目的:探讨YB-1和P53蛋白在结直肠肿瘤不同发展阶段表达水平的差异及其与结直肠癌临床病理特征的关系.方法:采用免疫组织化学方法检测结直肠20例正常组织、30例低级别上皮内瘤变组织、30例高级别上皮内瘤变组织和50例癌组织中YB-1和P53蛋白的表达.结果:YB-1蛋白在结直肠正常组织(NCM)、低级别上皮内瘤变组织(LGIN)、高级别上皮内瘤变组织(HGIN)和癌组织(CRC)中的强阳性率分别为0%、76.7%、80.0%、80.0%,低级别、高级别上皮内瘤变组织和癌组织中的强阳性表达率均与正常组织比较差异有统计学意义(P<0.05),且在癌组织中其强阳性表达与淋巴结转移和TNM分期有关(P<0.05);P53蛋白在结直肠正常组织、低级别上皮内瘤变组织、高级别上皮内瘤变组织、癌组织中表达分别为0%、6.7%、40.0%、60.0%,高级别上皮内瘤变组织和癌组织中其表达率均与正常组织和低级别上皮内瘤变组织比较差异有统计学意义(P<0.05),其在癌组织中的表达与分化程度、淋巴结转移和TNM分期有关(P<0.05).在癌组织中YB-1和P53的表达呈正相关性(r=0.306,P<0.05).结论:YB-1和P53蛋白在结直肠癌的发生发展、转移中起重要作用,YB-1和P53蛋白的联合检测可能为判断结直肠癌恶性程度和转移提供重要参考.  相似文献

收集26例肝癌患者手术新鲜癌组织标本及同次手术病理证实为正常的肝脏组织标本。用4%多聚甲醛固定、石蜡包埋;采用免疫组化染色法观察肝细胞核因子-1α(HNF-1α)在肝癌组织和正常肝组织中的表达及其在细胞中的定位。结果HNF-1α在正常肝组织表达高于肝癌组织(P〈0.05),HNF-1α在高、中分化肝癌组织中的表达高于低分化(P〈0.05)。提示HNF-1α低表达与肝癌的发生、发展密切相关,HNF-1α可在一定程度上提示肝癌预后。  相似文献

目的观察卵巢癌组织中增殖细胞核抗原(PCNA)、增殖细胞相关核抗原(Ki-67)的表达变化,并探讨其临床意义。方法采用免疫组化SP法检测40例卵巢癌、20例卵巢良性肿瘤组织中的PCNA、Ki-67。结果 PC-NA、Ki-67在卵巢癌组织中的阳性表达率明显高于卵巢良性肿瘤(P均〈0.05);PCNA、Ki-67的表达与卵巢癌FIGO分期、组织分级、淋巴结转移有关(P均〈0.05);卵巢癌组织中PCNA与Ki-67的表达呈正相关(r=0.929,P〈0.05)。结论卵巢癌组织中PCNA、Ki-67的表达增加,二者可作为评估卵巢癌临床分期、组织分级及淋巴结转移的参考指标。  相似文献

目的 检测FEZ1和增殖细胞核抗原(PCNA)蛋白在食管鳞癌组织中的表达,探讨其临床意义及相关性。方法采用免疫组化SP法检测100例食管鳞癌及100例正常食管组织中FEZ1和PCNA的表达情况。结果FEZ1蛋白在食管鳞癌组织中的阳性表达率明显低于正常食管组织,其表达与浸润深度及淋巴结转移密切相关;PCNA在食管鳞癌组织中的阳性表达率明显高于正常食管组织,其表达与组织分化程度、浸润深度及淋巴结转移密切相关。FEZ1和PCNA在食管鳞癌组织中的表达存在负相关。结论FEZ1的表达可能与食管鳞癌的发生有关,并且参于了其浸润、转移等过程。  相似文献

