目的 :探讨白细胞介素 10 (IL 10 )基因转染对小鼠心脏移植排斥反应中IL 12、IL 15、IL 18和IL 4表达的影响。方法 :采用小鼠颈部心脏移植模型 ,随机分为 3组 :对照组、移植组和IL 10组。于术后第 5天取移植心脏 ,用逆转录聚合酶链式反应 (RT PCR)法观察IL 12、IL 15、IL 18、IL 4及IL 10的表达情况。结果 :移植组IL 12、IL 15、IL 18表达与对照组比较明显升高 ,IL 10、IL 4表达显著降低 (均P <0 .0 1)。IL 10组IL 12、IL 15、IL 18表达与移植组比较明显降低 ,而IL 4及IL 10表达显著升高 (均P <0 .0 1)。结论 :IL 10基因转染抑制心脏移植排斥反应主要与其抑制IL 12、IL 15、IL 18等Th1型细胞因子的表达 ,促进Th2型细胞因子IL 4的表达 ,使免疫反应由Th1型向Th2型偏移有关  相似文献

目的 :研究白细胞介素 (IL) 1β在大鼠心脏移植排斥反应中心脏局部的表达变化情况 ,探讨其在心脏移植急性排斥反应诊断中的可能作用。方法 :建立大鼠颈部异位心脏移植模型 ,不同品系大鼠之间的移植供、受体分别为SD大鼠和Wistar大鼠 (共 5 0只 ) ,同品系大鼠之间的移植供受体均为Wistar大鼠 (共 5 0只 )。观察移植心脏自然生存时间 ,另于术后第 1、3、5、7、9、11天 ,分别切取移植心脏各 5只 ,提取总RNA ,进行逆转录 ,将目的引物和对照引物在同一管中扩增 ,以两者像素值比值均数作曲线图 ,观察排斥反应时IL 1βmRNA的动态表达情况。结果 :不同品系移植心脏的停跳高峰出现在术后第 6~ 10天。排斥反应时移植心脏局部IL 1βmRNA的表达水平明显增高 ,峰值在术后第 1天。结论 :IL 1β在心脏移植排斥反应中可能起重要的作用 ,IL 1βmRNA的动态检测可望成为早期诊断急性排斥反应的指标  相似文献

目的 研究不同细胞因子微环境下,白细胞介素(IL)-18对早期Ⅱ型胶原诱导性关节炎(CIA)的作用.方法 CIA小鼠,发病前给予每只重组小鼠腹腔注射IL-18(0.2 μg/d)/IL-10(0.1 μg/d),IL-18(0.2 μg/d)IL-4(0.1 μg/d),IL-18(0.2 μg/d)IL-12(0.1 μg/d),连用5 d,用关节炎评分评估关节炎病情的进展,用反转录聚合酶链反虚(RT-PCR)测定CIA小鼠髌骨及邻近滑膜组织的细胞因子表达水平;用组织学染色评价膝关节的软骨和骨破坏.采用Wilcoxon rank检验. 结果 IL-18/IL-4组关节炎评分轻度降低.IL-18/IL-10联合治疗有效抑制了C1A小鼠的关节炎症反应,免疫第38天治疗组关节炎评分(0.12±0.20)显著低于对照组(0.29±0.19,P<0.05),阻止了滑膜部位的炎细胞浸润,防止了软骨的破坏.滑膜和其附近软骨的Th1型细胞因子[IL-18(0.22±0.06),IL-12(0.14±0.05)]与炎症性细胞因子[IL-6(0.22±0.11)]水平与对照组相比显著降低(P<0.05),Th2型细胞因子[IL-10(6.35±0.12),IL-4(3.57±0.13)]水平与对照组相比明显升高(P<05). IL-18R(0.40±0.15)水平明显降低(P<0.05).T-bet水平降低,转录因子(GATA)-3水平(5.7l±0.11)显著升高(P<0.05).结论 IL-18/IL-10免疫干预尚未发病的CIA小鼠,可以抑制Th1型免疫反应,使免疫反应向Th2逆转,其机制可能为下调了IL-18/IL-18R水平,诱导了转录因子GATA-3的表达.  相似文献

