目的研究肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和干扰素γ(IFN-γ)以及细菌脂多糖(LPs)对CD68抗原在角质形成细胞株HaCaT细胞中表达的影响。方法RPMI1640培养液培养的HaCaT细胞随机分成自然增殖孔(无刺激组)、IFN一1刺激孔(组)、TNF-α刺激孔(组)、LPS刺激孔(组)、IL-6刺激孔(组)。培养24h后收集细胞,采用流式细胞仪、免疫组化和RT-PCR方法检测TNF-α、IL-6、IFN-γ和LPS作用HaCaT细胞后CD68表达情况。结果与无刺激组比较,不同刺激因素可使HaCaT细胞CD68阳性细胞数增多,但以TNF-α和IL-6的作用较强(t值为3.60和3.93,P值均〈0.01),IFN-γ和LPS的作用稍弱(t值为2.38和2.52,P值均〈0.05)。只有IL-6可以使HaCaT细胞CD68阳性细胞平均荧光强度增强(t值为8.34,P值〈0.01)。在IFN-γ和TNF-α、IL-6作用HaCaT细胞24h后均有CD68表达于胞质和胞膜,并且以TNF-α和IL-6作用较强。TNF-α和IL-6作用HaCaT细胞24h后可见CD68mRNA表达明显增强(t值为4.34和7.52,P值均〈0.01),IFN-γ作用后出现较弱表达(t=2.81,P〈0.05)。结论在IL6、TNF-α、IFN-γ和LPS的作用下HaCaT细胞CD68表达增强。  相似文献

Objective To investigate the effect of tacrolimus on the expression of nuclear factor-κB (NF-κB) in HaCaT cells stimulated by tumor necrosis factor-α(TNF-α),and on the expression of glucocorticoid receptor (GR)α and β in untreated HaCaT cells in vitro.Methods Cultured Ha CaT cells were treated with TNF-α(10μg/L) only,combination of TNF-α(10μg/L) and various concentrations (10-8mol/L, 10-7mol/L,10-6moL/L) of tacrolimus or tacrolimus of different concentrations only.After additional 12-,24-, 36- or 48-hour cnlture, Westem blot and immunofluorescenee-confocal laser scanning microscopy were used to detect the expressions of NF-κB,GRα and GRβ in HaCaT cells.Those untreated HaCaT cells served as the control.Results The relative protein expression level of NF-κB was increased in HaCaT cells after treatment with TNF-α for 24 and 48 hours zompared with untreated ceils (0.73±0.0316 and 0.8925±0.0171 vs 0.4988±0.03506,both P<0.05);however,the increase in NF-κB expression was inhibited by the combination treatment with tacrolimus,and the relative expression level of NF-κB protein was 0.6825±0.0263.0.6200±0.0163 and 0.5575±0.0299 in HaCaT cells treated with TNF-α plus tacrolimus of 10-8mol/L 10-7mol/L and 10-6mol/L,respectively;the difference was significant etween TNF-α-treated cells and those dealt with the combination of NF-α and tacrolimus of 10-7 or 10-6 mol/L (both P<0.05).No significant difference was observed in the expression of NF-κB by HaCaT cells between different time oints treated with tacrolimus of 10-8,10-7 or 10-6 mol/L.Also,there was no zignificant difference in the expression of GRα or GRβ between untreated HaCaT cells and those treated with tacrolimus of 10-8, 10-7 or 10-6 mol/L at any time point.Conclusions Tacrolimus ould inhibit the expression of NF-κB by TNF-α-stimulated HaCaT cells,but does not affect the expression of GRα or GRβ,in untreated HaCaT cells.  相似文献

Methods Neutrophils were isolated from venous blood samples of 25 patients with psoriasis vulgaris (including 13 cases of active psoriasis and 12 cases of inactive psoriasis) as well as 25 normal human con-trols, and cultured. Then, these neutrophils were grouped and treated with lipopolysaccharide (LPS, 100 g/L),CpG-A (50 mg/L), CpG-B (50 mg/L), and RPMI 1640 culture medium, respectively, for 24 hours followed by the collection of culture supematants. Human keratinocytes (HaCaT) were cultured in the presence of su-pematants of treated or untreated