目的 研究黄芪减轻新生儿窒息后血清诱导的HK-2细胞黏附中性粒细胞的作用及信号转导机制.方法 以HK-2细胞为研究对象,实验细胞分为对照、窒息和黄芪干预组3组,用含20%(体积比)窒息后血清作为攻击因素.采用倒置相差显微镜下观察HK-2细胞损伤及黏附中性粒细胞的情况,用生化法检测细胞悬液中髓过氧化物酶(MPO)活性;采用流式细胞术检测细胞间黏附因子-1(ICAM-1)的表达情况.结果 与对照组比较,窒息组HK-2细胞黏附较多中性粒细胞(MPO活性增加),ICAM-1表达增加,差异有统计学意义(P均<0.05);与窒息组比较,黄芪干预组HK-2细胞黏附中性粒细胞较少(MPO活性下降),ICAM-1表达减少,差异具有统计学意义(P均<0.05).结论 黄苠具有减轻窒息后血清诱导HK-2细胞黏附中性粒细胞的作用,其机制可能与抑制HK-2细胞表面ICAM-1表达有关.  相似文献

目的探讨细胞间黏附因子-1(ICAM-1)及其配体淋巴细胞功能相关抗原-1(LFA-1)对热性惊厥(FS)患儿的神经免疫调节作用。方法2004-11—2006-12将武汉市儿童医院60例FS患儿分为单纯性FS(SFS)组30例和复杂性FS(CFS)组30例;对照组30例,为年龄和性别相匹配的同期体检健康儿童。采用双抗夹心ELISA法检测血浆可溶性ICAM-1/LFA-1水平,流式细胞术检测外周血单个核细胞(PBMC)表面ICAM-1/LFA-1的表达水平。结果SFS组和CFS组患儿血浆ICAM-1水平分别为(21.54±11.09)ng/mL和(24.34±6.86)ng/mL,均明显低于对照组(29.73±12.39)ng/mL儿童(P<0.05);3组血浆LFA-1水平从高到低依次为CFS组(12.30±8.04)ng/mL、SFS组(12.09±8.83)ng/mL和对照组(9.51±8.07)ng/mL,组间比较差异无统计学意义(P>0.05)。SFS组的PBMC表面ICAM-1表达水平为(29.96±12.31)%,明显高于CFS组(22.50±8.19)%及对照组(14.21±11.31)%儿童(P<0.05),CFS组的PBMC表面ICAM-1表达水平明显高于对照组儿童(P<0.05);而LFA-1在PBMC表面的表达水平则不同,3组比较,SFS组最高为(50.89±21.36)%,CFS组最低为(34.35±11.45)%,对照组为(41.39±16.30)%,组间比较差异有统计学意义。结论ICAM-1/LFA-1作为早期应激状态下的协同刺激信号免疫因子,参与白细胞黏附及其黏附级联反应,使大脑神经元在对热应激不适应基础上呈过度兴奋状态,诱导惊厥的发生。且CFS的神经免疫病理过程要比SFS相对复杂,抑制FS的ICAM-1/LFA-1黏附活动可能为其防治开辟新的途径。  相似文献

目的 检测血管细胞间黏附分子-1(ICAM-1)在毛细支气管炎患儿血清和诱导痰上清液中的表达,探讨其在毛细支气管炎患儿发病中的作用和意义。方法 选取2015年7月至2017年1月确诊的毛细支气管炎住院患儿67例为研究对象,按诊断标准分为轻度组(n=22)、中度组(n=24)、重度组(n=21)。选取同期在我院行健康体检的20例婴幼儿为健康对照组。采用ELISA法检测各组毛细支气管炎患儿急性期及恢复期,以及健康对照组婴儿血清及诱导痰上清液中ICAM-1水平。结果 与健康对照组比较,急性期毛细支气管炎轻度组、中度组、重度组血清及痰液ICAM-1水平均显著升高(P < 0.01);与轻度组比较,急性期中度组、重度组血清及痰液ICAM-1水平均显著升高(P < 0.01);与中度组比较,急性期重度组血清及痰液ICAM-1水平显著升高(P < 0.01);与毛细支气管炎各组患儿急性期比较,恢复期轻、中、重度组毛细支气管炎患儿血清及痰液ICAM-1水平均显著下降(P < 0.01)。相关性分析结果显示,毛细支气管炎患儿急性期痰液与血清ICAM-1水平呈正相关(r=0.875,P < 0.001)。结论 ICAM-1参与了毛细支气管炎的发病过程,且和病情严重程度相关。  相似文献

