盐酸恩丹西酮缓释片的研制及体外释药特性

制备盐酸恩丹西酮缓释片并对影响释药的因素进行考察。以羟丙甲纤维素（HPMC）为骨架材料制备了盐酸恩丹西酮缓释片，通过正交设计试验优选最佳处方和工艺。对影响释药的因素，如HPMC的黏度和用量、填充剂的种类、制片工艺、润湿剂及释放介质pH等进行了考察。盐酸恩丹西酮缓释片体外释药符合Higuchi方程，HPMC的黏度、填充剂的种类及测定释放度的转速对该缓释片的释药几乎无影响，而制片工艺、润湿剂及释放介质pH值对缓释片释药影响较大。盐酸恩丹西酮缓释片在体外12h缓释效果较好。     

应用生物可降解材料玉米醇溶蛋白制备胃肠道生物黏附控释片

背景：胃肠道生物黏附控释制剂能延长药物制剂在胃肠道的停留时间,提高药物的生物利用度。 目的：制备5-氟尿嘧啶胃肠道生物黏附控释片。 方法：利用生物可降解性玉米醇溶蛋白为骨架材料和黏附材料,氟尿嘧啶为模型药,制备氟尿嘧啶胃肠道黏附控释片。对片芯工艺进行正交设计,优化包衣液的选择,观察生物黏附缓释片的体外黏附力及体内外相关性。 结果与结论：5-氟尿嘧啶玉米醇溶蛋白生物黏附片体外释放10 h内均符合零级释放特征,片剂具有较好的体外黏附力,且在2~8 h 内体内血药浓度较为平稳,没有明显峰谷现象,体内外释放吸收具有良好的相关性。     

法莫替丁颗粒、片剂药代动力学及生物利用度研究

本文报道8名成年男性健康受试者单剂口服法莫替丁颗粒、片剂药代动力学及相对生物利用度比较。8名受试者接受单剂量(40mg)交叉给药(颗粒剂和片剂)后所得药时曲线符合一室模型。其颗粒剂和片剂的平均药代动力学参数分别为:血浆药物峰浓度:110.29±23.59与112.95±23.36μg/L,药物达峰时间:1.66±0.59与1.97±0.53h,消除半衰期:3.53±0.33与3.18±0.53h,药—时曲线下面积:737.11±147.09与721.42+100.94μg/L·h。其相对生物利用度以片剂为100％,颗粒剂的相对生物利用度为102.96±20.05％。     

丙戊酸镁缓释片对精神分裂症兴奋状态的疗效观察

目的探讨丙戊酸镁缓释片联合抗精神病药治疗精神分裂症兴奋状态的疗效及安全性。方法将64例有兴奋状态的精神分裂症患者随机分为两组,丙戊酸镁缓释片联合抗精神病药组(合用组)32例,单用抗精神病药组(单用组)32例治疗4周。采用简明精神病量表(BPRS)和副反应量表(TESS)评定。结果治疗4周后合用组较单用组的BPRS评分显著降低(t=6.00,P<0.01),治疗显著而快,不良反应少(t=2.80,P<0.01)。结论丙戊酸镁缓释片联合抗精神病药治疗精神分裂症兴奋状态起效快,疗效好。     

国产和进口盐酸环丙沙星片剂人体药代动力学和生物利用度比较

本文报道8名成年男性健康受试者单剂口服环丙沙星片剂药代动力学及相对生物利用度比较。8名受试者接受单剂量(500mg)交叉给药(国产和进口)后所得药时曲线符合二室模型。其国产和进口片剂的平均药代动力学参数分别为:血浆药物峰浓度:2.91±0.29与2.80±0.20mg/L,药物达峰时间:1.22±0.19与1.46±0.28h,消除半衰期:3.95±0.57与3.68±0.63h,药-时曲线下面积:16.39±2.27与16.91±1.96mg/L·h。其相对生物利用度以进口片剂为100％,国产片剂相对生物利用度为96.97±8.71％。     

