目的:探讨川芎嗪对培养的类风湿性关节炎(RA)活动期、静止期外周血单个核细胞(PBMC) IL- 12 m RNA表达的影响。方法:应用细胞培养技术,采用RT- PCR方法,检测细胞培养液IL - 12 m RNA表达水平。结果:RA患者活动期、静止期IL - 12高度表达(P <0 .0 5 ) ,川芎嗪可以抑制RA活动期PBMC IL - 12合成,并对活动期PBMCIL - 12 m RNA有抑制作用(P<0 .0 5 )。结论:川芎嗪可通过抑制RA患者中PBMCIL - 12 m RNA的表达,减轻RA进展。  相似文献

目的 探讨类风湿性关节炎(RA)患者血清免疫球蛋白(IgG、IgA、IgM)和补体(C3、C4)水平变化及其临床意义.方法 选择初诊RA患者41例,其中活动期27例,非活动期14例,并选择同期体检健康者20例作为对照组,采用免疫比浊法检测3组血清免疫球蛋白(IgG、IgA、IgM)和补体(C3、C4)水平.结果 RA活动组、RA非活动组血清IgG水平均高于对照组,差异均有统计学意义(P<0.01,0.05),RA活动组与RA非活动组血清IgG水平差异无统计学意义(P>0.05);RA活动组血清C3水平均高于RA非活动组和对照组,差异均有统计学意义(P <0.05,0.01),RA非活动组与对照组血清C3水平差异无统计学意义(P>0.05);3组间血清IgA、IgM、C4水平差异均无统计学意义(P>0.05).结论 血清IgG可作为RA的辅助诊断指标,血清C3可作为观察RA活动状态的指标.  相似文献

为了解类风湿关节炎(RA)患者血清蛋白电泳图谱的改变,采用HYDRASYSLC(SEBIA)琼脂糖凝胶电泳技术,测定124例RA(活动期80例,非活动期44例)和70名健康成人(对照组)的血清蛋白;同时在DADE BEHRING BNP全自动特种蛋白分析仪上采用散射比浊法测定其C反应蛋白(CRP)、免疫球蛋白G(IgG)、免疫球蛋白A(IgA)、免疫球蛋白M(IgM)的含量。结果显示,活动期和非活动期RA患者血清蛋白电泳特征均为白蛋白比例减少,α1、α2、γ球蛋白比例增高,活动期较对照组差异更加显著(P<0.01)。活动期血清中CRP、IgG、IgA、IgM的含量明显高于对照组(P<0.01),两组间有显著差异;非活动期较对照组差异不如活动期显著(P<0.05)。结论:RA患者血清蛋白电泳的改变主要表现在白蛋白降低,α1、α2、γ球蛋白的比例增高,γ球蛋白比例增高与CRP、IgG、IgA、IgM的增高有较大相关性。血清蛋白电泳检测对于RA的诊断虽没有特异性,但对于RA活动期判断及疗效观察有重要的参考价值。  相似文献

目的:探讨血清免疫球蛋白IgM、IgG、IgA及补体C3、C4与类风湿性关节(RA)活动度的关系,并探讨其与类风湿性关节炎的活动性指标血沉(ESR)、C反应蛋白(CRP)、类风湿因子(RF)滴度比较有无相关性。方法:120例初诊RA患者根据疾病活动度评分28(DSA28)分为低度活动组、中度活动组、高度活动组,设立健康对照组30例,采用速率散射比浊法检测所有实验者的血清IgM、IgG、IgA及补体C3、C4水平。结果:中重度活动组RA患者的IgA、IgM、C3与对照组相比差异有显著性,IgA、IgM、C3与RA实验室活动指标ESR、CRP、RF、PLT有直线相关关系。结论:中重度RA患者的血清IgA、IgM、C3与RA病情活动度有相关性,可作为RA病情活动的观察指标之一。  相似文献

我院于1980年12月,应用南大生物系与泰州生化制药厂制备的猪胸腺素组分5(PTHF_5)洽疗活动期系统性红斑狼疮(SLE)16例与干燥综合征合并典型或确定的类风湿性关节炎(SS+RA)或典型类风湿性关节炎(RA)19例。并以同期收治的,在性别、年龄、病变程度与活动性方面相似的SLE15例和SS+RA16例(两组皆为活动期病例)作为对照,以观察其疗效及对免疫功能的影响。  相似文献

