目的 检测Ⅲ、Ⅳ型胶原在CP及正常胰腺组织中的表达水平,研究其在胰腺间质纤维化形成中的作用.方法 免疫组织化学方法检测16例CP标本及6例正常胰腺组织中Ⅲ、Ⅳ型胶原的表达.结果 16例CP中,Ⅲ型、Ⅳ型胶原表达均为轻度以上深染,其中重度深染50%(8/16),中度深染31.25%(5/16);而正常胰腺组织Ⅲ、Ⅳ型胶原表达均为浅染.正常胰腺组织中Ⅲ、Ⅳ型胶原表达于基底膜、间质纤维、导管壁、血管壁,Ⅲ型胶原以间质纤维表达最明显,Ⅳ型胶原以基底膜表达最明显.CP组织中Ⅲ型胶原表达于基底膜、间质纤维、导管壁、血管壁,以间质纤维最明显;Ⅳ型胶原表达于基底膜、间质纤维、导管壁、血管壁,以基底膜最明显.结论 Ⅲ、Ⅳ型胶原的表达增强在胰腺纤维化的形成中可能有重要作用.  相似文献

目的 研究动物骨质疏松对Ⅰ型胶原及Ⅰ型胶原mRNA的影响,为预防骨质疏松提供基础理论.方法 对正常与骨质疏松模型大鼠椎骨以图像分析系统观察Ⅰ型胶原免疫组化及Ⅰ型胶原mRNA原位杂交检测.结果 得出了各组试样的阳性染色分别面积、平均灰度值、阳性反应面积等指标.结论 模型组各项指标显著低于正常对照组(P<0.05).骨质疏松后胆原免疫组化指标及Ⅰ型胶原mRNA发生改变,易发生骨折.  相似文献

目的 观察通心络胶囊时链脲佐茵素(STZ)糖尿病大鼠心肌Ⅰ型和Ⅲ型肢原阳性表达及含量、细胞转化因子(TGF-β1)含量的影响.方法 制备STZ糖尿病大鼠38只,随机分为STZ对照组、胰岛素组、通心络组,另取同月龄Wistar大鼠10只(正常大鼠组),分别灌胃8周.制备心肌切片,免疫组化检测胶原Ⅰ、胶原Ⅲ阳性面密度比值代表其表达程度,取心肌匀浆,酶联免疫方法测定心肌Ⅰ型、Ⅲ型胶原及TGF-β1含量.结果 通心络组与STZ对照组血糖无明显变化.与正常大鼠组比较,STZ对照组心肌胶原Ⅰ阳性面密度比值明显增高(P(0.01),胶原Ⅲ阳性面密度比值明显降低(P(0.01),心肌匀浆胶原Ⅰ含量明显增高(P<0.01).胶原Ⅲ含量降低(P<0.05),心肌胶原Ⅰ/Ⅲ比值明显增高(P<0.01),TGF-β1含量降低(P<0.01);与STZ对照组比较,通心络组胶原Ⅰ阳性面密度比值降低趋势,胶原Ⅲ阳性面密度比值明显升高(P<0.05),胶原Ⅰ含量明显降低(P<0.01),胶原Ⅲ含量升高(P相似文献

目的探讨血管损伤修复中Ⅲ型胶原的表达规律及其与血管平滑肌细胞(VSMC)增生的关系.方法26只犬冠状动脉内置入过大钽丝支架建立再狭窄模型,分别于术后7d、14d和28d处死动物,用透射电镜及免疫组化染色技术观察新生内膜中Ⅲ型胶原表达和VSMC特征,并用图象处理技术对Ⅲ型胶原的染色密度作定量分析.结果支架置入后7d以VSMC移行增生为主,14d时VSMC增生达高峰并有较多胶原表达,28d时VSMC由合成型逐渐转变为收缩型并表达大量胶原,Ⅲ型胶原在不同时间阶段的表达量具有显著差异(P<0.05).结论Ⅲ型胶原可能在再狭窄形成的后期内膜增生中起重要作用.  相似文献

目的 探讨不同浓度血清、不同体外培养时间对L929系小鼠成纤维细胞表达产物Ⅰ、Ⅲ型胶原及其比值的影响.方法 采用双抗体夹心法检测细胞培养液中Ⅰ、Ⅲ型胶原的含量,并同时计算出Ⅰ/Ⅲ型胶原的比值.结果 除0%血清浓度组的Ⅰ、Ⅲ型胶原呈现无明显变化的微弱升高趋势外,其余各血清浓度组的Ⅰ、Ⅲ型胶原均处于增加状态,且在6~8 d时增加最为明显;另外,Ⅰ/Ⅲ型胶原比值随着血清浓度和细胞生长时间不同,表现出不同的变化趋势.其中20%和40%血清浓度组随着培养细胞生长时间延长,Ⅰ/Ⅲ型胶原比值逐渐增加;而其他血清浓度组,随着细胞培养时间的延长,Ⅰ/Ⅲ型胶原比值增加并不明显.结论 L929细胞在不同体外培养时间和不同浓度血清下其表达产物Ⅰ、Ⅲ型胶原含量及其比值有一定变化,为今后优化体外细胞培养条件提供了相关的实验数据.  相似文献

