Male adult rats were fed on diets containing 80 g/kg galactomannans with different galactose (G): mannose (M) ratios/kg. The galactomannans were compared with purified cellulose (Solkaflok) and the animal were also fed on a basal diet free from fibre. All diets contained cholesterol (10 g/kg) and sodium cholate (2 g/kg). The three galactomannans were fenugreek gum (1G:1M), guar gum (1G:2M) and locust-bean gum (1G:4M). In comparison with the fibre-free and Solkaflok diets, all three galactomannans lowered the concentrations of cholesterol in both liver and blood plasma. The galactomannans also decreased the rate of hepatic synthesis of cholesterol. Dietary galactomannans increased caecal volatile fatty acids, particularly propionic, increased the weight of the caecum and its contents and increased the amount of water in the faeces. The increase in propionic acid production was significantly related to a decrease in caecal pH, but not to changes in plasma cholesterol or hepatic cholesterol synthesis. These effects were significantly influenced by chemical composition and structure of the galactomannan; they were most evident when the proportion of galactose in the galactomannan was highest (i.e. fenugreek gum). The three galactomannans also differed markedly in their effects on the viscosity of the digesta, but the galactomannan which gave the highest viscosity was least effective in lowering plasma cholesterol. A separate experiment with perfused loops of small intestine in vivo showed that the most effective galactomannan, fenugreek gum, had no direct effect on cholesterol absorption.  相似文献

The role of dietary cholesterol in people with diabetes has been little studied. We investigated the effect of a hypoenergetic high-protein high-cholesterol (HPHchol) diet compared to a similar amount of animal protein (high-protein low-cholesterol, HPLchol) on plasma lipids, glycaemic control and cardiovascular risk markers in individuals with type 2 diabetes. A total of sixty-five participants with type 2 diabetes or impaired glucose tolerance (age 54·4 (sd 8·2)?years; BMI 34·1 (sd 4·8)?kg/m2; LDL-cholesterol (LDL-C) 2·67 (sd 0·10)?mmol/l) were randomised to either HPHchol or HPLchol. Both hypoenergetic dietary interventions (6-7?MJ; 1·4-1·7 Mcal) and total carbohydrate:protein:fat ratio of 40:30:30?% were similar but differed in cholesterol content (HPHchol, 590?mg cholesterol; HPLchol, 213?mg cholesterol). The present study was undertaken to evaluate the lipid-lowering effect of fenugreek leaves in diabetes mellitus. Albino Wistar rats were randomly divided into six groups: normal untreated rats; streptozotocin (STZ)-induced diabetic rats; STZ-induced rats + fenugreek leaves (0.5 g/kg of body weight); STZ-induced rats + fenugreek leaves (1 g/kg of body weight); STZ-induced rats + glibenclamide (600 microg/kg of body weight); and STZ-induced rats + insulin (6 units/kg of body weight). Rats were made diabetic by STZ (40 mg/kg) injected intraperitoneally. Fenugreek leaves were supplemented in the diet daily to diabetic rats for 45 days, and food intake was recorded daily. Blood glucose, total cholesterol, triglycerides, and free fatty acids were determined in serum, liver, heart, and kidney. Our results show that blood glucose and serum and tissue lipids were elevated in STZ-induced diabetic rats. Supplementation of fenugreek leaves lowered the lipid profile in STZ-induced diabetic rats.  相似文献

