 Objective To investigate the expression of NF-κB and epithelial-mesenchymal transition(EMT) related markers E-cadherin and Vimentin proteins in human pancreatic cancer tissues and its relation with the malignant features. Methods The expression of NF-κB、E-cadherin and Vimentin proteins in 62 cases of pancreatic cancer tissues were detected by using immunohistochemistry and compared with the clinicopathological data of pancreatic cancer. Results The positive expression rate of NF-κB was 81 % (50/62), Vimentin protein increased of expression was 61 % (38/62), and E-cadherin protein loss of expression was 55 % (34/62) in pancreatic cancer. The positive expression rate of NF-κB was significantly related with the lymph node metastasis (χ2=11.761, P <0.05), distant metastasis(χ2=9.225, P <0.05), the absent expression of epithelial marker E-cadherin protein (r =0.352, P <0.05) and the positive expression of mesenchymal marker Vimentin protein (r =0.343, P <0.05), but there was no relation with the patients gender, age, tumor location, tumor type and tumor differentiation (P >0.05). In addition, the significant correlation of E-cadherin expression loss and Vimentin expression with tumor lymph node metastasis and distant metastasis was found (χ2=6.914, 4.984, 7.753, 5.144, P <0.05). Conclusion The overexpression of NF-κB in pancreatic cancer may accelerate invasion and metastasis of pancreatic cancer through inducing EMT.  相似文献

Objective To investigate the expression of E-cadherin in nasopharyngeal carcinoma ( NPC) and its relationship with cervical lymph node metastasis. Methods The expression of E-cadherin in 80 patients with NPC was detected by immunohistochemistry. Results Lower expression of E-cadherin was associated with advanced N-stage of the tumor ( P = 0. 018 ). There was no significant correlation between the expression of E-cadherin and lymph node size ( P = 0.435 ). The expression of E-cadherin was higher in patients with cervical lymph node metastasis limited to a single area than that distributing in some scattered areas (P = 0. 000). There was a trend that the expression of E-cadherin in the cases with the tumor and lymph nodes in the same side was higher (56. 5% ) than that in the patients with bilateral lymph node metastases (32. 6% ) , however, the difference was not significant (P =0. 059). The expression rates of E-cadherin in patients with lymph node metastasis in levels Ⅱ , Ⅲ and Ⅴa were higher than that in levels Ⅰ , Ⅳ, Vb and Ⅵ, but with a non-significant difference (P = 0.059). Conclusion The expression of E-cadherin has influence on the lymph node metastasis in nasopharyngeal carcinoma. E-cadherin expression is negatively correlated with the numbers of the lymph node metastases and the metastasis distance, i. e. a lower expression of E-cadherin leads to an advanced N-stage. The lymph node metastasis of nasopharyngeal cancer from above to below is more considerably influenced by E-cadherin expression than the metastasis towards contralateral lymph nodes.  相似文献

