The chemical characteristics, fatty acid and conjugated linoleic acid (CLA) contents and cholesterol levels of some traditional Turkish cheeses that are consumed mostly in Turkey were determined in this study. The fatty acid and cholesterol contents and CLA amount of cheeses were analysed as methyl esters by gas chromatography. The aim of this study was to determine the nutritional profile of some Turkish cheeses produced by traditional methods. The major fatty acids of the cheeses were palmitic acid (C(16:0)) and oleic acid (C(18:1)). The saturated fatty acid content of samples changed between 60.80% and 76.57%, while the monounsaturated fatty acid content ranged from 21.42% to 34.05% and the polyunsaturated fatty acid content was between 1.47% and 3.59%. The CLA contents of the cheeses ranged from 0.44 to 1.04 g/100 g in fat. The cholesterol levels of the samples were determined as 44.6-147.69 mg/100 g in cheese. 相似文献

To know the effect of individual conjugated linoleic acid (CLA) isomers on lipid metabolism, male rats were fed diets containing either linoleic acid (LA, a control fatty acid), CLA containing mainly 9c,11t-isomer, 10t,12c-isomer (at the 0.8% level) or an equivalent mixture of these isomers (at the 0.4% level) for 26 days. Although there were no differences in food intake and body weight gain among the groups, the food efficiency was low in rats fed the CLA containing 10t,12c-isomer in comparison with those fed 9c,11t-CLA. The weight of white (epididymal and perirenal) adipose tissues decreased in all CLA groups, while that of brown adipose tissue increased in rats fed 10t,12c-CLA, the difference between 9c,11t- and 10t,12c-isomers being significant. The concentration of serum total cholesterol, triacylglycerol and phospholipid tended to be lower in rats fed CLA preparations containing 10t,12c-CLA, while there was a trend toward an increasing concentration of serum HDL-cholesterol in all CLA groups. The rate of fatty acid β-oxidation in the liver peroxisomes was slightly higher in rats fed CLA than in those fed LA, and the activity of mitchondrial carnitine parmitoyltranceferase (CPT) in the CLA-mix group significantly increased compared to the LA group. The effect of CLA on the concentration of spleen and serum IgE, IgG and IgA level was variable. No CLA effect was observed on the concentration of serum leptin and TNF-. Thus, individual isomers of CLA may have different effects on some metabolic parameters in rats. The results also suggested a probable interaction of different isomers on lipid metabolism. 相似文献

Pickle white cheeses were produced from whole milk with five different probiotic cultures (Enterococcus faecium, Lactobacillus paracasei, Bifidobacterium longum, Bifidobacterium bifidum and Lactobacillus acidophilus). Conjugated linoleic acid (CLA) content of cheeses ranged from 3.52 to 3.92 mg/g. Probiotic differences and storage process have not affected the CLA contents of the samples statistically. There was no correlation between the CLA content of all probiotic cheeses and saturated fatty acids. A positive correlation between the CLA and linoleic acid contents of L. paracasei and L. acidophilus cheeses was observed. 相似文献

Human breast milk is a complex mixture of organic and inorganic compounds. Some compounds, such as conjugated linoleic acid (CLA), come partly from the mother's diet and are produced by the mother's body and secreted into the milk. Although several studies have examined the effect of chronic CLA supplementation on breast milk CLA appearance, little is known about the transfer of food CLA to breast milk over the short term. The objective of this study was to conduct a preliminary analysis of the kinetics of CLA appearance in breast milk over the short term. Seven women expressed breast milk at 4- to 6-hour intervals for 2 days after eating either CLA-enriched (1912 mg CLA) or control (231 mg CLA) cookies. Milk samples were freeze-dried, fatty acid methyl esters were prepared using methanolic-potassium hydroxide (KOH), and CLA isomers were quantified by gas chromatography. Analysis revealed the following: (1) CLA enrichment of total fatty acids in the breast milk for 48 hours post ingestion of the CLA-enriched cookies was 2.9-fold above control; (2) total breast milk CLA content for 48 hours post CLA-enriched cookies ingestion was 46% greater than post CLA-moderate cookies ingestion; (3) after ingestion of the CLA-enriched cookies, breast milk CLA enrichment plateaued between 8 to 28 hours. This preliminary study suggests that breast milk fatty acids are enriched in CLA compared to control within 28 hours after the ingestion of a CLA-rich food product and invites further research on the extent and timing with which breast milk composition reflects dietary CLA content. 相似文献