The mechanisms implicated in the LDL-cholesterol (LDL-C)-lowering effects of the Mediterranean-type diet (MedDiet) are unknown. The present study assessed the impact of the MedDiet consumed under controlled feeding conditions, with and without weight loss, on surrogate markers of cholesterol absorption, synthesis and clearance using plasma phytosterols, lathosterol and proprotein convertase subtilisin/kexin-9 (PCSK9) concentrations, respectively, in men with the metabolic syndrome. The subjects' diet (n 19, 24-62 years) was first standardised to a baseline North American control diet (5 weeks) followed by a MedDiet (5 weeks), both under weight-maintaining isoenergetic feeding conditions. Soyfoods contain estrogenic isoflavones--namely, genistein (G) and daidzein (D)--that, like estrogens, display physiological effects in humans and animals. Previously we and others have demonstrated antioxidant and cardioprotective effects of orally ingested soy diets and soy isoflavones. The overall objective of this study was to test the effects of injected soy isoflavones, G and D, on liver lipids, liver free radical scavenger systems, and parameters of cardiovascular risk. Forty male rats were injected with G, D, estradiol (E), or a vehicle control (V) for 6 weeks. At the end of the study, body weight, food intake, feed efficiency ratio (FER), plasma glucose and cholesterol, abdominal fat pad weight, reproductive organ weight, liver weight, liver lipids, and liver free radical scavenger systems were compared. Food intake was significantly (P < .04) higher in the D-, G-, and E-treated animals compared with V-treated animals. FER was lower (P < .001) in D-, G-, and E-treated animals compared with the V- and the E-treated animals. Body weight, testis weight, and prostate weight were markedly (P < .001) lower in the E-treated animals compared with D-, G-, and V-treated animals. Intraabdominal fat pad weights were also significantly (P < .001) lower in the E group, although this effect was lost when corrected for body weight. Liver weights were considerably lower in the D-, G-, and E-treated animals versus the V group (P < .001). Total plasma cholesterol was reduced (P < .05) in D- and E-treated animals versus the V group. Liver lipids appeared to be unchanged by the isoflavones and slightly elevated by E treatment (P < .02). Liver catalase levels were numerically higher in the D- and E-treated animals compared with the V group (P < .1). Similarly, Cu/Zn superoxide dismutase (SOD) activity was significantly elevated in the D and E groups (P < .01), while G treatment (P < .03) elevated SOD to a lesser degree, versus the V group. These results suggest that subcutaneous injections of the naturally occurring soy isoflavone D and, to a lesser extent, G exert cardioprotective effects and stimulate antioxidant systems, while minimizing the undesirable effects elicited by E treatment.  相似文献

BACKGROUND: National guidelines recommend consideration of step down or withdrawal of medication in patients with well-controlled hypertension, but knowledge of factors that predict or mediate success in achieving this goal is limited. OBJECTIVE: To identify patient characteristics associated with success in controlling blood pressure (BP) after withdrawal of antihypertensive medication. DESIGN: The Trial of Nonpharmacologic Interventions in the Elderly tested whether lifestyle interventions designed to promote weight loss or a reduced intake of sodium, alone or in combination, provided satisfactory BP control among elderly patients (aged 60-80 years) with hypertension after withdrawal from antihypertensive drug therapy. Participants were observed for 15 to 36 months after attempted drug withdrawal. MAIN OUTCOME MEASURES: Trial end points were defined by (1) a sustained BP of 150/90 mm Hg or higher, (2) a clinical cardiovascular event, or (3) a decision by participants or their personal physicians to resume BP medication. RESULTS: Proportional hazards regression analyses indicated that the hazard (+/- SE) of experiencing an end point among persons assigned to active interventions was 75% +/- 9% (weight loss), 68% +/- 7% (sodium reduction), and 55% +/- 7% (combined weight loss/sodium reduction) that of the hazard for those assigned to usual care. Lower baseline systolic BP (P < .001), fewer years since diagnosis of hypertension (P < .001), fewer years of antihypertensive treatment (P < .001), and no history of cardiovascular disease (P = .01) were important predictors of maintaining successful nonpharmacological BP control throughout follow-up, based on logistic regression analysis. Age, ethnicity, baseline level of physical activity baseline weight, medication class, smoking status, and alcohol intake were not statistically significant predictors. During follow-up, the extent of weight loss (P = .001) and urinary sodium excretion (P = .04) were associated with a reduction in the risk of trial end points in a graded fashion. CONCLUSIONS: Withdrawal from antihypertensive medication is most likely to be successful in patients with well-controlled hypertension who have been recently (within 5 years) diagnosed or treated, and who adhere to life-style interventions involving weight loss and sodium reduction. More than 80% of these patients may have success in medication withdrawal for longer than 1 year.  相似文献

