涎腺肿瘤中细胞凋亡及其调控基因的表达   总被引:3,自引:0,他引:3  
目的:探讨涎腺肿瘤中细胞财亡及调控基因p53、bcl-2、c-myc间的关系。方法:采用脱氧核糖核酸末断转移酶介导的dUTP缺口末端标记(TUNEL)技术,免疫组织化学染色和HE染色对18例多形性腺瘤和52例恶性涎腺肿瘤中细胞凋亡和bcl-2、p-53、c-myc蛋白表达进行观察和比较。结果:恶性涎腺肿瘤的凋亡细胞指数明显高于多形性腺瘤(P<0.05),恶性涎腺肿瘤中bcl-2、p53和c-myc蛋白表达率及表达强度明显高于多形性腺瘤(P<0.05)。结论:细胞凋亡的调控基因异常在涎腺肿瘤中可能起重要作用,bcl-2和c-myc蛋白的共同作用可抑制细胞凋亡。  相似文献

[目的]观察青蒿琥酯对人乳腺癌MCF-7裸鼠移植瘤生长的作用,并探讨其抑制肿瘤的机制。[方法]建立人乳腺癌MCF-7裸鼠移植瘤模型,给予不同剂量青蒿琥酯治疗并观察其对移植瘤的抑制作用;电镜观察移植瘤细胞形态学改变;流式细胞术检测细胞凋亡率和细胞周期的变化:免疫组化检测移植瘤细胞p53、bcl-2、bax和caspase-3的表达情况,分析它们之间的相关性;Westernblot检测IGF—IR蛋白的表达:[结果]给药12d后,低剂量、高剂量青蒿琥酯组、CTX组和联合给药组抑瘤率分别为(24.39±10.20)%、(40.24±7.02)%、(57.01±5.84)%和(68.29±5.1)%;青蒿琥酯使肿瘤细胞阻滞于G0/G1期而使S期细胞减少;在青蒿琥酯组中,bcl-2的蛋白表达量明显下降,bax、caspase-3蛋白表达上调,p53蛋白表达量与对照组比较无差异:bcl-2/bax及bcl-2/caspase-3表达均呈负相关.IGF-IR蛋白表达下调。[结论]青蒿琥酯可显著抑制乳腺癌MCF-7细胞裸鼠移植瘤的生长,其机制可能与其影响细胞凋亡相关蛋白、诱导凋广、抑制细胞增殖有关.  相似文献

乳腺癌bcl-2表达与患者预后相关因素关系的研究   总被引:4,自引:2,他引:4  
目的:研究乳腺癌中bcl-2蛋白表达与患者生存期及其组织学分级、雌激素受体(ER)、p53、c-erbB-2等影响预后因素的分析。方法:采用免疫组化LSAB法检测bcl-2、ER、p53及c-erbB-2的表达,并分析其相关性及其与预后的关系。结果:bcl-2阳性表达率为50.27%(92/183),其表达与乳腺癌组织学分级、ER状态呈明显正相关,与p53及c-erbB-2表达呈明显负相关;bcl-2阴性者其总生存率及无瘤生存率均明显低于bcl-2阳性者。结论:bcl-2蛋白阴性的乳腺癌患者预后不好,这可能与bcl-2失表达肿瘤中,p53蛋白聚积、c-erbB-2过度表达以及ER的缺失有关;免疫组化检测bcl-2表达可为预测乳腺癌患者预后及选择内分泌治疗提供一个有实用价值的指标。  相似文献

