Some leafy vegetables were studied for their nutritional composition, antioxidant and free radical scavenging activities. The aerial parts of Coriandrum sativum, Spinacia oleracea, Trigonella corniculata and Trigonella foenum-graecum showed lower inhibitory concentration values (4.1–7.9 mg/ml), efficiency concentration values (178–321 mg/mg DPPH) and higher values of anti-radical power (0.31–0.51) as compared with their seeds. Thermal treatment reduced the total phenolic contents, antioxidant and free radical scavenging activities. The leaves of C. sativum were found with good amounts of caffeic acid, ferulic acid, gallic acid and chlorogenic acid.  相似文献

Phenols, a major group of antioxidant phytochemicals, have profound importance due to their biological and free radical scavenging activities. To identify their potential sources, extracts of some plants were studied for their total phenolic content (TPC), antioxidant (AOA) and free radical scavenging activities (FRSA) by different methods at multiple concentrations followed by specific phenolic composition. The amount of TPC varied from 2.8 mg/g (Withania somnifera, roots) to 107.8 mg/g (Cassia fistula, fruits) and the AOA from 24.2% (Curcuma zeoderia, leaves) to 96.9% (Trewia nudiflora, leaves). Bark of Azadirachta indica, fruits of C. fistula, and leaves and fruits of T. nudiflora were found to have high TPC (89.8- 107.8 mg/g) and high AOA (84.8-96.9%). Promising plant parts were studied for their FRSA and reducing power (RP), where the bark of A. indica, Casuarina equisetifolia and Cinnamomum zeylanicum, flowers of Indigofera tinctoria, fruits of Lawsonia inermis, and fruits and leaves of T. nudiflora showed a very low inhibitory concentration value ranging from 0.14 to 0.26 mg/ml, efficiency concentration value from 6.1 to 11.6 mg/mg DPPH and reducing power value from 0.6 to 2.8 ascorbic acid equivalents (ASE/ml), and reasonably high values (8.5 -16.2) of anti-radical power (ARP), indicating their strong FRSA. They also showed better inhibition of hydroxyl radical induced deoxyribose degradation than that of reference standard. Fruits of C. fistula with high phenols (107.8 mg/g) showed poor reducing power (5.9 ASE/ml) and ARP (4.7); in contrast, the bark of C. equisetifolia and fruits of L. inermis were with comparatively lower phenols (72.1 and 75.8 mg/g) but exhibited good ARP (16.2 and 14.4) and reducing power (0.7 and 0.6 ASE/ ml, respectively). Some of the plants were also found effective in protecting plasmid DNA nicking induced by hydroxyl radicals generated by Fenton's reaction. They were further assayed for their specific phenolic composition through high-performance liquid chromatography and MS/MS, where the amount of caffeic acid varied from 0.312 to 0.797 mg/g, chlorogenic acid from 0.018 to 2.109 mg/g, ellagic acid from 0.009 to 0.902 mg/g, ferulic acid from 0.036 to 0.078 mg/g, gallic acid from 0.192 to 3.597 mg/g, kaempferol from 0.011 to 0.910 mg/g, quercetin from 0.047 to 1.106 mg/g and rutin from 0.059 to 2.029 mg/g.  相似文献

Water and ethanol crude extracts from black pepper (Piper nigrum) were investigated for their antioxidant and radical scavenging activities in six different assay, namely, total antioxidant activity, reducing power, 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging, superoxide anion radical scavenging, hydrogen peroxide scavenging, and metal chelating activities. Both water extract (WEBP) and ethanol extract (EEBP) of black pepper exhibited strong total antioxidant activity. The 75?µg/ml concentration of WEBP and EEBP showed 95.5% and 93.3% inhibition on peroxidation of linoleic acid emulsion, respectively. On the other hand, at the same concentration, standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and α-tocopherol exhibited 92.1%, 95.0%, and 70.4% inhibition on peroxidation of linoleic acid emulsion, respectively. Also, total phenolic content in both WEBP and EEBP were determined as gallic acid equivalents. The total phenolics content of water and ethanol extracts were determined by the Folin-Ciocalteu procedure and 54.3 and 42.8?µg gallic acid equivalent of phenols was detected in 1?mg WEBP and EEBP.  相似文献

