目的 探究白细胞介素6(IL-6)、白细胞介素18(IL-18)在肥胖慢性牙周炎患者血清中的表达水平变化及意义。方法 选取2017年9月~2018年5月在佳木斯大学附属口腔医院牙周科门诊确诊为慢性牙周炎患者15例设为NP组,另选取同期慢性牙周炎伴肥胖患者15例设为FP组。收集两组患者外周血样本并记录牙周PD、CAL,采用双抗体夹心酶联免疫吸附法(ELISA法)测定样本中的IL-6、IL-18的表达水平,分析两组间指标的差异及相关性。结果 FP组血清中IL-6、IL-18分别为(9.16±2.35)pg/ml、(325.22±98.67)pg/ml,其表达水平分别高于NP组的(6.15±2.06)pg/ml和(241.52±78.82)pg/ml,差异有统计学意义(P<0.05);FP组中PD与血清中IL-6、IL-18呈高度正相关(r=0.894,P<0.01;r=0.819,P<0.01);CAL与血清中IL-6、IL-18呈正相关(r=0.885,P<0.01,r=0.828,P<0.01);NP组中PD与血清中IL-6、IL-18也有正相关关系(r=0.842,P<0.01,r=0.728,P<0.01);CAL与血清中IL-6、IL-18呈正相关(r=0.884,P<0.01,r=0.707,P<0.01)。结论 肥胖状态所导致的外周血中IL-6、IL-18水平的升高与CP的发生发展存在着密切联系。  相似文献

Periodontal disease (PD) and peri-implantitis (PI) are characterized by an immune response leading to destructive inflammation. The prominent impact of genetic factors on periodontitis has been previously evaluated and IL-17 has found to play a critical role in this process. This cytokine has a controversial behavior. This study aimed at finding out whether the polymorphism of this cytokine plays a significant role in chronic periodontitis (CP) and PI or it is just a pro-inflammatory regulatory cytokine. Fresh human blood samples were obtained and three main genotypes were traced carefully. The samples were transferred into 96-well plates and sent to KBioscience Institute in the United Kingdom for genotyping the polymorphism using Competitive Allele Specific PCR (KASP) technique. SPSS version19 software and chi-square and Kruskal–Wallis tests were used for statistical differences considering p-value less than 0.05. A significant difference was detected between the three groups in terms of specific SNP studied in this experiment (P = 0.00). The CC genotype of IL17 polymorphism (rs10484879) may contribute to the pathogenesis of peri-implantitis and periodontitis. The association of IL-17 polymorphism with PI and CP is a promising finding that may help in future similar studies on other ethnicities and larger study populations.  相似文献

目的 探讨IL 1β在成人牙周炎性牙龈组织的分布及与炎症细胞浸润的关系。 方法 免疫组织化学SP方法。结果  19例牙周炎组炎性牙龈标本中均有IL 1β蛋白阳性表达 ,阳性细胞数与正常对照组牙龈相比有非常显著差异 (P <0 0 1)。IL 1β阳性细胞主要为巨噬细胞。 结论 IL 1β可能参与了成人牙周炎的发生和发展。巨噬细胞是成人牙周炎炎性牙龈组织中表达细胞因子IL 1β的主要细胞类型 ,与成人牙周炎的发生和发展有关  相似文献

为探索IL-6(Interlmkin-6)是否在IDDM(insulin-depenednt diabetes mellitus.胰岛素依赖的糖尿病)的发病中起作用,本文报告了我们采用反转录多聚酶链反应(RT-PCR)对IDDM 病人,NIDDM(Non-insulim dependent diabetes Mellitus,非胰岛素依赖的糖尿病)病人,正常儿童(Normal chidren,NC)正常成人(Normal adults,NA)四组受试者的外周血单个核细胞(peripheral blood mononuclear cells,PBMNCs)中的IL-6 mRNA 的表达水平初步研究。以内源性β_2微球蛋白(β_2-microglobulim,β_2m)的mRNA 作为内对照,与靶序列IL-6 mRNA 在同一管内同时反转录及扩增,以特异的PCR 产物产量之比IL-6/β_2m 表示IL-6 mRNA 表达的相对水平,结果显示:在12例IDDM 病人,29例NIDDM 病人,23例正常儿童及12例正常成人的PBMNC_a中,IL-6 mRNA 的相对水平分别为0.91±0.19;0.10±0.06;0.43±0.08,0.01±0.07。这个结果提示IL-6很可能在IDDM 的发病过程中扮演重要角色。  相似文献