Conjugated linoleic acids (CLA) were identified in 1980''s, since then it has been intensively studied due to its various beneficial health effects such as anti-inflammatory, anti-atherogenic, anti-carcinogenic and anti-diabetic/obesity effects. Isomer specificity of a number of CLA isomers, especially predominant isomer 9Z,11E- and 10E,12Z-CLA, is now recognized. However, the less prevalent CLA isomers have not been well characterized. Recently, studies have reported the distinctively different effects of 9E,11E-CLA in colon cancer cells, endothelial cells, and macrophage cells compared to the rest of CLA isomers. In this review, various effects of CLAs, especially anti-inflammatory and anti-atherogenic effects, will be discussed with focusing on the isomer-specific effects and potential mechanism of action of CLA. At last, recent studies about 9E,11E-CLA in in vitro and animal models will be discussed. 相似文献

ProSafeBeef is a 5‐year integrated project funded by the European Commission under the sixth Framework Programme. The overall aim is to advance beef safety and quality across Europe and the work programme spans seven integrated “pillars”. Pillar 3 is concerned with producing safe beef and beef products with enhanced nutritional and eating quality characteristics. A particular focus is on the development of strategies to enhance the concentrations in beef of those fatty acids considered to be of benefit to human health, without causing a detrimental effect on the appearance, shelf‐life or eating quality of the beef. There is accumulating evidence of the importance of long‐chain n‐3 (omega‐3) polyunsaturated fatty acids (PUFAs) for human health and disease prevention, and also evidence from experimental studies that has shown anticarcinogenic, antiatherogenic, and anti‐obesity effects of two isomers of conjugated linoleic acid (CLA). Based on this evidence, the major focus of research efforts to improve the nutritional value of beef has been on increasing the concentration of the n‐3 PUFAs and CLA. Considerable progress has been made within pillar 3 of ProSafeBeef to meet this aim, primarily by manipulating the diet of cattle. Fundamental information on ruminal lipid metabolism and on the control and/or prevention of ruminal hydrogenation of dietary lipids arising from research within ProSafeBeef will facilitate the production of beef with a ‘healthier’ fatty acid profile. Moreover, strategies will be defined for industry on how to optimise nutritional and sensory properties and oxidative quality of beef products, by combining the nutritional enhancement made in the live animal together with target levels of functional ingredients to be added during processing. 相似文献

The aim of this research was to validate a method for the quantification of conjugated linoleic acid (CLA) isomers in the fat of ewe's milk. The isomers in the form of methyl ester (FAME) are separated by silver ion high performance liquid chromatography (Ag+HPLC) and the quantification was based on the measurement of the integrated area under the 231 nm peaks, using external CLA reference standards. The response linearity is maintained in a wide range of concentrations and the quantification limit estimated was of 0.005 μg/injection for the cis-9,trans-11 and trans-10,cis-12 isomers. Referred to the samples this limit would be 0.008 mg/g of fat in the usual working conditions. In the precision assay an RSDr of 2.14% for the isomer cis-9,trans-11 was calculated and for the rest of the CLA isomers varied between 2.08 and 7.5%. The recovery assays were performed adding CLA cis-9,trans-11 triglyceride and the mean recovery was 96%. The method proposed allows the determination of the individual content of different CLA isomers using one single technique, HPLC and supposes saving time and resources. 相似文献