The aims of this study are to determine and compare the antioxidant levels and activities (i.e. primary and secondary) between selected seeds of Malaysian tropical fruits - guava (Psidium guajava), mango (Mangifera indica L.) and papaya (Carica papaya L.). Seeds are among byproducts from the processing of fruitsbased products. Instead of discarding seeds as waste, seeds with high potential as antioxidants could be utilised for commercial purposes. Accordingly, the selected seeds of Malaysian tropical fruits were tested in this study for total phenolic content (TPC), free radical scavenging activity by 1, 1- diphenyl-2-picrylhydrazyl (DPPH) assay and metal ion chelating effect by ferrous ion chelating (FIC) assay. Extraction of antioxidant compounds from sample was done with 70% ethanol. TPCs of the seeds were expressed as gallic acid equivalents (GAE) in mg per 100 g fresh seed weight. TPC assay showed that mango seeds had the highest TPC (i.e. 32 ± 0.001 mg GAE) followed by guava seeds (i.e. 20 ± 0.001 mg GAE) and papaya seeds (8 ± 0.003 mg GAE). For DPPH assay, IC50 data showed that mango seed extract scavenged 50% DPPH radicals at the lowest concentration (0.11 ± 0.01 mg/mL) followed by the positive control BHA (0.13 ± 0.01 mg/mL), guava seed extract (0.26 ± 0.01 mg/mL) and papaya seed extract (0.34 ± 0.01 mg/mL). Interestingly, all seed extracts showed higher free radical scavenging activities than BHA after sample concentration of 0.60 mg/mL. However, FIC assay indicated that metal ion chelating effects of all seed extracts were weaker than BHA suggesting that the fruit seeds are not sources of good metal ion chelators. Overall, present results suggest that TPC of the seeds show strong negative correlation with their primary antioxidant activity (r= -0.985, R2= 0.970), and not all compounds in extracts which could scavenge DPPH radicals are good metal ion chelators. Mango seeds relatively showed the highest antioxidant level and primary antioxidant activity followed by guava seeds and papaya seeds.  相似文献

目的研究三种蛋白酶的蛋清蛋白酶解物(EWPH)的抗氧化及血管紧张素转换酶(ACE)抑制活性。方法分别采用Fenton体系、邻苯三酚自氧化体系和亚油酸自氧化体系测定胰蛋白酶、木瓜蛋白酶及alcalase碱性蛋白酶的EWPH清除羟自由基、超氧阴离子及抑制脂质过氧化的能力。利用高效液相色谱法测定ACE抑制活性。超滤分离上述三种蛋白酶的EWPH,检测各组分的抗氧化和ACE抑制活性。结果抗氧化活性最强的是木瓜蛋白酶的EWPH,在浓度10mg/ml时,对羟自由基和超氧阴离子清除率分别为53.14%和27.87%,在浓度0.8mg/ml时,对亚油酸自氧化的抑制率73.72%(D7)。ACE抑制效果最明显的是胰蛋白酶的EWPH,其IC50值为0.985mg/ml。低分子量(Mw3ku)组分酶解物的活性优于高分子量(Mw3ku)组分。结论三种蛋白酶的EWPHs均具有抗氧化和ACE抑制活性,但两种活性之间无明显的相关,且小分子组分的活性最强。  相似文献