This study was designed to investigate the possible beneficial effects of consuming a sodium-rich carbonated mineral water on lipoprotein metabolism and to determine whether consumption of this water influences endothelial dysfunction (ED) in postmenopausal women. Women included in the study were amenorrheic (>1 y), healthy, and not obese (BMI < 30 kg/m(2)). The subjects did not take estrogen replacement therapy; supplements of vitamins, minerals, and phytoestrogens; or other medications known to affect bone and lipid metabolism. The study consisted of 2 intervention periods of 2 mo each, during which women drank 1 L/d of a control mineral water (low mineral content) for 2 mo followed by the carbonated mineral water, rich in sodium, bicarbonate, and chloride, for 2 mo. Body weight, height, and blood pressure were measured, and BMI was calculated. Blood samples were taken from fasting subjects and serum was analyzed for total cholesterol, HDL-cholesterol, LDL-cholesterol, triacylglycerols, apolipoprotein AI, apolipoprotein B, soluble intercellular cell adhesion molecule-1 (sICAM-1), soluble vascular cell adhesion molecule-1 (sVCAM-1), and glucose. Blood pressure levels did not change throughout the study. Carbonated water intake decreased total cholesterol and LDL-cholesterol levels by 6.8% (P = 0.001) and 14.8% (P < 0.0001), respectively, whereas HDL-cholesterol concentration increased by 8.7% (P = 0.018), compared to the control period. Therefore, cardiovascular disease (CVD) risk indexes (total cholesterol/HDL-cholesterol and LDL-cholesterol/HDL-cholesterol) were markedly reduced (both P < 0.0001). Soluble ICAM-1 and sVCAM-1 levels decreased by 8.4% (P = 0.007) and 14.8% (P = 0.015), respectively. Fasting serum glucose concentration decreased by 6.7% (P < 0.0001). Triacylglycerol levels did not change. Consumption of this sodium rich carbonated water can play a beneficial role in the prevention of CVD and the metabolic syndrome.  相似文献

The effects of naringenin (Nar) on hepatic neutral and polar lipids were examined in rats that were fed either a high (27.98%) cornstarch (CornS) or a high (19.05%) coconut oil (CocoO) diet, with or without supplementation with 0.60% NAR, for 40 days. Liver weights and total, neutral, and polar lipids were significantly increased following the feeding of CocoO (P <.01) when compared with the CornS-fed rats. Furthermore, significant elevations in liver weight and total neutral lipids were observed (P <.01) with the CornS-Nar compared with the CornS diet; cholesterol esters (CE), CE:cholesterol (CE:C) ratio, triglycerides (TG), and phosphatidylethanolamine (PE) were significantly increased (P <.01). When Nar was added to the CocoO diet, liver protein, total lipids, and total neutral and polar lipids were significantly decreased (P <.01). Of the neutral lipids, CE, CE:C ratio, TG, free fatty acids (FFA), mono- and diglycerides (P <.01), and C (P <.05) were significantly decreased. CE and TG were more significantly decreased than C, while FFA decreased most markedly (P <.01). Of the hepatic polar lipids, lysophosphatidylcholine, sphingomyelin, phosphatidylcholine, phosphatidylserine and -inositol, PE, and phosphatidic acid were significantly elevated (P <.001) following the feeding of the high CocoO diet compared with the high CornS diet. When Nar was added to the CornS diet, PE was significantly elevated (P <.01), and the outer membrane to inner membrane lipids ratio was significantly decreased (P <.01). When Nar was added to the CocoO diet, all of the polar lipids examined were significantly decreased when compared with the CocoO diet (P <.01). Dietary Nar supplementation significantly reduced both hepatic neutral and polar lipids in rats fed a high CocoO diet, but not in the CornS-fed rats.  相似文献

Abstract Acute and chronic inflammation and dyslipidemia play a critical role in the development of various diseases, including cardiovascular disease. Green tea polyphenols possess potent antioxidative and anti-inflammatory properties that contribute to the beneficial effects on heart health. The present study was carried out to determine if administration of a green tea extract (Polyphenon(?) E [PPE]; Mitsui Norin Co., Ltd., Tokyo, Japan) at 0.2% in the diet reduces cardiovascular risk factors, including dyslipidemia, inflammation, adiposity, and oxidative stress, in rats fed an atherogenic (high fat, cholesterol, and sugar) diet with dextran sodium sulfate (DSS) in drinking water. DSS treatment increased serum total cholesterol, low-density lipoprotein (LDL)-cholesterol, C-reactive proteins (CRP), and markers of liver toxicity and decreased high-density lipoprotein (HDL)-cholesterol significantly. Adding PPE to the atherogenic diet (PPE-diet) was associated with lower total cholesterol and LDL-cholesterol (P<.001) and increased HDL-cholesterol (P=.001). In addition, the PPE-diet was associated with decreased serum CRP concentration (P=.023) and increased total antioxidant capacity (P=.016) and catalase (P=.001) and glutathione peroxidase (P=.050) activities. The PPE-diet significantly lowered epididymal fat pad weight (P=.009). Feeding the PPE-diet also ameliorated some of the DSS-induced lipid, inflammatory, and oxidative symptoms. In summary, green tea supplementation decreased several cardiovascular risk factors, including body composition, dyslipidemia, inflammatory status, and antioxidant capacity, in rats fed an atherogenic diet. This study supports green tea as an effective dietary component for sustaining cardiovascular health.  相似文献