Salivary, serum matrix metalloproteinase-8 (MMP-8), tissue inhibitor of matrix metalloproteinases-1 (TIMP-1), neutrophil elastase (NE), and myeloperoxidase (MPO) levels were investigated in generalized chronic periodontitis (GCP), generalized aggressive periodontitis (GAgP), and healthy groups. Whole-mouth clinical periodontal measurements were recorded. Salivary, serum concentrations of MMP-8, MPO, TIMP-1, and NE were determined by immunofluorometric assay or ELISA in 18 patients with GCP, 23 patients with GAgP, 18 individuals with healthy periodontium. We studied morphological characteristics of the regenerative process in gingival tissues during therapy of chronic periodontitis. Energostim stimulated macrophageal reaction and promoted neoangiogenesis in the inflammatory infiltrate. It was not observed after traditional drug therapy. Energostim promoted vascularization in regenerating tissues, normalized the structure at a greater area of the lamina propria of the gingiva, and prevented fibrous and sclerotic changes. The volume of histiolymphocytic infiltrates in regenerating gingival tissues decreased after application of intradental splints.  相似文献

Periostin, an extracellular matrix protein functioning as an important structural mediator and adhesion molecule, has been shown to be an important regulator of connective tissue integrity. This study aimed to evaluate the levels of periostin in chronic periodontitis (CP) and aggressive periodontitis (AgP) compared to non-periodontitis (NP). Individuals were submitted to gingival crevicular fluid (GCF) and saliva sampling. Periodontal examination consisted of plaque index (PI), gingival index (GI), probing depth (PD), bleeding on probing (BOP), and clinical attachment level (CAL) measurements. Assays for periostin were performed by an enzyme-linked immunosorbent assay. Periodontitis patients presented more severe clinical indices compared to the NP group (p?<?0.001). The mean GCF level of periostin was lowest in the AgP group as compared to the other groups and was lower in the CP group as compared to the NP group (p?<?0.001). Increased levels of periostin were observed in the saliva of patients with AgP as compared to the CP and NP groups (p?<?0.05). There was a negative relationship between GCF periostin levels and clinical parameters (p?<?0.01), whereas a positive correlation was observed between salivary periostin levels and full-mouth GI and CAL scores (p?<?0.01). To our knowledge, this is the first report investigating periostin levels in GCF and saliva in aggressive periodontitis. The results suggest that subjects with CP and AgP exhibit a different periostin profile. Periostin in GCF may have a protective role against periodontal disease. Furthermore, salivary periostin concentrations may have a promising diagnostic potential for the aggressive forms of periodontal disease.  相似文献

为利用实时监测定量PCR技术检测细胞因子mRNA的表达 ,从mRNA水平探讨IFN γ、IL 4mRNA的变化。对初诊SLE患者外周血IFN γ、IL 4的mRNA表达进行检测 ,并同时使用LPS及PHA作刺激剂研究其对SLE患者细胞因子mRNA表达的影响。结果是PHA刺激后 ,正常组IFN γmRNA的表达明显高于SLE组 (P =0 0 2 4 ) ;LPS对正常对照的IFN γ和IL 4的mRNA表达均能明显上调 ,但SLE患者仅表现为IFN γ升高 ,IL 4升高不明显 ;未刺激组SLE患者与正常对照的比较结果显示IFN γ的表达及IFN γ/IL 4有明显降低 (P =0 0 14 ,P =0 0 33)。故SLE患者与正常对照相比 ,IFN γmRNA的表达下降 ,同时SLE患者的IFN γ、IL 4mRNA表达对LPS及PHA的反应下降。由于不同的患者受累系统不同 ,临床表现不一 ,可能T细胞的异常并不相同 ,其免疫系统紊乱的机制也不一致  相似文献