The objective of this study was to evaluate the effects of a supplementation of pomegranate seed oil (PSO), being rich in punicic acid, on the biochemical parameters of healthy rats. PSO was given to the animals intragastrically for 40?days at concentrations of 1%, 2% and 4%. There were no changes in their total body weight gain, their serum biochemical markers, or in the oxidative stress in their tissues. However, the TBARS values were reduced in the brains of the animals, noting that no significant amounts of conjugated fatty acids were found in this tissue. Conjugated linoleic acid (CLA) was present in all the other tissues studied. The results obtained have demonstrated that punicic acid from PSO was metabolised and incorporated in the form of CLA in different rat tissues. It did not cause alterations in their lipid metabolism, nor did it participate in the processes of oxidation inhibition. 相似文献

The role of dietary fatty acids on cancer is still controversial. To examine the current literature on the protective role of conjugated linoleic acid (CLA) and marine long-chain fatty acids [eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA)] and the risk of breast and prostate cancer, data from 41 case-control and cohort studies and relevant in vitro and animal experiments were included in this 2000–2010 revision. Epidemiological studies on CLA intake or its tissue concentration related to breast and prostate tumorigenesis are not conclusive; EPA and DHA intake have shown important inverse associations just in some studies. OBJECTIVE: We investigated the effects of specific conjugated linoleic acid (CLA) isomers on glucose metabolism and insulin resistance and on mRNA levels of genes important in glucose and lipid metabolism. METHODS: Sprague-Dawley rats were fed for 8 wk on a high-fat diet (45% kcal from fat) or one of three CLA-supplemented diets (1% CLA) containing differing isomers of CLA, including a mixture of CLAs (CLA mix), cis-9, trans-11-CLA (C9,T11-CLA), or trans-10, cis-12-CLA (T10,C12-CLA). RESULTS: Compared with the high-fat group, all the CLA groups had enhanced glucose tolerance. Insulin resistance index was significantly lower in the CLA-treated groups. No significant difference could be observed in the level of serum lipids between groups and in the activities of phosphoenolpyruvate carboxylase, glucose-6-phosphatase, and glucokinase. However, C9,T11-CLA and T10,C12-CLA significantly increased acyl coenzyme A oxidase mRNA in skeletal muscle. In addition, C9,T11-CLA increased hepatic acyl coenzyme A oxidase mRNA and skeletal muscle uncoupling protein-2 mRNA. The CLA mix showed intermediate effects on the levels of these genes. CONCLUSIONS: The addition of all types of CLA to Sprague-Dawley rats fed a high-fat diet can decrease insulin resistance. Possible mechanisms are increased fat oxidation and energy expenditure by increasing acyl coenzyme A oxidase and uncoupling protein-2 mRNA in the liver and/or skeletal muscle. 相似文献

目的通过研究t10,c12-共轭亚油酸(t10,c12-CLA)对高游离脂肪酸环境中骨骼肌细胞脂肪酸代谢的影响,探讨t10,c12-CLA改善脂代谢的作用机制。方法体外培养成熟的C2C12骨骼肌细胞,通过施加软脂酸在体外模拟高游离脂肪酸环境,再给予不同浓度的t10,c12-CLA(20、30和40μmol/L),通过荧光免疫法利用激光共聚焦显微镜观察t10,c12-CLA对细胞脂肪酸转运蛋白(FAT/CD36)的影响,应用Western blot法检测t10,c12-CLA对C2C12细胞脂肪酸代谢相关酶及AMP激活的蛋白激酶(AMPK)的作用。结果t10,c12-CLA剂量依赖性的增加了高游离脂肪酸环境中细胞对脂肪酸的摄取,抑制了乙酰辅酶A羧化酶(ACC)的活性,增加了肉碱酰基转移酶-1(CPT-1)的表达,并且激活了AMPK。结论t10,c12-CLA可通过激活AMPK增强细胞对脂肪酸的氧化代谢。 相似文献

The effect of supplementation with 1% conjugated linoleic acid and 1% n-3 long chain polyunsaturated fatty acids (CLA/n-3) was assessed in rats. Food intake increased with no difference in body weights. White adipose tissue weights were reduced whereas brown adipose tissue and uncoupling protein-1 expression were increased. Plasma adiponectin, triglyceride and cholesterol levels were reduced while leptin, ghrelin and liver weight and lipid content were unchanged. Hypothalamic gene expression measurements revealed increased expression of orexigenic and decreased expression of anorexigenic signals. Thus, CLA/n-3 increases food intake without affecting body weight potentially through increasing BAT size and up-regulating UCP-1 in rats. 相似文献