This study aims to evaluate the antioxidant activity (total antioxidant and free radical scavenging activities) of seaweeds commercially available in the Malaysian supermarket. Four types of seaweeds namely Nori (Porphyra sp.), Kumbu (Laminaria sp.), Wakame (Undaria sp.) and Hijiki (Hijikia sp.) were used in the study. The extracts were prepared with water and ethanol, respectively. The b-carotene bleaching and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays were used to determine antioxidant properties of seaweeds by measuring the decrease in absorbance at 470 and 517 nm. In water extract, Kumbu showed the highest total antioxidant activity of 63% compared with other samples. Kumbu, Nori and Hijiki exhibited higher radical scavenging activity than Wakame when extracted with water. Wakame exhibited the highest antioxidant and free radical scavenging activities in ethanolic extract with 58% and EC50 = 0.42 mg/ml respectively. The results of ANOVA analysis show significant differences (p<0.05) in the means of total antioxidant and free radical scavenging activities of the seaweeds. The results showed that processed commercial seaweeds exhibited varying degrees of antioxidant properties.  相似文献

Waste obtained from six genotypes of tomato was subjected to evaluation as potential sources of hydrophilic antioxidants on the basis of content of total phenolics (TPh; from 11.7 to 18.6 mg/g), total flavonoids (TFl; from 7.62 to 12.1 mg/g) and ascorbic acid (AA; from 0.51 to 1.89 mg/g), and antioxidant activities. The antioxidant activities of tomato waste extracts were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferrous ion chelating activity (CA) and reducing power (RP) assay. The O(2) tomato waste extract was found to have the strongest antioxidant activity in scavenging DPPH radicals (effective concentration, EC(50) = 0.18 mg/ml). The Saint Pierre tomato waste extract showed the best performance in RP assay (EC(50) = 1.57 mg/ml). The Rutgers tomato waste extract exhibited a strongest CA (EC(50) = 1.49 mg/ml). The linear correlation analysis produced moderate-to-high correlation coefficients between the antioxidant activities and TPh, TFl and AA.  相似文献

We are attempting to develop the novel phenolic synthetic antioxidants aimed at retarding the effects of free radicals and oxidants. The phenolic compounds (7-12) were synthesized by Friedel-Crafts alkylation of isoeugenol (1) and phenol derivatives (2-6) and their structures were determined by spectroscopic methods. All the synthesized phenolic compounds (7-12) except 12 are new. Antioxidant and radical scavenging activities of synthesized compounds (7-12) were determined by using various in vitro assays such as 1,1-diphenyl-2-picrylhydrazyl free radicals (DPPH), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radicals (ABTS(+)), and superoxide anion radicals (O(2)(-)) scavenging, ferric reducing antioxidant power (FRAP) and total antioxidant activity by ferric thiocyanate. The antioxidant activities of compounds were compared with standard antioxidants such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and trolox as positive controls. The results showed that the synthesized compounds, especially 10 and 11, had better properties than standard antioxidants (BHT, BHA and trolox).  相似文献

The aim of this study is to examine the in vitro antioxidant and antimicrobial activities of the acetone, methanol, and aqueous extracts of the lichens Cladonia furcata (many-forked Cladonia), Hypogymnia physodes (puffed lichen), and Umbilicaria polyphylla (manyleaf navel lichen) and the antimicrobial activity of fumarprotocetraric acid, gyrophoric acid, and physodic acid isolated from the respective lichen species. Antioxidant activity was evaluated by five different methods: free radical scavenging, superoxide anion radical scavenging, reducing power, determination of total phenolic compounds, and determination of total flavonoid content. U. polyphyla exhibited more powerful antioxidant activities than the other lichen species examined: methanol extracts of U. polyphyla showed 90.08% free radical scavenging activities (the ratio was greater than those of standard antioxidants). Moreover, the extracts tested had effective reducing power and superoxide anion radical scavenging. Total contents of phenol and flavonoid in extracts were determined as pyrocatechol equivalents and as rutin equivalents, respectively. A significant strong relationship between total phenolic and flavonoid contents and their antioxidative activities was observed. The antimicrobial activity was estimated by determination of the minimal inhibitory concentration by the broth microdilution method against six species of bacteria and 10 species of fungi. In general, the lichen extracts tested and their respective acids had relatively strong antimicrobial activity against the microorganisms tested. The present study shows that the lichen species tested demonstrated strong antioxidant and antimicrobial activities. The results suggest that lichens may be used as good sources of natural antioxidants and for pharmaceutical purposes in treating various diseases.  相似文献

