Summary Rabbit cornea cells exhibited a sensitivity to 1 g aflatoxin b1 and 5 g rubratoxin B per ml of growth medium. No changes were observed in the bacteriophages tested in the presence of 25 g aflatoxin b1 or 100 g rubratoxin B per ml of medium by the plaque-forming unit method or single-step growth curves.  相似文献

Nitrous oxide (N2O) was assayed in 676 urine samples and 101 blood samples provided after exposure by operating theatre personnel from nine hospitals. The blood and urine assays were repeated in 25 subjects 18 h after the end of exposure. For 80 subjects, environmental N2O was also measured during intraoperative exposure. Mean urinary N2O in the 676 subjects at the end of exposure was 40 g/l (range 1–3805 g/l); in 10 of the 676 subjects, urinary N2O was in the range 279–3805 g/l (mean 1202 /l). The 98th percentile was 120 g/l. Mean blood N2O at the end of exposure, measured in 101 subjects, was 21 g/l (median 16 g/l, range 1–75 g/l). Blood and urine N2O (1.5 g/l and 4.9 g/l, respectively) in 25 subjects, 18 h after exposure, was significantly higher than in occupationally non-exposed subjects (blood 0.91 g/l, urine 1 g/l). Environmental exposure was significantly related to blood and urinary N2O (r = 0.59 andr = 0.64, respectively). Blood and urinary N2O were significantly related to each other (r = 0.71), and were equivalent to about 25% of the environmental exposure level. The mean urinary N2O of 1202 g/l in 10/676 subjects was not related to environmental exposure in the operating theatre. The highest urinary N2O levels measured in these 10/676 subjects could be explained by an asymptomatic urinary infection.  相似文献

The mycelial (25°C) and yeast-like (37°C) forms of Penicillium marneffei clinical and type strains were investigated for their in vitro susceptibility to amphotericin B (AmB), 5-fluorocytosine (5-FC), fluconazole (FLU) and itraconazole (ITZ), using Bacto antibiotic medium 3, yeast-nitrogen, Sabouraud's dextrose (pH 5.7) and high resolution (pH 7.1) broth media (1ml/tube), respectively. Results indicated that the minimal inhibitory and minimal fungicidal concentrations (MICs and MFCs) for the mycelial cultures of P. marneffei to AmB were in the range 0.78–1.56 and 0.78–3.125 g/ml, respectively, as against 3.125–25 g (MICs) for the yeast form cultures. The MFCs to AmB for the yeast form were one dilution higher. The MICs to FLU were generally lower for the yeast form (6.25–25 g) than the mycelial form (25–50 g/ml), whereas MFCs for the mycelial cultures were > 100 g as compared to 6.25–100 g for their yeast form. The MICs for the mycelial form to 5-FC ranged from < 0.195–0.39 g. Higher MICs (6.25 g) were recorded for their yeast form. The MFCs to 5-FC for the yeast form were 25–100 g/ml. The MICs for the mycelial form to ITZ ranged from < 0.195 to 3.125 g/ml. Higher values (< 0.195–50 g) were recorded for their yeast-like form. The MFCs to ITZ for mycelial and yeast forms ranged from < 0.195–0.39 and 25–100 g/ml, respectively. Results indicate that P. marnefei's yeast form is more sensitive to FLU and ITZ (8 of 10 strains) while the mycelial form displayed greater susceptibility to AmB and 5-FC. The MICs for ITZ remained steady in SD medium, pH 5.7 to 7.1. However, some strains gave higher MIC values (0.39–1.56 g/ml) when tested in the HR.  相似文献

Selenium contamination of aquatic ecosystems is rapidly becoming a concern throughout the United States, leading to a lowering of the freshwater selenium criteria by the U.S. Environmental Protection Agency from 35 g/L to 5 g/L measured as total selenium. However, considerable information indicates important differences in bioaccumulation and toxicity between the various selenium forms. We conducted a series of experiments to determine the comparative toxicity of selenite and selenate to the amphipodHyallela azteca. In 48 h, 96 h, and 240 h (10 d) LC50 tests, selenite was 2 to 4 times more toxic than selenate, the difference decreasing as exposure time increased. This difference in toxicity was even more pronounced in the 24 d reproductive bioassay, where selenite caused a significant decrease in number of young per female at 200 g/L while selenate had no effect up through 700 g/L (the highest concentration tested).  相似文献

