肿瘤的侵袭性和转移性是恶性肿瘤细胞的标志,通过肿瘤细胞与其周围组织的相互作用而体现。在这个病理过程中,有很多基因及其表达产物的参与。S100蛋白家族是一类EF-手型钙结合蛋白,通过对钙离子的调节及与靶蛋白的相互作用,发挥重要的生物学作用,参与细胞周期活动、细胞分化、肿瘤生长以及细胞外基质分泌活动等过程,且其多个成员在多种肿瘤中表达异常,特别是S100A4、S100A6的异常表达,与肿瘤的侵袭与转移相关。因此S100家族与肿瘤的发生发展关系密切。 相似文献

目的 多数前列腺癌有两个以上病灶,试图探索这些不同病灶是同源还是各自独立起源.方法 选用19例有两个以上病灶的前列腺癌标本,应用显微切割技术获取肿瘤组织,提取DNA.进行PCR扩增,检测染色体8p(D8S133、D8S136、D8S137)和17q(D17S855)4个位点的等位基因杂合性缺失(LOH).结果 在19例前列腺癌中发现40个病灶,其D8S133,D8S136,D8S137和D17S855 LOH 率分别为74%、38%、86%和46%.18例中有15例(83%)多发灶LOH不相同,显示其病灶各为独立起源.3例组织学结构显著不同的多发癌灶,LOH为单克隆性(同一来源).结论 多数前列腺癌同一肿瘤中不同病灶的起源不同. 相似文献

目的:探讨S100A8、S100A9在子宫颈鳞癌及正常宫颈组织中的表达及其意义。方法:采用半定量的RT-PCR技术和免疫组织化学的方法,从mRNA及蛋白水平检测32例子宫颈鳞癌和15例正常宫颈组织中S100A8、S100A9的表达,及与不同临床病理参数的关系。结果:子宫颈鳞癌组织中,S100A8、S100A9在mRNA及蛋白水平上均低表达(P<0.05),其中,低分化肿瘤的表达明显低于高、中分化肿瘤(P<0.05)。结论:S100A8、S100A9在子宫颈鳞癌组织中低表达,且可能与鳞状细胞的分化程度相关。提示S100A8、S100A9低表达可能与子宫颈鳞癌的发生发展有关系。 相似文献

S100A9是一种钙结合蛋白,主要限制性地表达于单核-巨噬细胞系、中性粒细胞以及特定病理状态下的角质化细胞中[1]。S100A9高选择性结合 Ca2+、Zn2+,以及花生四烯酸、角蛋白中间丝、晚期糖基化终产物受体( receptor for advanced glycation end products, RAGE)、Toll样受体4(Toll-like receptor 4,TLR4)、基质金属蛋白酶( matrix metalloproteinase,MMP)等,具有细胞内、外调节活性,参与炎症反应和肿瘤发展过程[2]。S100A9与感染性疾病、免疫性疾病、肿瘤等多种疾病相关,以S100 A9蛋白为治疗靶点的研究也成为热点。 相似文献

目的检测胰腺癌患者血清S100A8与S100A9的水平及探讨其临床意义。方法收集胰腺癌患者90例,同期纳入体检健康者50例为对照组。检测两组研究者血清中S100A8和S100A9的水平。分析胰腺癌患者血清S100A8和S100A9水平与其临床病理特征的相关性。对所有患者疗后随访6个月,比较死亡组与存活组患者血清中S100A8和S100A9水平。结果胰腺癌患者血清中S100A8和S100A9水平显著高于对照组(P<0.01)。胰腺癌患者血清中S100A8和S100A9水平与患者年龄、性别、肿瘤部位无显著相关(P>0.05),与其分化程度、临床分期呈明显相关(P<0.01)。6个月随访结果:27例死亡,18例存活,其中死亡组患者入院时血清S100A8和S100A9水平明显高于存活组,比较有显著性差异(P<0.01)。结论胰腺癌患者血清中S100A8和S100A9水平明显提高,且与其恶性程度密切相关,可能对临床评估胰腺癌预后有一定参考价值。 相似文献

目的 分析小核糖体亚基S2 9、18SrRNA和甘油醛 3 磷酸脱氢酶 (GAPDH)三种基因在手术切除的人肺肿瘤组织中的表达水平并作比较。方法 Northern分子杂交 ,结果采用密度扫描进行分析。结果 发现GAPDH基因在肺肿瘤组织 (包括良性肿瘤 )中表达调高 ,而S2 9基因和 18SrRNA基因的表达水平没有明显变化。结论 在分析肿瘤的基因表达状况时 ,不应用GAPDH作为标定mRNA量的内参照 ,而应当同时研究组织和培养细胞两方面。GAPDH的表达调高可能是肿瘤发生的一个指征。 相似文献