法莫替丁片剂的健康人体药代动力学及生物利用度研究

本文报道8名成年健康男性受试者单剂的口服两种法莫替丁片剂的药代动力学及相对生物利用度比较。实验结果表明:8名受试者接受单剂(40mg)交叉给药后所得药时曲线符合一室模型。两种片剂的药代动力学参数分别为:AUC:854.29与803.45μg·h/L,T_(1/2)ke:2.61与2.92h,Cmax:129.44与116.22μg/L,Tmax:2.48与2.52h。     

氨酚氯雷伪麻双层片的处方设计

目的 研究亲水凝胶型骨架片氨酚氯雷伪麻双层缓释片的处方工艺,并对缓释片制备过程中的几点影响因素进行了初步探讨.方法 以氯雷他定的含量均匀度为考察指标,通过正交试验筛选出最佳处方工艺,并考察羟丙甲基纤维素(HPMC)用量对氨酚氯雷伪麻缓释片体外释放度的影响.结果 按最佳处方制备的氨酚氯雷伪麻缓释片,对乙酰氨基酚和硫酸伪麻黄碱释放度达到要求,氯雷他定含量均匀度符合要求.结论 按优化处方制备的氨酚氯雷伪麻缓释片符合相关规定.     

载表柔比星的壳聚糖纳米粒的制备及其特性研究

目的 制备经聚乙二醇修饰的壳聚糖纳米粒(PEG/CS NP),并负载表柔比星(EPI),研究载表柔比星的壳聚糖纳米粒(PEG/CS-EPI NP)体外释药性能.方法 应用阴离子凝聚法制备PEG/CS-EPI NP,透射电镜观察纳米粒的形态特征,激光粒度分析仪测定粒径大小,紫外分光光度法测定纳米粒的载EPI量,动态透析法考察载EPI纳米粒的体外释放特性.结果 当壳聚糖与三聚磷酸钠质量比为6∶1,壳聚糖与EPI质量比为8∶1时,制备的PEG/CS-EPI NP呈圆形或椭圆形,分散性良好,平均粒径(322.1±14.4)nm,载EPI量为(13.9±1.1)%,包封率(74.2±1.8)%,72 h累积释药率达(82.0±2.1)%.结论 采用阴离子凝聚法制备的PEG/CS-EPI NP形状规则、粒度分布均匀,具有较高包封率和较好缓释性能.     

骨形态发生蛋白2/聚乳酸缓释微球的制备及表征

背景:聚乳酸具有良好的生物相容性,是优良的药物缓释载体。目的:制备重组人骨形态发生蛋白2/聚乳酸缓释微球,考察其理化特性。方法:采用复乳溶剂挥发法制备重组人骨形态发生蛋白2/聚乳酸缓释微球,进行扫描电镜、激光粒度、Zeta电位、溶胀性能检测及采用ELISA试剂盒检测包封率、载药率及体外释药率。结果与结论:扫描电镜见重组人骨形态发生蛋白2/聚乳酸缓释微球微球近似圆形,形态较规则,分散性较好,表面光滑。激光粒度分析重组人骨形态发生蛋白2/聚乳酸缓释微球微平均粒径839.6 nm,Zeta电位(-32.93±3.74)mV,微球溶胀系数1.157±0.059,包封率及载药率分别为(88.943±2.878)%,(0.026±0.001)%;微球在第1天释药约10.199%,随后释药较恒定,至第19天累计释药率为54.643%。说明制备出的重组人骨形态发生蛋白2/聚乳酸缓释微球的粒径达到中华人民共和国药典第10版二部关于亚微球的定义标准及包封率不低于80%的要求,并且在体外具有很好的缓释功能。     

甘草酸表面修饰万乃洛韦白蛋白纳米粒的制备及其肝靶向性研究

以万乃洛韦为模型药物 ,去溶剂化法制备普通载药纳米粒 ,结合高碘酸盐氧化法制备甘草酸 -万乃洛韦白蛋白纳米粒偶联物。对其表面甘草酸密度、形态、大小及其分布、体外释药特性、载药量、包封率、动物体内肝分布和体外肝细胞的摄取情况进行了研究。修饰纳米粒表面甘草酸密度为 9;平均粒径 d0 .5=2 6 8± 2 3nm;载药量1.35 % ;包封率 6 8.76 % ;体外释药符合双相动力学规律 ;对肝细胞具有选择靶向性。静注 15 min后 ,有 6 9.89%集中在肝脏 ,对照组为 6 4 .82 % ,二者之间存在显著差异 (P<0 .10 )。甘草酸表面修饰白蛋白纳米粒制备成功 ,为肝细胞靶向给药提供了新途径。     