Objective: The aim of our study was to observe the expressions and clinical Significance of E-cadherin, β-catenin and E-cadherin-catenins complex in breast cancer and precancerous lesions, and analyze the relationship between the expressions and clinicopathological features in breast cancer. Methods: Immunhistochemical UltraSensitiveTM S-P method was employed to detect the expression of E-cadherin, β-catenin and E-cadherin-catenins complex in 128 cases of invasive ductal carcinomas, 89 cases of ductal carcinoma in situ and 57 cases of atypical ductal hyperplasia, 53 cases of usual ductal hyperplasia breast tissues were selected as a control group. The express of E-cadherin, β-catenin and their relationship with mult biological parameters including histological grade, region lymph node metastasis, distant metastasis and recurrence on files were also assessed. Results: (1) The staining patterns character of E-cadherin, β-catenin and E-cadherin-catenins complex: In UDH breast tissues, E-cadherin and a-catenin were expressed on cell membrane of ductal and acinic cells, showing cellular contour and border among cells. The abnormal expression of the three proteins occurred in breast invasive ductal carcinomas, ductal carcinoma in situ and atypical ductal hyperplasia tissues, showing cytoplasmic or nuclear staining, decrease and loss of cytomembrane staining. (2) The abnormal expression rates of E-cadherin, β-catenin and E-cadherin-catenins complex in invasive ductal carcinomas were 53.91%, 65.63% and 81.25%, which were significantly higher than that in ductal carcinoma in situ, atypical ductal hyperplasia, usual ductal hyperplasia tissues (P 〈 0.01). Compared with usual ductal hyperplasia breast tissues group, the abnormal expression rates of E-cadherin, β-catenin and E-cadherin-catenins complex were significantly decreased (P 〈 0.01) in the breast cancer group. However, there was no significance of the abnormal expression rate between ductal carcinoma in situ and atypical ductal hyperplasia tissues groups (X2 = 0.76, P = 0.38; x2 = 0.14, P = 0.70; x2 = 0.81, P = 0.37; X2 = 2.19, P = 0.14) (P 〉 0.05). (3) There was a significantly difference in the mean E-cadherin, β-catenin and E- cadherin-catenins complex frequency between estrogen receptor & progesterone receptor positive IDC group and negative group, epidermal growth factor receptor type 2 (HER2/neu) positive and negative groups, Ki-67 proliferation index 〈 14% and 〉 14% groups, histological grade (I + II) and grade III invasive ductal carcinomas groups, with lymph node metastasis, distant metastasis and recurrence groups (P 〈 0.05) and without groups (P 〈 0.05). However, there was no difference in the mean E-cadherin, β-catenin and E-cadherin-catenins complex frequency between age (_〈 50 years vs 〉 50 years), tumor diameter (〈 2 cm vs 〉 2 cm) (P 〉 0.05). Conclusion: In breast cancer, the expressions of E-cadherin, β-catenin and E-cadherin-catenins complex are abnormally decreased and are correlated with pathology grade, differentiation disturbance and metastasis. E- cadherin and β-catenin may be as the predictors for prognosis. Combined detection may improve accuracy and sensitivity of predicting metastasis and prognosis of breast Cancer.  相似文献

Objective: To detect the expressions of E-cadherin, α-catenin and β-catenin and analyze the relationship between Ecadherin-catenin adhesion complex and clinicopathological features in breast cancer. Methods: The expressions of E-cadherin, α-cadherin and β-catenin in specimens of 54 breast cancer, 21 normal breast tissues around tumor, 15 breast hyperplasia of usual type and 15 breast atypical hyperplasia were detected by immunohistochemical method. Results: In 21 normal breast tissues, E-cadherin and α-catenin were expressed on cell membrane of ductal and acinic cells, showing cellular contour and border among cells. The staining character of the three proteins in breast hyperplasia of usual type was the same as that in normal breast tissue. In breast atypical hyperplasia, the abnormal expression rates of E-cadherin, α-catenin and β-catenin were 6.7%, 13.3% and 26.7%, respectively. The total abnormal expression rate of E-cadherin-catenin complex was 33.3%. In breast cancer, the abnormal expression rates of E-cadherin, α+catenin and β-catenin were 51.9%, 63.0% and 61.1%, respectively. The total abnormal expression rate of E-cadherin-catenin complex was 88.9%. Abnormal expression of E-cadherin and α-catenin were significantly correlated with histological grade. Abnormal expressions of α-catenin and β-catenin were significantly correlated with TNM staging, axillary lymph nodes metastasis and postoperative distant metastasis. Abnormal expression of E-cadherin-catenin complex was correlated with TNM staging, histological grade and axillary lymph nodes. Abnormal expression of β-catenin was negatively correlated with expression of HER-2. COX multiple factor analysis showed that E-cadherin or α-catenin or β-catenin was not independent prognostic indicator. Conclusion: Abnormal expressions of E-cadherin, α-catenin and β-catenin frequently occur in breast cancer. Abnormal expression of E-cadherin-catenin complex is correlated with differentiation disturbance and metastasis. Combined measurement of E-caherin, α-catenin and β-catenin may improve accuracy and sensitivity of predicting metastasis and prognosis of breast cancer.  相似文献

Objective To explore the expression and clinical significance of E-cadherin and β-catenin in osteosarcoma tissues. Methods From April 2012 to October 2015, 54 specimens of osteosarcoma tissues and 22 specimens of osteochondroma tissues were enrolled. Immunohistochemistry was used to detect the expression of E-cadherin and β catenin in those above tissues. The correlation of the expression of E-cadherin, β-catenin and clinical parameters of osteosarcoma were analyzed. Results In 54 specimens of osteosarcoma tissues, the positive rate of E-cadherin protein was 35.2%(19/54), significantly lower than 68.2%(15/22) in osteochondroma tissues (P<0.05). However the positive rate of β-catenin protein was 68.5%(37/54), significantly higher than 9.1%(2/22) in osteochondroma tissues (P<0.05). The expression of E-cadherin protein was associated with Enneking stage and metastasis (P<0.05). The expression of β-catenin protein was associated with tumor size, Enneking stage and metastasis (P<0.05). Moreover, the expression of E cadherin and β catenin was negatively correlated (r=-0.764,P<0.05). ConclusionE-cadherin and β-catenin are abnormally expressed in osteosarcoma tissues, and they are correlated to Enneking stage and metastasis. Both of them play a role in the development and progression of osteosarcoma.  相似文献

