Potentialities of the flow cytornetry method in studies of the qualitative and quantitative composition of endometrial immunocompetent cells are demonstrated. Allogeneic stem cells interact with T lymphocytes which are formed when intact thymocytes are cultured with syngeneic mononuclear phagocytes. It is shown that biologically active peptides inhibit the concanavalin A-induced generation of cells which may proliferate in response to exogenous recombinant interleukin-2. Determination of the number of CD25-containing cells showed that synthetic peptides are able to lower not only the number of CD25-containing cells but the level of its expression as well. The cytotoxic activity of natural killer cells and the intensity of conjugate formation are studiedin vitro in the natural cytotoxicity reaction against3H-uridine-labeled human erythromyeloleukotic cells K-562 in the presence of fibronectin, γ-globulin, and fibronectin/γ-globulin combination. It is demonstrated that fibronectin does not change natural cytotoxicity, γ-globulin increases the activity of human natural killer cells, and the fibronectin — γ-globulin combination increases both the intensity of conjugate formation and the cytotoxic activity of natural killer cells. The cytotoxicity of natural killer cells (NK) against3H-uridine-labeled target cells (TC, human erythromyeloleukosis cells K-562) and the intensity of conjugate formation in the NK:TC system in the presence of γ-interferon, C-reactive protein, and human fibronectin are studiedin vitro in 14 patients with multiple sclerosis. It is shown that γ-interferon and C-reactive protein decrease the cytotoxic activity of NK with a simultaneous stimulation of conjugate formation in the NK:TC system. The correlation between the studied parameters becomes weaker. Human fibronectin induces collateral changes in the activity of NK and in the number of effector:target conjugates formed in the natural cytotoxicity reaction. Study of the hypothalamus and adjacent areas of the brain in adult rats revealed that, along with the osmosensitivity of neurosecretory cells, the transport of vasopressin and oxytocin along their axons and their release from the terminals into the bloodstream are impeded. This is due to the absence of axonal bonds between the accessory groups of neurosecretory cells and the posterior lobe of the pituitary. It is shown that cultured Ehrlich ascitic carcinoma cells are a convenient test system for the investigation of the effects of various factors on DNA synthesis in the cells of this tumor. The application of this system markedly facilitates fractionation of a chalone-containing preparation, the purpose of this fractionation being the isolation of components affecting specific phases of the mitotic cycle. After separation of normal murine bone marrow cells in a Percoll density gradient cellular fractions with densities of 1.076 and 1.060 g/ml are capable of suppressing thein vitro growth of leukemia cells. The cytostatic activity of these fractions, however, does not surpass the level of antitumor antiproliferative activity intrinsic to intact bone marrow cells. These cells were found to be capable of joining the splenocytes, thymocytes, and lymph node cells in effector cytostatic cooperation and thus enhance the final antitumor effect. It is shown that the structure of Paneth cells in duodenum of the eastern gray squirrel and red-cheeked suslik is similar to that in other mammals. There are no statistically significant differences in the stimulation index for purified lymphocytes of control mini-pigs and mini-pigs with chronic alcohol intoxication. Autologous sera of control and experimental animals strongly suppress mitogen-induced blast transformation of lymphocytes without death of these cells. There are no statistically significant intergroup differences in the absolute number (per mm3) of T lymphocytes in peripheral blood. The subline Bro of human melanoma xenograft with a high metastasizing activity is studied. Cytosolic and plasma membrane receptors for progesterone and estradiol are studied in myomatous nodes (MN) and in histologically unaltered myometrium (HUM) against the background of myoma. The level of cytosolic receptors for both hormones is higher in the myoma cells than in the essentially healthy myometrium. In the plasma membranes the progresterone reception is reduced and the estradiol reception is unchanged compared with HUM. Monoclonal antibodies toS. typhimurium K-antigens representing IgG1 immunoglobulins with a “kappa”-type light chain are prepared and characterized. They are highly active in indirect enzyme immunoassay with purified K-antigens and whole fixedS. typhimurium cells and virtually do not cross-react with O-antigen of the same bacterial species or withE. coli antigenic structures. The kinetics of binding and ability to agglutinate whole bacterial cells are studied and epitope analysis is carried out, which shows that antigenic determinants ofS. typhimurium K-antigen qualitatively differ from those of O-antigen of the same bacterium. The proliferative activity of human natural killers (CD16+CD56+ cells) in the presence of 100 and 1000 IU/ml human recombinant interleukin-3 is investigatedin vitro. It is shown that recombinant interleukin-3 reliably enhances natural killer proliferation, causing a 9–15.2-fold increase of3H-thymidine uptake by CD16+CD56+ cells both in complete culture medium and in conditioned medium. The effect of the factor is 3.9–6.4 and 3.6–8.9-fold more potent than that of recombinant interleukin-2 and granulocyte-macrophage colony-stimulating factor, respectively, in the same doses. The number of neurons was counted in different functional neuronal complexes of the sensorimotor cortex in albino rats in the control and at different times after clinical death caused by asphyxia. A decrease of the number of neurons from 2.5×106 in the control to 1.56×106 toward the 30th day of the postresuscitation period was found in the sensorimotor cortex. The complexes of small neurons in the upper floor (layers II–IV) suffer badly, while the neurons of layer VI are damaged to a somewhat lesser degree and the cells of layer V are minimally altered.