目的探讨氨茶碱抗哮喘气道炎症的机制。方法采用卵蛋白(OVA)致敏的过敏性哮喘豚鼠为模型,诱发哮喘后24h利用放射免疫法测定肺组织白细胞介素2(IL-2)、IL-6含量,免疫组织化学方法检测肺组织细胞间黏附分子1(ICAM-1)蛋白表达。结果哮喘组肺组织IL-2、IL-6含量及ICAM-1阳性表达均显著高于正常对照组,氨茶碱组IL-2、IL-6及ICAM-1阳性表达较哮喘组明显降低。结论氨茶碱抗哮喘气道炎症机制可能与降低IL-2、IL-6合成或释放,降低肺组织ICAM-1阳性表达,从而抑制炎症细胞在气道浸润有关。  相似文献

目的探讨脂多糖(LPS)致大鼠脑水肿发生过程中,地塞米松作用下细胞间黏附分子-1(ICAM-1)蛋白表达水平。方法将实验大鼠随机分为9g/L盐水组(NS组)、LPS组、地塞米松组(DXM组)。各组按观察时间分为4、6、12、24及48h组。LPS诱导大鼠脑水肿发生。免疫组织化学染色法测定脑组织ICAM-1表达,并测定脑组织伊文思蓝(EB)水平。结果LPS和DXM组脑组织含水量和EB水平均较NS组明显增高(Pa〈0.05);LPS组各时间点ICAM-1蛋白表达量均高于NS和DXM组(Pa〈0.01,0.05);DXM组4、6、12、24hICAM-1蛋白表达量均较NS组增高(Pa〈0.05),48h时二组无显著差异(Pa〉0.05)。LPS组脑组织含水量和EB水平、ICAM-1蛋白表达量和脑组织含水量、ICAM-1蛋白表达量和EB水平均呈显著正相关(r=0.537,0.467,0.549Pa〈0.05)。结论ICAM-1参与LPS诱导的脑水肿的发生、发展过程。早期应用糖皮质激素可抑制炎性细胞因子的生成,减轻炎性反应与组织损伤。  相似文献

目的 建立肥胖大鼠模型,观察肥胖大鼠血清脂联素的变化及动脉血管内皮核因子(NF)-κB、血管内皮细胞黏附分子(VCAM)-1、细胞间黏附分子(ICAM)-1的表达,探讨其在血管病变发生发展中的作用机制.方法 4周龄雄性健康SD大鼠48只,随机分为对照组(18只)和高脂组(30只),给予高脂饲料制备肥胖大鼠模型.8周后,以高脂组大鼠的体质量超过对照组大鼠平均体质量的20%作为肥胖组大鼠.用酶联免疫法检测血清脂联素含量,应用免疫组化技术测定NF-κB、VCAM-1、ICAM-1在血管内皮的表达.结果 高脂组大鼠平均体质量明显高于对照组,(446.82 ± 10.05)g比(365.21 ± 13.20)g,P < 0.01;高脂组中有22只大鼠体质量超过对照组平均体质量20%,作为实验用肥胖大鼠(肥胖组).肥胖组大鼠血清脂联素水平明显低于对照组,(0.61 ± 0.13)ng/ml比(1.00 ± 0.17)ng/ml,P < 0.01,动脉血管内皮NF-κB、VCAM-1、ICAM-1阳性表达显著高于对照组,且肥胖组大鼠血清脂联素水平与血管内皮NF-κB、VCAM-1、ICAM-1表达均呈明显负相关(r = -0.938 ~ -0.891,P均 < 0.001).结论 高脂饮食引起的肥胖可使血清脂联素水平降低,使脂联素对NF-κB的抑制减弱,导致肥胖大鼠血管内皮VCAM-1、ICAM-1表达的增加,从而使动脉血管内膜增厚,血管结构发生改变导致血管疾病的发生.  相似文献

