目的:建立测定人血浆中双氯芬酸钠含量的超快速液相色谱-质谱联用(UFLC-MS/MS)方法。方法:血浆样品经乙腈沉淀蛋白后,以乙腈-2 mmol.L-1乙酸铵水溶液(45∶55)为流动相,用Ultimate C18(5μm,2.1 mm×150 mm)色谱柱,通过电喷雾离子化三重四级杆串联质谱,经多反应监测模式检测,监测的离子对为m/z 295.9→m/z 251.8(双氯芬酸钠),m/z250.8→m/z207.9(苯妥英钠,内标)。结果:双氯芬酸钠在1.955~195.5μg.L-1范围内线性关系良好,最低检测浓度为1.955μg.L-1。方法回收率(n=5)分别为73.92%,80.98%,86.28%,日内和日间精密度RSD均小于10%。结论:该法操作简单,灵敏度高,准确,重现性好,适用于该药的临床药代动力学研究。  相似文献

目的 :建立人血浆中双氯芬酸钠浓度的高效液相色谱测定方法。方法 :血样加乙腈去蛋白沉淀后 ,在Hypersil -BDS(250mm×4 6mm ,5μm)色谱柱上 ,以甲醇 -水 -10 %磷酸 (60∶40∶1)为流动相进行色谱分离 ,检测波长为276nm ,柱温为35℃。结果 :双氯芬酸钠浓度在20 50~2050 0ng/ml范围线性关系良好 (n=5,r=0 9998) ,最低检测浓度约为10ng/ml。低、中、高3种浓度的相对回收率为99 47 %~105 4 % ;日内相对标准差为3 77%~5 81 % (n=5) ,日间相对标准差为4 93 %~8 96% (n=5)。结论 :本法简便、快速 ,适合双氯芬酸钠药动学的研究  相似文献

目的制备双氯芬酸钠海藻酸钙凝胶微球,并对凝胶微球的性质进行分析。方法采用滴制法制备双氯芬酸钠海藻酸钙凝胶微球,紫外分光光度法测定药物的含量,对凝胶微球的粒度、圆整度、休止角和堆密度进行测定,并用扫描电镜观察凝胶微球的形态。结果制备的双氯芬酸钠海藻酸钙凝胶微球包封率较高,粉体学性质良好,球形均匀圆整。结论该方法适用于双氯芬酸钠海藻酸钙凝胶微球的制备。  相似文献

目的 建立大鼠微透析液和血浆中苦参碱的反相高效液相检测方法。方法 色谱柱为Diamonsil C18柱(4.6 mm×150 mm,5 μm);流动相为乙腈-0.1%磷酸(用三乙胺调pH至7.6~7.7)(微透析液测定:35∶65;血浆测定:30∶70);流速1.0 mL·min-1;检测波长220 nm;柱温35 ℃;进样量20 μL。结果 本方法测定苦参碱线性范围微透析液为0.10~25.00 μg·mL-1,血浆为0.32~81.80 μg·mL-1,相关系数均为1。微透析液的日内、日间精密度RSD≤6.95 %,血浆RSD≤10.75 %。微透析液的平均回收率为94.7~105.9 %,血浆为98.6~107.9 %,稳定性良好。结论 本方法准确、简单、专属性强,适用于苦参碱大鼠体内药动学研究中微透析液和血浆中药物浓度的测定。  相似文献

目的将双氯芬酸钠制备成明胶微球,考察其缓释效果。方法采用单凝聚法制备双氯芬酸钠明胶微球,并对微球的体内外释药进行考察。结果制备的微球粒径范围为48~100μm,平均粒径为70.70±11.29μm,载药量为35%。体内外释药实验结果表明双氯芬酸钠明胶微球有缓释作用。结论本法制备的双氯芬酸钠明胶微球能够起到明显缓释作用。  相似文献

本文结合脑内微透析技术与现代分析方法,考察川芎嗪对大鼠脑内乙酰胆碱释放的影响。采用脑内微透析技术进行取样,建立高效液相-串联四极杆质谱方法测定脑透析液中的乙酰胆碱含量。结果显示,川芎嗪皮下给药能够剂量相关地增加大鼠脑内不同脑区中乙酰胆碱的释放。该法能够准确反映药物对大鼠脑内乙酰胆碱释放量的影响,与传统方法相比,具有明显的优势。  相似文献

目的:对双氯芬酸钠微乳(DS-ME)的性质、皮肤刺激性及体外透皮吸收进行考察,探讨其局部外用给药的可行性。方法:采用紫外分光光度法测定DS-ME中DS的含量;采用激光粒度仪测定DS-ME的粒径分布;家兔皮肤刺激性实验考察DS-ME和空白ME对单次给药正常皮肤、多次给药正常皮肤和单次给药破损皮肤的影响;Franz扩散池法比较DS-ME与市售DS凝胶经小鼠离体皮肤的透皮参数。结果:所制DS-ME为O/W型微乳,粒径为(30.140±9.020)nm;与空白ME比较,DS-ME对家兔皮肤的刺激评分无明显差异;DS-ME与市售DS凝胶的稳态渗透速率分别为34.16、18.62μg/(cm~2·h),渗透系数分别为1.029、0.561 cm/h,滞后时间分别为0.124 2、0.367 2 h。结论:DS-ME的粒径小,无皮肤刺激性,可提高DS的透皮吸收速率。  相似文献

