Isomer-specific concentrations of polychlorinated biphenyls (PCBs) including planar, mono- and di-ortho congeners and concentrations of DDT were determined in striped dolphins affected by a morbillivirus epizootic in the western Mediterranean in 1990. Extremely high concentrations of PCBs ranging from 94 to 670 g/g (wet wt) were detected in the blubber. Similarly, DDT concentrations were high, between 22 and 230 g/g (wet wt). The concentrations of three non-ortho coplanar PCBs were 43 (3,3,4,4-T4CB), 6.8 (3,3,4,4,5-P5CB), and 7.8 (3,3,4,4,5,5-H6CB) ng/g (wet wt), respectively, the highest residue levels reported to date. The estimated 2,3,7,8-TCDD toxic equivalents of non-, mono- and di-ortho PCB congeners in striped dolphins were several times higher than those observed for other marine mammals and humans. Mono-ortho congeners contributed greater 2,3,7,8-TCDD toxic equivalents than non-ortho members. The higher ratio of 3,3,4,4,5,5-H6CB/3,3,4,4,5-P5CB (IUPAC 169/126) suggested a strong induction of mixed function oxidase enzymes and highlighted the possibility of using this ratio as an index for risk assessment of PCB contamination in marine mammals. Elevated concentrations of PCBs may have played a role in the immune depression in striped dolphins, ultimately leading to the development of morbillivirus disease.  相似文献

The emissions from 2- and 4-stroke motorcycles pollute the air of urban areas where motorcycle is a popular means of transportation. This study aimed to determine the endocrine-disrupting activity of motorcycle exhaust particulate (MEP) using MCF-7 human breast cancer cells and immature female Wistar rats treated with organic extracts of MEP. Treatments with 1, 10, and 50 g/ml MEP extract for 2 and 4 days produced dose-dependent inhibition of thymidine incorporation and cell growth, respectively, in untreated and 1 nM 17-estradiol (E2)-treated cells. Treatments of MCF-7 cells with MEP extract replaced [3H]E2 from the estrogen receptor in a time- and concentration-dependent manner. These antiestrogenic and receptor binding properties of MEP extract were blocked by cotreatment of the cells with 2 M -naphthoflavone, a cytochrome P450 inhibitor and aryl hydrocarbon receptor antagonist. E2 metabolism and HPLC analysis showed that treatment of MCF-7 cells with 50 g/ml MEP extract for 24 h increased E2 2- and 4-hydroxylation in microsomes. The MEP-mediated increase in E2 2-hydroxylation was inhibited by the addition of 1 M -naphthoflavone to MCF-7 microsomes. Cotreatment of immature female rats with 10 g/kg E2 and 10 mg/kg MEP extract intraperitoneally for 3 days decreased the E2-induced uterine weights. MEP extract alone showed no effect on rat uterine weight. The endocrine-disrupting activity of MEP extract was further confirmed in parallel experiments using MCF-7 cells and immature female rats treated with benzo(a)pyrene, an MEP constituent compound. The present findings demonstrate that MEP extract is antiestrogenic in vitro and in vivo and cytochrome P450 induction is an underlying mechanism.  相似文献

Polychlorobiphenyls (PCBs), DDT (p,p-DDT,p,p-DDD, andp,p-DDE and HCH isomers (hexachlorocyclohexanes including , , and isomers) were determined in a marine food chain, including seawater, zooplankton, myctophid, squid and striped dolphin collected from the western North Pacific waters. The concentrations of these pollutants increased in higher trophic levels, where the highest bioconcentration factors (concentration ratio in organisms to water), as much as 107, were obtained from both PCBs and DDT in striped dolphin. The proportions of highly chlorinated biphenyls,p,p-DDE and -HCH increased in higher-ranking predators. These observations suggest that the bioaccumulation mechanisms in lower trophic levels depend primarily on the physicochemical properties of pollutants, whereas those in higher trophic levels are affected by the biodegradability of the pollutants and the metabolic capacity and life-span of the organisms.  相似文献