目的研究p53mRNA及bcl-2、bax蛋白在直肠癌组织中的表达及其意义。方法应用组织芯片技术制作正常直肠、腺瘤组织标本各16例及80例直肠癌石蜡标本的组织芯片,采用原位分子杂交检测p53 mRNA,免疫组织化学SP法检测bcl-2、bax蛋白,脱氧核糖核苷酸末端转移酶介导的缺口末端标记技术(TUNEL)检测直肠癌细胞的凋亡指数。结果正常直肠组织平均p53mRNA阳性细胞率为15.26%±3.24%,腺瘤组织平均阳性细胞率为27.32%±5.54%,直肠癌组织平均阳性细胞率为75.38%±9.61%,三者比较,差异有统计学意义(P<0.01),直肠癌患者不同年龄、性别、肿瘤部位、大小、血清CEA水平、组织学类型的p53表达程度差异无统计学意义(P>0.05);不同分化程度、病理分期、淋巴转移、5年生存率的p53表达程度差异有统计学意义(P<0.05);直肠癌组织bcl-2蛋白表达水平明显高于正常直肠组织和腺瘤(P<0.01),bax蛋白表达水平低于正常直肠组织和腺瘤(P<0.01),直肠癌组织p53mRNA阳性细胞率与bcl-2蛋白表达呈正相关(P<0.01),与bax蛋白表达呈负相关(P<0.01);直肠癌p53mRNA表达程度不同的直肠癌细胞凋亡指数(AI)差异有统计学意义(F=49.85,P<0.01)。结论直肠癌中p53及bcl-2、bax的异常表达共同参与了肿瘤的发生发展和浸润转移,联合检测上述标志物有一定的临床意义。  相似文献

目的:探讨p16,bcl-2,bax蛋白在脑星形细胞瘤中的表达及其意义。方法:采用S-P免疫组织化学法,检测P16,bcl-2,bax蛋白在60例不同病理分级脑星形细胞瘤组织中的表达情况。结果:60例脑星形细胞瘤组织中,p16,bcl-2,bax蛋白表达阳性率分别为50.0%(30/60),58.3%(35/60),66.7%(40/60),其中P16蛋白阳性表达与病理分级有显著性相关(P<0.05),bcl-2,bax蛋白表达与病理分级无相关性(P>0.05),60例脑星形细胞瘤组织中,p16,bcl-2,bax蛋白共同表达率,I级为47.8%(11/23),Ⅲ-Ⅳ级为0(0/16),在bcl-2,bax蛋白表达的基础上,不同病理分组织中p16蛋白的缺失率:I级为4.3%(1/23),Ⅲ-Ⅳ级为50.0%(8/16),有非常显著性差异(P<0.01),结论:P16蛋白表达与脑星形细胞瘤的发生发展密切相关,在脑星形细胞瘤的发展过程中,bcl-2,bax,p16蛋白可能有某种协调作用。  相似文献

bcl-2、bax、c-myc和p53蛋白在卵巢上皮性肿瘤中的表达   总被引:1,自引:0,他引:1  
目的:研究bcl-2、bax、c-myc和p53基因的蛋白产物在卵巢上皮性肿瘤中的表达,相互关系及其意义。方法;应用免疫组化方法,以抗bcl-2、bax、c-myc、p53的单克隆抗体为一抗,对86例卵巢上皮性肿瘤组织进行检测。同时选择12例正常卵巢组织作对照组。结果:bcl-2、bax、c-myc和p53在卵巢上皮良性、交界性、恶性肿瘤中的阳性表达率分别为6.25%、10.23%、39.28%;21.88%、50.00%、53.57%;6.25%、61.54%、64.28%和3.13%、53.85%、60.71%;而正常卵巢组织中均为阴性。结论:四个基因均以不同的作用方式参与了卵巢上皮性肿瘤的发生、发展过程,具有不同的生物学特性,并对该肿瘤的病情发展、疗效评价和预后估计均有一定意义。  相似文献