OBJECTIVE: To assess the impact of increased consumption of milk, without other dietary advice, on older adults' energy and nutrient intakes, weight, cardiovascular risk factors (blood pressure, plasma lipid levels), and quality of life. SUBJECTS/SETTING: Two hundred four healthy men and women, aged 55 to 85 years, who consumed fewer than 1.5 dairy servings per day were chosen from six US academic health centers. DESIGN: Randomized, controlled open trial. INTERVENTION: Advice to increase skim or 1% milk intake by 3 cups per day (n = 101) or to maintain usual diet (n = 103) for 12 weeks after a 4-week baseline period. MAIN OUTCOME MEASURES: Changes in energy and nutrient intake assessed from 3-day food records, body weight, blood pressure, and plasma lipid levels. STATISTICAL ANALYSES PERFORMED: Group-by-time analysis of variance with repeated-measures, chi 2 test. RESULTS: Compliance with the intervention was good. Compared with controls, participants in the milk-supplemented group significantly increased energy, protein, cholesterol, vitamins A, D, and B-12, riboflavin, pantothenate, calcium, phosphorus, magnesium, zinc, and potassium intakes. Prevalence of nutrient inadequacy, assessed for nutrients with Estimated Average Requirements, decreased among women in the milk group for magnesium (40% at baseline vs 13% at 12 weeks, P < .001) and vitamin B-12 (6% vs 0%, P < .05) and tended to decrease (P < .10) for protein and thiamin (women) and magnesium and vitamin B-6 (men). The milk group gained 0.6 kg more than control group (P < .01); however, weight gain was less than predicted, which suggests some compensation for the added energy from milk. Blood pressure decreased similarly over time in both groups. Total and low-density lipoprotein cholesterol levels, and the ratio of total cholesterol to high-density lipoprotein cholesterol, were unchanged. Triglyceride levels increased within the normal range in the milk group (P = .002). Quality of life scores were high at baseline and remained high throughout. APPLICATIONS/CONCLUSIONS: Older adults can successfully increase milk intake, thereby meaningfully improving their nutrient intakes. Dietitians can play a key role in disseminating this advice.  相似文献

OBJECTIVE: To examine the relationship between lipid values and BMI (body mass index) on hospitalizations in hemodialysis (HD) patients. DESIGN: Retrospective (2-year) study. SETTING: Outpatient dialysis center in a large metropolitan city. PATIENTS: This study used 158 HD patients stratified on the basis of ethnicity (non-Black and Black) and diabetic status (nondiabetic and diabetic). INTERVENTION: Subjects were observed for 2 years. Body weight, BMI, lipid parameters, and hospitalization duration were determined 8 times (3-month intervals). MAIN OUTCOME MEASUREs: Body weight, BMI, lipid parameters (serum triglyceride concentration, serum total cholesterol, high-density lipoprotein [HDL]-, low-density lipoprotein [LDL]-, very low-density lipoprotein [VLDL]- cholesterol concentrations, serum Apo-protein A1 [Apo-A1] concentration, and serum Apo-protein B [Apo-B] concentration), and morbidity data were recorded. RESULTS: Hemodialysis subjects were hospitalized 2.3 +/- 1.6 times over the 2-year experimental period. Length of hospital stay averaged 6.6 +/- 0.5 days/hospitalization. Length of hospital stay was inversely related to HDL concentration (r = -0.21, P <.05, n = 89), but not significantly related to BMI in HD subjects. BMI was positively associated with LDL concentration (r = +0.28, P <.01, n = 97). Cholesterol concentration was directly associated with LDL concentration (r = +0.52, P <.01, n = 138), VLDL concentration (r = +0.47, P <.01, n = 139), and triglyceride concentration (r = +0.54, P <.01, n = 155). Mean concentration of HDL-cholesterol was inversely related serum triglyceride concentration (r = -0.43, P <.01, n = 140). Although Apo-A1 concentration was directly associated with HDL level (r = +0.39, P <.01, n = 139), Apo-B was inversely related to HDL level (r = -0.37, P <.01, n = 138) and directly related to cholesterol concentration (r = +0.71, P <.01, n = 138), VLDL concentration (r = +0.87, P <.01, n = 138), and triglyceride concentration (r = +0.81, P <.01, n = 138). CONCLUSION: Cardiac disease remains the primary cause of morbidity and mortality in HD patients, and results of the present study suggest that dyslipidemias present in the HD population negatively impact cardiovascular profiles which, in turn, influence the frequency/duration of hospitalizations. Among all lipid parameters analyzed in the present study, increased LDL and decreased HDL concentrations were more strongly related to length of hospital stay than was BMI.  相似文献