The toxicity of mercury (HgCl2), copper (CuCl2: 5 H2O), nickel (NiSO4: 6 H2O), lead (Pb(CH3COO)2: 3 H2O) and cobalt (CoCl2: 6 H2O) was studied under standardized conditions in embryos and larvae of the zebrafish,Brachydanio rerio. Exposures were started at the blastula stage (2–4 h after spawning) and the effects on hatching and survival were monitored daily for 16 days. Copper and nickel were more specific inhibitors of hatching than cobalt, lead, and mercury. Nominal no effect concentrations determined from the dose-response relationships (ZEPs, Zero Equivalent Points) for effect on hatching time were 0.05 g Cu/L, 10 g Hg/L, 20 g Pb/L, 40 g Ni/L and 3,840 g Co/L, and those for effect on survival time were 0.25 g Cu/L, 1.2 g Hg/L, 30 g Pb/L, 80 g Ni/L, and 60 g Co/L. The no effect concentrations for Ni, Hg and Pb are consistent with previously reported MATC values for sensitive species of fish. The no effect concentrations for copper are 1–2 orders of magnitude lower than previously reported values. The major reason for the latter discrepancy was considered to be the absence of organics that can complex copper ions in the reconstituted water that we used, which had a hardness of 100 mg/L (as CaCO3) and a pH of 7.5–7.7. Unexposed controls were started with embryos from different parental zebrafishes and the parental-caused variability in early embryo mortality, median hatching time and median survival time were estimated.  相似文献

Selenium has been found at elevated concentrations in water, sediments, and aquatic biota in the Elk River (British Columbia, Canada) and some of its tributaries downstream of several coal mines. Selenium water concentrations in those areas exceed Canadian and British Columbia guidelines and are above levels at which adverse effects to fish and waterfowl could occur. We compared selenium concentrations in the eggs of two riverine waterbirds, American dippers and spotted sandpipers, with measures of productivity: the number of eggs laid, egg hatchability, and nestling survival. In American dippers, the mean egg selenium concentration from the exposed areas, 1.10±SE 0.059 g/g wet weight, was indistinguishable from the reference areas, 0.96±SE 0.059 g/g wet weight. For spotted sandpipers, the mean egg selenium concentration in the exposed areas, 2.2±0.5 g/g wet weight, was significantly higher than in the reference areas, 1.2±0.14 g/g wet weight, but less than reported thresholds for waterfowl and other shorebirds. There were no significant differences in egg hatchability between dippers in reference and exposed areas, but reduced hatchability was apparent for sandpipers in exposed locations. Despite the slightly reduced hatchability in sandpipers, overall productivity was higher than regional norms for both species; thus, selenium did not affect the number of young recruited to local populations. We did not observe teratogenic effects in either species, although none was expected at these concentrations. Despite moderately high selenium concentrations in the water, mean egg selenium concentrations were less than predicted from uptake models. We hypothesise that the relatively low uptake of selenium into the eggs of the two waterbirds in this study is likely due to their lotic environments low biological transformation and uptake rates.  相似文献

Flow-through early-life-stage (ELS) toxicity tests were conducted with the pesticides aldicarb (Temik®) and fonofos (Dyfonate®) to determine their effect on the survival and growth of fathead minnows. Concentrations of 78g/L of aldicarb and 16g/L of fonofos did not affect survival and growth. However, 156g/L of aldicarb and 33g/L of fonofos were lethal to larval-juvenile exposed for 30 days post-hatch. The maximum acceptable toxicant concentration (MATC) of aldicarb and fonofos for the fathead minnow is estimated to be between 78–156g/L and 16–33g/L, respectively. The corresponding chronic values (geometric mean of MATC values) would be 110g/L and 23g/L. Acute toxicity tests gave 96-hr LC50 values of 1370g aldicarb/L and 1090g fonofos/L. The acute-chronic ratio (96-hr LC50/chronic value) is 12 for aldicarb and 47 for fonofos.  相似文献