目的:探讨p21、p27、bcl-2、bax蛋白在肺癌中的表达及其临床意义.方法:采用免疫组化S-P法,检测85例肺癌患者及21例良性肺疾病患者术后组织标本中p21、p27、bcl-2、bax的表达.结果:p21、p27、bcl-2、bax蛋白总阳性表达率分别为72.94%、37.65%、61.18%、41.18%.各种蛋白的阳性表达率在肺癌组织与对照组间均存在差异性(P<0.05).仅bcl-2蛋白表达与肺癌的不同病理类型有关,其中在小细胞肺癌中表达率最高、鳞癌次之、腺癌最低.p27、bcl-2、bax三者的阳性表达随病理分化程度的降低而降低(P<0.05).p21与bcl-2蛋白的阳性表达与肺癌临床TNM分期有关(P<0.05).bax蛋白的表达与有无淋巴结转移有关.p27、bcl-2、bax三者之间存在相关性.结论:p21、p27、bcl-2、bax蛋白与肺癌的发生、发展有密切关系.联合检测p21、p27、bcl-2、bax对于临床诊断肺癌及判断病情、预后有重要意义.  相似文献

目的:探讨多西紫杉醇联合曲格列酮(troglitazone,TGZ)对人前列腺癌裸鼠移植瘤的抑制作用,并初步探索其作用机制.方法:建立人前列腺癌裸鼠移植瘤模型,观察多西紫杉醇联合TGZ对移植瘤生长的抑制作用,FCM法检测移植瘤细胞的凋亡率,免疫组织化学法检测移植瘤组织中bcl-2和bax蛋白的表达情况.结果:与对照组比较, 多西紫杉醇联合TGZ可明显抑制人前列腺癌裸鼠移植瘤的生长,肿瘤体积和质量明显下降,细胞凋亡率明显上升;免疫组织化学法检测结果显示,移植瘤组织中bax蛋白的表达明显增强,bcl-2蛋白的表达明显减弱.结论:多西紫杉醇联合TGZ对前列腺癌有一定的抑制作用,其作用机制可能是通过上调促凋亡蛋白bax的表达及下调抗凋亡蛋白bcl-2的表达,导致肿瘤细胞加速凋亡而实现的.  相似文献

目的:探讨bcl-2、Ki-67、p53蛋白在乳腺癌中的表达及其关系。方法:应用免疫组化SP方法,观察56例乳腺癌中的bcl-2、Ki-67、p53的表达情况。结果:bcl-2、Ki-67和p53在乳腺癌中表达率分别为63%,68%,52%,与乳腺癌分级及淋巴结转移密切相关(P<0.05)。结论:bcl-2、Ki-67、p53的异常表达在乳腺肿瘤发生发展中起重要作用。  相似文献

[目的]研究金银花多糖对S180肉瘤的抑制作用与机制。[方法]建立小鼠S180实体瘤模型,分别给予低、中、高剂量金银花多糖,测定各组小鼠肿瘤生长抑制率、脾脏指数和胸腺指数;采用免疫组织化学法测定肿瘤组织中Bax和Bcl-2蛋白表达;酶联免疫吸附实验法(ELISA)测定小鼠血清肿瘤坏死因子(TNF-α)的含量。[结果]中、高剂量的金银花多糖对S180肉瘤的抑制率分别为23.95%和30.02%,与模型组比较有显著性差异(P<0.05);高剂量的金银花多糖可以显著提高荷瘤小鼠的脾脏指数;金银花多糖可以上调肿瘤组织中的Bax、Bcl-2蛋白表达;中、高剂量的金银花多糖实验组小鼠血清TNF-α的含量显著高于模型组小鼠(P<0.05)。[结论]金银花多糖具有抑制肿瘤生长的作用,并且对荷瘤小鼠的生长和免疫功能没有明显不良影响;其抑瘤的机制可能与调节Bcl-2/Bax凋亡通路及促进TNF-α的分泌有关。  相似文献

