Interplay between integrins and extracellular matrix is suggested to play an important role in malignant progression and tumor differentiation. The aim of the study was to determine the combined expression of integrin β3 and tenascin-c (TN-c) in breast cancer and examine whether integrin β3 and TN-c can activate urokinase-type plasminogen activator (uPA) through p38 mitogen-activated protein kinase (p38 MAPK). We detected the expression of integrin β3, TN-c, p-p38, and uPA in 80 cases of breast invasive ductal carcinoma by immunohistochemistry. In addition, we blocked integrin β3 and TN-c in the MDA-MB-231 breast cancer cells and detected the expression of p-p38 and uPA by Western blot. Integrin β3, TN-c, p-p38, and uPA showed high levels of expression in breast invasive ductal carcinoma. The expression of integrin β3, TN-c, and uPA was correlated with lymph node metastasis and TNM stage in breast cancer. Furthermore, correlations were noted between any two of the three proteins. The expression of p-p38 and uPA decreased in MDA-MB-231 cells after the addition of integrin β3 antibody and TN-c antibody. The expression of uPA decreased after addition of SB203580. Our results demonstrate that inhibition of the expression of integrin β3 and TN-c could decrease the expression of uPA through p38 MAPK in breast cancer, suggesting that the interaction between integrin β3 and TN-c serves an important role in breast cancer.  相似文献

The activation of glial cells in the CNS has been suggested to be involved in abnormal pain sensation after peripheral nerve injury. Previous studies demonstrated phosphorylation of p38 mitogen-activated protein kinase (MAPK) in spinal cord glial cells after peripheral nerve injury, and such phosphorylation has been suggested to be involved in the development of neuropathic pain. The aim of this study was to examine the dorsal column nuclei for phosphorylation of p38 MAPK following peripheral nerve injury and to explore a possibility of its contribution to neuropathic pain. Immunohistochemical labeling for phosphorylated p38 (p-p38) MAPK was performed in histological sections of the rat spinal cord and medulla oblongata after the fifth lumbar (L5) spinal nerve ligation (SNL). The number of p-p38 MAPK-immunoreactive (IR) cells was significantly increased in the L5 dorsal horn and the gracile nucleus ipsilateral to the injury at days 3-21 after SNL. Double immunofluorescence labeling with cell-specific markers revealed that p-p38 MAPK-IR cells co-expressed OX-42, suggesting their microglial identity. Increased immunofluorescence labeling for OX-42 indicated that microglial cells were activated by SNL in the L5 dorsal horn and the gracile nucleus ipsilateral to the injury. Continuous infusion of a p38 MAPK inhibitor into the cisterna magna for 14 days beginning on the day of SNL suppressed the development of tactile allodynia, but not thermal hyperalgesia induced by nerve injury. These results demonstrate that SNL activates p38 MAPK pathway in microglia in the gracile nucleus as well as in the spinal cord dorsal horn. Activation of p38 MAPK in medullary microglia may contribute to the pathogenesis of neuropathic pain.  相似文献

目的:探讨微小RNA-125a-5p(miR-125a-5p)通过GSK-3β/Snail信号通路对乳腺癌细胞上皮-间充质转化(EMT)的影响。方法:RT-qPCR检测人正常乳腺上皮细胞与乳腺癌细胞中miR-125a-5p的表达量,同时检测miR-125a-5p质粒在人乳腺癌MDA-MB-231细胞中的转染效率;趋化运动实验与Transwell侵袭实验检测趋化运动能力和侵袭能力;Western blot检测EMT相关标志物的变化,同时检测磷酸化糖原合成酶激酶3β(p-GSK-3β)的蛋白水平及Snail的转核情况。结果:乳腺癌细胞中miR-125a-5p的表达量明显低于人正常乳腺上皮细胞(P0.05);miR-125a-5p在转染miR-125a-5p质粒的MDA-MB-231细胞中表达水平明显增高;MDA-MB-231细胞的趋化运动能力在表皮生长因子(EGF)浓度为10μg/L时最强;在EGF刺激下,与MDA-MB-231/NC细胞组相比,MDAMB-231/miR-125a-5p细胞组的侵袭能力明显降低,上皮钙黏着蛋白(E-cadherin)表达量升高,波形蛋白(vimentin)和p-GSK-3β的蛋白水平明显降低,同时Snail转核受到明显抑制;与MDA-MB-231/miR-125a-5p+Con细胞组相比,MDA-MB-231/miR-125a-5p+GAB2细胞组的侵袭能力明显增强,E-cadherin表达量降低,vimentin和p-GSK-3β的蛋白水平明显升高,同时促进Snail转核。结论:miR-125a-5p可通过GSK-3β/Snail信号通路抑制乳腺癌细胞的EMT,进而抑制乳腺癌细胞的侵袭能力。  相似文献