Selected species of wild edible mushrooms were obtained from the interior areas of East Malaysia to determine the total phenolics and antioxidant properties, including free radical scavenging, reducing power and metal chelating activities. The in vitro antioxidant activities of petroleum ether (PE) and methanolic extracts of the edible wild mushrooms were comparable to the cultivated oyster mushroom. The radical scavenging activity was the highest in PE extract of Pleurotus porrigens (angel's wings) (85%), while methanolic extract of Hygrocybe conica (witch's hat mushroom) exhibited the highest (94%) chelating effect at 20 mg/ml. PE extracts were more effective than methanolic extract in scavenging ability on DPPH radicals, whereas methanolic extracts were more effective in reducing power and chelating ability on ferrous ions as evidenced by their lower EC50 values. Principal component analysis (PCA) indicated phenolic group was the primary factor contributing to the metal chelating ability for PE extract although phenolic was better correlated with reducing power in methanolic extracts.  相似文献

Peperomia pellucida leaf extract was characterized for its anticancer, antimicrobial, antioxidant activities, and chemical compositions. Anticancer activity of P. pellucida leaf extract was determined through Colorimetric MTT (tetrazolium) assay against human breast adenocarcinoma (MCF-7) cell line and the antimicrobial property of the plant extract was revealed by using two-fold broth micro-dilution method against 10 bacterial isolates. Antioxidant activity of the plant extract was then characterized using α, α-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging method and the chemical compositions were screened and identified using gas chromatography-mass spectrometry (GC-MS). The results of present study indicated that P. pellucida leaf extract possessed anticancer activities with half maximal inhibitory concentration (IC(50)) of 10.4 ± 0.06 μg/ml. The minimum inhibitory concentration (MIC) values were ranged from 31.25 to 125 mg/l in which the plant extract was found to inhibit the growth of Edwardsiella tarda, Escherichia coli, Flavobacterium sp., Pseudomonas aeruginosa and Vibrio cholerae at 31.25 mg/l; Klebsiella sp., Aeromonas hydrophila and Vibrio alginolyticus at 62.5 mg/l; and it was able to control the growth of Salmonella sp. and Vibrio parahaemolyticus at 125 mg/l. At the concentration of 0.625 ppt, the plant extract was found to inhibit 30% of DPPH, free radical. Phytol (37.88%) was the major compound in the plant extract followed by 2-Naphthalenol, decahydro- (26.20%), Hexadecanoic acid, methyl ester (18.31%) and 9,12-Octadecadienoic acid (Z,Z)-, methyl ester (17.61%). Findings from this study indicated that methanol extract of P. pellucida leaf possessed vast potential as medicinal drug especially in breast cancer treatment.  相似文献

The aim of the study was to investigate antioxidant activities of Ganoderma lucidum and Funalia trogii. Ethanol and water crude extracts from G. lucidum and F. trogii were investigated for their antioxidant capacity in some different assays, namely, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, metal chelating activity against ferrous ions, and plasma lipid peroxidation inhibitory. In addition, the amounts of total phenol, ascorbic acid, β-carotene, and lycopene components in the extracts were determined. Among the four mushroom extracts, G. lucidum water extract and G. lucidum ethanol extract showed the highest scavenging activity against DPPH radicals (50% inhibitory concentration?=?0.055?±?0.001?mg/mL). Total phenol was the major antioxidant component found in the mushroom extracts. These results showed that G. lucidum may be used in pharmaceutical applications because of its effective antioxidant properties.  相似文献