目的:探讨树突状细胞(DC)荷载α-半乳糖神经酰胺(α-GalCer)联合肿瘤特异性细胞毒 T 淋巴细胞(CTL)对小鼠 Heps 肝癌移植瘤生长的抑制作用。方法:诱导扩增小鼠骨髓来源的 DC 细胞和 T 淋巴细胞,培养成为具有肿瘤特异性的 CTL,DC 细胞体外荷载α-GalCer。建立 Heps 肝癌移植瘤模型,将36只模型鼠随机分为4组(n =9),分别尾静脉给予生理盐水(对照组)、CTL(CTL 组)、DC 荷载α-GalCer(DC 荷载α-GalCer 组)、DC 细胞荷载α-GalCer +CTL(联合治疗组)输注。每隔两天测量移植瘤体积,观察体积变化。于治疗后第14d,处死小鼠,取眼球血及瘤体组织。流式细胞术(FCM)检测外周血 CD4+、CD8+ T 细胞比例。免疫组织化学染色观察肿瘤组织中 Bcl -2/Bax 凋亡蛋白的表达。结果:与对照组相比,各治疗组均可抑制肿瘤的生长,差异具有统计学意义(P <0.01);联合治疗组较 DC 荷载α-GalCer 组及 CTL 组肿瘤体积明显减小(P <0.05)。联合治疗组小鼠外周血 CD4+、CD8+ T 细胞数量较另外三组显著升高(P <0.05);对照组、CTL组、DC 荷载α-GalCer 组小鼠外周血中 CD4+、CD8+ T 细胞含量无明显差异(P >0.05)。与对照组相比,各治疗组移植瘤内 Bax 阳性细胞数量明显增加(P <0.05),Bcl -2阳性细胞数量明显减少(P <0.05);与 CTL组和α-GalCer 组相比,联合治疗组移植瘤内 Bax 阳性细胞明显增加(P <0.05),Bcl -2阳性细胞明显下降(P <0.05)。结论:DC 荷载α-GalCer 与肿瘤特异性 CTL 联合应用能够对小鼠 Heps 肝癌移植瘤具有协同杀伤作用。  相似文献

目的:探讨多烯紫杉醇联合TNF-α抑制前列腺癌的作用,并进一步了解其作用机理。方法:建立人前列腺癌细胞的裸鼠皮下移植瘤模型,在多烯紫杉醇和TNF—α的联合作用下观察其对皮下移植瘤生长的抑制作用,并通过RT—PCR、免疫组化方法检测Bax、Bcl-2基因mRNA表达及p53蛋白表达。结果:与多烯紫杉醇组、TNF—α组及对照组相比,多烯紫杉醇和TNF—α联合治疗组可抑制前列腺癌皮下移植瘤生长速度,且肿瘤体积明显减少(P〈0.01)。BaxmRNA表达上调,而Bcl-2mRNA表达水平下降,免疫组化结果表明p53蛋白的表达水平下降。结论:多烯紫杉醇联合TNF—α对前列腺癌有一定的抑制作用,作用机理可能是通过引起促凋亡基因Bax表达增高,从而导致肿瘤细胞加速凋亡而实现的。  相似文献

应用免疫组化SP法观察47例人脑胶质瘤中Bcl-2和p53基因蛋白的表达。结果发现Bcl-2蛋白阳性率为14.9%(7/47),p53蛋白的表达率为36.2%(17/47),表达与肿瘤组织学分级和肿瘤复发有关。反应性胶质细胞增生和正常脑组织中bcl-2和p53均为阴性。结果表明bcl-2基因在脑胶质瘤发生过程中的作用有限;p53基因异常表达与胶质瘤的发生有关,且与肿瘤的恶性度和预后有关;Bcl-2和p53两种癌基因蛋白表达无明显关系。  相似文献

