目的:研究诃子乙醇提物的抗氧化作用及其机制。方法:测定诃子醇提物对FeSO4/Cys氧化体系对肝微粒体脂质过氧化的抑制率,对H2O2引起红细胞溶血的抑制效果、自氧化溶血的抑制率和清除DPPH自由基实验。结果:诃子乙醇提取物12.5ug/ml对肝微粒体脂质过氧化有显著抑制作用,能够抑制H2O2引起的红细胞溶血和自氧化引起的溶血,乙醇提取物浓度在3ug/ml对DPPH自由基有显著的清除作用。结论:诃子具有较强的抗氧化能力,具有很好的应用前景和开发价值。  相似文献

目的研究微孔草油的抗氧化活性。方法采用95%乙醇浸提工艺提取,初步分离后,经石油醚萃取,硅胶柱色谱进一步分离纯化得到3种提取物,提取物通过对羟基自由基、超氧阴离子自由基、DPPH自由基的清除能力测定进行抗氧化活性研究。结果微孔草油具有抗氧化活性。抗氧化性:提取物C<提取物B<提取物A;提取物对自由基的清除能力表现为:DPPH自由基<超氧阴离子自由基<羟基自由基。结论实验证明提取物A抗氧化活性最佳。  相似文献

目的: 研究马尾松等5种中药的乙醇提取物的体外抗氧化活性。 方法: 采用乙醇超声提取法获得各中药提取物,并以抗坏血酸(VitC)为阳性对照来考察其对超氧阴离子(O2-·)、羟自由基(·OH)、1,1-二苯基-2-三硝基苯肼自由基(DPPH·)的清除效果。 结果: 甘草、丹参、马尾松、当归、罗汉松对O2-·的抑制率分别为(浓度相当于原药材100 g·L-1):15.86%,11.68%,11.31%,7.88%,5.94%;在清除DPPH·的实验中各中药的IC50(g·L-1)分别为:丹参1.6,马尾松1.87,罗汉松3.11,甘草13.91,当归22.32;而在清除·OH的实验中各中药的IC50(g·L-1)则为:丹参1.6,罗汉松2.02,马尾松2.1,甘草2.77,当归4.03;而三大体系阳性对照的IC50分别为0.047,0.93,0.21 g·L-1结论: 5种中药乙醇超声提取物均具有一定的抗氧化活性,其中丹参、马尾松效果较好;但与阳性对照Vit C相比仍有一定差距。  相似文献

目的:抗氧化物质一般通过诱导抗氧化酶的表达或直接与自由基发生反应这两种方式发挥作用,银杏叶提取物(EGb)的抗氧化作用是通过哪种途径值得研究.在本研究中,我们在体外对两种途径进行了测试.方法:EGb作用于细胞,用Western-blot检测其对抗氧化酶GST-P1和GCLC的诱导作用;用相应的功能检测法测定EGb在体外直接清除超氧阴离子及羟自由基、抑制大鼠红细胞溶血及大鼠肝组织的脂质过氧化的作用.结果:EGb可诱导抗氧化酶GST-P1和GCLC的表达,并能直接清除超氧阴离子及羟自由基、抑制大鼠红细胞溶血及大鼠肝组织的脂质过氧化.结论:EGb具有诱导抗氧化酶表达及直接清除自由基的双重抗氧化作用.  相似文献

 目的 研究炒紫苏子提取物的抗氧化作用。方法 15月龄昆明种小鼠,随机分组,每日1次灌胃炒紫苏子醇提药物和水提药物,给药21次后,杀鼠取脑和血。采用MDA试剂盒、SOD试剂盒,测定血浆中MDA,SOD值,采用紫外分光光度法测小鼠脑匀浆中MAO。结果 炒紫苏子醇提物269,134 mg·kg-1剂量组均能显著降低MDA水平(P<0.001),提高SOD酶活性(P<0.05),而134 mg·kg-1剂量组能显著降低MAO水平(P<0.05);炒紫苏子水提物213 mg·kg-1剂量组均能显著降低MDA,MAO水平(P<0.001,P<0.01),提高SOD酶活性(P<0.05),而106 mg·kg-1剂量组能显著降低MDA水平(P<0.001),MAO水平(P(0.05)。结论 炒紫苏子醇提取物和水提取物具有较强的抗氧化作用。  相似文献

