建立^3H-精氨酸转化测定原生型和诱生型一氧化氮合酶的方法。方法 以二乙氨乙基(DEAE)纤维素柱层析和2、5-ADP琼脂糖亲合层析法纯化牛、猪及大鼠小脑一氧化氮合酶、用^3H-精氨酸转化法测定一氧化氮合酶活性。结果 猪、大鼠小脑组织匀浆一氧化氮合酶活性分别为669、688pmol.mg^-1.min^-1分别纯化了4460倍、2646倍。获得率分别为2.7%、27%。纯化后的猪和大鼠小脑一氧化氮  相似文献

四物汤刺激淋巴细胞^3H—TdR掺入及分泌IL—2   总被引:9,自引:0,他引:9  
实验表明四物汤能显著促进自发性和ConA刺激的小鼠淋巴细胞^3H-TdR掺入,显著促进正常小鼠,正常大鼠淋巴细胞和混合脾淋巴细胞产生白细胞介素-2(IL-2),但未能显著提升血虚大鼠脾淋巴细胞和混合脾淋巴细胞分泌IL-2的水平。结果说明 汤能显著增加机体的细胞免疫功能。  相似文献

This study was conducted in adult male Sprague-Dawley rats to determine thedistribution of [3~H]-nicotine in blood and tissues following a bolus injection and a constantinfusion of pure nicotine.The animals were anesthetized and injected with either 0.5 ml ofnicotine solution or given a constant infusion of the same nicotine solution with identicalamounts of radioactive nicotine.After sacrifice,blood,brain,trachea.salivery gland,esophagus,lung,heart,liver,fundus,antrum,spleen,pancreas,duodenum,jejunum,ileum,cecum,colon,kidneys,adrenal gland,and testes were collected and measured for radioactivityby scintillation counting.The distribution of nicotine was found highest in kidneys by bothroutes of administration.Higher accumulations were also found in salivary and adrenalglands,fundus,antrum,duodenum,jejunum,ileum and colon.Retention of nicotine viaconstant infusion was significantly higher in esophagus,fundus antrum,spleen,cecum,pancreas,testes,heart and muscle when compared with bolus injection.Six-fold  相似文献

SC1001 is a new Synthesized CNS depressant. In this paper, the absorption, distribution and excretion of 3H-SC1001 in mouse and rats were studied by radioactivity trace method. The experiment showed that SC1001 was absorbed rapidly after administering orally and intramuscularly. Maximum radioactivity was found at 0.13h and 0.33h. The rapidly and widely distribution was demonstrated after absorption, the level of radioactivity was the highest in the blood, then in liver, kidney, intestine lung, heart, spleen, and brain. 3H-SC1001 could easily pass through the blood-brain barrier. 3H-SC1001 was eliminated in faeces and then in urine. In 3 days the cumulative radioactivity excretion was 32.0% in urine and 74.04% in faeces, t1/2 of p.o. was 5.69h and t1/2 of i.m. was 6.91h.  相似文献

一种新的^3H—TdR掺入法抗癌药敏试验   总被引:1,自引:0,他引:1  
作者对琼脂-液体双层抗癌药敏试验系统加以改进,建立了更为简便有效的试验系统。经对84例临床肺癌患者活标本测试,成功率达85%。将肺癌细胞对长春新碱(VCR),丝裂霉素(MMC),顺铂(DDP)和阿霉素(ADM)的敏感界限,以1/10血液峰值浓度下细胞存活率表示,并分别定为50%、40%、30%和30%时,测试结果与文献报告的肺癌临床单药化疗有效率近似。本法与干细胞法作平行试验测试10个肺癌细胞系对4种药物的敏感性,两法符合率为83%。测试4例肺癌活检标本,两法符合第为100%。作者认为本试验系统具有临床推广应用价值。  相似文献

自然杀伤细胞(Natural Killer Cells,NK)活性是反应机体细胞免疫功能的重要指标之一。国内应用较多的检测方法为~(125)Ⅰ-尿嘧啶核苷(~(125)Ⅰ-UdR)18h释放法和~3H-胸腺嘧啶核苷(~3H-TdR)18h释放法。这两种方法均需要较长时间孵育,而且由于长时间孵育需要无菌操作,给检测工作带来很多不便。为缩短实验时间,简便操作,我们将~3H-TdR释放法的孵育时间缩短为4h,取得较满意的结果。  相似文献

观察内皮素等血管收缩剂对幼年大鼠卵巢,子宫和睾丸组织摄取^H-TdR的影响。将组织块分别放入含有^3H-TdR和不同试剂-内皮素-1、前列腺素F2α或去甲肾上激素的培养杯中孵育4h,令组织融化并测定其摄取能力。结果显示内皮素明显促进卵巢和睾丸细胞摄取^3H-TdR的能力,前列腺素和去甲肾上腺素则使子宫组织摄取能力有所增强。  相似文献

Mitoxantrone (DHAQ) we studied is a new semisynthetic antitumor drug prepared in China. This paper reports the pharmacokinetic studies of 3H-mitoxantrone in mice by liquid scintillation. The results showed that the decline of radioactivity in the plasma was a biphasic curve after intramuscular and intravenous injection in rats. The adsorption of 3H-DHAQ was ready and complete after intramuscular injection, it was widely distributed in body tissues. The concentration order of various organs was liver greater than intestine greater than kidney greater than lung greater than heart greater than muscles greater than brain. The elimination of the drug was slow, T1/2 was 42.56 h. In 72 h after administration the cumulative excretion of radioactivity in urine was 7% of the total dose, while that in feces was 43%. The main forms of the drug in urine were its metabolites.  相似文献

探讨^3H-TdR参入DNA技术在组织培养中的合理应用。测定16周龄SHR和WKY大鼠培养的胸主动脉血管平滑肌细胞的自然增长曲线以及接种后60h期间,每隔4-6h^3H-TdR参入的变化。SHR大鼠ASMC分裂增殖能力比WKY强,在15%FBS-1640培养基作用下,1-60h期间,SHR ASMC ^3H-TdR高峰参入率在20-24,50-55h之间,WKY的高峰参入率在40-50h之间。  相似文献

目的 研究乙醇对小鼠不同脑区脑片外源性摄取^3H-γ-氨基丁酸(^3H-GABA)功能的影响,方法 离体培养脑片法和同位素示踪法,结果 乙醇对小脑,中脑,间脑和大脑皮层脑片摄取^3H-GABA有明显的抑制效应(P〈0.01),戊四唑,青霉素,荷包牡丹碱加入乙醇后各脑区的摄取水平低于或处于正常水平,氨氧乙酸,印防己毒素,蝇草醇,戊巴比妥钠对各脑区脑片摄取^3H-GABA都有促进作用,其中印防己毒素囊  相似文献