目的:探讨p38 MAPK在顺铂诱导大鼠近端肾小管上皮细胞(RPTC)凋亡中的作用。方法:首先采用Western blot实验检测0、5、10和20μmol/L顺铂处理24 h对细胞凋亡的影响,确定最佳处理剂量;而后采用20μmol/L顺铂联合50 mg/L p38 MAPK抑制剂SB203580刺激RPTC,实验分为对照组、顺铂组及顺铂+SB203580(加入SB203580处理RPTC 1 h后再给予顺铂处理24 h)。采用相差荧光显微镜观察和流式细胞术分析顺铂处理后RPTC的凋亡情况;采用Ac-DEVD-AFC试剂盒检测RPTC裂解液中的caspase活性;Western blot实验检测p38、磷酸化p38、cleaved PARP和cleaved caspase-3等的蛋白水平;pH计检测顺铂处理后RPTC外环境pH值改变。结果:20μmol/L顺铂处理RPTC 24 h,可以明显诱导细胞凋亡;顺铂处理15 min后RPTC中p38 MAPK开始磷酸化并达到高峰。顺铂处理后12.08%的RPTC呈凋亡形态,具有增强的caspase活性,并且cleaved PARP和cleaved caspase-3水平明显升高(P0.05);p38 MAPK抑制剂SB203580可抑制p38的磷酸化,降低RPTC的凋亡率和caspase活性,并减少cleaved PARP和cleaved caspase-3的蛋白水平。同时,SB203580可逆转顺铂诱导的RPTC培养基pH值的改变。结论:p38 MAPK的磷酸化在顺铂诱导的RPTC凋亡中发挥作用。顺铂诱导RPTC凋亡后,可改变细胞外酸性环境,并可被p38 MAPK抑制剂SB203580所抑制。  相似文献

CAS proteins and Ezrin, Radixin, Moesin (ERM) family members act as intracellular scaffolds and are involved in interactions with the cytoskeleton, respectively. Both protein families have previously been associated with metastasis and poor prognosis in cancer. Our group recently reported on the overexpression of EZR/VIL2 and BCAR1 and their protein products in breast carcinoma effusions compared to primary breast carcinoma. In the present study, the role of these two proteins was studied in semi-normal MCF10A cells and metastatic MDA-MB-231 breast carcinoma cells cultured in tri-dimensional (3-D) conditions that were hypothesized to reproduce the in vivo conditions of breast cancer metastasis. MCF10A cells formed spheroid-shaped colonies without any Matrigel invasion, while MDA-MB-231 cells displayed an invasive phenotype and showed satellite projections that bridged multiple cell colonies in 3-D culture. E-cadherin was expressed in MCF10A, but not in MDA-MB-231 cells. The temporal expression of ezrin and BCAR1/p130Cas at the mRNA and protein level differed in the two cell lines upon 3-D culturing on Matrigel. Upregulation of BCAR1/p130cas was observed in the transition of MDA-MB-231 from attached to detached culture. Silencing of Ezrin and p130Cas in MDA-MB-231 cells by short hairpin RNA resulted in decreased invasive potential, and p130Cas silencing further resulted in smaller spheroid/colony formation. Our data show that MCF10A and MDA-MB-231 cells differ in their ability to form spheroids, in expression of E-cadherin and in the expression of Ezrin and BCAR1/p130Cas in 3-D cultures on Matrigel, suggesting a role in tumor progression in breast carcinoma.  相似文献