目的:探讨金鸡菊提取物体外抗氧化活性。方法:采用分光光度法,测定金鸡菊提取物对羟基(OH)自由基、超氧阴离子(O 2-.)自由基及1,1-二苯基-2-苦肼基自由基(DPPH)的清除能力,通过多元线性回归分析,考察金鸡菊提取物含量及其体外抗氧化活性的相关性,并同维生素(VC)进行了比较。结果:金鸡菊提取物对.OH自由基,O 2-.自由基,DPPH自由基均有较好的清除作用,在供试质量浓度范围内(0.2~1.0 g.L-1)对各自由基的清除效率与金鸡菊提取物浓度有一定量效关系,当质量浓度达1.0 g.L-1,其中50%乙醇洗脱物对OH自由基、O 2-.自由基和DPPH自由基的清除率可达86.19%,97.90%和71.85%,且清除能力与Vc相当。结论:金鸡菊提取物具有显著的体外抗氧化作用。  相似文献

目的分析半枝莲乙醇提取物及不同极性溶剂萃取后体外抗氧化活性。方法乙醇提取半枝莲黄酮类化合物,以石油醚、乙酸乙脂、正丁醇分别萃取得各部分化合物,200~750 nm吸收光谱扫描,进行ABTS+、羟自由基、DPPH自由基清除实验。结果半枝莲乙醇提取物具有明显抗ABTS+、羟自由基、DPPH自由基活性,不同极性溶剂萃取半枝莲黄酮对其抗氧化能力有不同程度影响。结论半枝莲乙醇提取物具有明显抗氧化活性,分类萃取可降低其抗氧化活性。  相似文献

目的 考察沙煲暗罗根乙醇提取物不同萃取物的抗氧化和抗菌活性.方法 研究沙煲暗罗根乙醇提取物不同萃取物对羟基自由基和DPPH的抗氧化效果,并与Vc对比,找出IC50值;同时研究其抑菌效果,测出最低抑菌浓度.结果 乙酸乙酯萃取物对羟基自由基和DPPH的清除效果最好,IC50值分别为110.8μg/mL和156.5 μg/mL;三氯甲烷萃取物有最好的抑菌效果,对大肠杆菌和枯草芽孢杆菌最小抑菌质量浓度均为20 mg/mL,对金黄色葡萄球菌的最小抑菌质量浓度为40 mg/mL.结论 沙煲暗罗根乙醇提取物中乙酸乙酯萃取物有较好的抗氧化,三氯甲烷萃取物有较好的抑菌活性.  相似文献

用月见草油给小鼠灌胃给药2、4m1/kg,可明显抑制乙醇诱导的小鼠肝脏脂质过氧化作用;显著增强正常小鼠血中过氧化氢酶的活力;月见草油的抗氧化作用与其所含大量不饱和脂肪酸有关。这一研究为进一步阐明月见草油的广泛药理作用的机理提供了实验基础。  相似文献

目的:探讨酢浆草提取物体外抗氧化活性。方法:采用DPPH法、FRAP法分别测定酢浆草水提取物清除DPPH自由基和总的抗氧化能力。结果:在实验的测定浓度范围之内,随着提取物浓度的增加,其对DPPH自由基的清除能力逐渐增强。2mg/mL酢浆草水提物对DPPH自由基的清除率达到67.144%。FRAP法测得20mg/mL酢浆草水提取液的总抗氧化能力为1.09mM。结论:酢浆草对DPPH自由基的清除效果良好,FRAP法测得酢浆草有一定的抗氧化能力,有进一步开发利用的价值。  相似文献

目的研究野甘草乙醇提取物对胃溃疡的治疗作用。方法通过醋酸烧灼法制成大鼠胃溃疡模型,按照同窝同性、体质量配对分为2组,实验组每只以野甘草乙醇提取物1 g/(kg.d)喂养,共喂30 d;对照组不喂药。30 d后剖腹取胃,测定溃疡体积。结果实验组大鼠胃溃疡体积小于对照组(P<0.05)。结论野甘草乙醇提取物对大鼠胃溃疡有治疗作用,应对其药理作用进行更深入的研究。  相似文献

Effect of garlic oil on ethanol induced gastric ulcers in rats   总被引:2,自引:0,他引:2  
Garlic oil was evaluated for gastroprotective activity against ethanol induced ulcers. Reactive oxygen species are involved in the pathogenesis of these ulcers. The possible involvement of garlic oil in restraining the oxidation process produced in gastric tissue was also investigated. The ulcer index, lipid peroxidation and antioxidant enzyme activity (GPx, catalase, SOD) were determined. Pretreatment with garlic oil in doses of 0.25 and 0.5 mg/kg, 30 min before administration of ethanol (1 mL of 100%) caused a decrease in ulcer index and lipid peroxidation and ameliorated the decrease in antioxidant enzyme levels caused by ethanol. The result suggests that garlic oil possesses antioxidant properties and provides protection against ethanol induced gastric injury.  相似文献

