目的:研究两种制备方法所制备的肺靶向地塞米松磷酸钠微球的特性,确定两种方法是否适合用于制备该微球。方法:对油/水型乳化-溶剂挥发法的工艺进行优化,以优化后的油/水型乳化-溶剂挥发法和水/油/水型乳化-溶剂挥发法制备微球,考察微球的大小、载药量、体外释药等性质。结果:两法所制的微球的平均粒径相近,载药量和体外释药特性各不相同。结论:两种方法都适合用于制备肺靶向的地塞米松微球。从载药量方面分析,优化后的油/水型乳化-溶剂挥发法较好。  相似文献

目的考察内相乙醇加入量及其他制备因素对聚丙交酯微球性质的影响。方法以水略溶性药物氟尿嘧啶和水溶性药物地基米松磷酸钠为模型药物,乳化/溶剂挥发法制备微球。维持其他制备条件不变,改变内相乙醇加入量、理论载药量、聚丙交酯浓度、溶剂挥发时间及聚丙交酯分子质量,考察微球载药量、包封率、粒径和释放的影响。结果随着内相乙醇加入量的增多,微球形成加快,微球的粒径减小,模型药物的载药量及包封率略有升高,但乙醇加入量太多时载药量及包封率均明显降低。内相聚丙交酯浓度越大,形成的微球粒径越大,载药量及包封率也愈大。理论载药量对微球粒径影响不大,地基米松磷酸钠的包封率随理论载药量的增加而减小,而氟尿嘧啶为理论载药量为15%时,包封率最大。溶剂挥发时间在0.5~3h,微球的性质无明显变化。分子质量愈小,微球的粒径愈小,载药量及包封率亦随之减小。结论内相中加入一定量乙醇可以提高药物的包封率,并减少有毒含氯溶剂的用量。  相似文献

目的:研究载羟基喜树碱的聚乳酸微球的制备方法并考察其体外释药性质。方法:以PLA为成膜材料,采用改良乳化-溶剂挥发法,制备载羟基喜树碱的聚乳酸微球并优化制备工艺;对载药微球进行表征;超声介导下进行载药微球的体外释药试验。结果:微球粒径在1~7μm,大小均一;羟基喜树碱浓度在10mg.mL-1下,载药微球包封率为62.2%,载药量为1.69%;药物体外释药符合Higuchi方程。结论:采用乳化-溶剂挥发法,以PLA为成膜材料可制得具有较高包封率的羟基喜树碱微球,有望实现降低羟基喜树碱给药量、减少不良反应,提高靶向性的目标。  相似文献

目的研制具有肺靶向性的紫杉醇壳聚糖微球,并对处方工艺进行优化。方法以壳聚糖为载体,采用乳化一化学交联法制备紫杉醇壳聚糖微球。单因素试验考察了油/水体积比、紫杉醇浓度、乳化时间、乳化剂量等因素,采用正交设计优化微球制备工艺,以HPLC法测定微球载药量、包封率。结果制得的微球显微观察形态圆整、表面光滑,无黏连;平均粒径为(8.23±0.25)μm,粒径在7~12μm平均占微球总数的84.2%,载药量为16.20%±1.15%,包封率为81.29%±1.62%。结论筛选的最佳处方工艺制备的微球粒径大小适宜,可满足肺靶向微球的要求并免除过敏试剂的加入。  相似文献

目的以环孢素A为模型药,使用生物可降解材料乳酸/羟基乙酸PLGA制备微球。并评价微球的体外释放。方法使用乳化溶剂挥发法制备环孢素微球,并对微球的外观,粒径和载药量进行评价。通过衡量温度,pH值和表面活性剂等因素筛选体外释放介质。结果制得的微球外观圆整,粒径统一,平均粒径50μm左右,载药量为13%。DSC结果表明环孢素A和PLGA有结合作用。体外释放实验表明微球的释放受温度,pH值和表面活性剂的影响,加入30%的异丙醇可使微球在2周内释放完全。结论乳化溶剂挥发法可制备得到质量符合要求的环孢素微球,含30%异丙醇的释放介质是微球体外释放评价的理想介质。  相似文献

