Objective To investigate the mutation of type Ⅱ human hair basic keratin (hHb) gene in a family with monilethrix.Methods Scanning electron microscopy was used to observe the structure of hair shafts.With informed consent,blood samples were drawn from affected and unaffected membets in this family,as well as from 50 healthy controls.Genomic DNA was isolated from these samples.The exon 1 and exon 7 of hHb1,hHb3 and hHb6 were amplified by polymerase chain reaction (PCR).All the PCR products were sequenced directly using ABI3730 automated sequencer.DNA sequence alignment was carried out with BLAST software.Results A typical beaded appearance was observed in affected hairs by using scanning electron microscopy.There were obvious longitudinal ridges and sulcuses in hair node.and hair cuticles were irregularly shaped.Most cortex and medullary substance were absent in affected hairs of a patient.After sequence alignment,a G1289A point mutation in exon 7 of hHb6 gene,which led to a substitution of arginine for glutamide at codon 430,was detected in affected members of this family,but not in unaffected family members or 50 unrelated human controls.No mutation was observed in exon 1 or exon 7 of hHb1 and hHb3 gene or exon 1 of hHb6 gene.Conclusion The missense mutation of R430Q is a novel mutation.which may be associated with the pathogenesis of monilethrix in this pedigree.  相似文献

目的 检测一个中国汉族人X-性连锁鱼鳞病家系的类固醇硫酸酯酶(STS)基因突变情况.方法 收集1个X-性连锁鱼鳞病家系的临床资料,提取外周血DNA,通过PCR扩增外周血基因组DNA类固醇硫酸酯酶基因的第1和第10外显子,以表型正常家系成员及50例健康人为正常对照.结果 家系内全部患者均存在STS基因的完全缺失,即10个外显子均缺失,家系中正常人及对照者未发现上述缺失.结论 STS基因的完全缺失可能为导致该家系临床表型的主要原因.
Abstract:
Objective To detect the steroid sulfatase (STS) gene mutation in a Chinese pedigree with X-linked ichthyosis (XLI). Methods Genomic DNA was extracted from the peripheral blood of 3 affected patients and unaffected members in this family and 50 unrelated healthy volunteers followed by the amplification of the exon 1 and exon 10 of STS gene by PCR. Results Complete deletion of the exon 1 to 10 of STS gene was detected in all the patients in this pedigree with XLI, while no mutation was found in this gene in unaffected members of this family or normal human controls. Conclusion The complete deletion of STS gene is likely to be the main cause of the phenotype of XLI in this family.  相似文献

目的 探讨一个中国汉族人表皮松解性掌跖角化病(EPPK)家系的角蛋白基因KRT1、KRT9、KRT10突变情况.方法 收集1个EPPK家系的临床资料,提取外周血DNA,通过PCR扩增角蛋白KRT1、KRT9、KRT10基因编码区的全部外显子及其侧翼序列并测序,以表型正常家系成员及50例健康人为正常对照.结果 发现家系内6例患者均存在KRT1基因错义突变c.1436T>C,导致第479位的异亮氨酸被苏氨酸取代(I479T),在家系中6例正常人及50例对照者未发现上述突变.结论 错义突变KRTI的c.1436T>C可能为导致该家系临床表型的主要原因.本例为国内首次发现的KRT1突变引起的EPPK家系.
Abstract:
Objective To analyze the mutations in keratin 1 (KRT1), KRT9 and KRT10 genes in a Chinese family with epidermolytic palmoplantar keratoderma (EPPK). Methods Clinical data were collected from a family with EPPK. Genomic DNA was extracted from the peripheral blood of 12 family members, including 6 patients and 6 unaffected members, as well as from 50 unrelated normal human controls. PCR was performed to amplify all the exons and flanking sequences of KRT1, KRT9 and KRT10 genes followed by DNA sequencing.Results A missense mutation C.1436T > C was found in the highly conserved helix termination motif of KRT1 gene of all the patients, resulting in a substitution of isoleucine by threonine at position 479 of the KRT1 protein. No mutation was found in the unaffected members or unrelated controls. Conclusions The missense mutation C.1436T > C in K.RT1 gene is likely to be the main cause of the phenotype of EPPK in this family.This is the first report of a pedigree with KRT1 gene mutation-induced EPPK in China.  相似文献