Schizophrenia in a North Swedish geographical isolate, 1900–1977. Epidemiology, genetics and biochemistry

Over 200 schizophrenic patients belonging to three major and interrelated pedigree complexes have been investigated over the past 30 years in a North Swedish geographically isolated population, presently numbering about 6,000. An intensive investigation of a number of biochemical correlates and genetic markers in a few selected families belonging to one of the major pedigrees has indicated new strategies for the current research program.
Schizophrenia, as defined operationally, is significantly associated with decreased activities of two enzymes (1) blood platelet monoamine oxidase, (2) plasma dopamine-β-hydroxylase, and (3) with the genetic marker Gc² (group specific antigen). Both enzymes are subject to genetic variation. A positive score for linkage between schizophrenia and low plasma DBH activity has been calculated, but, so far, available data are insufficient for discrimination between linkage and partial contribution of genetically controlled low plasma DBH to the pathogenesis of the disease. Alternatively, both mechanisms could be involved.
As a model for continued research, schizophrenia is explained as based on a double dominant-recessive genotype (Aabb), representing a vulnerability which in about 50 % of cases develops into clinical schizophrenia. It is suggested that the dominant mutation (A) operates on or affects MAO activity, and that the recessive genotype (bb) is instrumental in low variates of DBH activity and very likely such variates within the normal range of physiological variation. Moreover, it is suggested that the combined effects of MAO- and DBH-reduced efficiency on the metabolism of e.g. dopamine could be an essential pathogenic mechanism for the schizophrenic illness which is segregating in this population.     

HLA in North Colombia Chimila Amerindians

HLA-A,-B,-C,-DRB1 and -DQB1 alleles have been studied in Chimila Amerindians from Sabana de San Angel (North Colombian Coast) by using high resolution molecular typing. A frequent extended haplotype was found:HLA-A*24:02-B*51:10-C*15:02-BRB1*04:07-DQB1*03:02 (28.7%) which has also been described in Amerinndian Mayos Mexican population (Mexico, California Gulf, Pacific Ocean). Other haplotypes had already been found in Amerindians from Mexico (Pacific and Atlantic Coast), Peru (highlands and Amazon Basin), Bolivia and North USA. A geographic pattern according to HLA allele or haplotype frequencies is lacking in Amerindians, as already known. Also, five new extended haplotypes were found in Chimila Amerindians. Their HLA-A*24:02 high frequencies characteristic is shared with aboriginal populations of Taiwan; also, HLA-C*01:02 high frequencies are found in New Zealand Maoris, New Caledonians and Kimberly Aborigines from Australia. Finally, this study may show a model of evolutionary factors acting and rising one HLA allele frequency (-A*24:02), but not in others that belong to the same or different HLA loci.

Starting with the integument, we see many organs are contractile sacs or multiples thereof, which tubes or bags constitute the major part of the entire body. Recognition of this basic unit and its characteristics sheds new light, individually and collectively, on many disorders previously considered unrelated. Muscular tears and perforations develop in the walls of these chambers, being no way peculiar to those organs, wherein, hydrochloric acid occurs. So, it is not necessary to explain the absence of excessive acid from patients who exhibit holes in the gastric, uterine, aortic, duodenal, rectal, pulmonary, retina, and other walls. Muscle, not acid is the great common factor relating idiopathic disorders in the gastrointestinal tract to each other and to similar diseases in other systems. When the units are linked together, the lesions tend to appear as arthropathies, i.e. at the joints. Rephrasing common-place observations, frees us from conventional, conceptual cul-de-sacs. An observation is only as good as its interpretation, so all possibilities must be considered, otherwise, we will remain blinded by our misconceptions.     

