Methods: For 136 cases who were diagnosed of NSCLC, immunohistochemistry was used to detect protein expression of VEGF, EGFR and MMP-9 tumour and normal lung tissue specimens. Statistical analysis was performed on the relationship between protein expression of VEGF, EGFR and MMP-9 and clinico-pathological parameters and prognosis. Results: Expression of VEGF and MMMP-9 was mostly in the cytoplasm, while EGFR wasfound in both cytoplasm and membranes. In NSCLCs, the positive rate of VEGF protein was 79.4% (108/136), for EGFR was 75.0% (102/136) and for MMP-9 was 68.4% (93/136), significantly greater than the normal tissue values of 16.0% (8/50), 12.0% (6/50) and 8.0% (4/50), respectively (P<0.01). Expression of VEGF, EGFR and MMP-9 was related to pathology grading, lymph node metastasis and clinical staging in NSCLC (P<0.01), while being independent of other clinicopathologic parameters (P>0.05). There was an obvious positive correlation among the expression of VEGF and EGFR (r=0.25; P<0.01), VEGF and MMP-9 (r=0.28; P<0.01), EGFR and MMP-9 (r=0.19; P<0.05) in NSCLCs. Conclusions: Protein expression of VEGF, EGFR and MMP-9 is elevated higher in NSCLC, correlating with progression.  相似文献

目的:本研究阐明了非小细胞肺癌(NSCLC)组织及其细胞系内基因长链非编码RNA00673(long intergenic non-coding RNA,LincRNA00673)的表达水平,以及Linc00673 在肺癌中的临床意义及与患者预后的相关性。方法:利用逆转录-聚合酶链反应(qRT-PCR)的实验方法,检测正常肺细胞系与肺癌细胞系之间及肺癌组织与癌旁组织之间Linc00673 的表达水平差异,并分析Linc00673 与肺癌临床病理特征及预后之间的相关性。结果:肺癌细胞系比正常肺细胞系中的Linc00673的表达明显升高,同时肺癌组织比癌旁组织中的Linc00673 明显高表达(P<0.01);Linc00673的表达与肿瘤的分化程度(P=0.002)、淋巴结转移(P=0.001)、远处转移(P=0.005)和临床分期(P=0.001)相关。并且在吸烟的肺癌患者中Linc00673的表达水平高于非吸烟肺癌患者。肺癌的Linc00673表达水平增高与肺癌患者的预后不良相关(P<0.01)。结论:Linc00673 的表达与肺癌的发生发展及预后密切相关,有可能成为一个具有特异性的预测性肿瘤分子标志物。  相似文献

目的探讨丝/苏氨酸蛋白激酶1(AKT1)表达与非小细胞肺癌(NSCLC)病理学特征及预后的关系。方法选取经病理学检查证实的NSCLC患者90例,分别取其癌组织、癌旁组织为NSCLC组和癌旁组,采用免疫组化染色法检测2组标本中的AKT1蛋白表达水平,并分析AKT1蛋白与NSCLC患者TNM分期、淋巴结转移情况、肿瘤分化程度、病灶的大小及预后等指标的关系。结果NSCLC组标本中的AKT1蛋白阳性表达率(58.89%)显著高于癌旁组(11.11%),差异具有统计学意义(P<0.05)。高分化及中分化、TNM分期(Ⅰ期+Ⅱ期)、未发生淋巴结转移的NSCLC组中AKT1蛋白阳性表达率低于低分化、TNM分期(Ⅲ期和Ⅳ期)及发生淋巴结发生转移NSCLC组织(P<0.05),不同年龄、性别、病灶大小及病理学类型的NSCLC组织中AKT1蛋白阳性表达率差异无统计学意义(P>0.05)。NSCLC组标本中的AKT1蛋白阳性表达与阴性表达患者的1年、2年生存率差异无统计学意义(P>0.05),AKT1蛋白阳性表达患者的3年生存率(13.21%)显著低于AKT1蛋白阴性表达患者的37.84%(P<0.05)。结论NSCLC患者癌组织中的AKT1蛋白表达上调,并且与肿瘤进展及不良预后关系密切。  相似文献

目的: 检测人非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中成纤维细胞活化蛋白(fibroblast activation protein,FAP)mRNA的表达,探讨FAP mRNA与NSCLC临床特征及预后之间的关系。 方法: 2004年6月至2006年12月选取天津医科大学附属肿瘤医院组织库中247例NSCLC患者瘤组织标本和对应的48例癌旁组织标本,患者随访至2011年12月1日。采用实时荧光PCR检测NSCLC组织和正常肺组织中FAP mRNA。临床病理参数与FAP mRNA相对表达量的相关性分析采用Mann-Whitney U检验,以Kaplan-Meier法绘制生存曲线,用log-Rank检验比较患者的生存差异,采用Cox回归模型评估独立的预后因素。 结果: FAP mRNA在NSCLC组织中过表达。NSCLC组织中FAP mRNA表达水平与NSCLC患者KPS评分成正相关(P<0.05),而与肿瘤原发部位、肿块大小、淋巴结转移、临床分期、组织学类型等其他病理特征无明显关系(P>005)。高表达FAP mRNA 的NSCLC患者与低表达者相比,中位总生存时间(overall survival,OS)无明显差异(43 vs 39个月,P>0.05)。进一步以组织学类型分层分析显示,肺腺癌患者FAP mRNA表达量与临床分期成负相关(P=0.031),与KPS评分成正相关(P=0.041)。高表达FAP mRNA的肺腺癌患者与低表达者相比,中位OS时间显著延长(42 vs 26个月,P<005),多因素分析进一步证实FAP mRNA表达是影响肺腺癌患者预后的独立因素。 结论: FAP在NSCLC组织中高表达,FAP mRNA高表达与肺腺癌的临床分期呈负相关,并与肺腺癌患者的预后密切相关。  相似文献