Previous work has suggested that the ratio of copper residues to zinc in the gills of rainbow trout may indicate short-term exposure to increased levels of waterborne copper. However, the effect of exposure to a combination of increased copper and zinc concentrates in the water column was unknown. We exposed rainbow trout to 8 ± 2 g L–1, 40 ± 2 g L–1 and 90 ± 9 g L–1 of waterborne copper and 21 ± 3 g L–1, 129 ± 40 g L–1, and 202 ± 40 g L–1 of waterborne zinc in a 2-factor experiments and gill copper and zinc residues were examined. Other gill parameters analyzed included the concentrations of calcium, magnesium, sodium, and potassium, liver copper and zinc concentrations and plasma copper, calcium, sodium, and potassium are also reported here. Copper residues in the gill filaments were significantly higher in the highest level of copper exposure (high Cu, 4.06 g g–1; low Cu 2.41 g g–1; 0 Cu 2.01 g g–1; P = 0.001), whereas no differences were seen in zinc concentrations at any treatment level. Gill sodium and plasma calcium concentrations were also decreased at the highest waterborne copper concentrations. Although copper–zinc ratios in the gills were significantly different between the highest and lowest copper treatments (P = 0.002, F = 6.59), copper–sodium and copper–magnesium ratios were more sensitive to waterborne copper exposure (P = 0.001, F = 17.91 and P = 0.002, F = 15.45, respectively). Summary Yellow phosphorus (P4) contaminated cod muscle (P4, 4-11 g/g) and liver (P4, 194 g/g) were lethal when fed to brook trout. The symptoms of P4 poisoning were identical to those observed when this element is present in water (Fletcher et al. 1970), namely, external redness, hemolysis and reduced hematocrits. This study presents the distribution of blood lead levels and lead in various environmental samples (water, sediments, soils, and air) near the Shing-Yie storage battery recycling plant in Taiwan before (July 1990 to June 1991) and after (July 1992 to June 1993) amelioration. Before amelioration, the average blood lead levels in the neighborhood of the plant were in the range of 10.55±5.7 to 12.28±7.9 g/dl. After amelioration, relatively lower average concentrations of blood lead (range 8.35±3.0 to 9.13±2.5 g/dl) were generally found; however, these averages were still higher than that (7.79±3.5 g/dl) from other lead-unpolluted areas of Taiwan. An exceedingly high geometric mean (GM) lead concentration (128 g/L) was found in the downstream river water of the Tawulum River passing by the plant. The concentrations of lead (GM=372 and 418 g/g) in the downstream river sediments were higher than those (GM=123 and 158 g/g) in the upstream river sediments before and after amelioration, respectively. Furthermore, lead species in river sediments were analyzed by a sequential leaching technique. The sum of phases I, II, and III accounted for 83.7% of total lead at station R2 (nearest to the plant). Maximum lead concentration (GM=2402 g/g) in dust at the soil surface from station S1 (nearest to the plant) was much higher than those from the other stations by about 18 times before amelioration. However, the maximum value dropped to 1,155 g/g after amelioration. On the whole, the geometric mean concentration of lead in dust at the soil surface nearest to the plant was >1,000 g/g and decreased to <100 g/g in the 15–30 cm depth soil about 2 km away from the plant. Before amelioration, the geometric mean lead concentration of 4.57 g/m3 (range 0.102–37.6 g/m3) in the air near the plant was higher than that at the background locations, the geometric mean value of which was 0.08 g/m3.  相似文献