Blood toluene was measured in a group of 100 workers occupationally exposed to a mean 8-h environmental toluene concentration of 128 g/l (34 ppm), and in a group of 269 normal subjects without occupational exposure to toluene. The mean blood toluene of the workers at the end of the shift and the following morning, after 16 h, was 457 and 38 g/l, respectively. The normal subjects had a blood toluene level of 1.1 g/l. On the basis of the highly significant correlation between blood toluene and occupational exposure, it can be calculated that environmental toluene exposure of 188 and 377 g/l (50 and 100 ppm) gives end-of-shift blood toluene levels of 690 and 1390 g/l, respectively. The corresponding blood toluene levels on the following morning are 50 and 100 /l, respectively.  相似文献

Summary Concentrations of p,p'-DDE, p,p-DDD, and p,p-DDT were determined in 34 samples of human milk obtained 3–5 days after delivery and in 37 samples obtained at later times of lactation (up to 55 weeks). All samples contained p,p-DDE, but only several contained p,p-DDD and p,p-DDT. The concentrations of p,p-DDE were 31 g/l in the beginning of lactation and 53 g/l at later time intervals. The concentration ranges in both groups overlap almost completely and the difference in the mean values is not significant.Serum samples from 35 mothers and cord blood were also analyzed. All samples contained p,p-DDE, the concentrations being 18 g/l and 6.8 g/l in the mothers' and cord blood serum, respectively. Serum samples of 24 nonpregnant women contained the same amount of p,p-DDE (20 pg/1) as mothers' sera.All samples were collected in a continental town of Croatia (Yugoslavia) between 1977 and 1979. Isomer-specific concentrations of polychlorinated biphenyls (PCBs) including planar, mono- and di-ortho congeners and concentrations of DDT were determined in striped dolphins affected by a morbillivirus epizootic in the western Mediterranean in 1990. Extremely high concentrations of PCBs ranging from 94 to 670 g/g (wet wt) were detected in the blubber. Similarly, DDT concentrations were high, between 22 and 230 g/g (wet wt). The concentrations of three non-ortho coplanar PCBs were 43 (3,3,4,4-T4CB), 6.8 (3,3,4,4,5-P5CB), and 7.8 (3,3,4,4,5,5-H6CB) ng/g (wet wt), respectively, the highest residue levels reported to date. The estimated 2,3,7,8-TCDD toxic equivalents of non-, mono- and di-ortho PCB congeners in striped dolphins were several times higher than those observed for other marine mammals and humans. Mono-ortho congeners contributed greater 2,3,7,8-TCDD toxic equivalents than non-ortho members. The higher ratio of 3,3,4,4,5,5-H6CB/3,3,4,4,5-P5CB (IUPAC 169/126) suggested a strong induction of mixed function oxidase enzymes and highlighted the possibility of using this ratio as an index for risk assessment of PCB contamination in marine mammals. Elevated concentrations of PCBs may have played a role in the immune depression in striped dolphins, ultimately leading to the development of morbillivirus disease.  相似文献

The herbicides atrazine and linuron, found in Wisconsin's groundwater, were tested alone and in combination, both in vivo and in vitro, to determine their individual and combined genotoxic effects. Human lymphocytes exposed in vitro to either 1 g/ml linuron or 0.001 g/ml atrazine showed little chromosome damage, whereas significant chromosome damage was observed in lymphocytes simultaneously exposed to 0.5 g/ml linuron and 0.0005 g/ml atrazine, suggesting at least an additive model. In another experiment, mice were fed 20 g/ml atrazine, 10 g/ml linuron, or a combination of 10 g/ml atrazine and 5 g/ml linuron in their drinking water for 90 days, after which bone marrow cells and cultured splenocytes were examined for chromosomal damage. None of the treatment groups showed chromosome damage in bone marrow, whereas the cultured splenocytes demonstrated damage in all treatment groups. These experiments suggest that, prior to assessing the risk of a herbicide, it may be necessary to test it in combinations which mimic the mixtures which would occur under field conditions, such as in contaminated groundwater.  相似文献