PURPOSE: The aim of the current study is to compare clear cell ovarian carcinoma (CCOC) and papillary serous ovarian carcinoma (PSOC) with respect to their clinical features and expression of different regulators of cell cycle, apoptosis, and chemoresistance. EXPERIMENTAL DESIGN: Women with stage III CCOC (n = 9) and those with stage III, poorly differentiated PSOC (n = 21) seen between 1996 and 2000 and treated with cytoreductive surgery followed by paclitaxel and platinum chemotherapy were compared in their demographic features, tumor marker profile, surgical substage, results of cytoreductive surgery, thromboembolic complications, response to chemotherapy, and tumor recurrence. Tumor samples were compared in their expression of p53, Bcl(2), Bcl(x), Bax, p21, p-glycoprotein (PGP), multi-drug resistance-associated protein (MRP), lung resistance protein (LRP), and glutathione S-transferase (GST) using immunohistochemistry. RESULTS: Women with CCOC had significantly lower mean preoperative CA-125 values, lower surgical substage, less expression of p53, and more expression of p21 than women with PSOC (P = 0.037, 0.012, 0.008, and 0.009, respectively). Women with CCOC had less ascites, smaller amount of residual tumor, higher incidence of thromboembolism, chemoresistance, more expression of Bcl(2), and less expression of PGP than women with PSOC (P = 0.067, 0.078, 0.108, 0.114, 0.091, and 0.118, respectively). CONCLUSIONS: Women with CCOC exhibit certain clinical and molecular differences compared to stage- and grade-matched women with PSOC. Women with CCOC have a smaller tumor volume and manifest different expressions of p53, p21, and Bcl(2) than women with PSOC. Although further studies with larger number of patients are needed, our findings indicate that chemoresistance in CCOC is probably not p53-dependent.  相似文献

Radiation is known to induce DNA damage resulting in the onset of apoptosis. The apoptosis is modulated by p53, Bcl2 and Bax proteins. High level of wild type p53 is required for radiation induced apoptosis. The p53 status, therefore, may be a crucial determinant of radiosensitivity of tumor cells. Overexpression of Bcl2, however, inhibits apoptosis via hetero- and homodimeric interaction. Bax might function as a cell death effector molecule that is neutralized by Bcl2. The aim of the present study is to investigate the correlation between p53, Bcl2, Bax and c-myc levels and the clinical response of head and neck cancer patients to radiation. The base line and 30 GY gamma radiation induced values of p53, Bcl2, Bax and c-myc were estimated by Western blot in 40 biopsies of head and neck cancers. We found that the radiosensitivity of head and neck cancer patients depends on the ratio of p53, Bcl2 and Bax protein levels. High Bcl2 levels resulted in radioresistance of cancer patients. Overexpression of Bax and c-myc may ensure the radiosensitivity of head and neck cancer patients. Our studies indicate that prediction of radiation sensitivity of tumors could be based on the simultaneous evaluation of p53, Bax and Bcl2 levels.  相似文献

目的 探讨四硫化四砷(As4S4)对卵巢癌裸鼠皮下移植瘤的生长和瘤细胞凋亡的影响。方法 建立卵巢癌SKOV3裸鼠皮下移植瘤模型,观察As4S4对肿瘤生长的抑制情况;RT-PCR法检测As4S4对卵巢癌裸鼠皮下移植瘤组织中bcl-2和bax mRNA表达水平的影响;Western blot法检测As4S4对卵巢癌裸鼠皮下移植瘤组织中bcl-2和bax蛋白表达的影响。结果 As4S4作用于卵巢癌裸鼠皮下移植瘤后,肿瘤的体积和重量明显减小,而且随药物剂量的增加抑瘤率明显增加。As4S4作用后,RT-PCR可见bcl-2 mRNA表达强度减弱,bax mRNA表达强度增强;Western blot法可见bcl-2蛋白带变细,bax蛋白带增粗。结论 As4S4可以抑制卵巢癌裸鼠皮下移植瘤的生长,诱导肿瘤细胞凋亡,其作用机制可能与调节Bcl-2/Bax的表达有关。  相似文献