The role of bicarbonated mineral waters on lipid metabolism and lipoprotein concentrations in man has scarcely been investigated. The present study aimed to investigate whether drinking sodium bicarbonated mineral water affects postprandial cholesterol and triacylglycerol metabolism in postmenopausal women. In a three-way, randomised, crossover study, eighteen healthy postmenopausal women consumed two sodium bicarbonated mineral waters (bicarbonated mineral water 1 and bicarbonated mineral water 2) and a low mineral water (500 ml of each) with a standard fat-rich meal (4552 kJ; 75.3 g fat). The bicarbonated waters were rich in sodium and bicarbonate and bicarbonated mineral water 1 contained 5.7 times more fluoride than bicarbonated mineral water 2. Fasting blood samples and postprandial blood samples were taken at 30, 60, 120, 240, 360 and 420 min after the end of the meal consumption. Cholesterol and triacylglycerols were determined in serum and chylomicrons. A significant water consumption effect was observed in the total area under the curve (TAUC) of serum and chylomicron triacylglycerols (ANOVA, P=0.008 and P=0.027, respectively). TAUC of serum triacylglycerols for bicarbonated mineral water 2 was significantly lower compared to low mineral water (Bonferroni, P=0.039). Peak concentration of serum triacylglycerols showed a significant water effect (P=0.025). Changes in chylomicron cholesterol were not significantly affected by the type of water. Bicarbonated mineral waters 1 and 2 did not show any significant differences. Drinking sodium bicarbonate-rich mineral waters reduces postprandial lipaemia in healthy postmenopausal women compared to drinking a low mineral water.  相似文献

Telfairia occidentalis (fluted pumpkin) is one of the commonly consumed leafy vegetables in Nigeria. In order to justify its inclusion in herbal preparations in African traditional medicine, the possible hypolipidemic effect of this vegetable was investigated in rats fed a cholesterol-rich diet. The ability of Questran, a hypolipidemic drug, to attenuate hypercholesterolemia was also examined. Rats were fed with either a basal diet containing cholic acid (0.2%) or a supplemented diet with T. occidentalis at the 3% and 6% levels. Oral administration of cholesterol for 9 consecutive weeks resulted in a significant increase (P < .001) in the relative weight of the heart of cholesterol-fed rats. However, supplemented diets significantly (P < .001) ameliorated the cholesterol-induced enlargement of the heart. Rats fed on supplemented diets had a dose-dependent reduction in plasma and postmitochondrial supernatant fraction (PMF) cholesterol levels. In particular, supplemented diets containing 3% and 6% T. occidentalis decreased plasma and PMF cholesterol levels by 20% and 30% and by 30% and 45%, respectively. A similar decrease in plasma and PMF cholesterol levels was obtained in Questran-treated hypercholesterolemic rats. Furthermore, 3% and 6% T. occidentalis-supplemented diets significantly (P < .05) decreased the cholesterol-induced increase in plasma and PMF low-density lipoprotein (LDL) cholesterol levels by 24% and 48% and by 28% and 52%, respectively. In contrast, there was no significant difference (P > .05) in plasma and PMF triglyceride levels of rats fed on supplemented diets when compared with cholesterol-fed rats. There were significant decreases (P < .05) in lipid peroxidation levels in rats fed on the supplemented diets. Specifically, 3% and 6% T. occidentalis-supplemented diets decreased plasma and PMF lipid peroxidation by 24% and 20% and by 42% and 21%, respectively. This study demonstrates that T. occidentalis may be a useful therapy for hypercholesterolemia through reducing oxidative stress and cholesterol levels.  相似文献