目的 探讨血管球囊损伤后血清I型前胶原羧基端肽浓度和Ⅰ型胶原基因表达的变化及麝香保心丸对其影响。方法 将 4 5只实验兔随机分为 3组 ,每组 15只 :血管成形术组、血管成形术 +麝香保心丸组及对照组。应用球囊拉伤动脉内膜加高脂饲养的方法建立兔髂动脉硬化模型并行球囊扩张血管成形术 ,采用放射免疫法和核酸分子杂交技术对血清I型前胶原羧基端肽和髂动脉壁平滑肌细胞的Ⅰ型胶原基因的表达情况进行观察。结果 术后 1周时血管成形术组血清I型前胶原羧基端肽较对照组水平增加 (I型前胶原羧基端肽为 9.5 4± 1.4 9,P <0 .0 5 ) ,Ⅰ型胶原基因表达增加 ,经计算机图像辉度扫描定量分析结果为 (16 0± 12 ,P <0 .0 5 ) ,2、4周时显著增加 (P<0 .0 1) ,(血清I型前胶原羧基端肽为 14 .2 1± 2 .6 7和 15 .6 3± 6 .87,Ⅰ型胶原基因表达为 4 16± 16和 4 38± 17) ,血管成形术 +麝香保心丸组血清I型前胶原羧基端肽无显著增加、Ⅰ型胶原基因未表达。结论 血管内皮损伤可致Ⅰ型胶原合成增加 ,麝香保心丸对其有抑制作用  相似文献

为了解基质降解在慢性肝病中的作用,对慢性肝炎、肝硬化患者的血清Ⅳ型胶原降解酶活性进行了研究.结果表明:慢性肝炎、肝硬化患者的血清Ⅳ型胶原降解酶活性均显著高于正常对照组;慢性肝炎患者酶活性较肝硬化患者亦显著为高.血清Ⅳ型胶原降解酶活性与PⅢP、Laminin间均呈显著负相关.上述结果提示.Ⅳ型胶原降解酶可能参与了慢性肝病的病理发生;随着慢性肝病的进展,酶活性逐渐下降.  相似文献

目的 观察MSCs治疗紫外线致皮肤光老化模型过程中大鼠皮肤Ⅰ型、Ⅲ型胶原的表达变化.方法 采用15 w UVA和UVB紫外灯,置于大鼠背部裸露皮肤区的正上方35 cm处,每日照射2 h,共照射60 d.随机分为对照组、模型组及MSCs治疗组,每组10只.体外分离培养MSCs,在大鼠背部裸露皮肤区多点注射,1次/w,共8 W.取皮肤组织,利用RT-PCR进行检测.结果 治疗组Ⅰ型胶原表达明显低于模型组(P<0.05),治疗组Ⅰ型胶原表达接近于对照组(P>0.05).治疗组Ⅲ型胶原表达高于模型组(P<0.05),但接近于对照组(P>0.05).结论 MSCs对紫外线皮肤光老化模型大鼠有治疗作用,表现为Ⅰ型胶原相对含量减少,Ⅲ型胶原相对含量增加.  相似文献