目的 在本实验中 ,以生物可降解和生物相容性良好的聚乳酸 (PLA)或乳酸 -乙醇酸共聚物 (PLGA)为载体材料制备含有雌二醇药物的缓释微球 ,考察在制备过程中水溶性更强的四氢呋喃的加入对微球性质的影响。方法 以乙酸乙酯和四氢呋喃为有机溶剂、采用乳化 -溶剂萃取法制备含药微球 ,分别从成球性、粒径、包封率和体外释药等方面 ,进行制备工艺研究以及微球相关性质的研究。结果 在乙酸乙酯中加入四氢呋喃 ,使包封率降低 ,但在乙酸乙酯体积比大于 5 0 %时 ,增加四氢呋喃的用量对包封率无明显影响。相同条件下 ,在考察范围内 ,粒径随四氢呋喃用量比例的增大而增大 ,而载体的性质和微球含药量是影响微球释药的主要因素。结论 尽管在制备过程中的油相中加入了水溶性更强的四氢呋喃 ,但是通过控制制备工艺和条件 ,仍然可以得到球形态好、有适当粒径分布范围、载药量高并且突释程度小的雌二醇生物可降解缓释微球  相似文献

目的:考察处方工艺参数对微球体外释放度的影响.方法:采用O/O型乳化溶剂挥发法,以乳酸-羟基乙酸共聚物为载体,制备盐酸昂丹司琼(Ondansetron hydrochloride,OND)微球.采用紫外分光光度法测定微球的体外释放度.结果:选择对OND具有较好溶解能力的混合溶剂为内油相溶剂,可以降低突释;增加理论载药量,延缓正己烷加入的时间和减小粒径可以增加OND微球的释药速度.结论:通过对处方和工艺的调节可使OND微球的体外释药曲线符合Higuchi方程,2周的累积释放量在80%左右.  相似文献

目的 制备罗哌卡因 醋酸地塞米松聚乳酸羟基乙酸共聚物(PLGA)微球(简称微球)并研究其体外释药特性。方法以PLGA为载体,采用W1/O/W2双重乳化 溶剂挥发法制备微球,研究实验过程中有机相PLGA浓度、外水相/有机相体积比、内水相体积、外水相聚乙烯醇(PVA)浓度几项因素变化对罗哌卡因 醋酸地塞米松PLGA微球粒径、表面形态﹑载药量﹑包封率和突释行为的影响。结果有机相PLGA浓度在制备微球的过程中是一个关键性因素。随着PLGA浓度增加,微球粒径增大,载药量﹑包封率明显提高,突释降低;外水相/有机相体积比增大,微球粒径增大, 载药量﹑包封率明显提高,微球表面更加光滑﹑微孔减少,突释降低;随着内水相体积增加使得微球表面的微孔明显增多,突释增加,载药量﹑包封率降低;当外水相PVA浓度由0.5%增加到2%,微球粒径变小,突释效应增加。通过优化条件制备的微球形状为球形,外观光滑圆整,粒径分布均匀,其中>90%分布在20~70 μm。罗哌卡因载药量(7.48±0.33)%,包封率(70.97±2.36)%;醋酸地塞米松载药量(1.52±0.16)%,包封率(57.30±1.17)%。结论采用W1/O/W2双重乳化 溶剂挥发法成功制备罗哌卡因加醋酸地塞米松PLGA微球;以优化工艺制备的微球,在体外具有明显的缓释行为,释药曲线呈典型S形三阶段模式。  相似文献