nentrophils for 72 hours followed by the detection of cell proliferation with MTT assay. To determine the role of proinflammatory factors, SOD/CAT and monoclonal antibody to IL-8 and TNF-alpha of 400 u/mL were used to pretreat HaCaT cells 1 hour prior to the stimulation with super-natants of neutrophils. Results Compared with culture medium, the supematant of unstimulated neutrophils from normal controls or patients with inactive psoriasis had no significant effect on the proliferation of HaCaT cells (P > 0.05), but that from patients with active psoriasis markedly promoted the proliferation of HaCaT cells (t = 2.41, P < 0.05). ARe, stimulation by LPS, CpG-A and CpG-B, the supematant of active patient-derived neutrophils significantly promoted the proliferation of HaCaT cells compared with that of normal control-derived nentrophils (t = 3.11, 2.89, 2.29, respectively, all P < 0.05). In comparison with tmstimulated neutrophils, the supematant from LPS- and CpG-A stimulated nentrophiles significantly accelerated the pro-liferation of HaCaT cells. Furthermore, the proliferation of HaCaT cells induced by the supematants of LPS-,CpG-A-, CpG-B-stimulated neutrophils from psoriatic patients was statistically suppressed by the pretreat-ment with the monoclonal antibody to IL-8, TNF-alpha and SOD/CAT (all P < 0.05). 目的 检测白癜风患者血清中15种神经内分泌免疫因子(ANG-Ⅱ、EP-β、IFN-γ、sICAM-1、5-HT、IL-10、酪氨酸酶抗体IgM、TNF-α、IL-8、IL-6、酪氨酸酶抗体IgG、IL-2、NE、Cortisol、E),并结合维吾尔医理论探讨其与白癜风发病的关系.方法 采用ELISA测定175例白癜风患者血清中上述指标水平,并与60例正常人对照组比较.结果 175例白癜风患者血清ANG-Ⅱ、EP-β、IFN-γ、sICAM-1、5-HT、IL-10、酪氨酸酶抗体:IgG、NE、Cortisol、E的浓度显著性高于对照组,差异有统计学意义(P<0.01).而TNF-α、IL-8、IL-2、IL-6浓度低于对照组,差异有统计学意义(P<0.01).IgM浓度与对照组差异无统计学意义(P>0.05).稳定期患者ANG-Ⅱ、5-HT、IL-8、Cortisol浓度高于进展期患者,差异有统计学意义(P<0.01).其余指标差异无统计学意义.结论 白癜风发病过程是一个神经内分泌免疫网络的综合过程,与维吾尔医"kuwiti tabiiy"(自然力)失调致白癜风理论是否一致值得研究.
Abstract:
Objective To measure the serum levels of 15 neuroendocrine and immune factors, including angiotensin-Ⅱ (ANG-Ⅱ), endorphin-β (EP-β), interferon-γ (IFN-γ), soluble intercellular adhesion molecule-1 (sICAM-1), 5-thrombotonin (5-HT), interleukin-10 (IL-10), IgM, tumor necrosis-α (TNF-α), IL-8, IL-6, IgG,IL-2, norepinephrine, cortisol and adrenaline in patients with vitiligo, and to analyze their association with the development of vitiligo according to traditional Uyghur medicine theory. Methods Enzyme-linked immunosorbent assay (ELISA) was conducted to measure the serum levels of the above 15 neuroendocrine and immune factors in 175 patients with vitiligo and 60 normal human controls. Results There was a significant increase in the serum levels of ANG-Ⅱ , EP-β, IFN-γ, sICAM-1, 5-HT, IL-10, IgG, norepinephrine, cortisol and adrenaline, together with a decrease in those of TNF-a, IL-8, IL-2 and IL-6 in patients with vitiligo compared with the controls (all P < 0.01). No statistical difference was observed for the serum level of IgM between the patients and controls (P > 0.05). Increased serum levels of ANG-Ⅱ, 5-HT, IL-8 and cortisol were noted in patients with stable vitiligo in comparison with those with progressive vitiligo (all P < 0.01), while the levels of the other factors were similar between the stable and progressive vitiligo patients. Conclusions The development of vitilito is a comprehensive process with the involvement of various neuroendocrine and immune factors,and whether the conclusion agrees with the traditional Uyghur medicine theory that imbalanced "kuwiti tabiiy" ["Nature power" (of human body)] induces vitiligo derserves further studies.  相似文献

