[目的]探讨沙立度胺(thalidomide,TD)及其对映体(S-thalidomide)S-TD对人非小细胞肺癌系NCI-H460细胞增殖的影响。[方法]体外培养NCI-H460细胞,采用MTT比色法测定不同质量浓度的TD、S-TD对NCI-H460细胞增殖的影响;采用流式细胞仪检测对细胞周期的影响;DAPI染色观察诱导细胞凋亡作用。[结果]在10.0~200.0μg/ml范围内,TD、S-TD作用于NCI-H460细胞24h可抑制细胞增殖、48h显著抑制细胞增殖,而在高浓度时(200.0μg/ml)S-TD的抑制作用弱于TD;TD、S-TD作用NCI-H460细胞24h、48h、72h,TD的抑制率分别为25.69%、41.92%、7.57%,S-TD为14.77%、33.24%、8.46%。200.0μg/mlTD和S-TD处理NCI-H460细胞48h后,G2/M期细胞比例显著增高,S期细胞比例下降。DAPI染色结果发现细胞数量明显减少,但未出现核固缩、核边集等凋亡形态学改变。[结论]TD和S-TD在一定浓度范围对NCI-H460细胞具有抗增殖作用,但S-TD的抗增殖活性稍弱于TD。抑制增殖作用可能与阻滞G2/M期细胞周期有关,DAPI染色结果未显示诱导细胞凋亡作用。  相似文献

目的:探讨榄香烯乳对人肺腺癌A549细胞株凋亡蛋白表达的影响.方法:体外培养人肺腺癌A549细胞株,四甲基偶氮唑蓝比色试验法(MTT法)测定不同浓度榄香烯乳在不同作用时间下对A549细胞株的生长抑制作用.IC软件计算10%的细胞抑制浓度(IC10).免疫细胞化学染色观察榄香烯乳作用24h后A549细胞的Bcl-2、Bax、Survivin及VEGF蛋白的表达变化.结果:榄香烯乳对人肺腺癌A549细胞株增殖具有明显抑制作用,呈时间和剂量依赖性.榄香烯乳作用24h后IC10为43.49μg/ml,Bax蛋白表达增加,细胞质着色增强;Bcl-2蛋白表达下降,细胞质着色减弱;增殖蛋白Survivin和VEGF的表达下降,且具有药物浓度相关性.结论:榄香烯乳对人肺腺癌A549细胞株有明显的抑制作用,榄香烯乳可上调凋亡相关蛋白表达、诱导凋亡.  相似文献

目的:研究白藜芦醇(resveratrol,Res)对肺腺癌A549细胞增殖,黏附及侵袭的影响。方法:用不同剂量的Res作用于A549细胞,MTT法测定A549细胞的增殖水平(成纤维3T3细胞为对照),流式细胞仪检测A549细胞的细胞周期和凋亡,体外黏附实验测定A549细胞的黏附能力,Transwell实验测定A549细胞的侵袭能力,荧光免疫细胞化学方法测定A549细胞中MMP-2和TIMP-2蛋白的表达。结果:Res以剂量(20~80μmol/L)依赖和时间(0~72 h)依赖方式抑制A549细胞的增殖,同样条件对3T3细胞增殖无影响。10、20、40、80μmol/L Res作用后,A549细胞的凋亡率分别为(34.9±0.91)%、(41.33±0.13)%、(45.47±0.87)%和(59.46±0.59)%。经20μmol/L Res处理48 h后,S期A549细胞比例为(56.41±1.67)%,细胞周期阻滞在S期。20μmol/L以上的Res可引起A549细胞的黏附力下降、侵袭力降低(P<0.05);同时,A549细胞内MMP-2蛋白表达下调,而TIMP-2蛋白表达增加。结论:Res抑制肺腺癌A549细胞的增殖、黏附和侵袭,其机制可能涉及对MMP-2和TIMP-2表达的双向调控。  相似文献