Der Einfluß von Nahrungsmitteln und Rauchen auf die klinisch-chemische Diagnostik von Phäochromozytom,Neuroblastom und Karzinoid-Syndrom

Zusammenfassung Der Einfluß von verschiedenen Nahrungsmitteln auf Methoden zur Bestimmung von Adrenalin (AD), Noradrenalin (NA), Vanillinmandelsäure (VMS), Metanephrinen (MN), Homovanillinsäure (HVS) und 5-Hydroxyindolessigsäure (5-HIE) im 24 h-Harn zur Diagnose des Phäochromozytoms bzw. Karzinoid-Syndroms wurde untersucht. Die in die Untersuchung einbezogenen Nahrungsmittel waren: Tee, Kaffee, Mandeln, Ananas, Käse, Walnüsse, Vanillepudding, Bananen, Tomaten und Milchschokolade. Außerdem wurde der Einfluß des Zigarettenrauchens auf die Bestimmung von AD, NA, VMS und MN untersucht.Walnüsse führten zu einer starken Erhöhung der 5-HIE-Ausscheidung. Bananen erhöhten die Ausscheidung von AD, NA, VMS, MN und 5-HIE. Kaffee und Ananas bewirkten eine geringe Zunahme der MN-Werte. Rauchen von 20–30 Zigaretten/Tag beeinflußte keine der vier Variablen.Wenn die beschriebenen Methoden benutzt werden, sollte lediglich auf den Verzehr von Bananen und Walnüssen vor und während der Harnsammelperioden verzichtet werden, da die oberen Normgrenzen im Harn überschritten werden könnten. Ein Verzicht auf Kaffee und Ananas in normalen Mengen ist nicht erforderlich. Es besteht kein Anlaß, weiterhin die bisherigen umfangreichen Restriktionen der übrigen Nahrungsmittel beizubehalten.     

A comparative study of the effects of dimebon,obsidan, finoptin,and cordaron on the functional state of ischemic focus and size of necrotic zone in experimental myocardial infarction

Dimebon, an antihistamine agent, exerts a moderate antianginal effect, improving the function of ischemic focus in the myocardium and decreasing the necrotic zone in experimental myocardial infarction. Effects of sex steroid hormones on lipid peroxidation and glutathione antioxidant system in rat skin

Effects of estradiol and testosterone on the intensity of lipid peroxidation and contents of glutathione redox system components in the dermis and epidermis of rat skin were studied. Adhesion and growth of CaCo2 cells on surface-modified PEEK substrata

A series of surface-functionalized poly(ether ether ketone) (PEEK) films has been prepared by selective wet-chemistry; they are hydroxylated polymer (PEEK-OH) obtained by reduction, aminated polymer (PEEK-[]-NH2) prepared by coupling a diisocyanate reagent to PEEKOH (PEEK-[]-NCO) followed by hydrolysis, and carboxylated and aminocarboxylated polymers (PEEK-[]-GABA and PEEK-Lysine) resulting from the coupling of aminoacids to PEEK-[]-NCO. The aminated and carboxylated substrata promoted the adhesion and growth of CaCo2 cells in the presence of serum. Fibronectin (FN), an extra-cellular matrix protein, has been covalently fixed and/or adsorbed on various PEEK substrata, in the presence or not of a polymeric surfactant (Pluronic F68). The performances of the FN-grafted substrata (PEEK-[]-FN(1) and PEEK-[]-FN(2)) were significantly higher than those of reference substrata simply coated with FN (PEEK-OH(+FN)(1) and (2), PEEK-[]-NH2(+FN)(1) and (2)), considering the adhesion and spreading of CaCo2 cells in the absence of serum. Moreover, the stability of the adherent cells on the FN-adsorbed substrata dramatically depended on the experimental conditions applied during the PEEK coating with FN.     

THE ODYSSEY – A JUNGIAN PERSPECTIVE: INDIVIDUATION AND MEETING WITH THE ARCHETYPES OF THE COLLECTIVE UNCONSCIOUS

Homer's epic tale of the 20-year return of Odysseus from the Trojan War is investigated with particular reference to Jung's theory of individuation. Odysseus' meetings with 'the anima' in the form of goddesses, sirens and female monsters and his visit to Hades demonstrate the confrontation and humanization of aspects of the archetypal level of the psyche, central to Jung's theories of psychic growth and development. Jung's important ideas of the psychoid level and the transcendent function are explored and linked both to his investigations into medieval alchemy and with findings from contemporary neuroscience. The importance of Jung's constructive method of the interpretation of dreams and myths is shown to be central.