Cells cultured within a three-dimensional (3D) in vitro environment have the ability to acquire phenotypes and respond to stimuli analogous to in vivo biological systems. This approach has been utilized in tissue engineering and can also be applied to the development of a physiologically relevant in vitro tumor model. In this study, collagen I hydrogels cultured with MDA-MB-231 human breast cancer cells were bioengineered as a platform for in vitro solid tumor development. The cell-cell and cell-matrix interactions present during in vivo tissue progression were encouraged within the 3D hydrogel architecture, and the biocompatibility of collagen I supported unconfined cellular proliferation. The development of necrosis beyond a depth of ~150-200 μm and the expression of hypoxia-inducible factor (HIF)-1α were demonstrated in the in vitro bioengineered tumors. Oxygen and nutrient diffusion limitations through the collagen I matrix as well as competition for available nutrients resulted in growing levels of intra-cellular hypoxia, quantified by a statistically significant (p < 0.01) upregulation of HIF-1α gene expression. The bioengineered tumors also demonstrated promising angiogenic potential with a statistically significant (p < 0.001) upregulation of vascular endothelial growth factor (VEGF)-A gene expression. In addition, comparable gene expression analysis demonstrated a statistically significant increase of HIF-1α (p < 0.05) and VEGF-A (p < 0.001) by MDA-MB-231 cells cultured in the 3D collagen I hydrogels compared to cells cultured in a monolayer on two-dimensional tissue culture polystyrene. The results presented in this study demonstrate the capacity of collagen I hydrogels to facilitate the development of 3D in vitro bioengineered tumors that are representative of the pre-vascularized stages of in vivo solid tumor progression.  相似文献

 目的:探讨在ErbB2非过表达乳腺癌细胞MDA-MB-231中,神经调节因子(NRGs)/ ErbB2信号通路对突变型p53(mtp53)和缺氧诱导因子-1α(HIF-1α)的影响及意义。方法:免疫细胞化学法和Western blotting检测MDA-MB-231细胞中神经调节因子(NRG)的表达。应用rbB2受体功能抑制剂AG825处理MDA-MB-231细胞,MTT法检测细胞的增殖抑制作用;流式细胞术检测细胞周期和细胞凋亡; Western blotting检测mtp53、HIF-1α蛋白表达;RT-PCR检测HIF-1αmRNA的表达。结果:神经调节因子在乳腺癌细胞MDA-MB-231呈显著表达,Western blotting实验可见分子量44 kD的NRG抗体阳性反应条带。应用ErbB2受体功能抑制剂AG825后,MDA-MB-231细胞生长受到抑制,作用呈时效、量效依赖关系(P<0.01);细胞周期阻滞在G0/G1期;细胞凋亡增加(P<0.05);mtp53、HIF-1α蛋白表达减少(P<0.05);HIF-1α mRNA表达减少(P<0.05)。结论:ErbB2非过表达乳腺癌细胞MDA-MB-231存在神经调节因子分泌,可能通过神经调节因子自分泌或旁分泌配体作用方式使ErbB2受体信号转导处于激活状态,上调mtp53和HIF-1α的表达,促进肿瘤细胞的增殖、存活能力和抑制凋亡。  相似文献