目的:探索连翘花醇提物(FFE)对自由基所致的线粒体损伤的保护作用,并研究其抗氧化能力、超氧阴离子的清除活性及抗衰老机制.方法:采用硫代巴比妥酸(TBA)比色法测定丙二醛(MDA)含量、分光光度法测线粒体的肿胀度,并以还原型辅酶Ⅰ-氮蓝四唑-吩嗪硫酸甲酯(NADH-NBT-PMS)为超氧阴离子生成系统测定对超氧阴离子的清除能力.小鼠颈背部皮下注射D-半乳糖(D-gal)100 mg/(kg·d),连续给药7周造衰老模型,利用钼酸胺比色法、黄嘌呤氧化酶法、二硫代二硝基苯甲酸比色法、Fenton反应显色法分别测定FFE对小鼠肝匀浆过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及抗羟自由基活力的影响.结果:FFE可明显抑制肝匀浆及线粒体的脂质过氧化、线粒体肿胀,并呈剂量依赖关系,清除超氧阴离子的能力显著,且能明显增强CAT、SOD、GSH.Px及抗羟自由基活力.结论:FFE能通过清除活性氧自由基及提高抗氧化酶活力来保护线粒体,具有一定的抗氧化及抗衰老药用价值.  相似文献

The antibacterial and antioxidant activity of the ethanol extract from Paullinia cupana var. sorbilis Mart. (Sapindaceae) seeds, commonly called guarana, was assessed towards selected bacteria as well as in different antioxidant models. The extract, at a concentration between 16 and 128 microg/ml, showed a significant antibacterial effect expressed as minimum inhibitory concentration (MIC) against both Gram-negative and Gram-positive bacteria. In particular, Pseudomonas aeruginosa (MIC=16 microg/ml), Proteus mirabilis (MIC=32 microg/ml), Proteus vulgaris (MIC=32 microg/ml) and Escherichia coli (MIC=32 microg/ml) were the most inhibited. The antioxidant activity was determined by the malonyldialdehyde (MDA) test, measuring the MDA concentration in 3T3-L1 cells after induced cellular damage using ferric ammonium citrate (FAC). The reduction of lipid peroxidation was 62.5% using a guarana extract with a concentration of 2 microg/ml. This effect was dose/dependent. The ethanol extract from Paullinia cupana seeds was analysed by spectrophotometry to determine the concentration of catechol substances after treatment of the extract with p-aminophenol. The total phenolics content in the ethanol extract was also determined spectrophotometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE). The concentration of catechol equivalent was 6.06+/-0.13 mg/g (mean+/-S.D.), while the total phenolic content was 8.43+/-0.21 mg/g (mean+/-S.D.). The correlation index between antioxidant activity and catechol content was 0.96.  相似文献

目的:研究青龙衣的冷、热乙醇提取物对H22小鼠肿瘤细胞膜蛋白含量、膜脂流动性、膜封闭度的影响。方法:应用SDSPAGE电泳分析膜蛋白含量,按Skinitzky法测定膜脂流动性,按Zamudio法测定膜封闭度。结果:青龙衣的冷、热乙醇提取物可降低肿瘤细胞膜蛋白含量,降低肿瘤细胞膜脂流动性,降低肿瘤细胞膜封闭度。结论:青龙衣冷、热乙醇提取物改变了肿瘤细胞膜的生化物质及生化功能,其结果导致肿瘤细胞的解体和死亡,这可能是青龙衣抗肿瘤作用的机制之一。  相似文献