目的研究布比卡因缓释微球制备方法并对其体外释药特性进行评价。方法采用紫外分光光度法测定布比卡因微球载药量、包封率;采用HPLC法测定微球体外释放;通过正交设计优选微球制备工艺;以乳酸羟基乙酸共聚物为载体,使用乳化溶剂挥发法制备布比卡因微球;用扫描电镜观察所得微球的粒径和形态;通过体外释药实验考察布比卡因乳酸羟基乙酸共聚物微球的缓释作用。结果微球载药量、包封率和体外释放的测定方法符合方法学要求;按照优选处方制备所得的微球为圆整球体,表面多孔,呈蜂窝状,粒径50~100μm之间的微球占80%;体外释放符合Ritger-Peppas方程,t1/2=242.05 h。结论乳化溶剂挥发法适用于布比卡因乳酸羟基乙酸共聚物微球的制备,所制得的微球形态圆整,在体外具有明显缓释作用。  相似文献

Clinical and in vitro investigations were carried out to test the efficacy of gut lavage, hemodialysis, and hemoperfusion in the treatment of poisoning with paraquat or diquat. In a patient suffering from diquat intoxication 130 times more diquat was removed by gut lavage 30 h after ingestion than was removed by complete aspiration of the gastric contents.Determination of in vitro clearances for paraquat and diquat by hemodialysis showed that, at serum concentrations of 1–2 ppm, such as are frequently encountered in poisoning in man, toxicologically relevant quantities of herbicide cannot be removed from the body. At a concentration of 20 ppm, on the other hand, hemodialysis proved to be effective, the clearance being 70 ml/min at a blood flow rate of 100 ml/min. The efficacy of hemoperfusion with coated activated charcoal was on the whole better. Especially at concentrations around 1–2 ppm, the clearance values for hemoperfusion were some 5–7 times higher than those for hemodialysis.In a patient suffering from paraquat poisoning, both hemodialysis as well as hemoperfusion were carried out. The in vitro results could be confirmed: At serum concentrations of paraquat less than 1 ppm no clearance could be obtained by hemodialysis while by hemoperfusion with activated charcoal quite high clearance values were measured and the serum level dropped down to zero.
Zusammenfassung Klinische Untersuchungen und Laboratoriumsversuche wurden durchgeführt, um die Wirksamkeit von Darmspülung, Hämodialyse und Hämoperfusion bei Paraquat- und Deiquat-Vergiftungen zu prüfen.Bei einem Patienten wurde 30 Std nach Deiquat-Aufnahme durch Darmspülung 130mal mehr Deiquat entfernt als durch vollständige Aspiration des Mageninhaltes. In vitro-Versuche ergaben, daß bei Blutserumkonzentrationen von 1–2 ppm, die bei Vergiftungen oft gemessen werden, durch Hämodialyse keine toxikologisch relevanten Paraquat- oder Deiquat-Mengen entfernt werden können. Dagegen erwies sich die Hämodialyse bei 20 ppm und einer Blutumlaufgeschwindigkeit von 100 ml/min mit einer Clearance von 70 ml/min als wirksam. Die Hämoperfusion mit beschicheter Aktivkohle war in diesen Versuchen aber eindeutig überlegen, denn insbesondere bei Konzentrationen um 1–2 ppm waren die Clearance-Werte 5–7mal höher als bei der Hämodialyse.Die in vitro-Ergebnisse wurden bei einem Patienten mit einer Paraquat-Vergiftung bestätigt: Bei Konzentrationen unter 1 ppm war die Hämodialyse wirkungslos, während durch Hämoperfusion relativ hohe Clearance-Werte erreicht wurden, so daß der Serumspiegel rasch unter die Nachweisgrenze abfiel.

This study describes a new approach for organophosphorous (OP) antidotal treatment by encapsulating an OP hydrolyzing enzyme, OPA anhydrolase (OPAA), within sterically stabilized liposomes. The recombinant OPAA enzyme was derived from Alteromonas strain JD6. It has broad substrate specificity to a wide range of OP compounds: DFP and the nerve agents, soman and sarin. Liposomes encapsulating OPAA (SL)* were made by mechanical dispersion method. Hydrolysis of DFP by (SL)* was measured by following an increase of fluoride ion concentration using a fluoride ion selective electrode. OPAA entrapped in the carrier liposomes rapidly hydrolyze DFP, with the rate of DFP hydrolysis directly proportional to the amount of (SL)* added to the solution. Liposomal carriers containing no enzyme did not hydrolyze DFP. The reaction was linear and the rate of hydrolysis was first order in the substrate. This enzyme carrier system serves as a biodegradable protective environment for the recombinant OP-metabolizing enzyme, OPAA, resulting in prolongation of enzymatic concentration in the body. These studies suggest that the protection of OP intoxication can be strikingly enhanced by adding OPAA encapsulated within (SL)* to pralidoxime and atropine.  相似文献