To determine the role of proinflammatory factors, SOD/CAT and monoclonal antibody to IL-8 and TNF-alpha of 400 u/mL were used to pretreat HaCaT cells 1 hour prior to the stimulation with super-natants of neutrophils. Results Compared with culture medium, the supematant of unstimulated neutrophils from normal controls or patients with inactive psoriasis had no significant effect on the proliferation of HaCaT cells (P > 0.05), but that from patients with active psoriasis markedly promoted the proliferation of HaCaT cells (t = 2.41, P < 0.05). ARe, stimulation by LPS, CpG-A and CpG-B, the supematant of active patient-derived neutrophils significantly promoted the proliferation of HaCaT cells compared with that of normal control-derived nentrophils (t = 3.11, 2.89, 2.29, respectively, all P < 0.05). In comparison with tmstimulated neutrophils, the supematant from LPS- and CpG-A stimulated nentrophiles significantly accelerated the pro-liferation of HaCaT cells. Furthermore, the proliferation of HaCaT cells induced by the supematants of LPS-,CpG-A-, CpG-B-stimulated neutrophils from psoriatic patients was statistically suppressed by the pretreat-ment with the monoclonal antibody to IL-8, TNF-alpha and SOD/CAT (all P < 0.05). Conclusions In patients with psoriasis vulgaris, there is an abnormal secretion of IL-8, TNF-alpha and superoxide by neutrophils in peripheral blood, and these proinflammatory factors could promote the proliferation of HaCaT cells.  相似文献

目的 探讨中性粒细胞在银屑病发病中的作用.方法 采用脂多糖(LPS)或人工合成的CpG ODNs(CpG-A或CpG-B)刺激中性粒细胞,取其培养上清液处理人永生化角质形成细胞株(HaCaT),观察HaCaT细胞株增殖的变化,并通过抗IL-8、抗TNF-α单克隆抗体以及SOD/CAT预处理评价中性粒细胞分泌的炎性因子在银屑病发病中的作用.结果 与RPMI 1640培养液比较,无LPS、CpG-A和CpG-B刺激的正常人和静止期银屑病患者中性粒细胞培养上清液对HaCaT细胞株的增殖无明显促进作用(P>0.05),而进行期患者中性粒细胞培养上清液明显促进HaCaT细胞株的增殖(t=2.41,P<0.05).与正常人比较,进行期银屑病患者中性粒细胞受LPS、CpG-A和CpG-B刺激后的培养上清液显著增强HacaT细胞的增殖(t值分别为3.11,2.89,2.29,P<0.05).经LPS、CpG-A刺激的中性粒细胞培养上清液与无刺激组比较,均明显促进HaCaT细胞的增殖.与未予阻断剂组比较,予抗IL-8、抗TNF-α单克隆抗体以及SOD/CAT预处理的进行期银屑病组中性粒细胞经LPS、CpG-A和CpG-B刺激后的上清液诱导的HaCaT细胞增殖均显著减少(P<0.05).结论 寻常性银屑病患者外周血中性粒细胞存在炎性因子IL-8、TNF-α和SOD/CAT分泌异常,且这些异常分泌的炎性因子可以促进HaCaT细胞的增殖.  相似文献

目的 探讨核因子-κB(NF-κB)亚基P65磷酸化是否参与化学性缺氧模拟剂氯化钴(CoCl2)诱导的永生化人皮肤角质形成细胞(HaCaT)毒性作用及炎症反应.方法 用2 mmol/L CoCl2处理HaCaT细胞,建立化学性缺氧诱导HaCaT细胞损伤的体外模型.RNA干扰法下调NF-κB亚基P65的表达.细胞计数试剂盒-8比色法检测细胞存活率;ELISA法检测培养基中IL-6和IL-8的含量;Western印迹法检测总量P65及磷酸化P65的蛋白表达.结果 CoCl2处理HaCaT细胞0~4 h,可促进NF-κB亚基P65的磷酸化,在0.5 h时P65亚基开始磷酸化,1.5 h时P65亚基的磷酸化水平达到高峰,约为对照组的6.6倍,而4 h基本恢复到正常水平.CoCl2处理HaCaT细胞0~6 h,可时间依赖性地降低细胞活力,2、4和6 h的细胞存活率与对照组比较,P值分别<0.05、<0.01及<0.01.CoCl2处理6 h,还引起IL-6和IL-8的释放显著增加.RNA干扰法下调P65的表达后,CoCl2处理引起的HaCaT细胞毒性作用被明显减弱,即使细胞存活率升高了11%左右,下调P65表达还明显抑制了CoCl2处理引起的IL-6和Il-8释放增多.结论 磷酸化NF-κB亚基P65介导CoCl2诱导的HaCaT细胞毒性及炎症反应.