目的:探讨慢性牙周炎(chronic periodontitis,CP)患者龈沟液(gingival crevicular fluid,GCF)中骨黏连蛋白(osteoneetin,ON)、白细胞介素-18(interleukin-18,IL-18)和IL-23测定意义及临床价值.方法:选择30例CP患者分别设置为治疗前、规范牙周基础治疗4周及治疗8周三组进行围治疗期分组测定分析.ON、IL-18和IL-23测定均采用酶联免疫吸附试验(ELISA).结果:CP患者治疗前患牙临床检查记录数据显示,牙周袋深度(PD)、附着丧失(AL)和龈沟出血指数(SBI)都显著高于对照组(P均<0.01),治疗4周三项临床指标明显下降,但仍显著高于对照组(P均<0.05),治疗8周后前二项临床指标仍显著高于对照组(P均<0.05),仅SBI已恢复至正常人水平(P>0.05).GCF中炎性指标结果显示,治疗前三项炎性因子水平均显著高于对照组(P均<0.01),治疗4周三项临床指标明显下降,IL-18和IL-23已恢复至正常人水平(P均>0.05),但ON仍显著高于对照组(P<0.05);治疗8周三项临床指标均恢复至正常人水平(P均>0.05).炎性指标与临床指标的相关性分析结果显示,ON与IL-18、IL-23呈微弱相关(r=0.546、0.502,P<0.06、P<0.05),而与PD、AL和SBI呈显著正相关,尤以PD、AL为甚(r=0.602、0.560、0.813,P<0.05、P<0.05、P<0.05).IL-18与PD、AL、SBI呈显著正相关,且相关性甚以ON与之(r=0.602、0.560、0.813,P<0.05、P<0.05、P<0.05);IL-23与PD、AL、SBI也呈显著正相关(r=0.703、0.631、0.828,P<0.05、P<0.05、P<0.05).结论:患者GCF中炎性指标参与了CP的病理进展过程,其测定对于了解本病的病理进展机制和炎症反应的评估提供一定的帮助.  相似文献

目的:探讨牙周病患者血清hs-CRP、IL-8和IL-10水平的变化及临床意义。方法:应用放射免疫分析、酶联法和免疫比浊法对32例牙周病患者进行了血清hs-CRP、IL-8和IL-10检测,并与35名正常人作比较。结果:牙周病患者在治疗前血清hs-CRP、IL-8和IL-10水平均非常显著地高于正常人组(P〈0.01),经治疗1个月后与正常人组比较仍有显著性差异(P〈0.05)。结论:检测血清hs-CRP、IL-8和IL-10水平的变化,对探讨牙周病发病机制、预防和指导治疗均有一定的临床价值。  相似文献

目的:观察慢性牙周炎患者血浆β-防御素-2浓度的变化,揭示其与慢性牙周炎发病的相关性。方法:收集慢性牙周炎患者和同期牙周健康者各80例。ELISA检测血浆β-防御素-2浓度。统计分析其与慢性牙周炎发病的相关性。结果:经t检验,慢性牙周炎患者血浆β-防御素-2浓度(405.2±133.9)pg/ml显著低于牙周健康者(641.6±128.3)pg/ml(t=6.492,P<0.01)。经Spearman相关分析,慢性牙周炎患者血浆β-防御素-2浓度与牙周探诊深度(r=-0.482,P<0.01)、附着丧失(r=-0.498,P<0.01)和出血指数(r=-0.501,P<0.01)均呈显著负相关。Logistic回归分析显示,血浆β-防御素-2浓度(OR=0.415,95%CI=0.297~0.862,P<0.01)是罹患慢性牙周炎的独立危险因素。ROC曲线分析显示,血浆β-防御素-2浓度预测罹患慢性牙周炎有显著预测价值(曲线下面积=0.843,95%CI=0.774~0.894,P<0.01),且判定血浆β-防御素-2浓度<621.5 pg/ml,对预测罹患慢性牙周炎有81.3%的灵敏度和75.0%的特异度。结论:慢性牙周炎患者血浆β-防御素-2水平降低,血浆β-防御素-2水平降低可能是慢性牙周炎的危险因素。  相似文献

