Reversed immunofixation agar gel electrophoresis

A method has been elaborated for investigation of biological fluids on the basis of the theory of the immunofixation electrophoresis. In spite of the usual methods of the immunofixation electrophoresis it is the antibody which moves by the electrophoresis and not the antigen. The antibody stains are fixed with cellulose acetate strip soaked with a suitably diluted antigen solution after electrophoresis. This method uses only a small quantity of the immune serum.     

Reticuloendotheliosis antigen for the agar gel precipitation test

An antigen for gel precipitation was prepared from chick embryo fibroblast cultures inoculated with the Reticuloendotheliosis virus. The specificity of the reaction was confirmed with reference Reticuloendotheliosis and Spleen necrosis sera. No precipitation reactions occurred between this antigen and Marek's disease, Newcastle disease, Infectious bronchitis, Pasteurella Multocida and Haemorrhagic enteritis antisera.     

Cultivation of human peripheral blood leukocytes in agar gel

The use of a modified method of cloning hematopoietic cells in semisolid nutrient media showed that among human peripheral blood leukocytes there are committed precursor cells of the granulocytic series (CFU_c). In healthy adults and children (aged from 4 days to 10 years) the number of these precursors is 0.05–6.38 and 0.2–2.9/10⁵ nucleated cells respectively. The number of CFU_c in the blood of patients with infectious mononucleosis is within normal limits (0.5–14/10⁵ nucleated cells). In acute leukemia very low colony-forming activity of the nucleated blood cells is found (0–0.3/10⁵ cells).Laboratory of Culture and Transplantation of Bone Marrow, Central Institute of Hematology and Blood Transfusion. A cuvette for microelectrophoresis on agar and starch gel and for microimmunoelectrophoresis

Summary A simple cuvette is described which is to be used for serial analyses by means of modified micromethods of electrophoresis on agar and starch gel immune electrophoresis on agar.