PurposeThis study was designed to evaluate the protective effects of allicin against diclofenac sodium induced hepatonephro toxicity in rats.MethodsSixty male Wister albino rats were assigned into six groups. The control group received calcium carbonate and corn starch. 2nd group received diclofenac sodium (2 mg/kg bw orally) for 30 days. 3rd group received allicin (45 mg/kg bw orally) for 30 days. 4th group administrated diclofenac sodium as in the 2nd group and allicin (15 mg/kg bw orally) for 30 days. 5th group received diclofenac sodium as in the 2nd group and allicin (30 mg/kg bw orally) for 30 days. 6th group received diclofenac sodium as 2nd and allicin (45 mg/kg bw orally) for 30 days.ResultsDiclofenac sodium significantly elevated activities of serum aspartate aminotransferase and alanine aminotransferase and serum levels of creatinine and urea. In addition, it induced hyperglycemia, lipid peroxidation, pathological alteration and caspase 3 protein expression in hepatic and renal tissues. However, it decreased reduced glutathione concentration and proliferating cell nuclear antigen protein expression in hepatic tissues. In contrast, allicin modulated the diclofenac sodium induced alteration in liver and kidney functions and structures dose dependently.ConclusionThis study indicated that allicin had potential preventive effects against diclofenac sodium induced hepatonephro toxicity in rats.  相似文献

目的:评价双芬酸钠滴眼液在人工晶体植入手术前后的应用效果。方法:在人工晶体植入术前,术后应用0.1%双氯芬酸钠滴眼液42例,同时用生理盐水设对照组36例,分别观察两组病人的临床效果。结果:双氯芬酸钠滴 液可以有效地防止瞳孔缩小及减轻术后炎症反应,结论:在人工晶体植入术前,术后加用此药,能达到缩瞳抑制及抗炎的双重作用,值得推广。  相似文献

目的建立毛细管区带电泳法测定人体尿样中双氯灭痛、氯丙嗪含量的方法。方法采用弹性石英毛细管(31.5cm,25μmi.d.,360μmo.d.),以4.90×10-3mol/LNa2HPO4-7.80×10-3mol/LNaH2PO4(pH=7)为缓冲液,10kV分离电压,5kV电渗进样10s,碳纤维电极工作电极(长200μm,直径8μm),检测电势0.72V。结果双氯灭痛、氯丙嗪两种药物分别在9.90×10-6~5.00×10-4mol/L(r=0.9982)、5.0×10-7~1.0×10-4mol/L(r=0.9998)范围内表现出良好的线性,其回收率分别为104%,96.0%。结论该方法简便、快速、准确,可作为人体尿样中双氯灭痛、氯丙嗪的分离检测方法。  相似文献

Diclofenac sodium is widely used in the non-steroidal anti-inflammatory drug in the treatment of pain and inflammation. It is also particularly associated with its adverse effects on avian fauna and linked to environmental issues. The present study was aimed to assess the dose-dependent toxicity of diclofenac sodium on a male reproductive system of rats. Four groups of healthy adult fertile male rats were administered with saline (control) or 0.25?mg/kg, 0.50?mg/kg and 1.0?mg/kg of diclofenac sodium, respectively for 30 days. Alterations in body and organ weight, sperm and testicular cell population dynamics, serum biochemistry, histopathology, and hematology were investigated as per aimed objectives. Diclofenac sodium treatment significantly (p?≤?0.001) reduced weights of testis, epididymis, ventral prostate and seminal vesicle. Sperm count, sperm density (in epididymis and testis), sperm motility and testicular cell population dynamics were lowered in a dose-dependent manner. Administration of diclofenac exhibited varying degrees of degeneration testis, abnormal histo-architectures, and shrinkages in seminiferous tubules, particularly in higher doses. Diclofenac sodium treatments also altered hepatic and renal function parameters significantly. In conclusion, it may claim that diclofenac sodium treatment altered reproductive metabolic status, androgenic activities and histo-architecture of the testis of male rats and induced hepatotoxicity and renal toxicity.  相似文献

目的 比较研究双氯芬酸钾凝胶(DPG)和双氯芬酸钠凝胶(DSG)治疗局部风湿性疼痛综合征的疗效和安全性。方法 试验设计为多中心、随机对照、单盲、平行性的比较研究。试验组 60例,1%DPG外用,每日3-4次,每次2-4g,日总量不超过20g,疗程2周;对照组45例,1%DSG外用,使用方法同DPG。结果 2周时DPG的总有效率为 88%,DSG总有效率为87%,DPG组疗效明显高于 DSG组(P<0.05)。两药均能显著改善病人的临床症状和体征(P<0.05~0.001),两组相比DPG对关节肿胀数的改善优于DSG组(P<0.05),对其他各指标的改善两组间差异无显著性(P>0.05). DPG的耐受性良好,无1例病人发生不良反应;DSG组的不良反应发生率为2%,两组间差异无显著性(P>0.05).结论DPG治疗局部风湿性疼痛综合征的疗效优于DSG,且耐受性好,无明显不良反应。  相似文献

Summary In experimental animal models nonsteroidal anti-inflammatory drugs may influence gastrointestinal motility, but as evidence is lacking in man. The effect of diclofenac sodum 75 mg i.m. on the motor response of the upper gastrointestinal tract to food has been studied by manometry in 9 healthy volunteers. Diclofenac had no effect on the motor activity of the stomach, duodenum, or jejunum after a 605 kcal meal.  相似文献

目的观察双氯芬酸钠栓用于妇产科术后镇痛的效果。方法将妇产科手术患者1000例随机分为治疗组和对照组各500例,于术后麻醉作用消失后,治疗组给予双氯芬酸钠栓肛门塞入,对照组给予盐酸哌替啶针剂,比较2组患者术后镇痛情况。结果治疗组术后镇痛效果优于对照组,差异有统计学意义(P<0.05)。结论双氯芬酸钠栓用于术后镇痛效果满意,且价廉、方便,未发现不良反应,更适合经济困难者。  相似文献