Objective To evaluate the prognostic value of the expression of epidermal growth factor receptor (EGFR) and nm23 in patients with nasopharyngeal carcinoma (NPC). Methods From 2003 to 2006, 127 NPC patients who had undergone biopsy before radiotherapy were reviewed retrospectively. All patients received intensity-modulated radiotherapy using 6 MV X-rays combined with platinum-based chemotherapy. Immunohistochemistry SP method was adopted to detect the expression of EGFR and nm23 in NPC biopsy specimens . The relationship between the expression of EGFR and nm23 and survival was analyzed. Results The positive rate of EGFR and nm23 were 80.3% and 47. 2% respectively. The nm23expression was correlated with distant metastasis (χ2=7.03, P = 0. 008 ). The 5-year estimated local control, over-all survival (OS) and disease-free survival (DFS) were 58.3% ,53.5% and 46. 5%. Patients with negative expression of EGFR had a significantly better 5-year OS, DFS (χ2=8.23, P=0.004;χ2=5.25,P=0.022) than those with positive expression. Patients with positive expression of nm23 had a significantly higher 5-year OS (χ2=15.68, P = 0. 000) and DFS (χ2=14. 85, P = 0. 000) than those with negative expression. The clinical stage, EGFR and nm23 expression were independent prognostic factors shown by Cox proportional hazard model (χ2=23.03, 18.33, 39.92, P= 0.000, 0.000, 0.000).Conclusions The EFGR and nm23 expression were correlated with the prognosis in NPC patients.  相似文献

Objective To study the expression and clinical significance of VEGF and COX-2 in papillary thyroid carcinoma (PTC).Methods The expression of VEGF and COX-2 were investigated by immuonhistochemical S-P method in 78 cases of papillary thyroid carcinoma tissue and adjacent tissue.78 cases of PTC including 42 cases with intrathyroidal invasion, 36 cases with extrathyroidal invasion,and 38 cases with lymph node metastasis and 40 cases without lymph node metastasis.Adjacent tissues including 34 cases of nodular goiter(NG),25 cases of Hashimoto thyroiditis(HT)and 19 cases of normal tissue.Results There was a significant difference between thyroid cancer tissue and the tumor free tissue(P<0.05)and the expression levels of VEGF and COX-2 increased from the tumor free tissue,adjacent tissue to the thyroid cancer tissue(P<0.05).The expression of VEGF and COX-2 were not related with site of tumor(P>0.05),but were correlated to the degree of the infiltration and lymph node metastasis in PTC.Expression of COX-2 was correlated positively with VEGF.Conclusion The high expression of VEGF and COX-2 in PTC relates to the development,progression and metastasis of PTC,which may serve as one of the parameters for diagnosis of PTC and determing biological behavior and prognosis of PTC.  相似文献

Objective To investigate the expression of MMP-2,TIMP-2,TGF-β1 and TGF-βRI and the relationship among them in breast cancer.Methods The protein expression of MMP-2,TIMP-2,TGF-β1 and TGF-β1R1 was detected on tissue chips by S-P immunohistochemical staining in 160 cases of breast carcinoma.Results The positive rates of TGF-β1,TGF-β1 mRNA,MMP-2,MMP-9,TIMP-1 and TIMP-2 expression were 73.7%,56.2%,96.9%,95.0%,87.5% and 89.4%,respectively.Axillary lymph node metastasis and TNM staging(P <0.01 and P <0.01,respectively)were positively correlated to the expression of TGF-β1.Relase-free survival of TGF-β1 positive group was lower than that of TGF-β1 negative group(P = 0.023).The expression of M MP-2 or M M P-9 was positively correlated to that of TGF-β1 (r=0.170,P<0.05;r =0.221,P<0.01)and was negatively correlated to that of TGF-β1 mRNA(r =-0.126,P >0.05;r = 0.019,P > 0.05).Conclusion The expression of TGF-β1 may be closely correlated with the invasion and metastasis of breast cancer.TGF-β1-induced invasiveness and metastasis of breast cancer cells are mediated by MMP-2 and MMP-9.  相似文献