目的:通过与C反应蛋白(CRP)检测对比,探讨抗炎因子白细胞介素(IL)-10、促炎因子IL-6与急性冠状动脉综合征(ACS)的关系。方法:冠心病患者78例,其中ACS患者40例(ACS组),稳定型心绞痛(SAP组)患者38例,33例胸痛综合征(CPS组)患者作为对照。所有患者均行冠状动脉造影检查。采用ELISA法检测血浆IL-6、IL-10水平,采用免疫比浊法测定CRP值。结果:与SAP组及CPS组相比,ACS组血浆IL-10水平明显降低,IL-6、CRP水平明显升高,差异有统计学意义;SAP组IL-10水平低于CPS组,IL-6、CRP水平高于CPS组,差异无统计学意义;血浆IL-10水平与IL-6及CRP水平呈负相关,IL-6水平与CRP水平正相关。结论:血浆IL-10水平降低和IL-6、CRP值升高对患者ACS的发生有预测价值。抗炎因子和促炎因子水平的失衡是导致斑块不稳定、急性冠状动脉事件发生的重要因素。  相似文献

目的 观察苏木水提物对心脏异位移植大鼠血清IL-4与IL-10水平的影响,探讨苏木水提物的免疫抑制作用。方法采用大鼠腹腔同种异位心脏移植术,受体来源于SD大鼠,供体Wistar大鼠。实验分为苏木水提物大、小剂量(生药37.5、25.0g/kg)组,环孢素A(5.0mg/kg)组,苏木(25.0g/kg)+环孢素A(2.5g/kg)组,对照组(移植心脏但不给药)。术前2d给药,术后3、7d采血测定血清IL-4、IL-10水平,取大鼠心脏观察移植心脏心肌病理变化。结果与对照组比较,大剂量苏木仅在术后3d时,能减轻心脏的排斥反应(t=3.006,P<0,01)。苏木+环孢素A合用,术后3d和7d时,都能降低心脏的排斥反应(t=3.009,P<0.01;t=2.524,P<0.05),同时血清IL-4低于对照组(t=2.192,P<0.05;t=2.997,P<0.01)。苏木大剂量组血清IL-4仅是在7d时,较对照组降低(t=2,988,P<0.05)。与对照组相比,环孢素A组和苏木小剂量组在术后7d时,血清IL-10水平升高(t=2.198、t=2.173,P<0.05),苏木大剂量组仅在术后3d时IL-10升高(t=2.203,P<0.05);结论 苏木水提物可降低心脏移植后血清IL-4水平,升高移植后血清IL-10的水平,有较强的免疫抑制作用,是一种很有前途的心脏移植后免疫抑制剂。  相似文献

目的探讨白细胞介素18(IL-18)、白细胞介素10(IL-10)在老年亚健康状态发生发展中的作用及相互关系。方法应用酶联免疫吸附法测定45例亚健康状态老年人(≥60岁)血浆IL-18和血清IL-10水平,以42例亚健康状态老年前期(50~59岁)和30例健康老年人(≥60岁)作比较,分析它们之间的关系。结果亚健康状态老年人血浆IL-18水平显著高于健康老年人(P〈0.01),以及亚健康老年前期组(P〈0.05),血清IL-10水平则显著低于对照组(P〈0.01)和老年前期组(P〈0.05);血浆IL-18水平与血清IL-10水平之间呈负相关(r=-0.882,P〈0.01)。结论 IL-18和IL-10可能共同构成一局部网络,并可能参与亚健康状态的发生发展过程。  相似文献

Background: Monitoring of phenotypic characteristics of T-lymphocytes in peripheral blood is commonly performed to give the clinical parameters in the management of kidney transplant recipients.   Objective: To predict rejection in renal transplantation by immune parameters. Methods: 16 non-diabetic kidney transplant candidates (4 females and 12 males, age = 20-65 yr, first time transplant) were selected. The transplanted patients were divided into two groups based on the rejection during 3 weeks post transplant: group I (n = 9) without rejection and group II (n = 7) with a rejection episode. Immune parameters including lymphocytes subpopulations (by flowcytometry) and immunoglobulin classes (IgM, IgG, IgA and IgE by nephlometric assay) before and 45 days after transplantation were determined.   Results: The results of this investigation showed that the level of immunoglobulin IgG, IgM, IgA and IgE decreased post transplantation due to immunosuppressive drugs. CD3, CD4, CD8 T cells count, CD56 NK cells count and CD20 B cells count pre- and post-transplantation did not show any significant differences. The amount of IgE (220   vs. 462 IU/ml), CD3 (62% vs. 69.7%) and CD4 (35% vs. 41.3%) cells increased in group II during rejection episode pre-transplantation. In addition, IgA increased pretransplantation in group I those without rejection episode in comparison with group II with a rejection episode. Forty five days post transplantation IgA (209   vs. 152 mg/dl), IgG (1009 vs. 703 mg/dl) and CD20 (15%   vs. 10%) increased in group I patients. Conclusion: It is suggestive that pre-transplantation increases IgE, CD3 and CD4 are predictive of acute rejection.  相似文献