Toxicity of Cadmium (Cd), Chromium (Cr), Copper (Cu), and Mercury (Hg) on the early developmental stages of Ciona intestinalis was investigated. Developmental defects of larvae after exposure of gametes throughout their development to the larval stage were assessed. Gamete exposure to increasing metal concentrations resulted in a significant decrease of the percentage of normally hatched larvae, showing median effective concentrations (EC50) of 721 g/L (6.42 M) for Cd, 12772 g/L (226 M) for Cr, 36.6 g/L (0.576 M) for Cu, and 44.7 g/L (0.223 M) for Hg. Larval attachment was significantly affected when gametes were exposed to the metals throughout development. The EC50 reducing larval attachment by 50% were 752 g/L (6.7 M) for Cd, 15026 g/L (289 M) for Cr, 67.8 g/L (1.607 M) for Cu, and 78.1 g/L (0.389 M) for Hg. Therefore, on a molar basis Hg is three times more toxic than Cu, 20–30 times more than Cd, and 700–1000 times more toxic than Cr, for both responses.  相似文献

The antimicrobial susceptibility of clinical isolates of the Bacteroides fragilis group obtained from nine large Belgian university and community hospitals was studied. Of the -lactam antibiotics tested, none were active against 100% of the isolates. Piperacillin was active against 93% of the strains at a breakpoint of 64 lag/ml. The percentage of isolates inhibited at 16 g/ml (and 32 g/ml) for the 7-alpha-methoxy antibiotics was: cefoxitin 84 [94]; latamoxef 87 [93]; and cefotetan 64 [79]. B. fragilis tended to be more susceptible to all -lactams than the other members of the group.Among the non -lactam antibiotics, the resistance rate for minocycline, doxycycline, clindamycin and erythromycin (at a breakpoint of 4 g/ml) were l%, 4%, 14% and 34%, respectively. Like the -lactams, clindamycin was less active against non B. fragilis species of the group.Chloramphenicol, metronidazole and tinidazole were still active against 100% of the isolates at their breakpoint values (8 lag/ml).  相似文献

The addition of 100 (g/L of Aroclor® 1242 (A1242) or 2,5,2,5-tetrachlorobiphenyl (TeCB) during 10 week chronic toxicity tests withHyalella azteca resulted in complete mortality. There were no effects on survival, growth, or reproduction after addition of 30 g/L. Toxic effects were observed at tissue levels of between 30 and 180 g/g on a wet weight basis, and tissue levels appear to be a better indicator of toxicity than levels in water. No toxic effects were observed after additions of up to 2,700 g/L of the coplanar congener 3,4,3,4-TeCB.H. azteca has the ability to avoid accumulating in excess of 140 g/g 3,4,3,4-TeCB. The amount taken up was proportional to the amount added in water up to 100 g/L, but was constant at higher additions, possibly accounting for its relatively low toxicity. The low toxicity of the coplanar congener, as compared to the non-coplanar 2,5,2,5-TeCB, is in direct contrast to the high toxicity of coplanar PCB congeners to mammals and may be associated with slower rates of aromatic hydrocarbon metabolism in amphipods. Polychlorinated biphenyl levels measured in amphipods from Lake Ontario are approximately 100-fold below levels associated with toxicity inH. azteca, but are above levels which, through biomagnification up the food chain, lead to salmonid residues in excess of 2 g/g, a tolerance limit for human consumption.  相似文献

Bald eagles (Haliaeetus leucocephalus) nesting in the Apostle Islands National Lakeshore and along the Wisconsin shoreline of Lake Superior produced an average of 0.8 young/occupied nest and had an average nest success of 57% during 1983–1988, compared to 1.3 young/occupied nest and 77% nest success in inland Wisconsin. Contaminant levels in nestling bald eagle carcasses collected from nests near Lake Superior were higher than those collected inland, suggesting local contamination. Prey remains collected at nests consisted of fish (50%); birds, primarily herring gulls (Larus argentatus) (48.4%); and mammals (1.2%). Organochlorine and polychlorinated biphenyl (PCB) residues were present at low levels (DDE: ¯x=0.07 g/g wet wt, PCB:¯x=0.21 g/g wet wt) in fish. Herring gulls contained higher concentrations (DDEx=5.5 g/g wet wt, PCB¯x=16.95 g/g wet wt) and appear to be the major source of elevated contaminant levels in bald eagles nesting near Lake Superior.  相似文献