Objective: The purpose of this study was to evaluate cyclooxygenase-2 (COX-2) expression in nasopharyngeal carcinoma (NPC) and its correlation with clinicopathologic features, angiogenesis, and prognosis. Methods: The expressions of COX-2 and vascular endothelial growth factor (VEGF) and microvascular density (MVD) were determined with immunohistochemical methods in eighty-six NPC patients followed up over 5 years. Results: Sixty-three tumors (73.3%) were classified as COX-2 positive. COX-2 expression was positively related to VEGF expression (r=0.438, P〈0.01) and correlated with the tumor pathological grade, extent of primary lesion, lymph node metastasis, distant metastasis and shorter survival. Conclusion: Our results suggest that COX-2, being highly expressed and strongly correlated with angiogenesis in nasopharyngeal carcinoma, is apt to be used as a predictor of prognosis, including local recurrence and distant metastasis.  相似文献

Objective: To study the expressive characteristics of iNOS to the prognosis NSCLC patients. Methods: The expression in non-small cell lung carcinoma (NSCLC) and it's affection of iNOS was detected in 50 NSCLC tissues by Immunohistochemistry technology. Results: There was positive expression in NSCLC (positive rate 38.00%). The expression of iNOS had a correlation with lymph node metastasis (P〈0.05). There was no significant difference between positive expression of iNOS and lung cancer histological types, TNM stages, T stages (P〉0.05). But the data suggested that there was a direct correlation between expression of iNOS and clinical prognosis (P〈0.05). Conclusion: Positive expression of iNOS was observed in NSCLC, which correlated with postoperative survival time and lymph node metastasis. Although iNOS level had no significant relation with T stage, we find the positive rate in T2 stage was much higher than that in TI stage. The result suggested that positive expression of iNOS was related to bad clinical prognosis because of promoting tumor growth and metastasis.  相似文献

Objective:To investigate the expressions and correlations of Pin1,β-catenin and cyclin D1 in elderly lung carcinomas.Methods:The expressions of Pin1,β-catenin and cyclin D1 were examined in the specimens of 92 elderly lung carcinomas and 10 normal lung tissues by immunohistochemistry and explored the relationship between the expression levels and clinicopathological factors.Results:(1) The overexpression of Pin1 and cyclin D1 in lung carcinomas was 46 (50%)cases and 60 (65.22%) cases respectively and 56 (60.82%) cases showed positive immunoreactivity for β-catenin in the nuclear and (or) cytoplasmic fraction in tumor tissues,In normal tissue,the expressions of Pin1 and cyclin D1 were negative,the expression of β-catenin was lied in cell membrane.(2) In lung carcinomas the expressions of Pin1,β-catenin and cyclin D1 correlated with tumor differentiation (P<0.05).The pesitive expression rate and intensity of Pin1 correlated with tumor stage (P=0.032) and lymph node positive disease (P=0.041).The expression of β-catenin correlated with lymph node positive disease (P=0.012).(3) High expression levels of Pin1 correlated with aberrant β-catenin expression (P=0.000) but did not show a correlation with cyclin D1 (P=0.157).Conclusion:In elderly lung carcinomas,the positive expression of Pin1 causes abnormal accumulation of β-catenin and actives its target gene,however,this target gene was not cyclin DI.The detection of Pin1 expression had some clinical significance in estimating prognosis of elderly patient with lung carcinomas.  相似文献

We aimed to detect cytokeratin 19 (CK19) and polymorphic epithelial mucin 1 (MUC1) expression in peripheral blood of thyroid cancer patients, and investigate the clinical value of it as a diagnostic marker for circulating blood micrometastases. Methods: The flow cytometry (FCM) was used to detect and analyze CK19 and MUC1-expressing cells in peripheral blood of 491 thyroid cancer patients. Results: CK19 and MUC1 expression showed no statistically significant difference with gender and age in thyroid cancer patients (P 〉 0.05), while had statistically significant difference with tumor size, lymph node stage and distant metastasis (P 〈 0.01). The expression of CK19 and MUC1 were positively correlated (r = 0.628, P = 0.00). Conclusion: CK19 is closely related to MUC1 expression, tumor size, extent of invasion and distant metastasis in peripheral blood of thyroid cancer patients. The circulating blood CK19 and MUC1 tests can help predict thyroid cancer micrometastases and prognosis.  相似文献

