目的:为了探讨人参、西洋参和三七中人参皂苷的资源含量,以确保人参、西洋参和三七中人参皂苷资源充分被利用,为开发以人参皂苷为主的创新药物提供科学数据。方法:采用高效液相色谱法对人参、西洋参和三七不同部位中人参皂苷的含量进行测定。色谱条件为:Agilent 1100 Series 高效液相色谱仪;色谱柱为德国 Nucleosil-C_(18)(4.6 mm×150 mm,5μm);流动相为水-乙腈梯度洗脱,流速1.5 mL·min~(-1);A为水,B为乙腈;梯度洗脱程序为:0~16 min,56%B;16~20 min,56%→100%B;20~38 min,100%B;38~45 min,100%→56%B;45~60 min,56%B;60~70 min,56%B。所有组分均70 min 内出完。检测波长203 nm,柱温35℃,灵敏度为0.02AUFS。线性关系考察r=0.9994;精密度试验 RSD=0.26%,平均回收率为99.88%,重现性试验 RSD=2.0%,分离度为R=3.042。结果:人参须根含有较高的人参皂苷 Rb_1(1.082%),人参茎叶则含有较高的人参皂苷 Rc(1.002%)、Re(3.430%)和 Rg_1(1.303%);西洋参根含有较高的人参皂苷 Rb_1(2.213%)和 Re(0.9188%),西洋参芦头中含有较高的人参皂苷 Rb_1(2.840%)和Re(1.224%);三七根含有较高的人参皂苷 Rb_1(2.163%)和 Rg_1(2.633%),三七芦头中含有较高的人参皂苷 Rb_1(4.376%)和 Rg_1(4.145%)。结论:高效液相色谱法分离、分析人参皂苷效果好、准确、迅速、简便,也可作为评价人参属植物质量的有效分析方法。建议对人参、西洋参和三七中含量较高的人参皂苷进行提取分离,直接用于创新药物的开发。  相似文献

抗心律失常新药关附甲素(Guan-fu base A,GFA)为一种 C_(20)-二萜类生物碱,1966年由我国首次从黄花乌头(Aconitum coreanum(Levl.)Raipaics)的块根中分离得到。GFA与三氟醋酐(TFAA)反应,迅速定量地生成关附甲素双-三氟乙酸酯(GFA-TFAA),其结构经 GC-MSD 分析得到证实。GC-ECD 法对 GFA 的检测线性范围宽(10—20000ng/ml,r=0.9984),血浆中平均回收率为97.52%。日间及日内差的变异系数分别为9%及7%以下。该法用于 GFA 的临床前药理研究。GFA 三种剂量静脉给药6小时内的家兔血药浓度范围是:0.338±0.014至13.459±5.392μg/ml(10mg/kg),0.133±0.038至2.546±0.480μg/ml(2mg/kg)以及45.25±10.34至535.86±24.36 ng/ml(0.4 mg/kg)。  相似文献

Presently available GC and HPLC methods for analysis of piracetam, require large samples and suffer from interference. A micro scale, isocratic high-performance liquid-chromatographic method is described for the determination of piracetam in plasma (25 μL) or cerebrospinal fluid (10 μL) using ultraviolet absorbance at 215 nm. The limit of quantitation is 4 μg mL?1 and the within-batch and between-batch coefficients of variation are less than 10%. No interference from other commonly prescribed antimyoclonic or antiepileptic drugs was observed and thus the method can be used to monitor piracetam in patients on polytherapy antimyoclonic or antiepileptic drug regimens. Because of the sensitivity and rapidity of the method it is suitable for pharmacokinetic and mechanistic studies and for analysis of paediatric samples.  相似文献

China has a great resource of Cannabis. Research on the taxonomy and morphology of Chinese Cannabis has been carried out, but so far no molecular genetic research has been published. Random amplifiedpolymorphic DNA (RAPD) is a suitable technique for molecular genetic research on Cannabis. In tills experiment,using Cannabis herbarium specimens as a source of genetic materials, the correlative conditions of the Polymerasechain reaction (PCR), (i.e., gradient density of Mg^2 , dNTPs, Taq DNA polymerase, anneal temperature, anneal timeand reaction cycles) were examined separately. 中国有丰富的大麻资源。目前,在我国对于大麻的研究多集中在形态和化学成分等方面,而分子遗传水平的研究至今尚未见报道。RAPD分子标记技术是适合于大麻分子水平研究的首选方法。本文以采集多年的大麻植物干标本的叶为研究材料,分别对Mg~(2 ), dNTPs, Taq DNA 酶的浓度,和退火温度和退火时间以及反应循环数等条件进行了检测,得到了适合大麻RAPD研究的理想条件。实验结果表明,用干标本来进行一定的分子遗传研究是可行的,且结果也相对稳定。  相似文献

Purpose. To develop and evaluate a simple, yet well defined, method to measure diffusion in semi-solids, i.e. polymeric materials. Methods. The method was based on a concept where equivalent discs of polymeric films were cut and stacked on top of each other. The diffusion process was allowed to proceed unidimensionaly through the stack of films perpendicular to the film surface. After an appropriate time, the stack was analysed disc by disc with respect to solute content and from the concentration profile so obtained the diffusion coefficient was calculated. Results. An all-in-one device was developed, manufactured in stainless-steel, that cuts circular discs and stores each one successively in a stack in the cell compartment. Conclusions. Data from a silicone based system shows that the method, although simple, is accurate and reproducible.  相似文献