Objective: Dystrophic epidermolysis bullosa is an autosomal dominant or recessive disorder caused by mutations in the gene encoding type Ⅶ collagen (COL7A1 gene). To detect the mutation of COL7A1 in a DEB family. Methods: We performed full exon sequencing on DNA of the proband, and verified the COL7A1 mutation site of her sister by Sanger sequencing. Results: The proband and her sister had the same COL7A1 genotype, and the point mutation on exon 86 was c.6761G>A (p.Gly2254Glu). Methods Fifty patients with psoriasis vulgaris (37 cases of active psoriasis and 13 cases of stable psoriasis) and 24 healthy human controls were included in this study. PBMCs were isolated from blood samples obtained from all patients and controls. The mRNA expressions of IFN-gamma receptor and TNF-aipha receptor in PBMCs were detected by RT-PCR. The disease severity in patients was evaluated by psoriasis area and severity index (PASI). Results The mRNA expressions of IFN-gamma receptor and TNF-alpha receptor were observed in the PBMCs of all subjects. The mRNA expression levels of IFN-gamma receptor were 0.72 ± 0.17 in healthy controls, 1.11 ± 0.55 in all patients with psoriasis, 1.13 ±0.57 in patients with active psoriasis and 1.03 ± 0.52 in patients with stable psoriasis, respectively. A signifi-cant increase was observed in the expression levels of IFN-gamma receptor mRNA in all psoriatic patients and in patients with active psoriasis compared with those in healthy controls (both P < 0.05), but there was no significant difference between the healthy controls and patients with stable psoriasis (P > 0.05). The expres-sion levels of TNF-alpha receptor mRNA were 2.05 ± 1.34 in healthy controls, 2.70 ± 3.80 in all psoriatic patients, 2.90 ± 4.40 in patients with active psoriasis, 2.14 ± 1.05 in patients with stable psoriasis, respectively;there was no significant difference between psoriatic patients and healthy controls (P > 0.05). However, no correlation was found between the mRNA expression of IFN-gamma receptor, that of TNF-alpha receptor,and disease severity in psoriatic patients. Objective To assess the relationship of IL-4 -590C/T polymorphism and TLR2 Arg753Gln polymorphism with genetic susceptibility to syphilis. Methods The study included 56 patients with early symptomatic syphilis, 38 patients with early latent syphilis and 102 normal human controls. Real-time quantitative fluorescent PCR was used to detect the IL-4 -590C/T polymorphism, and DNA sequencing to analyze the TLR2 Arg753Gln polymorphism, in these subjects. Results The frequency of C and T alleles at 590 position of IL-4 gene was respectively 0.229 and 0.771 in patients with early syphilis, 0.216 and 0.784 in the normal controls, 0.232 and 0.768 in patients with early symptomatic syphilis, 0.224 and 0.776 in patients with early latent syphilis. GG genotype was observed at position 753 in TLR2 gene in all the subjects. No significant difference was observed for the frequency of C or T alleles at position 590 in IL-4 gene or the frequency of A or G alleles at position 753 in TLR2 gene between patients with early syphilis and normal controls or between patients with early latent syphilis and those with early symptomatic syphilis (all P > 0.05).Conclusion There seems no evident association between the susceptibility to syphilis and IL-4 -590C/T polymorphism or TLR2 Arg753Gln polymorphism.  相似文献

目的 分析寻常性银屑病患者外周血中T细胞受体β可变区(TRBV)的优势表达状况,探讨其在银屑病发病中的作用.方法 以人功能性TRBV家族设计33个上游引物,共同的T细胞受体β恒定区(TRBC)基因设计下游引物,在T细胞受体α恒定区(TRAC)设计引物作为内参,分析寻常性银屑病患者、正常人对照外周血样本各10例,检测各PCR反应管荧光强度,以大于正常人外周血T细胞相应TRBV基因相对表达量(-x+3s)筛选TRBV优势表达基因家族.结果 TRAC扩增产物阈值循环数(Ct)值集中于21~24,银屑病组TRBV2扩增产物Ct与TRAC产物Ct差值平均数银屑病患者为2.98,TRBV5-7为3.24,TRBV6-6/6-9为2.52,TRBV12为2.04,TRBV24为3.56,TRBV29为4.12,其表达与正常人比较均明显增高(P值均<0.05),其中TRBV6-6/6-9、TRBV12、TRBV29在银屑病患者呈优势表达.结论 银屑病患者外周血TRBV基因家族存在优势表达,可能在银屑病T细胞异常免疫反应中起重要作用.
Abstract:
Objective To assess the preferential expressions of peripheral blood T cell receptor beta chain variable region (TRBV) subfamilies in patients with psoriasis vulgaris(PV), and to estimate their role in the pathogenesis of psoriasis. Methods Thirty-three upstream primers were designed to target the human functional TRBV genes, downstream primers to target the common T cell receptor beta constant (TRBC) gene,with T cell receptor alpha constant (TRAC) gene as the internal reference. Total RNA was extracted from the peripheral blood T cells of 10 health human controls and 10 patients with PV, and transcribed into cDNA.Then, TRBV genes were amplified by real-time fluorescence quantitative PCR (RFQ-PCR) and the fluorescence intensity of each samples was detected. The expression levels of TRBV genes in the control group were used to calculate the cut-off values (mean expression levels of TRBV subfamilies in the 10 normal controls + 3 standard deviations). When the expression level of a TRBV subfamily from patients with PV was equal to or higher than the cut-off value, it was considered as the preferentially expressed TRBV subfamily. Results The threshold cycle (Ct) value varied from 21 to 24 for TRAC gene. The difference in the Ct value between TRBV subfamily genes and TRAC gene in patients with PV was 2.98 for TRBV2 gene, 3.24 for TRBV5-7 gene, 2.52 for TRBV6-6/6-9 gene, 2.04 for TRBV 12 gene, 3.56 for TRBV 24 gene, and 4.12 for TRBV 29 gene, and the expression levels of these subfamily genes were significantly higher than those in the normal controls (all P < 0.05). According to the above standard, TRBV6-6/6-9, TRBV12 and TRBV29 were considered to be preferentially expressed subfamilies. Conclusions There is a preferential expression of TRBV gene subfamilies in peripheral blood of patients with psoriasis vulgaris, which may play a vital role in the abnormal T cell-mediated immune responses in psoriasis.  相似文献

