Considerable regional differences in the phospholipid composition of mitochondrial membranes are found in the brain of cats in the terminal phase of hemorrhagic shock. The most prominent alteration is noted in the medulla oblongata and consists in a progressive elimination of phosphatidylcholine. Changes in the main phospholipids in mitochondrial membranes of the cerebral hemispheres are less pronounced and consist in a drop of phosphatidylinositol and phosphatidylethanolamine. Accumulation of lysophosphatidylcholine and lysophosphatidylethanolamine is a regular feature of the studied mitochondria. Accumulation of lysophosphatidylserine is found primarily in mitochondrial membranes of the medulla oblongata. Progressive elimination of phosphatidylcholine from the hepatocyte plasma membranes is observed during the development of hemorrhagic shock in cats. The phosphatidylinositol content decreases 30 min after the start of bleeding and then gradually increases. The phosphatidylethanolamine content is increased on the 30th and 60th min of bleeding, while the content of phosphatidylserine increases 1 h after the start of bleeding. At the peak of hemorrhagic shock the major shifts are observed in the phosphatidylcholine and phosphatidylinositol contents. It is established that buprenorphine (0.3 mg/kg) induces considerable alterations in the phospholipid composition of hepatocyte plasma membranes as a result of phosphatidylserine accumulation and a considerable loss of sphingomyelin and lysophosphatidylserine. When administered in a dose of 0.03 mg/kg, buprenorphine facilitates normalization of the phosphoinositol turnover in hepatocyte plasma membranes. Experiments on cats indicated that the use of phosphatidylcholine liposomes in hemorrhagic shock may reduce the intensity of free-radical processes in the liver, stabilize the phospholipid bilayer of plasma membranes in hepatocytes, decrease the severity of pathomorphological changes in the target organs, and raise systemic arterial pressure with its stabilization at a subnormal level. The use of phosphatidylcholine liposomes in cats with hemorrhagic shock resulted in a considerable prolongation of their survival. Relations between rheologic, hemodynamic, and metabolic indices during hemorrhagic shock were studied in experiments on dogs. The mechanical properties of the blood were found to change immediately after blood loss parallel with the macrohemodynamic disorders, and they preceded the development of metabolic acidosis. Hemorrhagic shock is shown to disturb the behavior of rats in the open field test. Changes in the liver mitochondria of rats after bilateral subdiaphragmatic vagotomy were studied. Two stages were distinguished in the dynamics of the response of the mitochondrial system to denervation. During the first stage (0.5–3 days after vagotomy) reversible functional disturbances due to postoperative stress took place in the mitochondria. The second stage (7–60 days after denervation) is characterized by more marked structural and functional changes with some common features with those observed in hypoxia and resulting from vagotomy itself.Department of Histology, Therapeutic Faculty, N. Hyperbaric oxygen therapy (3 atm O2, 60 min) of hemorrhagic shock in cats (blood pressure 60/50 mm Hg) prevents the sharp increase, characteristic of hypoxia, in the concentration of ammonia, urea, alanine, and -aminobutyric acid in the brain and activates glutamine formation. 为探讨654-2抗休克的细胞机制,本工作在家兔晚期失血性休克模型上,观察654-2对休克动物肝细胞线粒体镁含量改变的影响,结果发现,失血性休克动物血浆镁浓度进行性升高与动物血压的降低呈负相关关系;休克动物肝线粒体镁含量明显减少;而用654-2治疗时,显著抑制失血性休克时血浆镁和肝线粒体镁含量的改变,明显改善休克动物的预后。实验结果提示654-2可能具有稳定休克动物肝线粒体的作用。  相似文献

Effect of intraperitoneal administration of spermin on oxidative phosphorylation and calcium capacity of isolated liver mitochondria was studied in normo- and hypothermic rats. Hypothermia stimulates mitochondrial respiration without decreasing the contingency and increases calcium capacity. Spermin suppresses mitochondrial respiration, the effect being stronger in hypothermia. In high doses spermin prevents stimulating effect of hypothermia on respiration and reduces the increase in calcium capacity. It is shown that apoC-III, but not other apoC proteins, components of very low density lipoproteins (apoC-I, apoC-II, apoC-III), reduced the rate of mitochondrial respiration in various metabolic states. This effect depended on the dose of apoprotein, type of oxidized substrate, and the presence of Ca ions in the incubation medium. ApoC-III completely blocked oxidative phosphorylation during oxidation of palmitoyl carnitine by mitochondria, while the respiration rate in metabolic state 4 remained unchanged. Lipid peroxidation (LPO) processes and the phospholipid (PL) content and composition in the homogenate and in the mitochondrial and supramitochondrial compartments of brain cells of adult male rats were studied 3 and 6 months after thymectomy. Thymectomy was found to markedly increase the rate of LPO and to reduce antiradical activity in all cellular compartments. A reliable increase of the level of total phospholipids in the homogenate and mitochondria was observed by the third month after thymectomy and a reduction of its control levels during the sixth month postoperation. On the other hand, a reduction of the content of total phospholipids was observed in the supramitochondrial fraction by the sixth month of the experiment. An increase of the phosphatidylethanolamine (PE) and phosphatidylserine (PS) contents was found to be mainly responsible for the changes in the level of total phospholipids. Changes in the cardiodynamics and contractile function of the myocardium were studied in experimental shock arising in 20 anesthetized dogs in response to intracoronary injection of anticardiac cytotoxic serum. Besides signs of a disturbance of left ventricular function (a fall in the systolic index, the working stroke index of the left ventricle, the rate of rise of intraventricular pressure, the indices of myocardial contractility, and the ejection fraction) a decrease was observed in the end-diastolic volume and pressure in the left ventricle. These results indicate that disturbances of the cardiodynamics are due to both cytotoxic injury to the heart and a disturbance of the blood flow to the left heart.Department of Experimental Cardiology, A. Modeling of hemorrhagic shock in rats adapted to immobilization stress required the removal of greater volumes of blood than that in control rats. Respiration of rat liver mitochondria after the addition of ischemic toxin to the incubation medium was compared with respiration of mitochondria isolated from ischemic rat liver. The changes in respiration in both cases could be prevented by preliminary addition of dithiothreitol and reduced by subsequent addition of cytochrome c or dithiothreitol to the incubation medium.