[目的]探讨血清超敏C反应蛋白(hs-CRP)、血小板活化因子(PAF)及抗环瓜氨酸肽(CCP)抗体等对类风湿性关节炎(RA)活动期中的水平变化及临床意义.[方法]采用酶联免疫吸附法检测68例活动RA患者(A组)血清PAF含量及抗-CCP抗体,采用速率散射比浊法检测血清hs-CRP水平,并与70例非活动期RA患者(B组)、45例健康体检者(C组)比较.[结果]活动期RA患者血清PAF含量为(117.26±23.52)pg/ml,hs-CRP为(14.12±1 1.37)mg/L,明显高于健康对照组;与非活动期组RA患者比较,CCP抗体阳性率明显较高,血清PAF水平差异有极显著性(Jp<0.01),但hs-CRP水平差异无显著性(P>0.05).[结论]活动期RA患者血清PAF、hs-CRP及CCP抗体水平明显升高,可作为反映RA患者风湿活动的评估指标,但hs-CRP对评估活动期RA与非活动期RA效果不明显,无明显鉴别诊断意义.  相似文献

目的分析类风湿性关节炎(RA)疾病特点,评价免疫球蛋白[免疫球蛋白A(IgA)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)]、抗血清抗环瓜氨酸肽(CCP)抗体在RA病理进展中的诊断价值。方法选取80例RA患者作为观察组,另选取同期健康体检者80例作为对照组;观察组RA患者根据28个关节疾病活动度评分(DAS28评分)分为高度组(>5.1分,15例)、中度组(3.2~5.1分,25例)、低度组(2.6~3.1分,20例)以及缓解组(<2.6分,20例)。测定所有受检者血清中抗CCP抗体、免疫球蛋白(IgA、IgG、IgM)水平。比较观察组和对照组血清免疫指标(抗CCP抗体、IgA、IgG、IgM)水平,不同活动度RA患者免疫指标水平。结果观察组患者的抗CCP抗体、IgA、IgG、IgM水平分别为(64.50±19.80)U/ml、(4.70±0.99)g/L、(18.28±5.05)g/L、(4.06±1.02)g/L,均高于对照组的(8.80±2.30)U/ml、(1.28±0.45)g/L、(10.58±3.58)g/L、(1.18±0.40)g/L,差异具有统计学意义(P<0.05)。高度组、中度组、低度组以及缓解组患者的血清抗CCP抗体、IgA、IgG、IgM水平均呈降低趋势,且组间两两比较差异具有统计学意义(P<0.05)。结论RA患者血清中免疫球蛋白、抗CCP抗体水平升高明显,与病情活动度关系密切,是RA疾病早期诊治的主要监测指标。  相似文献

目的探讨口服雷公藤(Tripterygium Wilfordii Polyglycosidium,TWP)治疗大鼠类风湿性关节炎(rheumatoid arithritis,RA)的效果。方法通过建立大鼠类风湿性关节炎模型,对正常对照组、致病组、TWP治疗组大鼠关节炎症指数、影像学、病理学等相关指标进行评估。结果口服雷公藤能明显降低类风湿性关节炎的关节评分;能明显抑制大鼠的继发性多关节炎的关节症状,还可明显减少RA大鼠的关节外表现。结论口服雷公藤能有效减轻类风湿性关节炎大鼠的关节病变,具有很好的抗炎作用。  相似文献

目的检测三型类风湿因子(RF)定量指标,分析与抗环瓜氨酸多肽抗体(ACCP)及炎性指标血沉(ESR)、C反应蛋白(CRP)的关系,探讨其对类风湿性关节炎(RA)诊断及病情评价的意义。方法选择2007年3月至7月我科住院患者50例,其中RA患者30例,非RA结缔组织病患者20名,同时选择健康成人20例为对照。以速率散射比浊法定量测IgM型类风湿因子(IgM-RF),ELISA法检测IgA及IgG型类风湿因子(IgA-RF及IgG-RF)采用方差分析及相关分析进行统计学分析。结果RA患者、非RA结缔组织病患者及正常人中三型RF检测水平差别有统计学意义,P<0.05。RA患者中IgM-RF及IgG-RF与ACCP、ESR及CRP水平呈明显正相关,IgA-RF与ACCP及CRP水平呈明显正相关,P<0.05。结论三型类风湿因子对RA诊断有意义,可协助RA的早期诊断,并对疾病病情评估有意义。  相似文献