目的探讨褪黑素在体外共培养条件下对肺腺癌A549细胞诱导血管内皮细胞增殖的影响及相关机制。方法建立肺腺癌A549细胞和人脐静脉内皮细胞(HUVECS)ECV-304共培养模型,实验共分成四组:Con组(对照组),nM组(褪黑素终浓度1 nM/L),μM组(褪黑素终浓度1 μM/L),mM组(褪黑素终浓度1 mM/L)。采用BrdU法观察不同浓度褪黑素对血管内皮细胞(ECV-304)增殖的影响,细胞迁移实验观察褪黑素对血管内皮细胞活力的抑制作用,ELISA法检测褪黑素对肺腺癌细胞VEGF蛋白表达的影响。结果低浓度的MLT(1 nM/L)对HUVECs增殖无明显影响,但高浓度 MLT(1 mM/L)明显抑制HUVECs增殖,并有浓度依赖倾向; 高浓度的褪黑素明显降低肺腺癌A549细胞培养液中VEGF的表达。 结论高浓度的褪黑素可以抑制肺腺癌细胞诱导的血管内皮细胞的增殖及迁移,其作用机制可能与褪黑素抑制肺腺癌细胞VEGF的表达有关。  相似文献

目的:探讨HuR siRNA 对人肺腺癌A549 株细胞增殖、迁移及侵袭的影响及其可能机制。方法:HuR siRNA 瞬时转染肺腺癌A549 细胞24 h 后,CCK-8 实验、集落形成实验检测细胞的增殖能力,划痕实验检测细胞迁移能力,Transwell 实验检测细胞侵袭能力,qPCR和Western blotting 检测HuR及基质金属蛋白酶(MMP)-9 基因mRNA和蛋白质表达。结果:HuR siRNA能够抑制人肺腺癌A549 细胞的增殖、迁移及侵袭;HuR siRNA 显著降低A549 细胞HuR、MMP-9 mRNA和蛋白表达(P<0.01)。结论:下调HuR能抑制肺腺癌A549细胞增殖、迁移及侵袭,其机制与下调MMP-9 有关。  相似文献

目的 通过研究VEGF、MMP-9之间的关系,初步探讨VEGF促进肺癌细胞侵袭性的自分泌机制,以期为肿瘤的抗VEGF治疗提供一定的理论依据.方法 在体外用条件培养基培养人肺腺癌细胞A549,经细胞免疫化学染色、侵袭实验方法来测定VEGF对肺腺癌细胞A549的MMP-9表达及侵袭性的影响.结果 (1)各实验组的A549细胞在不同浓度的rh VEGF165和不同时间的作用下,细胞内的MMP-9的表达有差异;(2)A549细胞侵袭数在和照组间差异有显著性(P<0.001).结论 (1)VEGF能上调A549细胞内的MMP-9表达,且促进作用具有时间-剂量依赖性;(2)VEGF能促进A549细胞的侵袭性;(3)肺癌细胞中存在有自分泌机制,VEGF可通过自分泌机制上调肺癌细胞的MMP-9表达,进而促进肿瘤的浸润转移.  相似文献

目的:探讨人参皂苷CK对人肺腺癌A549细胞增殖的影响及其作用机制。方法:不同浓度的人参皂苷CK作用于人肺腺癌A549细胞株,MTT比色法测定细胞增殖抑制率;倒置显微镜观察细胞形态学变化;流式细胞术分析细胞周期及细胞凋亡;ELISA法检测细胞内源性VEGF含量的变化。结果:人参皂苷CK对A549细胞增殖有抑制作用,其抑制作用呈浓度和时间依赖关系;倒置显微镜下可观察到细胞明显的形态学改变;流式细胞仪可检测到实验组细胞出现明显的凋亡峰,且细胞周期被阻滞在G0/G1期;实验组细胞培养液中的VEGF低于对照组(P<0.05),且随着人参皂苷CK浓度增加和作用时间的延长,VEGF的含量降低(P<0.05)。结论:人参皂苷CK可抑制人肺腺癌A549细胞的增殖并诱导其凋亡,并能抑制其内源性分泌VEGF。  相似文献

