针对结核杆菌菌体含有蜡质的特点和传统Ziehl Neelsen抗酸染色法存在的不足 ,笔者在以前工作的基础上对染色液的配方进行改良〔1,2〕,并结合微波辐射技术建立了一种快速、敏感、简便的痰涂片抗酸染色法。1 材料与方法1.1 材料  5 3例结核杆菌痰培养阳性的痰液 ,每例涂片 2张 ,95 %乙醇固定 15~ 30min ,分别用Ziehl Neelsen改良染色法和传统的抗酸染色法染色 ,同时设阳性对照、阴性对照。1.2 染液配制  (1)改良的Ziehl Neelsen染色液 :碱性复红无水乙醇饱和液 (相当 6 %浓度 ) 10ml,活性剂 5ml,蒸馏水85ml,先将碱性复红溶于无水乙醇…  相似文献

本文采用PCR技术检测结核杆菌的DNA(TB-DNA),其中临床诊断为结核或可疑结核者9例,肿瘤6例,结节病3例,淋巴结炎4例,共22例。病理诊断确诊为不典型结核者14例,不能除外结核者4例,结节病2例,瘤样病变2例。所有病例经抗酸染色均未查到结核杆菌。PCR检测结果表明,22例中18例呈阳性(占81.82%),4例呈阴性。PCR技术敏感、特异,对于不典型结核病例检查TB-DNA片段用于辅助诊断颇  相似文献

Bodian染色法[1] 于 1936年首创 ,是一种用于观察神经元、轴突和树突内神经原纤维变化的浸银染色法。基本原理是把甲醛固定后的石蜡切片浸于银溶液中 ,再用还原液处理后 ,使银颗粒沉积于神经纤维上呈现为棕色或棕黑色。传统染色液的配制是在蛋白银溶液内放入金属铜。但此方法存在着以下几个缺陷 :(1)背景色深 ,血管和胶原纤维有共染现象 ,影响对神经纤维的观察。 (2 )染色时间难于掌握。 (3)容易脱片。我们对经典Bodian染色法进行了改良 ,建立了一种去铜改良型Bodian染色法 ,并对其应用进行了探索。一、材料与染色步骤1.…  相似文献

目的 建立改良抗酸染色法并分析其在盆腔结核中的诊断价值.方法 对48例盆腔结核患者盆腔积液用离心涂片法、改良抗酸染色法和BACTEC MGIT 960培养法进行检测,并对结果进行分析.结果 离心涂片法、改良抗酸染色法和BACTEC MGIT 960培养法阳性率分别为2.08% (1/48)、39.58% (19/48)和37.50% (18/48),改良抗酸法分别与离心涂片法及BACTEC MGIT 960培养法比较,改良抗酸染色法的阳性检出率最高,其与离心涂片法差异有统计学意义(P<0.001),但与BACTEC MGIT 960培养法差异无统计学意义(P>0.05).结论 改良抗酸染色法较离心涂片法阳性率明显提高,虽然和BACTEC MGIT 960培养法阳性率无明显差异,但是改良抗酸染色法可以快速诊断盆腔结核,对于临床工作有重要意义.  相似文献

CD1d presentation of alpha-galactosyl ceramides to natural killer T cells has been a focal point of the study of regulatory T cells. KRN7000, an alpha-galactosyl ceramide originally generated from structure activity studies of antitumor properties of marine sponge glycolipids, is currently the most commonly used agonist ligand and is used to stain NKT cells. However, this glycolipid suffers from poor solubility and availability. We have developed an alpha-galactosyl ceramide with improved solubility over KRN7000 that effectively stains NKT cells, both mouse and human, and stimulates cytokine release at low concentrations.  相似文献

目的评价一种改良的网织红细胞染色液。方法对改良网织红细胞染色液进行显微镜镜下观察试验、染色时间试验、有效期试验、准确度试验和精密度试验。结果改良网织红细胞染色液染色后的标本在显微镜镜下具有不易褪色、沉渣少、视野清晰、背景明亮等特点,且改良网织红细胞染色液染色时间短(5min),有效期长(30个月),准确度与煌焦油蓝盐水染色液一致(P〉0.05),精密度好于煌焦油蓝盐水染色液。结论改良网织红细胞染色液是一种优秀的网织红细胞染色液,值得推广应用。  相似文献

In the serum of a healthy blood donor, Mar, antibodies were identified that agglutinate blood group M erythrocytes only after their preincubation in glucose-containing solution. This agglutination was inhibited by blood group M glycoprotein preincubated in glucose solution but not by untreated M and N glycoproteins or N glycoprotein pretreated with glucose. A similar "activating' effect on M erythrocytes or M glycoprotein was achieved when glucose was replaced by mannose or N-acetylglucosamine, whereas several other replaced by mannose or N-acetylglucosamine, whereas several other sugars were ineffective. The "activating' effect of glucose on M antigen was time-, temperature-, pH-, and concentration-dependent. The binding of glucose to M glycoprotein was demonstrated under the preincubation conditions used in the above mentioned agglutinations. The results obtained suggest that the antibodies of serum, Mar, react with blood group M determinants modified by formation of a glycosylamine linkage between amino groups of the glycoprotein and glucose or other hexoses with the same configuration at asymmetric carbons 3, 4 and 5 as glucose.  相似文献

Renal pathology uses a battery of stains to allow proper assessment of all renal components. One of the most useful of these stains is the silver methenamine with a Masson trichrome counterstain (SMMT). The SMMT stain uses thin 1-um sections to identify immune complexes deposits on the basement membranes and in the mesangium of the glomerulus and can provide an excellent and inexpensive method. Problems can, however, occur when the stain is not performed by a technician or the stain needs to be performed in large numbers. A variation to the SMMT method has proved to provide a better, more robust and reproducible counterstain. The new method substitutes Light Green SF, used in the traditional Masson trichrome stain, with Methyl Green, a dye with much smaller molecule size, allowing better penetration and a stronger bonding to the tissue components.  相似文献