Objective:To investigate the expressions of caveolin-1,E-cadherin and β-catenin in gastric carcinoma,precancerous gastric and chronic non-atrophic gastritis tissues,and evaluate the correlation of these expressions with the development of gastric cancer.Methods:The expressions of caveolin-1,E-cadherin and β-catenin were detected by biotin-streptavidin-peroxidase(SP) immunohistochemistry on 58 gastric cancer tissues,40 precancerous gastric tissues and 42 chronic non-atrophic gastritis tissues.The correlation between the expressions of caveolin-1,E-cadherin and β-catenin,and the clinicopathologic parameters of gastric cancer was analyzed retrospectively.Results:The positive rates of caveolin-1 and E-cadherin expressions in gastric carcinoma were significantly lower than precancerous gastric and chronic non-atrophic gastritis tissues(P<0.01).An abnormal rate of β-catenin expression in gastric carcinoma was higher than precancerous gastric and chronic non-atrophic gastritis tissues(P<0.01).Moreover,low expressions of caveolin-1,E-cadherin and β-catenin correlated with tumor size,depth of invasion,lymph node metastasis and TNM stage(P<0.05).The positive rates of caveolin-1 and E-cadherin expressions decreased(P<0.01),while an abnormal rate of β-catenin expression increased inversely,with the degree of atypical hyperplasia(P<0.01).Caveolin-1 expression correlated positively with E-cadherin(r=0.41,P<0.05).Caveolin-1(r= 0.36,P<0.05) and E-cadherin(r= 0.45,P<0.05) expressions negatively correlated with abnormal β-catenin expression.Conclusion: These results suggested that dysregulated expressions of caveolin‐1, E‐cadherin and β‐catenin correlated with the development of gastric cancer and its biological behavior.  相似文献

OBJECTIVE To examine the possibility of human sodium iodide symporter (hNIS) protein expression in lung cancer cells. METHODS Human lung A549 cancer cells were thawed and cultured in vitro. The cells were divided into an experimental group transfected with a recombinant pcDNA3-hNIS plasmid and a control group transfected only with a pcDNA3 plasmid. The recombinant plasmid vector encoding the hNIS gene (pcDNA3-hNIS) was amplified, purified and identified. The hNIS gene was followed by DNA sequencing. A Western blot and an immunohistochemical assay were applied to detect the hNIS protein expression in the transfected human lung A549 cancer cells. RESULTS Restriction enzyme digestion and DNA sequencing results showed the size and direction of the inserted gene in the recombinant pcD- NA3-hNIS plasmid was correct. The Western blot method and immunohistochemical analysis showed a positive NIS protein expression in the experimental group. The NIS protein was detected mainly in the cell membranes showing a positive rate up to 70.6% with no expression of the NIS protein in the control group. There was a significant difference between two groups (P=0.000). CONCLUSION The hNIS gene was transfected effectively into human lung A549 cancer cells mediated by Lipofectamine 2000, and was expressed with its protein in vitro.  相似文献

Objective To investigate the expression of COX-2 and VEGF in clear cell renal cell carcinoma(CCRCC)and their correlation with tumor angiogenesis.Methods Envision immunohistochemistry was used to determine the expression of COX-2 and VEGF,and microvessel density(MVD)was marked by CD34 in 80 CCRCC tissues and 20 normal kidney tissues.The relationship between the above mentioned markers were analyzed.Results Expressions of COX-2 and VEGF were noted in both CCRCC and normal kidney tissues.The positive rates of COX-2 and VEGF were significantly higher in CCRCC than in normal kidney(P <0.05);The expression of COX-2 was correlated with TNM stage(P <0.05),histological grade (P <0.05)and lymph node metastasis(P <0.05)in CCRCC,but not with age(P = 0.663)and diameter of tumor(P =0.528).Both COX-2 expression(r = 0.851,P < 0.01)and VEGF expression(r = 0.736,P < 0.01)were significantly associated with MVD in CCRCC.There was a positive correlation between expression of cox-2 and that of VEGF in CCRCC.Conclusion COX-2 expression is correlated with tumor angiogenesis in CCRCC.It is likely that VEGF is one of the most important mediators in the COX-2 angiogenic pathway.  相似文献