Antioxidant compounds are abundantly available in plants and play an important role in scavenging free radicals, thus providing protection to humans against oxidative DNA damage. Mentha spicata Linn., commonly called spearmint, belongs to the family lamiaceae. It was selected in the present study because Mentha extracts have antioxidant properties due to the presence of eugenol, caffeic acid, rosmarinic acid and alpha-tocopherol. Four solvent fractions (hexane, chloroform, ethyl acetate and water) of ethanolic extract of dried leaves powder of M. spicata were analyzed for total antioxidant activity (TAA) and relative antioxidant activity (RAA) and compared with standard antioxidants such as Quercetin, beta-carotene, L-ascorbic acid and glutathione using ABTS*+ decolorization assay (ABTS/Potassium persulphate). The antioxidant activity was assumed to be from the total phenolic content of the ethanolic extract. Total phenolics are found to be highest in ethyl acetate fraction (54 mg/g) and least in hexane fraction (13 mg/g) and more or less similar in water and chloroform fractions (30-32 mg/g). TAA is found to be less in hexane and chloroform fractions (<53% at 50 microg/ml) and highest in ethyl acetate (95% at 20 microg/ml) and water (84% at 30 microg/ml) fractions. The RAA of ethyl acetate fraction is 1.1 compared to quercetin (at 5 microM/ml), but greater when compared to beta-carotene (15 microM/ml), L-ascorbic acid (15 microM/ml) and glutathione (15 microM/ml). The RAAs with these antioxidants are in the range of 1.31 -1.6. The values of RAAs for water fraction also show similar trend and are in the range of 1.0-1.4. The antioxidant activities of the solvent factions are closely related to the content of total phenolics present in them.  相似文献

The antioxidant activities of methanolic extracts of far-infrared irradiated rice hull (FRH) and non-irradiated intact rice hull (IRH) were determined. The antioxidant effects against reactive oxygen species (ROS) were evaluated by measuring scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide, hydrogen peroxide (H2O2), and nitric oxide radical and capacity for chelating metals. Except for H2O2 scavenging activity, FRH showed higher scavenging activity than IRH; for example, the 50% inhibitory concentration (mg/mL) values for DPPH radical scavenging of FRH and IRH were 0.067 and 0.085, respectively, as compared with 0.362 and 0.012 for butylated hydroxytoluene and alpha-tocopherol, respectively. The effect of rice hull extract on DNA damage induced by H2O2 in human lymphocytes was also evaluated by comet assay. The protective effect of rice hull extract increased as its concentration increased from 12.5 to 50 microg/mL, as indicated by DNA strand breakage decreasing from 38% to 22% with FRH and from 49% to 28% with IRH as compared with H2O2-treated positive controls. When human lymphocytes were post-incubated with rice hull extract for 30 minutes after exposure to H2O2, the protective ability of the rice hulls remained unchanged. These results suggest that methanol extracts of rice hulls possess significant ROS scavenging and metal chelating activities and protective effect against oxidative DNA damage.  相似文献

The antioxidant activity, ascorbic acid and phenolic content were studied in 10 exotic fruits from Brazil: abiu, acerola, wax jambu, cashew, mamey sapote, carambola or star fruit, Surinam cherry, longan, sapodilla and jaboticaba. The ascorbic acid was determined by 2,6-dichloroindophenol titrimetic methods and total phenols were measured colorimetrically using the Folin-Ciocalteu reagent. The antioxidant activity was investigated with three different methods: hypochlorous acid scavenging activity, 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation decolorization assay, and 2,2-diphenyl-1-picrylhydrazyl radical scavenging method. The highest content of vitamin C (1,525.00 mg/100 g pulp) occurred in acerola. The total phenol content was higher in abiu, acerola, Surinam cherry and sapodilla. In relation to antioxidant activity, acerola has showed the great values in all three different methods tested. It was found that the fruits have a significant antioxidant effect when tested by each method, respectively, and these antioxidant capacities are promising. The sample concentration also influenced its antioxidant power.  相似文献

