Extract of Ehrlich's ascites tumor cells (chalone) and its cell-free fluid have a marked inhibitory action on proliferation of these tumor cells 4 h after injection. The effect is tissue specific, it is more marked in the extract, and it depends on the dose of the agent. Mitotic activity in the tumor 8 h after injection of the extract or cell-free fluid is higher than in the control, evidence of a short-term effect of the chalone on the G2 phase of the mitotic cycle and on synchronization of cell division.Department of General Biology and Genetics, Medico-Biological Faculty, N. Chalone from Ehrlich's ascites tumor has a short and reversible inhibitory action on proliferation of cells of this tumor when administered as both one and two injections. A considerable increase in mitotic activity in the tumor cells compared with the control is observed 10 h after one or two injections (the second injection being given at 6 p.m.) of the chalone, indicating that it acts on the G2-cell population in the mitotic cycle and that it synchronizes cell division. If the second of two injections of chalone is given at 9 p.m. it leads to more prolonged inhibition of the cells and to a more marked synchronization wave of the G2-cell population. The duration of inhibition of cells in the G2-phase of the mitotic cycle after two injections of the chalone thus depends on the state of the cell population on which the chalone acts.Department of Biology and Genetics, Medico-Biological Faculty, N. Hyperdiploid or hypotetraploid strains of Ehrlich's ascites tumor were transplanted into noninbred male albino mice. The results observed on the 4th, 5th, and 6th days of development of the Ehrlich's tumor show fluctuations in the mitotic index both in the hypotetraploid strain, under natural lighting conditions, and in the hyperdiploid strain with a regular 12-h photoperiod. Maximal values of the mitotic index were observed in the morning and afternoon. The results indicate that fluctuations in the mitotic index and labeling index during the 24-h period are not synchronized in different strains and under different conditions of illumination. It is concluded that the temporal organization of cell reproduction in Ehrlich's ascites tumor cells differs from that in normal tissue cells.Department of Biology and Genetics, Medico-Biological Faculty, N. Diurnal changes in the number of normal and colcemid mitoses were studied in Ehrlich's ascites carcinoma cells in mice receiving colcemid after preliminary partial synchronization of cell division produced by injection of dibutyryl cyclic AMP and also in animals after injection of colcemid or of dibutyryl cyclic AMP alone. Synchronization of cell division in the tumor was shown to increase by 2.6 times the number of tumor cells blocked by colcemid and to lead to more rapid arrest of colcemid mitoses.Department of Biology, N. Intraperitoneal injection of various interferons (native cultural, concentrated and partially purified cultural, and also serum) significantly inhibits the development of Ehrlich's ascites carcinoma in noninbred mice. Because of the comparatively low activity of serum interferon, the effect of normal mouse serum on the development of this tumor and its action on the effect of native cultural interferon were investigated. Normal mouse serum was shown to depress the action of cultural interferon and to accelerate the development of Ehrlich's ascites carcinoma.N. F.