Growing pigs were fed diets, similar in protein and fat content, based on soy protein isolate and soybean oil (soy) or ground beef containing 21% fat (beef). Ground corn, vitamins and minerals completed the diets. Egg yolk was added to balance the intake of cholesterol. No significant differences were seen in total plasma cholesterol or liver cholesterol concentrations. Soy-fed pigs had significantly greater concentrations of cholesterol in the viscera excluding the liver (p<.001), aorta (p<.04), carcass (p<.001) and whole body (liver + other viscera + carcass) (p<.001). No evidence of fatty streaking or plaque formation was seen in Sudan IV-stained aorta segments of either treatment group. Although pigs fed the soy diet gained weight at a slower rate, their carcasses contained a significantly greater (p<.02) concentration of fat.Our data show that the soy diet, as compared to the beef diet, increased the proportion of fat in the carcass and increased tissue cholesterol deposition.  相似文献

Ninety-six 14-d-old male broiler chicks were divided into three dietary groups and fed a corn-soybean meal diet, a barley diet with beta-glucanase and that diet without beta-glucanase. All diets contained 4 g cholesterol/kg. Average daily body weight gain, plasma total cholesterol concentration, LDL cholesterol concentration and digestibility of lipids and protein were lowest (P < 0.05) in the chicks fed the barley diet without beta-glucanase and highest (P < 0.05) in the chicks fed corn-soybean meal diet. Supplementation of the barley diet with beta-glucanase resulted in greater (P < 0.05) average daily weight gain, plasma total and LDL cholesterol concentrations and digestibility of lipids. Viscosity of small intestinal digesta was greatest in chicks fed barley, lowest in those fed the corn-soybean diet and intermediate in chicks fed enzyme-treated barley. Significant (P < 0.01) negative correlations occurred between viscosity of the small intestinal contents and average daily weight gain, plasma total and LDL cholesterol concentrations, and digestibility of lipids and protein. A lower concentration of insoluble beta-glucans in small intestinal digesta of the chicks fed barley supplemented with beta-glucanase compared with the chicks fed the unsupplemented barley diet reflects hydrolytic activity of the supplemental beta-glucanase in the diet.  相似文献

Fenugreek (Trifolium foenum graecum) a leguminous herb, is extensively cultivated in India, Mediterranean region and North Africa. It is used for culinary and medicinal purposes and also for fodder in various parts of the world. The hypoglycemic effect of fenugreek seeds and leaves was tested in normal and diabetic subjects. Six protocols A, B, C, D, E and F involved the acute administration (single dose of 25 g of seeds, 5 g of gum isolate and 150 g of leaves) of whole fenugreek seeds, defatted fenugreek seeds, gum isolate, degummed fenugreek seeds, cooked fenugreek seeds and cooked fenugreek leaves to healthy subjects. The rise in plasma glucose after a dose of glucose or meal was prevented by fenugreek seeds. The serum insulin levels were also modified (P<0.05). The reduction in area under glucose curve was greatest with whole seeds (42.4%), followed by gum isolate (37.5%), extracted seeds (36.9%), and cooked seeds (35.1%) in that order. The degummed seeds and fenugreek leaves showed little effect on glycemia. Fenugreek seeds were also administered for 21 days to diabetic subjects. The effects of active immunisation with gastric inhibitory polypeptide (GIP) or (proline3)GIP-ovalbumin conjugates on insulin resistance, metabolic dysfunction, energy expenditure and cognition were examined in high-fat-fed mice. Normal mice were injected (subcutaneously) once every 14 d for 98 d with GIP-ovalbumin conjugates, with transfer to a high-fat diet on day 21. Active immunisation resulted in GIP antibody generation and significantly (P < 0·01 to P < 0·001) reduced circulating non-fasting plasma insulin concentrations compared to high-fat control mice from day 70 onwards. The glycaemic responses to intraperitoneal glucose or nutrient ingestion were significantly improved in all treated mice, with corresponding stimulated plasma insulin levels depressed compared to high-fat controls. These changes were associated with substantially (P < 0·001) improved glucose-lowering responses to exogenous insulin and decreases of muscle and fat TAG, pancreatic insulin, circulating total and LDL-cholesterol levels (P < 0·01 to P < 0·001). Treatment with GIP-ovalbumin conjugates was not associated with alterations in energy expenditure, indirect calorimetry or aspects of cognitive function. The observed changes were almost identical in GIP and (Pro3)GIP immunised mice and were independent of any effects on food intake or body weight. Further tests established that coupling of GIP peptides to ovalbumin abolished any intrinsic insulin-releasing or glucose-lowering activity. These results suggest that induction of GIP-neutralising antibodies with GIP-ovalbumin conjugates is an effective means of countering the metabolic abnormalities induced by high-fat feeding and does not adversely have an impact on a marker of cognition function or energy expenditure.  相似文献