Objectives To study the monitoring of rejection after orthotopic heart thansplantation. Methods From 1998 to 2005, 10 othotopic heart thansplans were performed, and acute rejection was monitored by endomyocardial biopsy as well as by clinical features, ECG, ultrasonocardiography and blood serum determination of Tropin I, and by the combination of these methods, we analysed the monitoring of acute rejection after the heart transplantation. Results With the combination of clinical features, ECG, ultrasonocardiography and blood serum test, 5 occurences of acute rejection were judged in the postoperative course, which were comfirmed by endomyocardial biopsy to be 2 acute rejections inⅠb degree, 3 acute rejections inⅢa degree. Endomyocardial biopsy were routinely performed 21 times postoperatively in which there were 1 acute rejection inⅠa degree and 5 acute rejections inⅠb degree. Conclusions Acute rejection is an important factor influencing the postoperative course of heart transplantation, so it is imperative to have an in-time, effective and planned monitoring procedure for acute rejection. Endomyocardial biopsy is a sensitive and reliable method in diagnosis of acute rejection, but it is invasive and probable for some complications. The noninvasive method such as clinical features, ECG, ultrasonocardiography and blood serum test can be used as additive means in the diagnosis of acute rejection. Endomyocardial biopsy should be combined with some noninvasive methods in monitoring acute rejection after the heart transplantation.  相似文献

Background/Aims

The aim of this study was to investigate whether genetic variations at positions -1082, -819, and -592 in the interleukin (IL)-10 promoter affect IL-10 production in children with irritable bowel syndrome (IBS).

Methods

Ninety-four children with IBS and 102 children as healthy controls (HCs) were enrolled. Genomic DNA was extracted, and IL-10 -1082, -819, and -592 polymorphisms were detected by direct sequencing from all participants. Peripheral blood mononuclear cells (PBMCs) from 46 IBS children and 38 HCs were isolated and cultured with and without 5 ng/mL Escherichia coli lipopolysaccharide (LPS). IL-10 levels in the culture supernatants were measured by enzyme-linked immunosorbent assay.

Results

There were no significant differences in the distribution of IL-10 -1082, -819, and -592 polymorphisms or in the allele and haplotype frequencies between IBS children and HCs. PBMCs from children with IBS had significantly lower IL-10 levels after LPS stimulation than PBMCs from HCs (p=0.011); however, LPS-induced IL-10 levels in PBMCs with different genotypes of -819 and -592 polymorphisms were not significantly different between IBS patients and HCs.

Conclusions

Although significantly lower LPS-induced IL-10 production by PBMCs was noted, it is unlikely that IL-10 production was fully genetically determined in our IBS children. ClinicalTrials.gov identifier: NCT01131442.  相似文献

Continued advances in surgical techniques and immunosuppressive therapy have allowed liver transplantation to become an extremely successful treatment option for patients with end-stage liver disease. Beginning with the revolutionary discovery of cyclosporine in the 1970s, immunosuppressive regimens have evolved greatly and current statistics confirm one-year graft survival rates in excess of 80%. Immunosuppressive regimens include calcineurin inhibitors, anti-metabolites, mTOR inhibitors, steroids and antibody-based therapies. These agents target different sites in the T cell activation cascade, usually by inhibiting T cell activation or via T cell depletion. They are used as induction therapy in the immediate peri- and post-operative period, as long-term maintenance medications to preserve graft function and as salvage therapy for acute rejection in liver transplant recipients. This review will focus on existing immunosuppressive agents for liver transplantation and consider newer medications on the horizon.  相似文献