背景与目的侵袭转移是人肺腺癌死亡的主要原因,表皮生长因子(epidermal growth factor,EGF)可通过活化ERK和PI3K-Akt信号通路,促进人肺腺癌A549细胞侵袭。本实验旨在研究白藜芦醇抑制EGF诱导体外A549细胞侵袭的作用,并初步探讨其机制。方法采用MTT法确定白藜芦醇对A549细胞无明显毒性的浓度范围,并以该浓度范围内的白藜芦醇作用EGF诱导的A549细胞,Transwell小室法检测其抑制细胞侵袭作用,明胶酶谱法分析细胞的MMP-2活性变化,Westernblot检测EGF信号通路相关蛋白表达。结果30μM内的白藜芦醇无明显的细胞毒性,20μM处理的受EGF诱导的A549细胞侵袭能力明显降低,MMP-2分泌减少,胞内p-ERK1/2和PI3K(0.5h-6h)含量下降。结论20μM白藜芦醇可有效抑制A549细胞侵袭,其机制可能是通过抑制ERK通路和PI3K-Akt通路中相关蛋白激活,最终降低MMP-2的分泌。  相似文献

背景与目的 Amorphigenin是从紫穗槐属植物的种子中分离提取的鱼藤酮类化合物,研究发现amorphigenin对多种肿瘤细胞具有增殖抑制作用。本研究拟探讨amorphigenin对人肺腺癌耐顺铂细胞株A549/DDP的抗肿瘤作用及其可能的分子机制。方法采用CCK-8法测定A549/DDP细胞的增殖;克隆形成实验测定A549/DDP细胞的克隆形成;流式细胞术检测细胞的凋亡率;Western blot技术检测caspase-3、PARP和LRP蛋白的表达。结果Amorphigenin可抑制A549/DDP细胞的增殖48 h[半数抑制浓度(half maximal inhibitory concentra-tion, IC50)]为(2.19±0.92)μmol/L、抑制克隆形成及诱导细胞凋亡。此外,Amorphigenin与顺铂联合可协同地抑制A549/DDP细胞生长和促进凋亡;降低耐药蛋白LRP蛋白的表达。结论 Amorphigenin可抑制A549/DDP细胞增殖和促进细胞凋亡;amorphigenin可能是通过抑制耐药蛋白LRP蛋白表达,进而与顺铂对A549/DDP细胞产生协同抑制作用。  相似文献

[目的]探讨重组人凋亡素2配体(Rh-Apo2L)联合长春瑞滨(NVB)对人肺腺癌A549细胞的杀伤作用及机制。[方法]人肺腺癌A549细胞分成4组:对照组、Rh-Apo2L组、NVB组、联合组。用MTT法检测不同浓度Rh-Apo2L和NVB对A549细胞的抑制率,绘制细胞生长曲线,流式细胞仪测定细胞周期分布,RT-PCR方法检测死亡受体DR5基因mRNA的表达。[结果]MTT结果显示不同浓度Rh-Apo2L和NVB对人肺癌A549细胞的体外抑制作用均呈剂量效应关系,Rh-Apo2L处理48h后的IC20为3.48μg/ml、IC50为197.92μg/ml,NVB处理48h后的IC20为0.29μg/ml、IC50为3.44μg/ml。联合组对A549细胞的抑制作用高于单药组(P=0.001)。流式细胞分析显示含NVB药物两组在24hG2/M期比率高于对照组。3个实验组在24h、48h、72h后DR5基因mRNA相对表达量均较对照组多(P<0.05),但各实验组间均无统计学差异(P>0.05)。[结论]Rh-Apo2L与NVB联合应用能协同增强对人肺腺癌细胞株A549的体外抗肿瘤活性,其协同作用机制可能与细胞周期阻滞在G2/M期有关,但与DR5基因mRNA的表达无相关性。  相似文献