This study examined the effects of a pre-exercise meal and a carbohydrate-electrolyte solution on endurance running capacity. Ten men performed 3 treadmill runs at 70% VO2max to exhaustion after consuming (a) a carbohydrate meal 3 h before exercise and a carbohydrate-electrolyte solution during exercise (M + C); or (b) the carbohydrate meal 3 h before exercise and water during exercise (M + W); or (c) a liquid placebo 3 h before exercise and water during exercise (P + W). Exercise time was longer in M + C (125.1 +/- 5.3 min; mean +/- SE) and M + W (111.9 +/- 5.6 min) compared with P + W (102.9 +/- 7.9 min; p < .01 and p < .05, respectively), and longer in M + C compared with M + W (p < .05). Serum insulin concentration at the start of exercise and carbohydrate oxidation rates during the first hour of exercise were higher, whereas plasma FFA concentrations throughout exercise were lower in M + W and M + C than in P + W (p < .01). A carbohydrate meal before exercise at 70% VO2max improved endurance running capacity; however, the combination of the meal and a carbohydrate-electrolyte solution during exercise further improved endurance running capacity.  相似文献

We evaluated changes in low-density lipoprotein (LDL) receptor and 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase gene expression in women with metabolic syndrome and elevated plasma LDL cholesterol (LDL-C). We hypothesized that expression of these 2 genes would be modulated by our dietary intervention. Twenty-five women were instructed to follow a Mediterranean-style low-glycemic-load diet for 12 weeks. Quantitative real-time polymerase chain reaction was used to measure messenger RNA (mRNA) abundance of the LDL receptor and HMG-CoA reductase in mononuclear cells, which were used as a proxy of liver expression of these 2 genes. All women experienced favorable impacts on metabolic syndrome variables, with decreases in waist circumference (P < .001), plasma triglycerides (P < .05), and systolic blood pressure (P < .05) compared with baseline. Furthermore, participants had reductions in LDL-C (P < .01), plasma insulin (P < .001), and homeostatic model assessment score for insulin resistance (P < .001) over time. In addition, significant decreases were found in plasma tumor necrosis factor α (P < .01), which might have contributed to the improvements observed in insulin resistance. Although no changes in LDL-receptor mRNA levels were observed, HMG-CoA-reductase gene expression was reduced (P < .001) after 12 weeks. The reductions in plasma insulin correlated with changes in HMG-CoA-reductase mRNA levels (r = 0.45, P < .01). In conclusion, the observed reductions in plasma insulin may have affected the expression of a key regulatory gene of cholesterol synthesis, HMG-CoA reductase. The decreased HMG-CoA-reductase expression may be related to lower secretion of very low density lipoprotein (VLDL)-cholesterol, which, in turn, would account for the reductions in LDL-C.  相似文献

Carbohydrate-restricted diets (CRDs) have been shown to reduce body weight, whereas whole egg intake has been associated with increased satiety. The purpose of this study was to evaluate the effects of additional dietary cholesterol and protein provided by whole eggs while following a CRD on insulin resistance and appetite hormones. Using a randomized blind parallel design, subjects were allocated to an egg (640 mg/d additional dietary cholesterol) or placebo (0 mg/d additional dietary cholesterol) group for 12 weeks while following a CRD. There were significant reductions in fasting insulin (P < .025) and fasting leptin concentrations (P < .01) for both groups, which were correlated with the reductions in body weight and body fat (P < .05 and P < .01, respectively). Both groups reduced insulin resistance as measured by the homeostatic model assessment of insulin resistance (P < .025). There was a significant decrease in serum glucose levels observed after the intervention. We did not observe the expected increases in plasma ghrelin levels associated with weight loss, suggesting a mechanism by which subjects do not increase appetite with CRD. To confirm these results, the subjective measures of satiety using visual analog scale showed that both groups felt more “full” (P < .05), “satisfied” (P < .001), and “wanted to eat less” (P < .001) after the intervention. These results indicate that inclusion of eggs in the diet (additional dietary cholesterol) did not modify the multiple beneficial effects of CRD on insulin resistance and appetite hormones.  相似文献