Lethal and sublethal responses to the herbicides 2,4-D, DEF®, propanil, and trifluralin of various life history stages of the Dungeness crab,Cancer magister, were examined to estimate maximum acceptable toxicant concentrations (MATC) of each compound for this species. Zoeae were found, in long term tests, to be the most sensitive stage. A new sublethal toxicity test was developed in this study to measure the effect of copper and lead on the motility of coral larvae. Larval motility was significantly affected by copper and lead doses immediately after dosing. The copper EC50 values for motility of Goniastrea aspera brain coral larvae (12 h, 21 g/L; 24 h, 16 g/L; 48 h, 22 g/L) were much lower than the copper LC50 values for G. aspera larval survival (6 h, 260 g/L, and 24 h, 121 g/L, for 5-day-old larvae and 6 h, 248 g/L, and 24 h, 136.64 g/L, for 6-day-old larvae) during the early part of the experiments. However, at later times, the LC50 values (48 h, 40 g/L, for 5-day-old larvae and 48- h, 87 g/L, for 6-day-old larvae) were similar to the EC50 values for larval motility. The lead 72-h EC50 value for G. aspera larval motility (2900 g/L) was much lower than the lead 72-h LC50 value for larval survival (9890 g/L). The results show that larval motility can be a useful parameter to measure in order to determine the sublethal effects of trace metals on coral larvae.  相似文献

Two chronic toxicity tests were conducted in which Daphnia magna were either continuously or intermittently exposed to bromoxynil octanoate (BO; as Buctril®) for 28 d. In the intermittent exposure test, daphnids were exposed to daily pulses of BO with 24-h mean concentrations equal to those in the continuous exposure test, and the peak concentrations were three times the 24-h mean values. After 28 d of continuous exposure to BO, survival of daphnids was reduced at 80 g/L, whereas mean number of young per adult, intrinsic rate of natural increase, and mean weight of adults were all reduced at 40 g/L. Intermittent exposures to daily pulses of BO for 28 d caused reduced survival of daphnids at 24-h mean concentrations 40 g/L and reduced mean number of young per adult, intrinsic rate of natural increase, and mean weight of adults at 24-h mean concentrations 20 g/L. The estimated geometric mean-maximum acceptable toxicant concentrations of BO based on 24-h mean nominal values were 28 g/L for continuous exposures and 14 g/L for intermittent exposures. These results demonstrated that continuous-exposure studies may not be adequate in assessing herbicide toxicity to aquatic biota when concentrations fluctuate temporally.  相似文献

Cadmium (Cd) toxicity and amelioration of Cd toxicity by Mn were measured inChlorella pyrenoidosa, using turbidostat culture. The responses were measured in terms of the maximum specific growth rate, max, of the populations. In turbidostat culture max is a dependent variable that can be measured continuously. Cd (as CdCl2· 2.5 H2O) was added to control populations at a concentration of 1.8 M Cd. Toxicity was expressed after a 5 generation lag and resulted in a max steady state 62% lower than the initial control after 2 generations. With continued Cd exposure, Mn (as MnCl2 · 6H2O) was then added stepwise to a concentration of 10.4 M Mn which caused a rapid, immediate increase in max followed by linear increase until a steady-state plateau was reached at a max 90% of control. The ameliorative response spanned 20 culture generations. After addition of Mn (10.4 M), cellular Cd concentration did not change and cellular Mn concentration increased. Increase in mean cell size accompanied Cd exposure and was significantly decreased when supplemented with 10.4 M Mn. Possible mechanisms of the amelioration are discussed.  相似文献

This study determined the acute and chronic toxicity of the organophosphate insecticide fonofos to standard freshwater aquatic organisms under laboratory conditions. Fonofos was acutely toxic to bluegill (Lepomis macrochirus), Daphnia (D. magna), and midge (Chironomous riparius) at 5.3, 2.7, and 39 g/L, respectively. Three fonofos formulations (technical, 94.8% A.I.; 20G, field granular 20% A.I.; and 4E, field liquid 4#/gal A.I.) exhibited similar acute toxicities to bluegill. Exposure to fonofos delayed reproduction and decreased the intrinsic rate of increase of Daphnia during 21-d chronic exposure at the lowest tested concentration (0.08 g/L). The no observable effect concentration (NOEC) for Daphnia survival was 0.42 g/L; 0% survival occurred at the lowest observable effect concentration (LOEC) of 1.45 g/L. The NOEC for midge emergence was 3.42 g/L; only 34% emergence occurred at the LOEC of 8.24 g/L. Chronic 30-d exposure of juvenile bluegills decreased growth and survival at 5.65 g/L (LOEC), but no effects occurred at 2.33 g/L (NOEC). The relative hazard of fonofos to aquatic life is similar to other carbamate and organophosphate corn insecticides.  相似文献