Summary Five men were exposed to toluene diisocyanate (TDI) atmospheres for 7.5 h. The TDI atmospheres were generated by a gas-phase permeation method, and the exposures were performed in an 8-m3 stainless-steel test chamber. The mean air concentration of TDI was ca. 40 g/m3, which corresponds to the threshold limit value (TLV) of Sweden. The inhaled doses of 2,4- and 2,6-TDI were ca. 120 g. TDI in the test chamber air was determined by an HPLC method using the 9-(N-methyl-aminomethyl)-anthracene reagent and by a continuous-monitoring filter-tape instrument. After hydrolysis of plasma and urine, the related amines, 2,4- and 2,6-toluenediamine 2,4-, and 2,6-TDA), were determined as pentafluoropropionic anhydride (PFPA) derivatives by capillary gas-chromatography using selected ion monitoring (SIM) in the electron-impact mode. The urinary elimination of the TDAs showed a possible biphasic pattern, with rapid first phases for 2,4-TDA (mean t 1/2 for the concentration in urine, 1.9 h) and for 2,6-TDA (mean t 1/2 for the concentration in urine, 1.6 h). The cumulative amount of 2,4-TDA excreted in urine within 28 h ranged from 8% to 14% of the estimated dose of 2,4-TDI, and the cumulative amount of 2,6-TDA in urine ranged from 14% to 18% of the 2,6-TDI dose. The average urinary level of 2,4-TDA was 5 g/l in the 6 to 8-h sample (range 2.8–9.6 g/l), and the corresponding value for 2,6-TDA was 8.6 g/l (range, 5.6–16.6 g/l). Biological monitoring of exposure to 2,4- and 2,6-TDI by analysis of 2,4- and 2,6-TDA in urine is feasible.  相似文献

Chlorophenols are transformedin vitro to polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/ Fs) by peroxidase-catalyzed oxidations. This is demonstrated with bovine lactoperoxidase as well as horseradish peroxidase, and with 3,4,5- and 2,4,5-trichlorophenol (TrCP). The yield of total PCDD/Fs with lactoperoxidase was 11 g per g 345-TrCP and 10 g per g 245-TrCP, of which 2,3,7,8-substituted PCDD/Fs constituted 8.5 and 2.2 g/g, respectively, corresponding to 0.85 and 1.2 g/g of Nordic TCDD-equivalents.  相似文献

Summary To investigate the mechanism for bone disease caused by exposure to cadmium serum samples were collected from 5 itai-itai disease patients, 36 Cd-exposed residents with renal tubular damage and 17 non-exposed individuals and analyzed for 1,25-dihydroxyvitamin D[1, 25(OH)2D], parathyroid hormone, 2-microglobulin, calcium and inorganic phosphorus. Measurement of percentage tubular reabsorption of phosphate (%TRP) were performed only on the Cd-exposed subjects. Serum 1, 25(OH)2D], levels were lower in itai-itai disease patients and cadmium-exposed subjects with renal damage than in non-exposed subjects. Parathyroid hormone and 2-microglobulin concentrations in serum were higher in the Cd-exposed subjects. Decrease in serum 1,25(OH)2D levels were closely related to serum concentrations of parathyroid hormone, 2-microglobulin and %TRP. This study suggests that cadmium-induced bone effects were mainly due to a disturbance in vitamin D and parathyroid hormone metabolism, which was caused by the cadmium-induced kidney damage.  相似文献

A new sublethal toxicity test was developed in this study to measure the effect of copper and lead on the motility of coral larvae. Larval motility was significantly affected by copper and lead doses immediately after dosing. The copper EC50 values for motility of Goniastrea aspera brain coral larvae (12 h, 21 g/L; 24 h, 16 g/L; 48 h, 22 g/L) were much lower than the copper LC50 values for G. aspera larval survival (6 h, 260 g/L, and 24 h, 121 g/L, for 5-day-old larvae and 6 h, 248 g/L, and 24 h, 136.64 g/L, for 6-day-old larvae) during the early part of the experiments. However, at later times, the LC50 values (48 h, 40 g/L, for 5-day-old larvae and 48- h, 87 g/L, for 6-day-old larvae) were similar to the EC50 values for larval motility. The lead 72-h EC50 value for G. aspera larval motility (2900 g/L) was much lower than the lead 72-h LC50 value for larval survival (9890 g/L). The results show that larval motility can be a useful parameter to measure in order to determine the sublethal effects of trace metals on coral larvae.  相似文献