Abstract:
Objective To explore whether the phosphorylation of NF-κB P65 subunit is involved in the cytotoxicity to and inflammation in an immortal human keratinocyte cell line HaCaT during cobalt chloride (CoCl2-induced chemical hypoxia. Methods HaCaT cells were treated with CoCl2 of 2 mmol/L to set up a chemical hypoxia-induced cell model of injury. Then, RNA interference was used to down-regulate the expression of P65 in CoCl2-induced HaCaT cells. After additional culture, cell viability was tested by cell counting kit8 (CCK-8), the levels of interleukin 6 (IL-6) and interleukin 8 (IL-8) were detected by ELISA kits, phosphorylated and total P65 protein was measured by Western blot. Results The exposure of HaCaT cells to 2 mmol/L CoCl2 for 0 to 4 hours enhanced the phosphorylation of P65, which began at 0.5 hour, peaked at 1.5 hours, and restored to the normal level at 4 hours, and the level of P65 phosphorylation was about 6.6 times that in the untreated control group. The CoCl2 of 2 mmol/L decreased the cell viability of HaCaT cells in a time dependent manner, and a significant difference was observed in the viability of HaCaT cells between CoCl2-treated and untreated HaCaT cells at 2, 4, and 6 hours (P < 0.05, 0.01, 0.01 ). The release of IL-6 and IL-8 from HaCaT cells was also promoted by CoCl2 treatment. The knockdown of P65 expression with siRNA markedly suppressed the CoCl2-induced cytotoxicity to and increase in the release of IL-6 and IL-8 from HaCaT cells,despite of an increment in cell viability by about 11%. Conclusion The phosphorylated P65 subunit mediates CoCl2-induced cytotoxicity and inflammatory injury to HaCaT cells.  相似文献

目的 探讨Toll样受体2(TLR2)在寻常痤疮发病中的作用及意义.方法 选择轻、中、重度痤疮患者各30例,采用流式细胞仪检测其外周血CD14+单核细胞TLR2的表达,并采用双抗体夹心酶联免疫吸附法(ELISA)检测血清IL-8和TNF-α的水平.另取30例正常人作为对照组.结果 轻、中、重度组痤疮患者外周血单核细胞表面TLR2的表达均显著高于正常人对照组(P<0.01).各组患者血清中IL-8和TNF-α的水平也显著高于正常人对照组(P<0.01).直线相关分析显示,各组患者TLR2的表达与IL-8、TNF-α水平均呈正相关性(r分别为0.382、0.517、0.436、0.641、0.725、0.593,P<0.05).痤疮严重程度与TLR2的表达呈正相关性(r=0.406,P<0.01).结论 TLR2表达水平及相关细胞因子浓度与痤疮严重程度密切相关.
Abstract:
Objective To explore the role of Toll like receptor 2 (TLR2) in the pathogenesis of acne vulgaris. Methods Venous blood was collected from 30 normal human controls and 90 patients with acne vulgaris. The patients were equally divided into three groups, i.e., mild, moderate and severe groups, according to disease severity. Flow cytometry was performed to detect the expression of TLR2 in PBMCs, and double antibody sandwich ELISA to measure the levels of serum interleukin-8 (IL-8) and tumor necrosis factor-α (TNF-α). Results A significant increase was observed in the expression of TLR2 in PBMCs, serum level of IL-8and TNF-α in the three groups of patients with acne vulgaris compared with the normal human controls (all P <0.01). The expression of TLR2 was positively correlated with the expression of IL-8 (r=0.382, 0.517,0.436,respectively, all P<0.05) and TNF-α(r=0.641, 0.725, 0.593, respectively, all P<0.05) in the patients with mild, moderate and severe acne vulgaris, respectively, and with the severity of acne vulgaris (r = 0.406,P<0.01 ). Conclusion The expression level of TLR2 and related cytokines seems closely correlated with the severity of acne vulgaris.  相似文献