目的 研究乌梢蛇Ⅱ型胶原对关节炎大鼠的免疫调节作用,以了解乌梢蛇治疗类风湿关节炎(RA)的机制.方法 关节炎大鼠随机分成3组,即乌梢蛇Ⅱ型胶原组、牛Ⅱ型胶原组和关节炎对照组,另设健康埘照组,每组7只,乌梢蛇Ⅱ型胶原组和牛Ⅱ型胶原组分别灌胃乌梢蛇Ⅱ型胶溶液1 ml(Ⅱ型胶原15 mg/kg)和牛Ⅱ型胶溶液1 ml(Ⅱ型胶原15 mg/kg),另两组灌胃不含胶原的胶原稀释液1 ml,每日1次,连续15 d后,取大鼠外周血测抗Ⅱ型胶原抗体、淋巴细胞亚群、肿瘤坏死因子(TNF)-a、白细胞介素(IL)-10、IL-1和IL-4水平.结果 关节炎埘照组:抗Ⅱ型胶原抗体、TNF-a水平和CD4+T/CD8+比值与健康对照组相比明显升高(P<0.05),IL-10水平明显下降(P<0.01);乌梢蛇Ⅱ型胶原组和牛Ⅱ型胶原组:TNF-a水平和CD4+T/CD8+比值与关节炎对照组相比明显下降(P<0.05),与健康对照组比较差异无统计学意义(P>0.05),抗Ⅱ型胶原机体滴度较关节炎对照组下降(P<0.05),与健康埘照组差异有统计学意义(P<0.01),IL-10水平较关节炎对照组升高(P<0.01),与健康对照组差异有统计学意义(P<0.05),口服Ⅱ型胶原两组间各项免疫指标比较差异均无统计学意义(P>0.05);外周血IL-1和IL-4水平,4组差异无统计学意义(P>0.05).结论 乌梢蛇Ⅱ型胶原与牛Ⅱ型胶原具有相同的免疫调节作用,两者能减低关节炎大鼠外周血TNF-a水平、抗Ⅱ型胶原抗体滴度和CD4+T/CD8+比值,提高IL-10水平的作用,推测乌梢蛇Ⅱ型胶原可能是乌梢蛇治疗RA的活性药理成分.  相似文献

Abstract: A tight, physiological link between the pineal gland and the immune system is emerging from a series of experimental studies. This link might reflect the evolutionary connection between self-recognition and reproduction. Pinealectomy or other experimental methods which inhibit melatonin synthesis and secretion induce a state of immunodepression which is counteracted by melatonin. In general, melatonin seems to have an immunoenhancing effect that is particularly apparent in immunodepressive states. The negative effect of acute stress or immunosuppressive pharmacological treatments on various immune parameters are counteracted by melatonin. It seems important to note that one of the main targets of melatonin is the thymus, i.e., the central organ of the immune system. The clinical use of melatonin as an immunotherapeutic agent seems promising in primary and secondary immunodeficiencies as well as in cancer immunotherapy. The immunoenhancing action of melatonin seems to be mediated by T-helper cell-derived opioid peptides as well as by lymphokines and, perhaps, by pituitary hormones. Melatonin-induced-immuno-opioids (MHO) and lymphokines imply the presence of specific binding sites or melatonin receptors on cells of the immune system. On the other hand, lymphokines such as -γ-interferon and interleukin-2 as well as thymic hormones can modulate the synthesis of melatonin in the pineal gland. The pineal gland might thus be viewed as the crux of a sophisticated immunoneuroendocrine network which functions as an unconscious, diffuse sensory organ.  相似文献

Abstract: The abundance of gap junctions between rat pineal astrocytes formed by connexin43 (Cx43) was studied during development. Levels and distribution of Cx43 were measured by immunoblotting and indirect immunofluorescence, respectively. The amount of Cx43 in cells located within the gland was low until about the 7th postnatal day and increased to adult values between the 14th and 21st days postpartum. Although astrocytes, recognized by their vimentin immunoreactivity, were scarce before birth, they were abundant by the 7th postnatal day suggesting that the low levels of Cx43 found at this age corresponded to a low expression of this protein. Localization of the immunoreactivity to Cx43 and vimentin showed a close correlation, indicating that mature or immature pineal astrocytes form gap junctions made of Cx43. Since Cx43 levels attained their adult values at about the time the innervation and the functional state of the gland reached maturity (2–3 weeks after birth), it is proposed that astrocyte gap junctions are involved in the function of the adult rat pineal gland.  相似文献

Duodenal diverticula are a relatively common condition. They are asymptomatic, unless they become complicated, with perforation being the rarest but most severe complication. Surgical treatment is the most frequently performed approach. We report the case of a patient with a perforated duodenal diverticulum, which was diagnosed early and treated conservatively with antibiotics and percutaneous drainage of secondary retroperitoneal abscesses. We suggest this method could be an acceptable option for the management of similar cases, provided that the patient is in good general condition and without septic signs.  相似文献

Abstract: Herein we documented the response of pineal melatonin production to electrolytes known to be effective on pineal function in view of a possible circadian stage dependence. We studied the release of melatonin by perifused rat pineal glands at 2 different circadian stages corresponding to the middle of the light and dark periods, i.e., respectively, 7 and 19 HALO (Hours After Light Onset, L:D = 12:12). The initial efflux rates were, as expected, much higher in the perifusates of glands removed from rats sacrificed during the dark phase than of those removed during the light phase. After 3 hr of perifusion, melatonin release reached similar levels which were found constant up to the 8th hr of perifusion, whatever the circadian stage. Perifusion of the glands with physiological concentrations for the rat of calcium (5.2 mmol/1) and magnesium (1.34 mmol/1) resulted in a stimulatory effect on the pineal glands removed from rats sacrificed in the middle of the dark period (19 HALO), whereas no effects were observed on the pineal glands removed from rats sacrificed during the light (7 HALO). Lithium (0.28 and 0.55 mmol/1) was ineffective on melatonin release in pineal glands removed 7 and 19 HALO. Our results show differences in the initial efflux rates of melatonin and in the response of perifused pineal glands to calcium and magnesium according to the circadian stage.  相似文献