 目的:探讨右美托咪啶(dexmedetomidine, Dex)对异氟醚(isoflurane, Iso)诱导的新生大鼠海马神经元凋亡的影响及其与p38丝裂原活化蛋白激酶(p38 mitogen-activated protein kinase,p38)和c-Jun 氨基末端激酶(c-Jun N-terminal kinase, JNK)蛋白活化的关系。方法:48只出生后7 d的SD大鼠随机分为对照组(Con组)、Dex组、Iso组和Iso+Dex组。前2组吸入空气,后2组吸入0.75% Iso 6 h。Dex组和Iso+Dex组在麻醉前20 min和麻醉开始后2 h、4 h经腹腔注射25 μg·kg-1 Dex;Con组和Iso组则在相同的时点注射150 μL生理盐水。麻醉结束后使用TUNEL法检测海马神经元凋亡; Western blotting检测海马组织激活型caspase-3、p38、磷酸化p38(p-p38)、JNK和磷酸化JNK(p-JNK)蛋白表达的变化。结果:(1) Iso组海马CA1区TUNEL阳性细胞数较Con组增加447.57%(P<0.01),Dex抑制Iso诱导的TUNEL阳性细胞增加达75.18%(P<0.01)。(2) Iso组海马组织激活型caspase-3的表达比Con组增加126.29% (P<0.01);Dex显著抑制了Iso诱导的caspase-3活化(P<0.01)。(3) Iso组海马p-p38/p38和p-JNK/JNK比值均较Con组明显增加(P<0.01);Dex显著抑制了Iso诱导的p-p38 和p-JNK表达增加(P<0.01)。结论:Dex能通过减少海马神经元凋亡来减轻Iso对新生大鼠的脑毒性。抑制p38和JNK的磷酸化可能是Dex发挥保护作用的机制之一。  相似文献

Crohn's disease and ulcerative colitis are inflammatory bowel diseases (IBD) characterized by chronic relapsing mucosal inflammation. Tumour necrosis factor (TNF)‐α, a known agonist of the mitogen‐activated protein kinase (MAPK) pathway, is a key cytokine in this process. We aimed first to determine whether p38 MAPK is activated in IBD inflamed mucosa, and then studied the effect of four different p38α inhibitory compounds on MAPK phosphorylation and secretion of proinflammatory cytokines by IBD lamina propria mononuclear cells (LPMCs) and organ culture biopsies. In vivo phospho‐p38α and p38α expression was evaluated by immunoblotting on intestinal biopsies from inflamed areas of patients affected by Crohn's disease and ulcerative colitis, and from normal mucosa of sex‐ and age‐matched control subjects. Both mucosal biopsies and isolated LPMCs were incubated with four different p38α selective inhibitory drugs. TNF‐α, interleukin (IL)‐1β and IL‐6 were measured in the organ and cell culture supernatants by enzyme‐linked immunosorbent assay. We found higher levels of phospho‐p38α in the inflamed mucosa of IBD patients in comparison to controls. All the p38α inhibitory drugs inhibited p38α phosphorylation and secretion of TNF‐α, IL‐1β and IL‐6 from IBD LPMCs and biopsies. Activated p38α MAPK is up‐regulated in the inflamed mucosa of patients with IBD. Additionally, all the p38α selective inhibitory drugs significantly down‐regulated the activation of the MAPK pathway and the secretion of proinflammatory cytokines.  相似文献

We studied p38 phosphorylation and its intracellular localization during p53 and Puma (a p53 upregulated modulator of apoptosis) apoptotic signaling pathway in bone marrow granulocytes in mice irradiated in vivo and the role of the radioprotector amifostine in ameliorating these responses. Sixty-four C57BL mice were randomly assigned in two non-irradiated (Ami−/rad− and Ami+/rad−) and two irradiated (Ami−/rad+ and Ami+/rad+) groups. Animals received 400 mg/kg of amifostine i.p. 30 min prior to a single whole body radiation dose of 7 Gy. The experiments were performed using immunohistochemistry for caspase-3, cleaved caspase-3, p53, p-p53 (Ser 15), Puma, p38 and p-p38 (Thr 180/Tyr 182) protein expression. In addition transmission electron microscopy was used for ultrastructural characterization of apoptosis. Data showed that: (i) amifostine significantly reduced the number of apoptotic cells, (ii) p-p53 and Puma proteins were strongly immunostained in granulocytes after irradiation (Ami−/rad+), (iii) amifostine decreased the immunostaining of the proteins (Ami+/rad+), (iv) p38 was immunolocalized in physiological conditions in the nucleus and cytoplasm of granulocytes and neither radiation nor amifostine changed the protein immunostaining or its subcellular distribution, but influenced its activation, (v) radiation-induced p38 phosphorylation and its cytoplasmic accumulation during apoptosis signaling in granulocytes after whole body high radiation dose and amifostine markedly reduced these effects.  相似文献