Serum and cerebrospinal fluid (CSF) of 45 patients with Alzheimer's disease (AD) and 24 healthy controls were examined in order to investigate the possibility of an intrathecal synthesis of immunoglobulins in AD. The group of AD patients showed higher IgG and IgM indices than the control group. Five (11%) AD patients had an intrathecal IgG synthesis and 7 (15%) an intrathecal IgM synthesis. In total, 12 (26%) AD patients had an intrathecal immunoglobulin synthesis, while there was none among the controls. This finding supports the hypothesis that immunological mechanisms may play a role in the etiology and/or pathogenesis of Alzheimer's disease, at least in a subgroup of patients.  相似文献

Macrophage migration inhibitory factor (MIF) has a multitude of biological activity and is associated with a number of inflammatory and immune diseases, including rheumatoid arthritis (RA). The increased serum levels of MIF in patients not only suggest this protein as a marker for disease progression, but also as a potential therapeutic target. The aim of this study is to develop a novel electrochemical method to more precisely and conveniently measure MIF in patient sera. An IgM murine monoclonal antibody (mAb) against human MIF was prepared and used in the electrochemical immunosensor, with modified gold electrode coated with compounds of gold nanoparticles, titanium dioxide nanoparticles and thionine (NGP-NTiP-Thi) followed by adsorption of anti-MIF antibodies with IgM or IgG1 isotype. The IgM immunosensor recognized MIF in a linear relationship in the range of 0.03 and 230 ng/mL with the lower limit (S/N=3) of 0.02 ng/mL. The measurement showed considerable levels of sensitivity, selectivity, stability and long-term maintenance of bioactivity, as shown by testing with serum MIF in RA patients as compared to healthy donors. The performance of the IgM immunosensor was also superior to IgG1 sensor. Thus, we have developed a novel measurement approach for serum MIF, which may have great potential in the clinic for monitoring the course of diseases associated with increased MIF.  相似文献

Lenalidomide (len) is an analog of thalidomide (thal), and both are used in the treatment of a diverse group of medical conditions. A common finding in this group is the detection of immunoglobulin in skin lesions, or high levels of immunoglobulin or myeloma protein in serum and urine. While their mechanism(s) of action is not known, the drugs are noted for their ability to modulate monocyte, lymphocyte, and natural killer cell functions; suppression of immunoglobulin synthesis could offer an explanation for their effectiveness in treating multiple myeloma (MM). Our objective was to determine if, on an equimolar basis, thal, len or dexamethasone (dex) could affect pokeweed (PWM)-induced synthesis of IgG, IgM and IL-2. When peripheral blood mononuclear cells were stimulated with PWM, len surpassed thal in suppressing IgM and IgG, and enhancing IL-2. Dex enhanced IgG, and suppressed IL-2. When the stimulated cells were treated with len (an effective promoter of IL-2 and suppressor of IgM and IgG) plus dex (an effective suppressor of IL-2 and enhancer of IgG), the net result was suppression of IgM and IgG. The synthesis of IgM and IgG by putative PWM-stimulated B cell blasts is significantly blocked by len. This suggest that the B-lymphocyte is a targeted cell for len, and that suppression of the synthesis of IgG and IgM could provide an explanation for the mechanism by which len effectively treats MM.  相似文献

目的观察类风湿关节炎灭活滑膜液单个核细胞对非灭活滑膜液单个核细胞分泌IL-10水平的影响。方法抽取类风湿关节炎(RA)患者关节液,分离关节液单个核细胞,分为对照组:2×10~6的非灭活单个核细胞,培养条件为37℃,5%CO_2孵育箱培养48h;实验组:细胞数为2×10~6个非灭活单个核细胞与甲醛灭活后的单个核细胞按1:10混合培养.用ELISA法测定培养上清中IL-10的水平.结果实验组较对照组IL-10水平明显升高(P<0.01).结论灭活滑膜液单个核细胞对非灭活滑膜液单个核细胞IL-10分泌有促进作用,提示未经克隆选择的RA滑膜液混合细胞可能具有诱导分泌IL-10CD_4调节性T细胞的生成作用.  相似文献