Summary The influence of the lead content of drinking water on the transplacental transfer of lead was investigated in 70 pregnant women living in a rural area of Belgium. The mothers were divided into 2 groups: group A: morning water lead below 50 g/liter; group B: morning water lead above this value. In group A, the mean lead content of water was 11.8 g/liter and in group B it amounted to 247.4 g/liter.The difference in the mean lead concentration between the two groups were for maternal blood: 3.2 g/100 ml, for umbilical cord blood: 3.3 g/100 ml, and for placenta: 3.6 g/100 g. These differences are statistically significant.There were significant correlations between water lead and lead concentration in blood (mother, newborn) or placenta. An increment of water lead concentration from 50 to 500 g/liter increases blood lead concentration in mother and in newborn by about 3 g/100 ml and in placenta by about 2.5 g/100 g (wet weight).  相似文献

Microencapsulated permethrin (penncapthrin) was evaluated under laboratory conditions for its toxicity toward several nontarget aquatic invertebrates. Average LC50 estimates for selected lotic invertebrates, based on a one hour dosing regime, were: 2.71 mg/L forSimulium vittatum, 4.59 mg/L forHydropsyche spp., and 13.41 mg/L forIsonychia bicolor. In acute static tests withDaphnia magna, there was no significant difference (p0.05) between the toxicity of penncapthrin at 96 h (LC50 range: 6.80–22.5 g/L) and the EC formulation at 72 h (LC50 range: 0.6–21 g/L). Comparatively, the toxicity of microencapsulated methyl parathion (penncap-m) was not significantly different from that of penncapthrin towardD. magna, the former having LC50 estimates ranging form 0.3–12.25 g/L. LC50 estimates associated withDaphnia pulex ranged from 19 to 131 g/L. The toxicity of penncapthrin and penncap-m towardD. pulex was difficult to determine because of frequent control mortality due to food deprivation resulting from the need to run tests for longer than 48 h. In successful tests, LC50 estimates ranged from 19 to 28 g/L for penncapthrin and 0.08 to 25 g/L for penncap-m after 72 h exposure. In long term toxicity tests, 95% of D. magna at 1 g/L, 44% at 10 g/L, and 20% at 15 g/L survived after 39 days exposure. Less than 15% ofD. pulex survived over the same concentration range following 32 days exposure. Despite some drawbacks, long-term toxicity tests were more appropriate than short-term tests for evaluating microencapsulated sticides because of reduced variability in LC50 estimates and lower control mortality.  相似文献

Summary A cross-sectional study was performed in order to investigate the influence of chronic lead-exposure on the peripheral nervous system. We examined 148 male workers of a storage battery manufacturing plant, who had been exposed to lead metal and inorganic lead compounds for 1 to 28 years (mean 11 years). Fifteen workers with non-occupational risks of peripheral neuropathy (former diseases, alcohol abuse, medication) were excluded from the study. The investigation program comprised: case history, physical examination, analyses of blood- and urine-samples and determination of maximal motor, mixed and sensory conduction velocity (NCV) of the ulnar and median nerve of the right forearm. Objectively no worker showed any signs of health effects related to lead exposure. The Biological Monitoring included the determination of (1) Blood-lead level (Pb-B), (2) Free erythrocyte porphyrins (FEP), (3) -Aminolevulinic acid dehydratase (ALA-D) and (4) -Aminolevulinic acid in urine (ALA-U). Further time-weighted-average (TWA)-values of Pb-B were calculated on the basis of several determinations over the period 1975–1981. The following actual (TWA) median values resulted: Pb-B 53 g/dl (54 g/dl), ALA-U 5.6 mg/l (8.4 mg/l), FEP 2.0 mg/l (2.0 mg/l). The Biologischer Arbeitsstoff Toleranz Wert (BAT) of 70 g//dl for Pb-B was exceeded in 15 workers (11%), and of 15 mg/l for ALA-U in 30 cases (23%). In comparison with age-matched controls, the lead workers showed a mild slowing of NCV with mean values between 0.8 and 2.0 m/s. Multiple stepwise regression analyses revealed statistically significant correlations between the four NCV and age as well as Pb-B. There were better correlations by using TWA than actual data of Pb-B. Consideration of the results of the regression analyses, together with an evaluation of the individual neurophysiological status as a function of internal lead exposure, a dose-effect-relationship was found only in the case of Pb-B exceeding 70 g/dl. From our study it is concluded that chronic lead exposure resulting in blood-lead levels of below 70 g/dl is no occupational risk causing a functionally significant slowing of nerve conduction velocities.With Grants from the Deutsche Forschungsgemeinschaft, Bonn (Project no.