目的:探讨IL-6 mRNA与子宫内膜异位症发病的关系。方法:利用大鼠子宫内膜异位症(内异症)动物模型,采用逆转录聚合酶链反应(RT-PCR)技术,检测子宫内膜白细胞介素-6(IL-6)mRNA表达情况。结果:正常对照组,各时点间(2、4、6和8周)IL-6 mRNA表达无显著差异(P>0.05)。内异症模型组,在成模后(2、4、6和8周)其在位、异位子宫内膜IL-6 mRNA的表达呈增高趋势(P<0.01)。在各时点,内异症异位子宫内膜>内异症在位子宫内膜>正常子宫内膜IL-6 mRNA的表达(P<0.01)。结论:IL-6 mRNA可能与内异症的进展有关。IL-6 mRNA可能促进异位内膜的种植和生长。  相似文献

目的:观察IL-35 在慢性鼻-鼻窦炎伴鼻息肉的表达与意义,完善慢性鼻-鼻窦炎伴鼻息肉的发病机制。方法:随机选取2016 年5 月~2017 年4 月在我科住院的慢性鼻-鼻窦炎伴鼻息肉患者临床资料及术中鼻息肉组织。主观症状采用视觉类比法(VAS)进行评分;纤维鼻咽喉镜检查结果采用Lanza-Kennedy 评分;鼻窦CT 采用Lund-Mackay 评分,鼻息肉组织采用Real-time PCR 方法检测IL-35 mRNA(EBI3 mRNA 和IL-12A mRNA)的表达水平和ELISA 方法检测息肉组织中IL-35 及IL-17 和IL-10 蛋白的表达水平。结果:共收集51 例慢性鼻-鼻窦炎伴鼻息肉患者临床资料及术中鼻息肉组织,其中男性30 例,女性21 例,平均年龄(43.3±15.1)岁,症状评分(5.5±1.7),鼻内镜评分(6.4±2.0),CT 评分(11.7±3.8)。较正常鼻黏膜组织相比,鼻息肉组织中IL-35 mRNA 和蛋白均呈低表达水平,差异有统计学意义(P<0.05),而IL-17 和IL-10 蛋白呈高表达水平,差异均有统计学意义(P<0.05)。结论:IL-35 可能在慢性鼻-鼻窦炎伴鼻息肉的病理机制中起着关键的作用。  相似文献

Objective: Chronic periodontitis is a bone-destructive disease affecting periodontal support structures. Although leptin has a protective effect against periodontitis, the underlying mechanism remains unclear. Therefore, this study aimed to investigate the possible role of leptin by examining its relationship with OPG and RANKL in human gingival tissues obtained from patients with chronic periodontitis.Method: Twenty-two patients with chronic periodontitis were enrolled (10 with moderate periodontitis and 12 with severe periodontitis) in the experimental group, and 12 healthy individuals were enrolled in the control group. Gingival tissue samples were collected, and the protein levels and localization of leptin, OPG, and RANKL were studied using immunohistochemistry (IHC). The staining intensities of leptin, OPG, and RANKL were correlated with the periodontal clinical index. Moreover, real-time quantitative PCR (RT-qPCR) was used to determine OPG and RANKL mRNA levels in gingival fibroblasts stimulated with gradient concentrations of leptin protein in vitro.Result: Leptin, OPG, and RANKL were located in the cytoplasm of gingival epithelial cells and the connective tissue. Leptin was widely and significantly expressed in the control group, whereas it was lightly stained in the severe group. RANKL was lightly stained in the control group, whereas it was widely and significantly expressed in the severe group. The control and the moderate groups had similar OPG levels, which were significantly higher than that in the severe group. Leptin was positively correlated with OPG(r = 0.905, p < 0.01) and negatively correlated with RANKL (r = -0.635, p < 0.01). In vitro low concentrations of leptin led to an increased OPG/RANKL mRNA ratio, whereas the opposite effect was observed at high concentrations.Conclusion: Leptin can regulate OPG and RANKL expression in gingival fibroblasts and may thus play a role in the development of chronic periodontitis by modulating the OPG/RANKL ratio.  相似文献