The literature suggests that religiosity helps cope with illness. The present study examined the role of religiosity in functioning among African Americans and Whites with a cancer diagnosis. Patients were recruited from an existing study and mailed a religiosity survey. Participants (N = 269; 36% African American, 56% women) completed the mail survey, and interview data from the larger cohort was utilized in the analysis. Multivariate analyses indicated that in the overall sample religious behaviors were marginally and positively associated with mental health and negatively with depressive symptoms. Among women, religious behaviors were positively associated with mental health and negatively with depressive symptoms. Religiosity was not a predictor of study outcomes for men. Among African Americans, religious behaviors were positively associated with mental health and vitality. Among Whites, religious behaviors were negatively associated with depressive symptoms. These findings suggest a mixed role of religious involvement in cancer outcomes. The current findings may have applied potential in the areas of emotional functioning and depression.  相似文献

We used a rat model to study the effects of renal irradiation on the pharmacology of methotrexate (MTX) and cisplatinum (cis-Pt). Unanesthetized rats were given bilateral kidney irradiation (20 Gy in 9 fractions). At 9 months after irradiation, 3% of the animals had died and survivors showed moderately impaired renal function. At 15 months, 30% of the animals had died and survivors showed severely impaired renal function. Some animals were given i.v. MTX 1 week to 15 months after irradiation. In irradiated rats, the area under the MTX plasma clearance curve equaled that of controls through 6 months, and was significantly above controls from 9 months on. Other animals were given i.p. cis-Pt 1 week to 9 months after irradiation. The acute toxicity of cis-Pt was the same in control and irradiated rats when cis-Pt was given immediately before or after irradiation. Beginning 3 months after irradiation there was a progressive increase in cis-Pt toxicity and a simultaneous decrease in urinary platinum excretion. Irradiated animals that survived cis-Pt treatment showed increased radiation nephritis; the greatest effect occurred when cis-Pt was given 3 months or more after irradiation. MTX and cis-Pt clearance decreased when renal dysfunction was first observed and changes in renal function preceded changes in drug clearance and toxicity.  相似文献

目的:探讨VEGF和KDR在大肠腺瘤和大肠腺癌中的表达及临床病理特征的关系。方法:大肠腺瘤和大肠腺癌组织标本各100例,采用免疫组织化学染色法检测VEGF和KDR在标本中的表达情况。结果:VEGF和KDR在大肠腺癌组中的阳性表达明显高于大肠腺瘤组(P〈0.05);在正常大肠黏膜均未见VEGF和KDR表达的阳性染色;VEGF阳性表达组中KDR的阳性表达率为70%,显著高于VEGF阴性表达组中KDR的阳性表达率16%,两组比较有统计学意义(P〈0.01)。结论:大肠腺癌组织中KDR的表达与肿瘤大小、转移情况、浸润深度密切相关;VEGF和KDR在大肠腺瘤中的表达与患者的年龄、性别及分型均无相关性,而与增生程度相关(P〈0.05)。在大肠腺癌患者中VEGF及KDR表达更高,二者具有协同效应。  相似文献

Morphine is an analgesic widely used to alleviate cancer pain. In addition, the perioperative management of pain in cancer surgery patients most often includes opioids. However, there are reports that these drugs may alter cancer recurrence or metastasis. Several mechanisms have been proposed, such as the modulation of the immune response or cellular pathways that control the survival and migratory behavior of cancer cells. The published literature, however, presents some discrepancies, with reports suggesting that opioids may either promote or prevent the spread of cancer. It is of great importance to determine whether opioids, in particular the most widely used, morphine, may increase the risk of metastasis when used in cancer surgery. This review examines the available data on the effects of morphine which influence cancer metastasis or recurrence, including immunomodulation, tumor cell aggressiveness, and angiogenesis, with special emphasis on recently published clinical and laboratory based studies. We further discuss the parameters that may explain the difference between reports on the effects of morphine on cancer.  相似文献