目的 观察过敏性紫癜患儿血清可溶性细胞间黏附分子-1(soluble intercellular adhesion molecul-1.sICAM-1)的含量变化,探讨其在儿童过敏性紫癜中的作用及临床意义。方法 采用酶联免疫吸附双抗体夹心法(ELISA)检测观察组63例过敏性紫癜患儿不同时期血清sICAM-1含量,其中合并紫癜性肾炎41例,未合并紫癜性肾炎22例,分别与健康儿童对照组24例比较。结果 观察组患儿早期血清sICAM-1含量均较对照组升高,恢复期下降(t=4.335,5.448,P均〈0.01);未合并紫癜性肾炎患儿早期和恢复期血清sICAM一1含量与对照组比较无显著性差异(P〉0.05);紫癜性肾炎患儿早期血清。ICAM-1含量较未合并紫癜性肾炎患儿和对照组明显增高(P均〉0.01),恢复期仍高于对照组(P〈0.05)。结论 ICAM-1可能通过血管内皮炎症性变化机制参与过敏性紫癜的病理生理过程,血清。ICAM-1含量变化与损害程度有关,急性期血清sICAM-1含量增高可作为儿童过敏性紫癜肾脏损害的早期指标。  相似文献

目的探讨脑白质损伤(CWMD)早产儿早期血清白细胞介素-8(IL-8)和细胞间黏附分子(ICAM-1)变化及其与预后的关系。方法对2009年3月至2012年6月收治并获得随访的102例CWMD早产儿(CWMD组)和42例正常早产儿(对照组),分别在生后48~72 h采集血清并检测IL-8和ICAM-1含量,12个月时采用Bayley婴幼儿发育量表对婴儿运动发育指数(PDI)及智能发育指数(MDI)进行评估,分析IL-8和ICAM-1水平与预后的相关性。结果轻、中、重CWMD组及对照组间血清IL-8和ICAM-1水平的差异有统计学意义(P0.05),且随着CWMD程度加重,IL-8和ICAM-1水平逐渐递增,差异有统计学意义(P均0.05)。轻、中、重CWMD组的PDI、MDI的差异有统计学意义(P0.05),且随着CWMD程度加重,逐渐递减,差异也有统计学意义(P均0.05)。CWMD组的血清IL-8和ICAM-1水平与MDI之间呈显著负相关(r=-0.64、-0.66,P均0.05),与PDI之间也呈显著负相关(r=-0.70、-0.71,P均0.05)。结论 CWMD早产儿出生后血清IL-8和ICAM-1水平显著增高,并且与其12月龄时的MDI及PDI呈显著负相关。  相似文献

Intercellular adhesion molecule 1 (ICAM-1), a ligand of leukocyte function associated antigen 1 (LFA-1), is present on many cells, including monocyte/macrophages. ICAM-1 is considered to play an important role in the induction and maintenance of inflammatory responses by permitting leukocyte adhesion. Its expression is inducible on endothelial and epithelial cells exposed to various inflammatory cytokines (preceding expression of HLA-DR) and is maturation dependent in certain cell lines. The distribution of ICAM-1 in decidua and placenta was evaluated using peroxidase-antiperoxidase immunohistochemistry. In decidua of first and third trimesters, scattered ICAM-1-staining cells were observed. In placentas of first and third trimesters, all types of trophoblasts stained negatively for ICAM-1. Prior to 10 weeks of gestation, the villous stroma was uniformly ICAM-1 and HLA-DR unreactive. Beginning in the chorionic plate at approximately 10 weeks, scattered ICAM-1-positive stromal cells were observed, whereas stromal cells of the terminal villi revealed no ICAM-1. By 14-16 weeks, approximately 40-50% of the terminal villous stromal cells were ICAM-1 staining. This parallels the 40-50% of the villous stromal cells that share other immunohistochemical markers, such as EB-11, with monocyte/macrophages. The lack of functional maturation of the villous stromal macrophage may explain the rarity of chronic villitis early in gestation.  相似文献