Effects of tributyltin (TBT) on survival, growth, morphometry, and RNA-DNA ratio in larval striped bass(Morone saxatilis) were assessed in three experiments. In Experiment I, 13 day old larvae were exposed to 0, 0.067, 0.766, or 2.284 g TBT/L for 6 days. All larvae exposed to 2.284 /L died by day 5; exposure to 0.766 g/L significantly reduced survival relative to controls (59.8% vs. 81.8%). Significant, concentration-dependent decreases in body depth occurred in larvae exposed to 0.067 and 0.766 g/L. In Experiment II, all 16 day old larvae exposed to 1.498 g/L died by day 6. Survival, weight, and morphometry parameters were not significantly different in larvae exposed to 0, 0.052, or 0.444 g/L for 7 days. In Experiment III, survival was similar in 21 day old larvae exposed to 0, 0.055, 0.218, or 0.514 /L for 7 days. Notochord length and dry weight decreased significantly in larvae exposed to 0.514 g/L. Weight and morphometry parameters were more sensitive indicators of sublethal stress than RNA-DNA ratio. Maximum TBT concentrations reported in Chesapeake Bay marinas are likely to cause increased larval mortality. Longer-term studies are needed to assess effects at <0.050 g/L, which may be more representative of habitat conditions.  相似文献

In 1984 every citizen of Tessin has been reached by an average of 24 preventive messages.

Nitrous oxide (N2O) was assayed in 676 urine samples and 101 blood samples provided after exposure by operating theatre personnel from nine hospitals. The blood and urine assays were repeated in 25 subjects 18 h after the end of exposure. For 80 subjects, environmental N2O was also measured during intraoperative exposure. Mean urinary N2O in the 676 subjects at the end of exposure was 40 g/l (range 1–3805 g/l); in 10 of the 676 subjects, urinary N2O was in the range 279–3805 g/l (mean 1202 /l). The 98th percentile was 120 g/l. Mean blood N2O at the end of exposure, measured in 101 subjects, was 21 g/l (median 16 g/l, range 1–75 g/l). Blood and urine N2O (1.5 g/l and 4.9 g/l, respectively) in 25 subjects, 18 h after exposure, was significantly higher than in occupationally non-exposed subjects (blood 0.91 g/l, urine 1 g/l). Environmental exposure was significantly related to blood and urinary N2O (r = 0.59 andr = 0.64, respectively). Blood and urinary N2O were significantly related to each other (r = 0.71), and were equivalent to about 25% of the environmental exposure level. The mean urinary N2O of 1202 g/l in 10/676 subjects was not related to environmental exposure in the operating theatre. The highest urinary N2O levels measured in these 10/676 subjects could be explained by an asymptomatic urinary infection.  相似文献

Summary Concentrations of -HCH, HCB, p,p-DDE, p,p-DDT and PCBs were determined by gas chromatography in 50 human milk samples collected from the general population during 1981/1982 in a continental town in Croatia (Yugoslavia). The samples were collected between one and 22 weeks after delivery from mothers breast-feeding one child only. The mothers' age was 18 to 31 years, and they were nursing their first or second child. All 50 samples contained -HCH, HCB, p,p-DDE and PCBs, whereas p,p-DDT was present in 37 samples. The concentrations of -HCH, HCB, p,p-DDE, p,p-DDT and PCBs expressed on a whole milk basis were (median in g/kg): 11, 7.1, 67, 7.3 and 22 respectively, and expressed on a fat basis (median in mg/kg): 0.28, 0.21, 1.9, 0.18 and 0.62 respectively. The fat content was 3.7% (median). PCBs were present in a mixture containing penta-, hexa- and heptachlorobiphenyls.  相似文献