目的 探讨原发性肝癌患者血清S100A8和S100A9的表达水平及其与临床病理特征的相关性.方法 收集原发性肝癌患者共90例,同期选择在我院进行体检健康者50例作为对照组;检测2组血清S100A8和S100A9的水平;分析肝癌患者血清S100A8和S100A9水平与临床病理特征的相关性;比较随访期间死亡组与存活组患者血清S100A8和S100A9表达.结果 与对照组比较,肝癌组患者血清S100A8和S100A9均显著升高(P<0.01).肝癌患者血清S100A8和S100A9的高表达与患者的年龄、性别、大小、病理分型无关(P>0.05),而与TNM分期、分化程度、淋巴结转移以及血行转移有关(P<0.01).所有患者6个月定期随访,31例死亡,59例存活;死亡组患者入院时血清S100A8和S100A9表达均显著高于存活组,差异有统计学意义(P<0.01).结论 原发性肝癌患者血清S100A8和S100A9水平均显著升高,与肝癌的恶性程度关系密切,有望成为原发性肝癌预后的重要评判指标. 相似文献

We report the use of Alemtuzumab (Campath-1H) as salvage treatment in three patients with advanced mycosis fungoides/Sézary
syndrome who had previously been treated with conventional chemotherapy. Two patients (case 1 and case 2), aged 42 and 68
yr, respectively, were heavily pretreated (more than three prior therapy regimens, including autologous transplant in case
2) and refractory to conventional chemotherapy, and the third patient (case 3), aged 80 yr, who had refused any chemotherapy,
had been resistant to treatment with cyclosporine and steroids. Campath-1H was administered intravenously, after an escalating
dose from 3 to 10 mg, at the dose of 30 mg, three times weekly, to a total dose of 1080, 223, and 480 mg, respectively. The
patients with Sézary syndrome (case 2 and case 3) showed clearance of circulating Sézary cells and clinical improvement of
the skin lesions after 2 wk of treatment. Two patients (case 1 and case 3) completed the treatment (12 and 6 wk) without significant
toxicity, the former achieving a partial response and the latter a clinical complete response. The patient (case 2), who suffered
from ischemic cardiopathy and diabetes, quickly achieved a clinical improvement of the Sézary syndrome, but he died because
of a myocardial infarction after 3 wk of treatment.
Our report shows that the treatment with Campath-1H is active even in patients with advanced refractory mycosis fungoides/Sézary
syndrome. Further clinical observations on a larger cohort of patients are needed to establish if Campath-1H may have a role
as first line therapy in addition to conventional therapy including chemotherapy. 相似文献

PURPOSE OF REVIEW: Mycosis fungoides and the Sézary syndrome represent a heterogeneous group of good-to-intermediate-risk non-Hodgkin lymphomas that have recently been identified as distinct histopathologic and clinical entities by the World Health Organization and European Organization for Research on the Treatment of Cancer lymphoma classification systems. Significant progress has been made in identifying and categorizing patients based on clinical prognostic factors, but there is little information regarding the etiology, molecular biology, and molecular genetics of these diseases. This review outlines recent advances in clinical diagnosis and prognosis as well as novel therapeutic approaches. RECENT FINDINGS: A number of reports have further defined clinical prognostic subgroups among early-stage patients and those with circulating Sézary cells. The recent availability and demonstrated efficacy of the oral RXR retinoid, bexarotene, has altered the treatment paradigm of early-stage patients who would not otherwise be exposed to systemic therapies. Novel targeted agents and receptor-directed therapies, including the fusion toxin, denileukin diftitox, histone deacetylase inhibitors, and novel nucleoside analog therapies, have demonstrated promising activity and are undergoing further clinical evaluation. The evolution of immunotherapy has been augmented by studies demonstrating the efficacy of peptide-loaded dendritic cells as well as the use of photopheresis to generate an anti-idiotype cytotoxic T-cell response. SUMMARY: This review will enumerate the most recent findings with respect to clinical staging, prognosis, and treatment of patients with mycosis fungoides and the Sézary syndrome. Novel treatment options will be reviewed and treatment paradigms will be outlined. 相似文献

目的:研究Dickkopf相关蛋白3(Dickkopf-related protein 3,Dkk3)、分泌型卷曲相关蛋白1(secreted friz-zled-related protein 1,SFRP1)和SFRP4在子宫内膜癌(endometrial carcinoma,EC)发生、发展中的作用及病理相关性。方法收集EC标本58例,子宫内膜正常组织30例。用RT-PCR、Western blot法检测组织中Dkk3、SFRP1和SFRP4的表达变化。结果Ⅱ、Ⅲ、Ⅳ期子宫内膜癌组织和正常组织比较,Dkk3、SFRP1和SFPRP4的mRNA及蛋白表达水平降低,并随着组织分期程度的升高而降低(均P<0.05)。结论 Dkk3、SFRP1和SFRP4与EC的分期呈负相关,可考虑作为EC的分期标志。 相似文献