目的 缺血缺氧性肾损伤的发生过程有炎症反应参与 ,而这种炎症反应的发生与细胞粘附分子有关 ,其中细胞间粘附分子 1(ICAM 1)可上游调节并介导白细胞与内皮细胞的粘附而致肾损伤。该文旨在探讨宫内急性缺血缺氧及再灌注时ICAM 1在胎鼠肾脏炎症反应发生中的作用。方法 通过钳夹孕 2 1日龄大鼠供应子宫的血管制备胎鼠宫内不同程度缺血缺氧模型和再灌注不同时间模型 ;利用免疫组织化学技术动态观察ICAM 1的变化 ,同时应用HE染色观察病理学改变。结果 ICAM 1在假手术组胎鼠肾组织即有少量表达 ,表达部位主要在近曲小管。比较肾皮质近曲小管ICAM 1的表达情况显示 :宫内缺血缺氧后 ,在 35min和 4 5min表达增强 ,差异有显著性 (P <0 .0 5 )。与假手术组相比 ,宫内缺血缺氧 15min后再灌注 2h ,ICAM 1表达即明显增强 (P <0 .0 1) ,15h达高峰 ,再灌注 30h时表达仍强于假手术组 (P <0 .0 5 )。病理学改变 :缺血 15min再灌注 15h病理改变最明显 ,肾组织普遍充血和渗出 ,可见中性粒细胞浸润 ,肾小管普遍空泡变性 ,伴细胞核模糊 ,细胞崩解 ,基底膜阶段性断裂 ,尤以近曲小管明显。结论 宫内急性缺血和再灌注后胎肾存在炎症反应 ;宫内急性缺血缺氧可以导致ICAM 1表达增强 ,其表达变化与病理学改变一致 ,提示ICAM 1  相似文献

Surgical stress induces systemic endocrine-metabolic responses that influence the function of endothelial cells (EC) to cause various systemic reactions. Intercellular adhesion molecule (ICAM)-1 is an adhesion molecule that plays an important role in inflammation, and increased expression of ICAM-1 on EC is a reflection of EC activation. In this study, we investigated the ICAM-1 response to surgical stress in neonates undergoing major surgery. Fifteen neonates (mean age at surgery: 3.5±1.2 days) were divided into two groups according to indications for surgery: Group I: Congenital diaphragmatic hernia without persistent pulmonary hypertension (n=5); Group II: Gastrointestinal surgery [n=10: duodenal atresia (n=3), intestinal atresia (n=6), and esophageal atresia (n=1)]. Serum samples were obtained preoperatively, immediately after completion of surgery (time zero), and 24, 48, 72, 96, and 120 h after surgery to measure ICAM-1 levels using an enzyme-linked immunosorbent assay, C-reactive protein (CRP), and white blood cell count (WBC). Postoperative recovery was uneventful in all cases. ICAM-1 levels in both groups increased significantly within 24 h of surgery (Group I: P=0.0038, Group II: P=0.0320). In Group I, ICAM-1 peaked 72 h postoperatively while in Group II it continued to rise until 96 h postoperatively. The difference between peak levels reached was not significant. CRP was first detected 24 h postoperatively in both groups and continued to increase until 48 h postoperatively. Again, the difference between peak levels reached was not significant. No significant changes in WBC were observed in either group. We found that ICAM-1 increases in response to surgical stress in neonates, although there was no significant difference in levels. However, surgical stress as represented by serum ICAM-1 would appear to last longer with intestinal surgery than with non-intestinal surgery. Further research is required to establish the usefulness of ICAM-1 as an easily detectable substance associated with endothelial damage that reflects the host’s response to major surgical stress.  相似文献

目的研究新生儿窒息后血清诱导的人肾小管细胞粘附中性粒细胞的作用及机制。方法以HK 2细胞株为研究对象, 用含20%的窒息后血清作为诱导因素。实验分空白对照组、窒息组(窒息后1,3,7 d组)和吡咯烷二硫氨基甲酸(PDTC)干预组。分别检测各组细胞悬液中髓过氧化物酶(MPO)活性;检测HK 2细胞膜ICAM 1和细胞核内NF κB的表达。结果窒息后1 d血清组细胞悬液MPO值显著高于各PDTC干预组、对照组及窒息后3,7 d血清组(P<0.01)。窒息后1 d血清组HK 2细胞表面ICAM 1表达和HK 2细胞核NF κB表达亦显著高于各PDTC干预组、对照组及窒息后3,7 d血清组(P<0.01)。结论 窒息后血清可以诱导肾小管上皮细胞活化进而粘附中性粒细胞,其作用机制可能涉及到窒息后血清活化了NF-κB,从而使肾小管上皮细胞膜表面ICAM-1表达增加,进而粘附中性粒细胞。  相似文献