Renal tubular acidosis(RTA) encompasses many renal tubular disorders characterized by hyperchloremic metabolic acidosis with a normal anion gap. Untreated patients usually complain of growth failure, osteoporosis, rickets, nephrolithiasis and eventually renal insufficiency. Fanconi-Bickel syndrome(FBS) is an example of proximal RTA due to a single gene disorder; it is caused by defects in the facilitative glucose transporter 2 gene that codes for the glucose transporter protein 2 expressed in hepatocytes, pancreatic β-cells, enterocytes and renal tubular cells. It is a rare inherited disorder of carbohydrate metabolism manifested by huge hepatomegaly [hence it is classified as glycogen storage disease(GSD) type XI; GSD XI], severe hypophosphatemic rickets and failure to thrive due to proximal renal tubular dysfunction leading to glucosuria, phosphaturia, generalized aminoaciduria, bicarbonate wasting and hypophosphatemia. The disorder has been reported from all parts of Europe, Turkey, Israel, Arabian countries, Japan and North America. Many mutant alleles have been described, its exact frequency is unknown and there is no single mutation found more frequently than the others. The presence of consanguinity in affected families suggests an autosomal recessive pattern of inheritance. New cases of FBS have been recently reported in the Middle and Far East in collaboration with specializedcenters. Two novel mutations have been discovered in two unrelated Egyptian families. The first was two bases deletion, guanine and adenine,(c.253_254del GA) causing a frameshift mutation(p. Glu85fs) and the second is mutation in exon6 in splicing acceptor site with intron5(c.776-1GC or IVS5-1GA). Moreover, a new different mutation was described in a 3 year old Indian boy.  相似文献

Objective To assess the DNA methylation status in peripheral blood mononulclear cells (PBMCs)from patients with systemic lupus erythematosus (SLE) using a constructed methyl-CpG-binding domain (MBD) proteins.Methods A recombinant plasmid MBD-pET30b+ was transformed into E.coli (DH5a)for clonal expansion followed by sequencing.Then,plasmids with correct sequence were transformed into E coli BL21 (DE3) for the expression of MBD proteins under the induction of isopropy-β-D-thiogalactoside (IPTG).The expression products were purified by Ni-NTA chromatography and identified by sodium dodecyl sulfate polyacrylamide gel electropheresis (SDS-PAGE)and Western blotting.Cultured 293T cells and isolated PMBCs from 21 patients with SLE and 15 normal human controls were immunostained with the expressed protein,and analyzed by fluorescence microscopy and flow cytometry.Results The sequence of recombinant plasmid was proved to be consistent with the expectation.SDS-PAGE and Western blotting demonstrated that the molecular weight of expressed tetrameric-protein was 46000,with the presence of N-terminal His-tag and C-terminal HA-tag.As fluorescence microscopy showed,the MBD proteins could specifically bind to intracellular CpG DNA in 293T cells.Patients with SLE had a significantly lower methylation level than the controls did(9.32±1.33 vs 11.66±1.04,t=5.68,P<0.05),and the DNA methylation level negatively correlated、with SLE disease activity index(r= -0.78,P<0.01).Condusions There is a lower DNA methylation in patients with SLE than in normal human controls,and the methylation level negatively correlates with SLE disease activity index.  相似文献

Objective To observe the clinical efficacy and safety of gahapentin in the treatment of postherpetic neuralgia. Methods A multicenter, randomized, double-blind, placebo-controlled, parallel design, 6-week study was performed. Patients with postherpetic neuralgia were recruited into this study and randomly divided into two groups to receive gabapentin or placebo 1800 mg daily in three divided doses with a forced titration schedule, respectively. The primary efficacy measure was change in the pain score based on a visual analogue scale from baseline to the final week of therapy, and secondary measure was the improvement in sleep quality scored on a 5-point severity scale. Efficacy and safety evaluation was performed at baseline, and 1, 3, and 6 weeks atter the treatment. Results One hundred and forty-one patients were recruited in four clinical centers, and 125 patients completed the trial, of whom 66 were in the treatment group and 59 in the control group. An improvement was observed in both pain scores and sleep scores on week 1, 3 and 6 in both two groups, and the improvement was greater in gabapentin-treated group than that in the control group. The response rate was 29.58% and 57.75%, respectively in gabapentin-treated group on week 1 and 3, com-pared to 13.04% and 40.58%, respectively, in the control group (t = 5.94, 4.12, respectively, both P <0.05).Gabapentin was well tolerated, and the most common adverse events were dizziness, vertigo, somnolence and transient abnormality of hepatic function. Conclusion Gabapentin could markedly reduce pain intensity and improve sleep quality with a low incidence of adverse events in patients with postherpetic neuralgia.  相似文献