BACKGROUND: Mediastinal teratomas are the most frequent mediastinal germ cell tumor. Whereas mature teratomas are benign tumors, immature teratomas are malignant. The purpose of this study was to find characteristics that could be used to distinguish between the growth and prognosis of the two teratoma types. METHODS: Twenty-four mediastinal teratomas (18 mature and 6 immature) were examined for apoptosis by 3'-end labeling of DNA and stained immunohistochemically for proliferating cell nuclear antigen, Bcl-2, Bax, p53 protein, and alpha-fetoprotein (AFP) expression in formalin fixed, paraffin embedded specimens. RESULTS: AFP was expressed in both immature teratomas and mature teratomas. Whereas p53 protein was expressed by most teratomas, p53 gene mutation was observed in only one patient with an immature teratoma in which the same mutation occurred in all tumor tissue components tested. Bax protein expression was relatively diffuse in mature teratomas but was focally expressed in immature teratomas. Bcl-2 protein was expressed focally in both mature and immature teratomas. Although the proliferative index was significantly higher in immature teratomas compared with mature teratomas (P < 0.001), the apoptotic index (AI) was significantly higher in mature teratomas compared with immature teratomas (P < 0.05). All patients except one in this study remain alive and disease free after undergoing tumor resection. CONCLUSIONS: The relatively high AI in mature teratomas may be due to the overexpression of the p53 protein. In contrast, immature teratomas exhibited higher proliferative activity and lower rates of apoptosis, which may explain the more aggressive behavior of these tumors. However, patients with immature mediastinal teratomas have a good prognosis if the tumor is resected completely after chemotherapy.  相似文献

目的:探讨光动力作用(PDT)对人乳腺癌细胞Bax和Bcl-2的影响。方法:体外培养的人乳腺癌细胞MDA—MB-231,分别在PDT作用不同时间后,用免疫组织化学法检测促凋亡蛋白Bax和抗凋亡蛋白Bcl-2蛋白表达情况。结果:Bax蛋白表达在所有组无明显差异(P〉0.05),Bcl-2蛋白表达自PDT后6h起显著下降(P〈0.01);PDT后6hBax/Bcl-2比值依次递增,且有统计学意义(P〈0.05)。结论:光动力作用可诱导人乳腺癌细胞发生凋亡,其机制可能与Bax和Bcl-2蛋白表达的比值有关。  相似文献

This study describes the incidence of Bax protein expression in a series of 106 cases of breast cancer including 56 cases of ductal carcinoma in situ (DCIS) and 50 cases of invasive ductal carcinoma (IDC). Relationships of Bax expression to the histological grades of DCIS & IDC, and to the expression of Ki67, ER, p53, cerbB2 & Bcl2 are described. The expression of Bax, Ki67, ER, p53, cerbB2 and Bcl2 proteins is determined immunohistochemically. Cases were regarded positive for Bax, Bcl2 and cerbB2 when they showed either moderate or strong staining for these markers. The nuclear stains (Ki67, ER, and p53) were quantified in terms of percentage positive cells and cases for ER and p53 were considered positive when more than 10% cells were labelled. DCIS were graded histologically as well (n=18), intermediately (n=18), and poorly differentiated (n=20) Invasive ductal carcinoma was graded as grade I (well-differentiated) n=7, grade II (intermediate) n=24 and grade III (poorly differentiated) n=19. 65/106 cases (61%) were Bax positive including 37/56 (66%) of DCIS and 28/50 (56%) of IDC. Bax expression did not correlate to increasing histological grades of either DCIS or IDC. It did not correlate to Ki67, ER, p53 or cerbB2 but positive correlation was seen with Bcl2 (p=0.003). Bcl2 immunostaining displayed a negative correlation with increasing histological grades both of DCIS and IDC (p=0.026), (p=0.041) respectively. There was a trend of negative correlation of Bcl2 with Ki67 (p=0.062). It correlated positively with Bax (p=0.003) and ER (p<0.0001). Results suggest that the regulation of apoptosis is important in ductal carcinoma in situ of the breast as well as invasive ductal carcinomas. Bcl2 is associated with good prognostic markers in both DCIS and IDC, whereas the regulation of Bax is complex and does not necessarily correlate with mutant p53.  相似文献
