目的探讨使用纳米羟基磷灰石(Nano-Hydroxyapitite,n-HA)作为骨组织工程支架材料的可行性。方法以成骨条件培养液培养兔骨髓基质细胞(BMSCs)至第3代并以1×106/cm2的密度与n-HA复合培养后回植于骨缺损区(1.0cm×0.8cm×0.5cm)。术后6周、12周处死动物,取缺损区及周围骨质进行肉眼、X线片、免疫组化染色观察及计算机图像分析。结果所有植入物无排除或感染。6周及12周通过肉眼及免疫组化观察实验组修复效果明显优于对照组,新骨面积百分比高于对照组(P<0.05),差异有统计学意义。结论n-HA作为骨组织工程支架材料具有可行性。  相似文献

目的 探讨骨髓基质细胞(BMSC)对骨缺损的修复作用.方法 实验动物为新西兰大白兔,抽取2 ml骨髓制成细胞悬液,体外培养2周.采用外科手术的方法在兔双侧桡骨处形成1.5 cm的骨膜-骨缺损,将培养2周的骨髓基质细胞注射到桡骨缺损处,于移植后第1、2、3、4 周末,用钼靶X线分别检查双侧桡骨缺损处的骨形成情况,并取第4周末的骨痂进行组织学检查.结果 移植后实验侧骨痂形成快于对照侧;第4周末,实验侧缺损处全部为骨性骨痂连接,对照侧缺损处仍未形成骨痂连接.结论 BMSC可以促进骨缺损的修复.  相似文献

The literature on osteoporotic bone marrow defect (OBMD) is reviewed, and an analysis of 20 additional cases is presented. The findings in this study are consistent with previous reports that the radiographic appearance may be indistinguishable from that of pathologic conditions; in some cases, the condition may be confused with aggressive tumors. It may also be significant that, in 16 of our 20 cases, the defects were found in regions where there had been previous extractions.  相似文献

目的:应用分离的骨髓干细胞(MSC)进行牙周骨缺损的自体移植动物实验,观察MSC用于牙周再生治疗的可行性。方法:体外分离狗髂骨的MSC,体外培养,移植前进行MSC的体外钙化实验,以胶原膜为移植载体,将MSC移植到狗下颌后牙的根分叉骨缺损中,覆盖国产聚四氟乙烯膜,30d后进行组织学观察。结果:分离的MSC在体外均出现了不同程度的钙化;动物实验结果发现,MSC移植组达到了骨缺损的完全修复,无根面吸收;  相似文献

狗牙周缺损处移植自体骨髓干细胞的骨化实验观察   总被引:20,自引:0,他引:20  
目的 对应用自体骨髓干细胞移植的动物骨化实验的观察进行评价。方法 将6只成年狗的36颗牙,分为实验组和对照组(各18颗牙)。有人工制造的牙周缺损中进行体外培养的自体骨髓干细胞移植,结合引导组织再生(guided tissue regeneration,GTR)方法(实验组)和单纯GTR方法(对照组)。6周后对切片行牙周组织学观察。结果 实验组新牙槽骨骨化效果明显好于对照组。实验组建立了正常钙化骨结构,Masson染色为红色;对照组虽然建立了正常骨结构,但钙化程度极低,基本为胶原性骨结构,Masson染色为蓝绿色。结论 应用自体骨髓干细胞移植结合聚四氟乙烯膜引导牙周组织再生可促进牙周组织的再生、加快正常骨组织结构的建立,并缩短修复再生时间。  相似文献

OBJECTIVE: To evaluate the effect of calcification of autogenous bone marrow stem cell transplantation in periodontal tissue regeneration. METHODS: Bone marrow stem cells derived from the same dog were cultured with alpha-MEM. 1 x 10(7) cells of first passage were allowed to attach to the collagen membrane for 24 hours. The membrane-cells were transplanted into periodontal defect in the same dog. Then the defects were covered with e-pTFE membranes. The defects covered only with e-pTFE without membrane-cells were served as control. Eighteen teeth of 6 dogs for every group were studied. The dogs were sacrificed after 6 weeks. RESULTS: The results showed that new bone formation in test group was significantly higher than that of control group. The calcification of new bone in test group was better than control group. CONCLUSIONS: The results suggested that autogenous bone marrow stem cell transplantation with guided tissue regeneration technique could enhance periodontal tissue regeneration and could form new bone tissue fast and could shorten times of periodontal tissue regeneration in dogs.  相似文献

目的应用骨髓间充质干细胞(BMSCs)细胞片层构建组织工程骨修复犬下颌骨缺损,探讨细胞片层在成骨中的作用。方法采用密度梯度离心法分离和培养BMSCs,将BMSCs向成骨细胞诱导培养后,制备细胞片层。将细胞片层包裹到聚乳酸羟基乙酸共聚物(PLGA)支架表面,将其植入犬左侧下颌骨全层缺损中,对侧下颌骨植入无细胞片层包裹的支架复合体作同体对照。将16只犬分为4组,每组4只。术后4、8、12、16周分别处死1组,取材行大体及组织学观察。结果实验侧成骨好于对照侧,术后16周,实验侧骨缺损大部分被新生骨替代,舌侧形成与正常骨相似的密质骨,与正常骨断端骨性愈合。实验侧新生骨光密度值大于对照侧,两者差异有统计学意义(P<0.05)。实验侧可见较多哈弗氏系统及红骨髓,大量板层骨;对照侧哈弗氏系统较少。结论利用细胞片层技术可以构建出含板层骨结构的组织工程骨。  相似文献

目的:探讨以多孔矿化Bio- Oss骨胶原为载体的自体骨髓基质细胞移植修复下颌骨缺损的效果。方法:标准成年新西兰白兔16只随机分2组,一组抽取骨髓基质细胞进行培养,矿化诱导后,细胞悬液与凝胶混匀滴入Bio- Oss胶原骨块中, 静置于37℃, 50ml/L的CO2 培养箱中温育4 ~6h,加入DMEM成骨条件培养液培养。然后修复下颌骨极限骨缺损。另组单纯Bio -Oss胶原块,修复下颌骨极限骨缺损。分别于术后8周和12周取材,分别行大体、X射线、组织学观察骨缺损的修复情况。结果:术后12周, (1)BMSC 凝胶 Bio -Oss胶原组,基本上由新生骨组织所修复。(2)单纯Bio- Oss胶原组大部分由纤维组织填充,周围骨形成明显,中央少量骨痂形成。结论: (1)Bio -Oss骨胶原有较强的诱导成骨能力; ( 2 )Bio -Oss骨胶原为载体的自体骨髓基质细胞移植能有效修复骨缺损,是骨组织工程良好的支架材料。  相似文献

Focal osteoporotic bone marrow defects usually appear as asymptomatic radioluencies in the edentulous posterior mandible of middle-aged women. The exact causative factor in the majority of focal osteoporotic bone marrow defects is still unknown. Because of their radiological similarity with many intraosseous lesions, accurate diagnosis is possible only with histopathological examination. A focal osteoporotic bone marrow defect that occurred 2 years postoperatively apical to an implant is presented with clinical, radiographic, and histopathologic features. According to the literature scan, this is the first case report of this phenomenon caused by a dental implant. Int J Oral Maxillofac Implants 2011;26:e1-e4.  相似文献