Objective
Little evidence exists concerning the effects of trans-10,cis-12 conjugated linoleic acid (CLA) under energy restriction. Thus, the effects of this CLA isomer on adipose tissue size, liver composition, as well as on expression and activity of carnitine-palmitoyl transferase I (CPT-I) and acyl CoA oxidase (ACO), in hamsters fed an energy-restricted diet were analyzed.Methods
Hamsters were fed a high-fat diet for 7 wk and then subjected to 25% energy-restricted diets supplemented with 0.5% linoleic acid or 0.5% trans-10,cis-12 CLA for 3 wk. Serum insulin, free-triiodothyronine and non-esterified fatty acid levels, liver triacylglycerol, protein and water contents, and CPT-I, ACO, and Peroxisome proliferator-activated receptor alpha (PPARα) expressions and enzyme activities were assessed.Results
Energy restriction reduced liver size, serum levels of insulin, free-triiodothyronine, and non-esterified fatty acid and increased CPT-I activity. Liver composition was not modified. No differences were found between both restricted groups, with the exception of CPT-I and ACO oxidative enzyme activities, which were greater in hamsters fed the CLA diet.Conclusions
Energy restriction does not cause trans-10,cis-12 CLA to induce liver hyperplasia. Although this CLA isomer increases liver CPT-I and ACO activities, this effect does not result in reduced hepatic triacylglyerol content or decreased adipose tissue size. Consequently, this CLA isomer seems not to be a useful tool for inclusion in body weight loss strategies followed during obesity treatment. 相似文献16.

topics covered.
Aims of the study The aim of the present study was to assess the effects of the trans-10,cis-12 isomer of CLA on cholesterolaemia and on several metabolic pathways involved in cholesterol metabolism in hamsters.
Methods Animals were fed atherogenic diets supplemented with 0.5% linoleic acid, 0.5% trans-10,cis-12 CLA or 1.0% trans-10,cis-12 CLA, for 6 weeks. Serum lipoproteins were separated by FPLC. Cholesterol in serum and liver, as well as triacylglycerols
and phospholipids in liver were assessed by spectrophotometry. 3-Hydroxy-3-methylglutaryl coenzyme A reductase (HMGCoAR),
acyl-coenzyme A:cholesterol acyltransferase (ACAT) and cholesteryl ester hydrolase (CEH) activities were measured by radiometry,
and LDL receptors were determined by Western blot.
Results
trans-10,cis-12 CLA feeding did not modify food intake nor final body weight. Although serum total cholesterol remained unchanged, when
cholesterol fractions were analyzed a significant decrease in VLDL-cholesterol was observed in CLA-fed animals, without changes
in HDL-cholesterol or LDL-cholesterol. trans-10,cis-12 CLA decreased cholesterol ester content and increased free cholesterol in liver. The activity of HMGCoAR was not modified.
In contrast, ACAT activity was reduced by both CLA doses and CEH was increased by the high CLA dose. LDL receptors were significantly
reduced by trans-10,cis-12 feeding when expressed as arbitrary units per mg of protein, however, the total receptor mass remained unchanged.
Conclusions These results suggest that, under the present experimental conditions, trans-10,cis-12 CLA feeding reduces cholesterol esterification in liver and decreases the minority serum VLDL-cholesterol fraction, but
it does not show a hypocholesterolaemic effect. A dose–response effect was not observed. 相似文献