Intercellular adhesion molecule-1 (ICAM-1) is a glycoprotein that is necessary for the transendothelial migration of leucocytes. This study was undertaken to elucidate the role of ICAM-1 in the pathophysiology of Hirschsprung's disease (HD)-related enterocolitis. Ganglionic and aganglionic portions of bowel from 18 patients with HD who did not have clinical or histological evidence of enterocolitis and 5 patients with HD who developed enterocolitis before or after a pull-through operation were stained using monoclonal antibody against ICAM-1. The bowel specimens obtained from 2 children with imperforate anus at the time of colostomy closure and 3 children at the time of bladder augmentation were similarly stained to act as controls. The ganglionic portion of bowel from patients with HD without enterocolitis and controls showed either no ICAM-I staining or occasional staining of the endothelial lining of submucosal vessels with no staining of the glandular crypt epithelium. In contrast, both ganglionic and aganglionic bowel from patients with enterocolitis complicating HD demonstrated strong ICAM-1 staining in the endothelium of submucosal vessels. Strong expression of ICAM-1 in the glandular crypt epithelium was seen in only 2 patients who had developed enterocolitis before pull-through operations. This study illustrates the importance of endothelial cell activation in the pathogenesis of HD-related enterocolitis.  相似文献

目的:探讨毛细支气管炎(简称毛支炎)患儿细胞间黏附分子-1(ICAM-1)和激活调节正常T细胞表达和分泌因子(RANTES)检测的意义及相关性。方法:采用流式细胞术检测28例毛支炎患儿、23例支气管肺炎患儿及24例健康婴儿外周血淋巴细胞ICAM-1表达情况,ELISA法测定其血清RANTES水平,并进行相关性分析。结果:毛支炎组ICAM-1表达(35.0±10.3%)高于肺炎组(29.9±8.6%; P<0.05)及对照组(24.6±6.9%; P<0.01),肺炎组ICAM-1表达高于对照组(P<0.05)。毛支炎组和肺炎组RANTES水平分别为32.1±6.0和 30.6±6.2 ng/mL,高于对照组的27.1±5.1 ng/mL(分别P<0.01,<0.05),毛支炎组与肺炎组差异无统计学意义。ICAM-1与RANTES在毛支炎组呈正相关(r=0.675,P<0.01)。结论:ICAM-1和 RANTES参与了毛支炎的发病过程且具有协同作用。[中国当代儿科杂志,2010,12(3):181-183]  相似文献

Atherosclerosis begins in childhood and progresses from fatty streaks to raised lesions in adolescence and young adulthood. This process is accelerated in children with risk factors for coronary artery disease (CAD). Cell adhesion molecules (CAMs) are supposed to play important roles in the initial development of atherosclerosis, which may suggest that the expression of CAMs is increased in children more than in older subjects or in CAD patients. To determine whether risk factors for CAD are associated with an increased expression of CAMs, we investigated the relationships of the serum levels of soluble vascular cell adhesion molecule-1 (VCAM-1), soluble intercellular adhesion molecule-1 (ICAM-1) and soluble P-selectin (P-selectin) with lipid and apolipoprotein parameters in children (40 boys and 45 girls). We also examined the relationships between soluble CAMs and the fractional esterification rate of cholesterol in HDL (FERHDL), particle size of LDL and lipoprotein containing apoA-I, but no apoA-II (LpA-I). In children, soluble VCAM-1 levels were correlated with the levels of triglyceride (in boys) and apoB, the ratio of apoB to apoA-I and FERHDL (in girls). Similar associations were found for soluble ICAM-1. Furthermore, the soluble ICAM-1 level was inversely correlated with LpA-I level, LDL size (in boys) and HDL cholesterol level (in girls). Soluble P-selectin levels were not correlated with these parameters. Conclusion Our data indicate that intervention to normalize risk factors for coronary artery disease should be started at a young age to prevent increased expression of cell adhesion molecules.