目的 观察脂多糖(LPS)对类风湿关节炎(RA)患者外周血单核细胞Toll样受体4(TLR4)、髓样分化因子88(MyD88)蛋白表达的影响。方法 采用活动期RA患者外周血进行单核细胞的分离培养,并与健康志愿者做对照。分为以下5组观察:健康对照组、健康+LPS组、RA对照组、RA+ LPS组、RA+LPS+TAK-242组,并采用Western blot法检测各组TLR4、 MyD88蛋白的表达。结果 健康+LPS组或RA+LPS组TLR4、MyD88蛋白表达与健康对照组或RA对照组比较均明显升高(均P<0.01);RA对照组或RA+LPS组TLR4、MyD88蛋白表达与健康对照组或健康+LPS组比较均明显升高(均P<0.01);RA+LPS+TAK-242组TLR4、MyD88蛋白表达与RA+LPS组比较明显降低(均P<0.01)。结论 RA活动期单核细胞可能存在TLR/MyD88信号通路的激活,且对LPS的刺激反应性增强。  相似文献

Trachoma is recognized as one of most important origins of blindness in developing countries and inclusion conjunctivitis is associated with STD in developed countries. We evaluated the diagnostic value of serological tests for the screening of eye diseases associated with Chlamydia trachomatis infection. We determined serum IgG, IgA and IgM antibodies to C. trachomatis from 53 Japanese patients with active inflammatory trachoma (aged more than 60 years) and from 107 adult patients (aged 20 to 50 years) with acute inclusion conjunctivitis by ELISA test kit. We detected serum IgG antibodies from 22 out of 53 (42.5%) patients with trachoma and from 40 out of 107 (37.4%) patients with acute inclusion conjunctivitis. We also detected serum IgM antibodies from 7 out of 53 (13.2%) patients with trachoma and from 35 out of 107 (32.7%) patients with acute inclusion conjunctivitis. The prevalence of serum IgM antibodies to C. trachomatis in patients with acute inclusion conjunctivitis was significantly higher than that in patients with active trachoma (p < 0.05). Serological tests are also thought to be useful for screening of chlamydial eye diseases.  相似文献

目的:动态监测SARS患者恢复期抗SARS病毒IgG和IgM抗体变化,探讨其临床意义。方法:采用酶联免疫吸附测定法(ELISA),检测SARS患者发病后5,10,14个月末血清IgG、IgM抗体水平,并与临床诊断进行比较。同时观察感染SARS后抗体的持续维持水平及效能。结果:IgG诊断与临床诊断符合程度不一致。随访中3个时期患者恢复期IgG抗体整体水平下降,部分患者IgG抗体阴转,随访后期少数患者IgM抗体水平仍为阳性。结论:SARS患者的血清学IgG抗体检测诊断与临床诊断不一致,SARS病毒抗体变化规律与一般传染病抗体产生规律不同。  相似文献

An important effect of the trichothecene mycotoxins is the impairment of the immune function, but immunotoxicity studies have mainly been conducted on the mouse model. In the present study, the effect of deoxynivalenol (DON) on the proliferation of ConA stimulated porcine peripheral blood lymphocytes (PBL) was assessed in vitro after adding of 70-560 ng DON per ml medium, and in vivo after chronic and acute (one single dose) dietary DON exposure (5.7 mg/kg). Immunoglobulin (IgA, IgG, IgM) concentrations were measured by ELISA in supernatants and serum of pigs. The proliferation rate was estimated with two different assays (BrdU incorporation and MTT cleavage). In vitro the ConA stimulated proliferation was inhibited to 50% (IC50) at 200 and 309 ng DON/ml for the BrdU and MTT assay, respectively, indicating a higher sensitivity of DNA synthesis to DON. Immunoglobulin concentrations in the supernatant after in vitro proliferation of PBL with increasing DON concentrations for 72 h were significantly decreased, with IC50 values of 120.6, 84.1 and 71.7 ng DON/ml for IgA, IgM and IgG, respectively. In vivo significant inhibition of lymphocyte proliferation was observed only in the DON acute group using the MTT assay, but values tended to be decreased in the BrdU assay and after chronic DON exposure. Immunoglobulins (IgA, IgM and IgG) in the supernatant of cultured lymphocytes were not significantly affected after dietary DON exposure. Serum IgA of pigs showed no significant differences between the groups, whereas IgM and IgG were significant increased in the DON acute group.  相似文献
