本文作者利用金黄地鼠胚胎细胞(sHE)检测了氚水(Hto)和氚一胸腺嘧啶核苷(3H—TdR)和氚一脲嘧啶核苷(3H—udR)及戊酸雌二醇(E)体外诱发其细胞的转化能力, 上述棱素在体外直接作用于细胞持续6天, 其结果表明HTO及3H—TdR能在体外直接诱发SHE细胞恶性转化, 表现在细胞形态学符合转化细胞的特征、刀豆素A凝集试验均呈阳性反应.在半琼脂培养基中转化细胞具有形成细胞集落的能力, 其结果还表明3H-UdR没能引起细胞转化, 表明3H可能是通过对胞核DNA损伤而不是通过对胞浆内大分子的损伤诱发细胞转化的。  相似文献

用叙利亚金黄地鼠胚胎(SHE)细胞的体外恶性转化方法,检测出偏二甲基肼(UDMH)可诱导SHE细胞的形态学恶性转化。转化细胞嗜碱性强,核浆比例增大,排列方向紊乱,可交叉重叠生长。从转化集落分离的细胞生长旺盛并具有异常的细胞生物学特性,细胞寿命明显延长,可被较低浓度的植物凝集素凝集,在软琼脂内非贴壁依赖生长,接种同种免疫抑制动物可长出肿瘤,以上结果说明,UDMH具有诱导SHE细胞恶性转化的作用,预示  相似文献

研究电离辐射在离体时诱发人细胞恶性转化。方法选用α辐射源诱发SV40永生化的人胎气管成纤维细胞恶性转化,观察在细胞恶性转化过程中细胞生物学特性的变化。结果经α粒子照射后细胞形态发生转化;细胞相对存活率与剂量呈依赖性,随着照射剂量的增加,存活率呈指数下降,存活曲线呈单击单靶型;早期传代细胞染色体数出现非整倍体和非稳定性畸变,同时有两个标记染色体;软琼脂克隆形成率随着照后传代次数的增加而明显增高;第40代细胞接种裸鼠长出肿瘤结节,病理诊断为纤维肉瘤。结论α粒子可诱发人胎永生化气管成纤维细胞体外恶性转化。  相似文献

作者对5例胶质二氧化钍症时胆管细胞癌及1例肝细胞与胆管细胞癌混合型癌作了影像学研究。确诊胶质二氧化钍症到发生肿瘤时间平均为44年5个月。CT对6例恶性肿瘤中5例能作出正确诊断,但确诊时肿瘤已增大到不能切除。US对6例中3例能作出诊断。单纯腹部平片无诊断价值。肿瘤标记阴性的胆管细胞癌US及CT对肝右叶前区至肝左叶内侧的镶嵌状胶质二氧化钍肿块能作出诊断,血管造影见右前枝血管呈圆弧状受压及不规则血管增生,并有肿瘤染色。1例肝细胞癌、胆管细胞癌混合型,CT于肿瘤标记阳性初期无异常所见,肿瘤标记迅速升高时,CT见胆囊内有钙乳样胆汁,呈  相似文献

目的 研究BEP2D细胞经辐射诱发恶性转化过程中,作为SMAD蛋白家族的抑制分子,Smad7对TGF-β/SMAD介导信号通路的调控。方法 用Northem blot检测TGF-β刺激之后,永生化BEP2D细胞及辐射诱发恶性转化的BERP 35T2细胞中Smad7 mRNA的表达水平。人工合成SMAD结合元件(SBE)重复序列,同报告基因碱性磷酸酶融合。构建好的载体同Smad7真核表达载体共转染,TGF-β刺激,通过报告基因的表达丰度来检测Smad7对TGF-β/SMADs介导的信号通路的调控。结果 Northern杂交结果表明,永生化细胞Smad7基因对TGF-β刺激的应答正常,恶性化细胞的应答降低。基因转染的结果表明,永生化细胞中SBE的基础活性较恶性化细胞高;TGF—β刺激之后,永生化细胞中SBE活性显著增强,恶性化细胞变化不明显;同Smad7真核表达载体共转染之后,两种细胞中SBE活性均显著降低。结论 在辐射诱发细胞发生恶性转化过程中,Smad7基因对TGF-β信号通路的反馈调节作用发生紊乱,使TGF-β信号通路持续处于抑制状态,TGF-β对细胞的负性调控作用减弱,细胞进一步向恶性化发展。  相似文献

本文报道了白云鄂博稀土铁矿工人肺内天然钍沉积基及其对健康影响的六年研究结果(1980~1985)。作者用负高压静电收集氧子体装置,对该矿200多名接尘矿工进行了600多人次呼出气中氧浓度的测定和肺内天然钍沉积量的推算。结果表明,大约有l/5的接尘矿工肺内天然钍的水平在20pCi以上,最高值为120pCi。根据线性无阈假说,推算了2072名矿工由于吸入天然钍和氧子体对肺组织所造成的累积剂量,估计了可能增加的肺癌例数,结合流行病学调查的结果,目前在该矿接尘矿工中,尚未观察到在1958至1984年期间有肺癌死亡率肯定增加的事实。然而,观察到了部分破碎车间工人肺内天然钍沉积量>41pCi,含钍稀土混合粉尘总量>940mg时,X线胸大片表现为0期(可疑)尘肺,肺功能指标,FEV1%,MBC,MMEF出现轻度障碍。  相似文献