This study was aimed at evaluating the antioxidant activity and efficacy of the ethanolic extract of the endemic plant species Kitaibelia vitifolia in inhibiting the growth of selected fungi and bacteria. Antimicrobial activity was tested using the broth dilution procedure for determination of minimum inhibitory concentration (MIC). MICs were determined for eight selected indicator strains. The highest susceptibility to K. vitifolia ethanolic extract among the bacteria tested was exhibited by Bacillus subtilis ATCC 6633, Staphylococcus aureus ATCC 25923, and Klebsiella pneumoniae ATCC 13883 (MIC=15.62 μg/mL), followed by Escherichia coli ATCC 25922 and Proteus mirabilis ATCC 14153 (MIC=31.25 μg/mL), and Proteus vulgaris ATCC 13315 (MIC=62.50 μg/mL). Of the fungi, Candida albicans ATCC 10231 (MIC=15.62 μg/mL) showed the highest susceptibility, and Aspergillus niger ATCC 16404 (MIC=31.25 μg/mL) had the lowest. Results showed that K. vitifolia extract possesses antioxidant activity, with total antioxidant capacity of 75.45±0.68 μg of ascorbic acid/g and 50% inhibition concentration values of 47.45±0.55 μg/mL for 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity, 35.35±0.68?μg/mL for inhibitory activity against lipid peroxidation, 95.25±0.52 μg/mL for hydroxyl radical scavenging activity, and 31.50±0.35 μg/mL for metal chelating activity. Total phenolics, flavonoids, condensed tannins, and gallotannins were 85.25±0.69 mg of gallic acid (GA)/g, 45.32±0.55 mg of rutin/g, 54.25±0.75 mg of GA/g, and 41.74±0.55 mg of GA/g, respectively. The phenolic composition of K. vitifolia extract was determined by high-performance liquid chromatography. Rosmarinic acid was found to be the dominant phenolic compound of the extract.  相似文献

The phenolic contents of methanolic extracts from five varieties of date palm (Phoenix dactylifera) fruits from Algeria, Deglet Nour (DN), Degla Baidha (DB), Ghars (Gh), Tamjhourt (Tam), and Tafezauine (Taf) were measured in this work. The antioxidant activities of these extracts were also evaluated. Total phenolic content ranged from 41.80 to 84.73 mg gallic acid equivalents (GAE)/100 g and the total flavonoid content varied from 7.52 to 14.10 mg rutin equivalents (RE)/100 g. The antioxidant activities of methanolic extracts were evaluated in vitro using scavenging assays of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS) radical ion (ABTS+) and potassium ferricyanide complex as reducing power assay. Effective scavenging concentration (IC50) on DPPH radical ranged from 10.83 to 21.27 mg/l, the IC50 values decreased in the order DN > Gh > Tam > DB > Taf. ABTS radical cation scavenging activity (trolox equivalent 1.66–3.35 mM), the trolox equivalent antioxidant capacity values decreased in the order of DN > Gh > DB > Tam > Taf. In the potassium ferricyanide complex assay, the antioxidant capacity of the extracts ranged between 2.06 and 4.21 mM ascorbic acid equivalents and the ascorbic acid equivalents antioxidant capacity values of the extracts decreased in the order Gh > Tam > DB > DN > Taf. These results suggest that all date varieties constitute a good source of natural antioxidants and could potentially be considered as a functional food.  相似文献

Hot water extract (LN), partially purified polysaccharides (LP) and hot alkali extracted polysaccharides (LNa) obtained from fruiting bodies of the wild basidiomycete Laetiporus sulphureus were examined for their antioxidant activities. LNa was the most active antioxidant, as shown by the median effective concentrations (EC50 values) of 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity (0.5 ± 0.2 mg/ml), reducing power (4.0 ± 0.3 mg/ml) and ferrous ion-chelating ability (1.5 ± 0.1 mg/ml). LNa contained the highest level of α-glucan (17.3 ± 1.2 g/100 g dw), whereas LP contained mostly β-glucans (66.8 ± 1.3 g/100 g dw). The prevalent monosaccharide in all extracts was glucose. The EC50 values of all three antioxidant activity assays were well-correlated with the α-glucan content. Strong and significant correlation was found between total phenolic compounds and DPPH scavenging ability and also reducing power. The three investigated extracts (at concentrations of 0.1–10 mg/ml) were not toxic to HTR-8/SVneo trophoblast cell line.  相似文献