目的探讨氧化应激条件下CD200如何参与调控MG(d'胶质细胞)激活后的炎性反应。方法对原代培养的MG进行氧化应激处理,并利用急性分离的皮质神经元进行干预。“对照组”和“H2O2组”分别采用生理盐水和H2O2(100μmol/L)进行处理,“H2O2+CD200组”在H2O2(100μmol/L)处理之前30min采用急性分离的神经元(约含1×10^6细胞)进行预处理。于处理后第0.5h、1h、2h、3h和4h时,采用Western blotting法对MG内p-p38 MAPK和IκB-α的蛋白表达量进行检测;以及第0h和72h时,采用ELISA法对培养介质内的TNF-α和IL-1β的含量进行检测。结果p-p38 MAPK和IκB—α的蛋白表达:“对照组”各时间点均无明显变化;“H2O2组”和“H2O2+CD200组”0.5b时表达量亦基本相同;而3h时“H2O2组”p-p38 MAPK蛋白表达量明显高于“H2O2+CD200组”;4h时“H2O2组”IκB-α的蛋白表达量明显低于“H2O2+CD200组”。TNF-α和IL-1β含量:各组培养介质内0h时均未检出含有TNF-α和IL-1β;第72h时两细胞因子含量高低次序相同,即“对照组”〈“H2O2+CD200”〈“H2O2组”。结论氧化应激条件下,CD200分子通过抑制MG胞质内p38 MARK的磷酸化,进而抑制了NF—κB的激活和炎性因子表达水平。  相似文献

Mitogen-activated protein kinases (MAPKs) are part of an intracellular signaling machinery consisting of three known distinct pathways, each leading to activation of a different protein kinase: p38, ERK (extracellular signal-regulated kinase), or JNK (c-Jun N-terminal kinase). We investigated the role of the p38 MAPK pathway in the phenomenon of lung endotoxin "priming": incubation of perfused rat lungs with lipopolysaccharide (LPS) for 2 hours results in drastically enhanced cyclooxygenase-2-dependent and thromboxane synthase-dependent vasoconstriction and bronchoconstriction, including edema formation in response to a second inflammatory stimulus, such as arachidonic acid application. Two unrelated selective inhibitors of p38 (SB203580 and SC-68376) dose dependently suppressed the arachidonic acid-induced pulmonary artery pressor response, edema formation, and bronchoconstrictor response in both control lungs and lungs that underwent preceding endotoxin priming. In parallel, thromboxane, but not prostacyclin, released into the lung perfusate was dose dependently inhibited. Using immunohistochemical techniques in combination with quantitative microdensitometry, p38 was detected in nearly all cell types in control lungs, whereas the activated form p-p38 was only expressed in certain cell types, eg, bronchial epithelial cells, endothelial cells, alveolar macrophages, and vascular smooth muscle cells (SMC) of small vessels. In response to endotoxin, p-p38 expression was additionally observed in septal cells, bronchial SMC, and vascular SMC of larger pulmonary vessels and was increased in most other cell types including small-vessel SMC. We conclude that both immunolocalization of p38 activity and pharmacologic interventions support a strong role of the p38 MAPK pathway in establishing an active cyclooxygenase-2/thromboxane synthase axis in vascular and bronchial SMC, with up-regulation of this signaling cascade occurring in LPS priming and being responsible for enhanced pulmonary artery pressor response, edema formation, and bronchoconstriction. Moreover, LPS induces or increases phosphorylation of p38 in other lung cell types. The physiologic consequences of these events remain to be established.  相似文献