Abstract: The use of antisera raised against bovine growth hormone (GH) and ovine prolactin (PRL) enabled the detection of related immunoreactive (ir) sequences of proteins in ovine pineal tissue. The isolation of PRL-like ir-material was accomplished using a 0.25 M ammonium sulphate (pH 5.5) extraction followed by ethanol precipitation, whereas the resulting 2.0 M ammonium sulphate (pH 7.0) precipitate contained a GH-like immunoreactivity. Gel chromatography of the GH-like immunoreactivity (Sephadex G-100) indicated the presence of several GH-like fragments ranging in the Mr range of 7,000 to 55,000. Analyses of the PRL-like ir-material found in pineal tissue on HPLC using a TSK 545-DEAE column led to the resolution into a single peak of immunoreactivity. A single peak of activity was also observed following chromatofocusing and hydrophobic interaction chromatography of the ir-peak from the TSK 545-DEAE column. The PRL-like ir-material inhibited the binding of [125I]ovine PRL-S14 to anti-ovine PRL antibodies without showing an affinity for binding to anti-rat PRL or anti-bovine GH antibodies. Scatchard analysis of the binding of pineal PRL-like ir-material and pituitary ovine PRL-S14 to liver membranes from day-20 pregnant rats revealed similar affinity constants (Ka of 4.7 ± 0.2 × 109 M-1). In addition, the replication of Nb 2 Node rat lymphoma cells was stimulated by pineal PRL-like ir-material, an effect known to be specific for lactogenic hormones. The pineal PRL-like immunoreactivity appeared on sodium dodecyl sulfate polyacrylamide gels as a single major band of Mr 24,000. The functional status of PRL-and GH-like ir-material in the ovine pineal remains to be determined, but evidence is presented that the overall protein synthesis rate of the rat pineal responded to circulating concentrations of PRL.  相似文献

PURPOSE: Individuals who are seropositive for the human immunodeficiency virus are at high risk for opportunistic infection and anorectal disorders. Little prospective information is available regarding anorectal pathogens in these patients. METHODS: One hundred sixty-three HIV-seropositive patients presented to the colorectal clinic between 1989 and 1992. Forty-seven (29 percent) patients were thought to have an infectious process and were prospectively studied using a standardized multiculture protocol. RESULTS: Mean age was 33 (range, 19–59) years. All were male; high-risk behavior accounted for 87 percent of HIV transmissions. Presenting complaints included anorectal pain (79 percent), pus per anum (28 percent), and blood per anum (26 percent). Examination revealed perianal tenderness (60 percent), condyloma (38 percent), perianal ulcers (38 percent), and anal fissures (34 percent). Sixty-six sets of cultures were performed; 28 patients had one set, 15 had two sets, and 4 had three sets. Thirty-two of these 47 patients (68 percent) had positive cultures including herpes (50 percent), cytomegalovirus (25 percent),Neisseria gonorrhoeae (16 percent), chlamydia (16 percent), acidfast bacilli (2 percent), and others (9 percent). Six of 32 patients with positive cultures had more than one organism cultured. Sixteen (50 percent) patients with positive cultures were treated medically, 8 (25 percent) were treated surgically and 8 (25 percent) were treated with both modalities. Sixty-one procedures were performed on 17 patients for condylomata. Eighteen patients had 20 procedures for abscesses, 50 percent of whom had positive cultures for other than common bowel flora; all improved. Fourteen patients underwent 33 procedures for perianal fistulas.Mycobacterium fortuitum was cultured from one patient who required 13 procedures for abscesses and fistulas. Forty-five (96 percent) patients were followed for an average of 12.5 months ±2.9 SEM (range, 1–94 months). Symptoms were improved or resolved in 22 of 32 (69 percent) patients with positive cultures and in 11 of 13 (84 percent) with negative cultures. CONCLUSIONS: Specific pathogens may often be identified in human immunodeficiency virus-seropositive patients with anorectal disorders if aggressively sought. Although patients without specific pathogens identified may be expected to improve with planned empiric treatment, positive identification allows more directed therapy.  相似文献