Abstract

The uptake of metals from food and water sources by insects is thought to be additive. For a given metal, the proportions taken up from water and food will depend both on the bioavailable concentration of the metal associated with each source and the mechanism and rate by which the metal enters the insect. Attempts to correlate insect trace metal concentrations with the trophic level of insects should be made with a knowledge of the feeding relationships of the individual taxa concerned. Pathways for the uptake of essential metals, such as copper and zinc, exist at the cellular level, and other nonessential metals, such as cadmium, also appear to enter via these routes. Within cells, trace metals can be bound to proteins or stored in granules. The internal distribution of metals among body tissues is very heterogeneous, and distribution patterns tend to be both metal and taxon specific. Trace metals associated with insects can be both bound on the surface of their chitinous exoskeleton and incorporated into body tissues. The quantities of trace meals accumulated by an individual reflect the net balance between the rate of metal influx from both dissolved and particulate sources and the rate of metal efflux from the organism. The toxicity of metals has been demonstrated at all levels of biological organization: cell, tissue, individual, population, and community. Much of the literature pertaining to the toxic effects of metals on aquatic insects is based on laboratory observations and, as such, it is difficult to extrapolate the data to insects in nature. The few experimental studies in nature suggest that trace metal contaminants can affect both the distribution and the abundance of aquatic insects. Insects have a largely unexploited potential as biomonitors of metal contamination in nature. A better understanding of the physico-chemical and biological mechanisms mediating trace metal bioavailability and exchange will facilitate the development of general predictive models relating trace metal concentrations in insects to those in their environment. Such models will facilitate the use of insects as contaminant biomonitors.  相似文献

The precocity and efficacy of the vaccines developed so far against COVID-19 has been the most significant and saving advance against the pandemic. The development of vaccines has not prevented, during the whole period of the pandemic, the constant search for therapeutic medicines, both among existing drugs with different indications and in the development of new drugs. In order to find out the values of the steroid resources for the future use. the compositions and contents of steroidal sapogenins from 13 domestic plants have been investigated. As a result,Dioscorea nipponica, D. quinqueloba andSmilax china were found to have large amount of diosgenin. Advances in the molecular biological knowledge of neuronal nicotinic acetylcholine receptors (nAChRs) have led to a growing interest by the pharmaceutical industry in the development of novel compounds that selectively modulate nAChR function. The ability of (-)-nicotine, an activator of nAChRs, to enhance attentional aspects of cognition in animals and humans, to exert neuroprotective and anxiolytic-like effects, and presumably to mediate the negative correlation between smoking and Alzheimer's (and Parkinson's) Disease, has focused interest on the potential therapeutic utility of modulators of nAChR function for treatment of some of the deficits associated with these progressive, neurodegenerative conditions. Numerous compounds are known which activate nAChRs and which might serve as lead compounds toward the development of such agents. The pharmacologic diversity of neuronal nAChR subtypes suggests the possibility of developing selective compounds which would have more favourable side-effect profiles than existing agents. This broader class of agents, collectively called cholinergic channel modulators (ChCMs), is anticipated to encompass compounds which would have more favourable side-effect profiles than existing agents, which generally exhibit low selectivity. This selectivity may be achieved by preferentially activating some subtypes of nAChRs (i.e., Cholinergic Channel Activators, ChCAs) or inhibiting the function of other subtypes (Cholinergic Channel Inhibitors, ChCIs). An overview of the biology of nAChRs and the rationale for the use of ChCMs for the treatment of dementia related to neurodegenerative diseases are presented, followed by a discussion of lead compounds and compounds under consideration for clinical evaluation.  相似文献