A total of 52 Italian and French commercial cheeses were analysed for fatty acid composition and conjugated linoleic acid (CLA) concentration. The cheeses were first grouped according to the ruminant species of the milk used in manufacturing, then according to the following criteria: (1) cheeses obtained from milk of the same ruminant species but using different production technologies; (2) cheeses produced from milk of different ruminant species but through similar cheesemaking processes. Comparative studies of fatty acid composition and CLA levels in the different cheese classes were carried out. In our study, the arrangement of the ruminant species according to the increasing CLA concentration in cheese was: goat = cow < sheep. Sheep cheeses had the highest levels of CLA (9.86 mg/g fat), α-linolenic acid (0.75%) and trans-vaccenic acid (1.63%), and the lowest contents of linoleic acid (1.80%) and oleic acid (16.83%). The comparison of cheeses obtained from milk of the same ruminant species but through different production technologies reported statistically significant differences in the fatty acid profiles that could be due to a different degree of lipolysis in the cheeses compared. Nevertheless, statistically significant differences in CLA concentration were not found; this suggests that the factors involved in the cheesemaking process generally do not affect the CLA content in milk fat. 相似文献

Conjugated linoleic acid (CLA) purportedly alters body composition, glucose tolerance, hepatic function, lipoprotein distributions, and other markers of health. Results are often inconclusive or contradictory, and presently, no studies have investigated the effects of naturally incorporated CLA from pasture-fed beef and dairy products on human health. We hypothesized that a diet comprised of foods naturally enriched with CLA from pasture-fed cattle would result in improved insulin sensitivity, body composition, circulating lipids, and other disease risk factors when compared to a diet comprised of commercial foods naturally low in CLA from grain-fed cattle. Eighteen healthy women 20 to 39 years of age consumed one of these 2 diets for 56 days. Balanced nutritionally complete diets comprised of 31% energy from lipid, 13% from protein, and 54% from carbohydrate were administered, with the primary difference being CLA content (CLA diet: 1.17 g/d; control diet: 0.35 g/d). The CLA diet did not result in any differences in insulin sensitivity, body composition, circulating blood lipids, or other measured disease risk factors as compared with the control diet. Thus, we conclude that a diet naturally enriched with over a 3-fold increase in CLA from pasture-fed cattle did not significantly alter selected health risk factors in healthy, premenopausal women as compared with a similar diet composed of foods from grain-fed cattle. 相似文献

A study was conducted to compare cholesterol and fatty acid levels of beef from three cattle breeds raised on natural pasture. Thirty-four, twenty-nine and fourteen steers each of Nguni, Bonsmara and Angus, respectively, were slaughtered at 18 months. There were no (P > 0.05) breed differences for the cholesterol and intramuscular fat (IMF) levels. The fatty acid profiles were similar (P > 0.05) among the three breeds except that beef from Nguni steers had higher (P < 0.05) docosahexaenoic acid content than beef from the other two breeds while beef from Bonsmara steers had the highest (P < 0.05) heptadacenoic acid content. It can be concluded that there were no breed differences on cholesterol and most fatty acids. 相似文献

The fatty acid composition of phospholipids plays a key role in the structural and functional properties of cellular membrane. In this study, it was hypothesized that conjugated linoleic acid (CLA) isomer supplementation changes the unsaturation degree of the fatty acids of neutral lipids (NLs) but not those of polar lipids (PLs). Thus, the main goal was to determine the pattern of fatty acid incorporation into hepatic PL and NL fractions. Wistar male rats were fed cis(c)9,trans(t)11 and t10,c12 CLA isomers, separately or as a mixture. Whereas the t10,c12 isomer incorporation in the PL fraction was similar when supplemented either individually or as a mixture, the c9,t11 isomer reached the highest values of incorporation when combined with t10,c12. In the PL fraction, the linoleic acid did not change; but the arachidonic acid decreased, especially in the rats given the mixture. Also in this fraction, the t10,c12 isomer, either separately or as a mixture, decreased the amounts of n-6 long-chain (LC) polyunsaturated fatty acids (PUFA) and increased those of the n-3 LC PUFA relative to the control. In the NL fraction, linoleic acid incorporation followed the diet composition, whereas the arachidonic acid was similar among treatments. Facing CLA isomer supplementation, the present study suggests that fatty acid incorporation into phospholipids, through the balance between n-6 and n-3 LC PUFA, is dependent upon maintaining the unsaturation degree of cellular membrane. 相似文献