The Knee injury and Osteoarthritis Outcome Score (KOOS) is a self-administered instrument measuring outcome after knee injury at impairment, disability, and handicap level in five subscales. Reliability, validity, and responsiveness of a Swedish version was assessed in 142 patients who underwent arthroscopy because of injury to the menisci, anterior cruciate ligament, or cartilage of the knee. The clinimetric properties were found to be good and comparable to the American version of the KOOS. Comparison to the Short Form-36 and the Lysholm knee scoring scale revealed expected correlations and construct validity. Item by item, symptoms and functional limitations were compared between diagnostic groups. High responsiveness was found three months after arthroscopic partial meniscectomy for all subscales but Activities of Daily Living.  相似文献

Objective To investigate endovascular treatment of traumatic direct carotid-cavernous fistulas (CCF) and their complications such as pseudoaneurysms. Methods: Over a five-year period, 22 patients with traumatic direct CCFs were treated endovascularly in our institution. Thirteen patients were treated once with the result of CCF occluded, 8 twice and 1 three times. Treatment modalities included balloon occlusion of the CCF, sacrifice of the ipsilateral internal carotid artery with detachable balloon, coll embolization of the cavernous sinus and secondary pseudoaneurysms, and covered-stem management of the pseudoaneurysms. Results All the direct CCFs were successfully managed endovascularly. Four patients developed a pseudoaneurysm after the occlusion of the CCF with an incidence of pseudoaneurysm formation of 18.2% (4/22). A total number of 8 patients experienced permanent occlusion of the ICA with a rate of ICA occlusion reaching 36.4% (8/22). Followed up through telephone consultation from 6 months to 5 years, all did well with no recurrence of CCF symptoms and signs. Conclusion Traumatic direct CCFs can be successfully managed with endovascular means. The pseudoaneurysms secondary to the occlusion of the CCFs can be occluded with stent-assisted coiling and implantation of covered stents.  相似文献

Acute limping may be the result of multiple pathologies in children. The differential diagnosis varies based on the age of the child. Irrespective of age, the initial imaging work-up includes AP and frog leg radiographs of the pelvis and ultrasound; MRI may sometimes be helpful. In children less than 3 years, infections and trauma are most frequent. MRI is the imaging modality of choice when osteomyelitis is clinically suspected. Between the ages of 3 and 10 years, transient synovitis of the hip and Legg-Calvé-Perthes disease are main considerations but infection, inflammation and focal bony lesions are also considered. In response to the ENFSI and EDNAP groups’ call for new STR multiplexes for Europe, Promega® developed a suite of four new DNA profiling kits. This paper describes the developmental validation study performed on the PowerPlex® ESI 16 (European Standard Investigator 16) and the PowerPlex® ESI 17 Systems. The PowerPlex® ESI 16 System combines the 11 loci compatible with the UK National DNA Database®, contained within the AmpFlSTR® SGM Plus® PCR Amplification Kit, with five additional loci: D2S441, D10S1248, D22S1045, D1S1656 and D12S391. The multiplex was designed to reduce the amplicon size of the loci found in the AmpFlSTR® SGM Plus® kit. This design facilitates increased robustness and amplification success for the loci used in the national DNA databases created in many countries, when analyzing degraded DNA samples. The PowerPlex® ESI 17 System amplifies the same loci as the PowerPlex® ESI 16 System, but with the addition of a primer pair for the SE33 locus. Tests were designed to address the developmental validation guidelines issued by the Scientific Working Group on DNA Analysis Methods (SWGDAM), and those of the DNA Advisory Board (DAB). Samples processed include DNA mixtures, PCR reactions spiked with inhibitors, a sensitivity series, and 306 United Kingdom donor samples to determine concordance with data generated with the AmpFlSTR® SGM Plus® kit. Allele frequencies from 242 white Caucasian samples collected in the United Kingdom are also presented. The PowerPlex® ESI 16 and ESI 17 Systems are robust and sensitive tools, suitable for the analysis of forensic DNA samples. Full profiles were routinely observed with 62.5 pg of a fully heterozygous single source DNA template. This high level of sensitivity was found to impact on mixture analyses, where 54–86% of unique minor contributor alleles were routinely observed in a 1:19 mixture ratio. Improved sensitivity combined with the robustness afforded by smaller amplicons has substantially improved the quantity of data obtained from degraded samples, and the improved chemistry confers exceptional tolerance to high levels of laboratory prepared inhibitors.  相似文献