The aim of this paper is to evaluate the effect of green tea on the erythrocyte antioxidant system of ethanol‐intoxicated rats, as well as its efficacy in the prevention of lipid peroxidation. Rats (2, 12 and 24 months old) were fed on a control or an ethanol Lieber‐DeCarli diet with and without green tea (7 g/L) for 5 weeks. Examination included the activity of antioxidant enzymes and the level of both non‐enzymatic antioxidants and lipid peroxidation marker in rat erythrocytes. It was shown that ageing was accompanied by changes in the antioxidant enzymes activity – increase in the SOD and CAT activity and decrease in GSSG‐R and GSH‐Px activity, as well as in the level of non‐enzymatic antioxidants – GSH, vitamin A and vitamin E. The increase in the level of lipid peroxidation marker – MDA – was also observed. Green tea consumption partially prevented lipid peroxidation process, especially in erythrocytes of 2‐ and 12‐month‐old rats. It was proved that ethanol administration caused a statistically significant decrease in the activity/level of the examined antioxidants in all age groups (the most significant in the case of 24‐month‐old rats) of rats, as well as an increase in the MDA level. However, ingestion of green tea by ethanol‐intoxicated rats partially prevented the decrease in activity/level of all examined antioxidant parameters, as well as protected lipids against peroxidation in all age groups of rats.  目的研究中草药有效成分氯原酸、连翘酯苷对活性氧(HO·,H2O2,O2)的清除作用及结构与活性的关系。方法邻菲罗啉、鲁米诺化学发光、NBT法以及量子化学计算[PM3,HF/6-31G(d)]。结果氯原酸、连翘酯苷对活性氧具有较强的清除能力;连翘酯苷结构中的A环酚羟基和B环酚羟基对活性氧的清除能力相当。结论连翘酯苷是一种有效的天然抗氧化剂。  相似文献

该文采用乙醇分级沉淀法提取不同浓度乙醇层的黄芪多糖,并探讨黄芪多糖相对分子质量、构效及活性之间的关系。采用不同浓度的乙醇(30%,50%,70%,75%,80%,90%)沉淀,以获得不同相对分子质量的黄芪多糖;采用红外及电子扫描显微镜观察各层多糖的形态特征;采用DPPH自由基清除率,还原力和β-胡萝卜素/亚油酸体系检测各层的抗氧化活性。结果显示该法所得黄芪多糖随乙醇浓度增加,相对分子质量降低,抗氧化活性增强,90%乙醇沉淀层多糖表面光滑,相对分子质量较小,三股螺旋链构象特异,抗氧化活性最强(P<0.05)。  相似文献

The present work was done to investigate the possible effects of Nigella sativa oil (NSO) on gastric secretion and ethanol-induced ulcer in rats. Thirty two adult male rats were used in this study (four groups) and several parameters were determined to assess any degree of protection. It was found that the administration of NSO in rats produced a significant increase in mucin content and glutathione level and a significant decrease in mucosal histamine content. Ethanol administration produced a 100% ulcer induction with an ulcer score of 12.62+/-1.35 (mean+/-S.E., n=8). It caused a significant reduction in free acidity and glutathione level while it produced a significant increase in mucosal histamine content. When animals were pretreated with NSO before induction of ulcer, there was a significant increase in glutathione level, mucin content and free acidity and a significant decrease in gastric mucosal histamine content with a protection ratio of 53.56% as compared to the ethanol group. It can be concluded that NSO imparted a protective action against ethanol induced ulcer in rats.  相似文献

Ethanol is one of the most widely used and abused drugs, increasing lipid levels in humans and experimental animals. Heating of oil rich in polyunsaturated fatty acids (PUFA) produces various lipid peroxidative end products that can aggravate the pathological changes produced by ethanol. In the present communication, the effect of Cuminum cyminum was investigated on alcohol and thermally oxidized oil induced hyperlipidaemia. The results showed increased activity of aspartate transaminase (AST), alkaline phosphatase (ALP) and gamma glutamyl transferase (GGT) and increased levels of cholesterol, triglycerides and phospholipids in the plasma of rats given alcohol, thermally oxidized oil and alcohol+thermally oxidized oil when compared with the normal control group. The levels of tissue (liver and kidney) cholesterol and triglycerides were increased significantly in rats groups given alcohol, thermally oxidized oil and alcohol+thermally oxidized oil when compared with the normal control rats. The levels were decreased when cumin was given along with alcohol and thermally oxidized oil. The level of phospholipids decreased significantly in the liver and kidney of groups given alcohol, thermally oxidized oil and alcohol+thermally oridized oil when compared with the normal control rats. The level increased when cumin was administered along with alcohol and thermally oxidized oil. The activity of phospholipase A and C increased significantly in the liver of groups given alcohol, thermally oxidized oil and alcohol+thermally oxidized oil when compared with the normal control rats, whereas the activity was decreased with the cumin treatment. The results obtained indicate that cumin can decrease the lipid levels in alcohol and thermally oxidized oil induced hepatotoxicity.  相似文献