Summary Blood styrene was measured by a gas chromatography-mass spectrometry method in 81 normal people and in 76 workers exposed to styrene. In the normal subjects, styrene was also tested in alveolar and environmental air. Styrene was found in nearly all (95%) blood samples. Average styrene levels in the normal subjects were 221 ng/1 in blood (Cb), 3 ng/1 in alveolar air (Ca) and 6 ng/1 in environmental air (Ci). Styrene levels did not differ significantly between smokers and non-smokers, 95% of values being below 512 ng/1 in Cb, 7 ng/1 in Ca and 15 ng/l in Ci. In workers with an average exposure to styrene of 204 g/l, at the end of the workshift, mean blood styrene concentration was 1211 g/l. In blood samples collected at the end of the Thursday shift, styrene levels were significantly higher (1590 g/1) than those found at the end of the Monday shift (1068 g/l. A similar difference was found in samples taken the morning after exposure (60 and 119 g/l, respectively). Significant correlations between blood and environmental styrene were found both at the end of the shift and the morning after exposure (r=0.61 and 0.41, respectively). In workers occupationally exposed to styrene, 16 h after the end of the workshift, blood styrene (94 g/l) was significantly higher than that found in the normal subjects (0.22 g/l). The half-life of blood styrene was 3.9 h.  相似文献

Fresh water and seawater acclimated (FWA and SWA) killifish (Oryzias latipes) were exposed to pentachlorophenol (PCP) at various salinities (1.2–18.7). The PCP accumulation in the fish decreased with increase of salinity. As the salinity increased [0 (freshwater) –18.6], the PCP clearance rate increased from 0.0097 to 0.0170 (h–1). A substantial increase of the clearance rate was observed when the salinity was over 9.3. The PCP uptake rate decreased from 20.8 to 10.6 (ml–1 g–1 fish h–1) with the increase of salinity. The uptake rate decreased at a lower salinity (4.7). The excretion of PCP glucuronide (PCP-G) increased markedly at the salinity of 9.3, although that of PCP sulfate (PCP-S) did not change. The increase of PCP-G excretion may correspond to the increase of clearance rate. Abrupt transfer of FWA and SWA killifish to a PCP solution with a different salinity decreased the PCP accumulation and increased the PCP metabolite excretion with the increase of the salinity. However, the amount of PCP accumulated by FWA was greater than that by SWA killifish.  相似文献

Lead and cadmium levels in blood and deciduous teeth (shed incisors only) of 6-year-old German children were determined in 1991 in a large epidemiological study carried out in rural and urban areas of western Germany (Duisburg, Essen, Gelsenkirchen, Dortmund, Borken) and eastern Germany (Leipzig, Halle, Magdeburg, Osterburg, Gardelegen, Salzwedel). In total, blood lead and cadmium levels of 2311 German children and tooth lead and cadmium levels of 790 German children were analyzed. Blood lead levels were generally low in all study areas with geometric means between 39.3 g/1 and 50.8 g/l in the western German and between 42.3 g/1 and 68.1 g/l in the eastern German study areas. The mean blood lead level of Turkish children (n = 213) living in the western German study areas was 50.1 g/l and thus 5.6 g/1 higher than the overall geometric mean of the western German children. The higher exposure may be explained by a higher oral uptake from food and different living conditions. These children were excluded from multiple regression analysis because they were all living in the western study areas. The mean tooth lead levels ranged between 1.50 and 1.74 g/g in the western and between 1.51 g/g and 2.72 g/g in the eastern study areas. Thus, they show a distribution pattern similar to blood. Blood and tooth lead levels were higher in urban than in rural areas and higher in the eastern German than in the western German study areas. With regard to the blood and tooth cadmium concentrations, no significant differences between the study areas could be found. The mean cadmium levels in blood ranged between 0.12 g/1 and 0.14 g/l and the mean tooth cadmium concentrations between 20.8 ng/g and 27.8 ng/g. Blood and tooth lead and cadmium levels of the eastern and western German children were thus mainly at a relatively low level in all rural and urban study areas. The study demonstrates and confirms that blood and tooth lead levels are influenced by several demographic, social, and environmental variables.