PKZ, protein kinase containing Z-DNA binding domains, is a novel member of eIF2α kinase in fish. PKZ can be up-regulated in response to Poly I:C or heat stress, and it has a typical eIF2α kinase activity. However, the relationship between fish PKZ and apoptosis remains unclear. In the present study, effects of PKZ on apoptosis were explored. Initially, we found that PKZ and PKR were up-regulated by poly I:C in a time-dependent manner. Q-PCR analysis showed that the change of Caspase-3 mRNA was consistent with that of PKZ under the stimulation of poly I:C or transfection with PKZ siRNA in CIK cells. It suggested that PKZ might mediate the induction of apoptosis. Knockdown and overexpression assays indicated that a significant increase of apoptotic cell number was observed in the wild type PKZ (PKZ-wt) transfected PKZ-deficient cells, while mutant PKZ-K198R lost this ability. MTT also showed that overexpression of PKZ-wt in CIK cells resulted in a striking decrease of cell viability rate to about 32.5%, whereas the viability of cell with PKZ-K198R was about 88.1%. Likewise, when transfected eIF2α-wt or phosphomimetic eIF2α (S51D), CIK cells displayed higher apoptotic rate than the controls. In contrast, overexpression of phosphodeficient eIF2α (S51A) blocked induction of apoptosis. In addition, levels of eIF2α phosphorylation were significantly related to apoptosis in these CIK cells. Furthermore, when mutant eIF2α (S51A) was transiently transfected into the PKZ/PKR knockdown cells that transfected with wildtype PKZ previously, the apoptotic rates and levels of eIF2α phosphorylation were dramatically decreased. Overall, these results suggested that PKZ could facilitate apoptosis via eIF2α phosphorylation.  相似文献

目的:研究下调非洲爪蟾驱动蛋白样蛋白2靶蛋白(TPX2)对直肠癌细胞凋亡的影响及机制。方法:用TPX2小干扰RNA(si RNA)转染直肠癌HR-8348细胞,记为TPX2 si RNA组;以不做转染的细胞作为正常对照(control)组;以转染si RNA阴性对照(si RNA-NC)的细胞作为si RNA-NC组;用p38 MAPK抑制剂处理敲减TPX2表达后的直肠癌HR-8348细胞记为TPX2 si RNA+SB203580组。RT-qPCR和Western blot测定TPX2的表达水平,MTT法测定细胞存活率,流式细胞术测定细胞凋亡,Western blot测定细胞中p38 MAPK、p-p38 MAPK、cleaved caspase-3和Bcl-2的蛋白水平。结果:TPX2 si RNA转染后HR-8348细胞中TPX2的m RNA和蛋白表达水平显著下降(P 0. 05),而转染si RNA-NC对HR-8348细胞中TPX2的m RNA和蛋白水平没有影响。敲减TPX2表达后的直肠癌HR-8348细胞存活率降低,凋亡率升高,细胞中的cleaved caspase-3、p-p38 MAPK/p38 MAPK蛋白水平明显升高,Bcl-2水平水平降低,与control组比较,差异有统计学意义(P 0. 05)。与TPX2 si RNA组相比,TPX2 si RNA+SB203580组的HR-8348细胞凋亡率、cleaved caspase-3水平和p-p38 MAPK/p38 MAPK蛋白水平明显降低,存活率明显升高(P 0. 05)。结论:TPX2表达下调可以通过激活p38 MAPK促进直肠癌HR-8348细胞凋亡。  相似文献

目的:研究c-Met对三阴性乳腺癌细胞株MDA-MB-231活力及对阿霉素耐药性的影响。方法:构建阿霉素耐药的MDA-MB-231/ADR细胞系,实时荧光定量PCR及Western blotting技术检测不同细胞系中c-Met mRNA及蛋白的表达。脂质体转染c-Met-siRNA及表达质粒或AKT-siRNA,Western blotting检测转染效率;四甲基偶氮唑法(MTT法)检测细胞的活力及对阿霉素的敏感性。结果:对阿霉素耐药的MDA-MB-231/ADR细胞中cMet的mRNA及蛋白表达均显著高于对照的MDA-MB-231细胞,转染高表达c-Met的质粒可增加MDA-MB-231细胞的活力并降低其对阿霉素的敏感性,而利用siRNA抑制耐药细胞株中c-Met的表达后,可以逆转MDA-MB-231/ADR细胞对阿霉素的耐药。此外,c-Met可以磷酸化激活细胞中的AKT,并通过该信号分子增加MDA-MB-231细胞活力并诱导耐药。结论:c-Met可作为一个重要的靶点应用于三阴性乳腺癌的治疗。  相似文献