目的了解黑色素瘤抗原(MAGE)-A1基因在肝癌组织中的表达状况,探讨该基因是否存在单核苷酸多态性(SNP).方法用RT-PCR方法检测49例原发性肝细胞性肝癌(HCC)患者肝癌组织MAGE-A1基因mRNA的表达情况,同时提取其中43例HCC患者的肝癌组织、癌旁组织及外周血细胞基因组DNA,PCR扩增MAGE-A1 DNA,并对上述PCR产物进行测序.结果49例患者中MAGE-A1 mRNA阳性22例,阳性率44.9%.43例HCC患者癌组织、癌旁组织和外周血细胞中扩增的KAGE-A1 DNA序列测定显示MAGE-A1基因存在两类变异:TAG有16例,变异发生率为37.2%,包括3个位置的变异:C159T,A272G,G393A,GTG变异有7例,发生率为16.3%,亦包括3个位点的变异即:A272G,C991T,A1125G.因为C991T没有改变编码的氨基酸,A1125G不在编码区,故仅A272G引起一个氨基酸的替换(T32A).结论MAGE-A1在HCC中高表达,表达率为44.9%.MAGE-A1 mRNA的表达与AFP无关.MAGE-A1基因存在三种SNP,但不影响其mRNA的表达.  相似文献

目的研究多药耐药相关蛋白P-糖蛋白(gp)和拓扑异构酶II(Topo-II)在原发性肝癌组织中的表达及临床意义。方法采用免疫组化S-P法检测49例原发性肝癌组织P-gp和Topo-II的表达。结果P-gp和Topo-II在原发性肝癌中阳性表达率分别为61.22%(30/49)和32.65%(16/49),P-gp主要在细胞膜和细胞浆中表达,Topo-II在细胞核内表达,P-gp和Topo-II表达与Edmonson分级有关,Topo-II还与转移有关。P-gp、Topo-II在原发性肝癌中表达呈负相关(P<0.05)。结论肝癌组织P-gp和Topo-II表达可能是肝癌对化疗不敏感的原因之一。  相似文献

增殖细胞核抗原(PCNA)也称周期蛋白。是反映细胞增殖分数和所处周期的客观指标,可用于判断肿瘤的恶性程度及预后,亦能较好地反映肿瘤细胞的增殖状态。P^53基因是一种重要的肿瘤抑制基因,其突变与肿瘤的发生发展密切相关。2002年1月至2003年12月,我们用免疫组织化学染色技术对49例口腔鳞癌组织的PCNA和P^53基因蛋白进行检测和分析,旨在探讨两项指标与此类肿瘤的诊断、鉴别诊断及病理分级的关系。  相似文献

目的探讨高尔基体糖蛋白73(GP73)表达诊断肝癌的价值。方法采用免疫组织化学染色SP法检测60份肝癌组织、癌旁组织及24份正常肝组织中GP73表达;分析GP73表达与肝癌临床病理特征的关系。结果肝癌组织、癌旁组织GP73阳性率显著高于正常肝组织(P<0.05)。GP73表达与Child-Pugh分级呈正相关(P<0.05),在肝外转移和HBV感染中表达亦显著增强(P<0.05),与性别、年龄、AFP、肿瘤大小、肝硬化和门脉癌栓无明显相关性。结论 GP73与肝癌的发生发展有关。  相似文献

目的研究Caveolin.1和Caspase-3蛋白在肝细胞癌中的表达及意义。方法采用免疫组织化学检测30例肝细胞癌,相应癌旁及10例正常肝组织中Caveolin-1和Caspase-3蛋白的表达。结果(1)Caveolin-1在正常肝组织、癌旁肝组织、肝细胞癌组织中的阳性表达率分别为90%、13.33%、10%。Caspase-3在正常肝组织、癌旁肝组织、肝细胞癌组织中阳性表达率分别为90%、70%、40%。Caveolin.1和Caspase-3在不同性质组织间的表达,差异均有统计学意义(P〈0.05);(2)Caveolin-1与Caspase-3的表迭在肝细胞癌中无明显相关性(r=0.181,P=0.337)。结论Caveolin-1与Caspase-3在肝细胞癌组织中表达均呈下降,但两者间无明显相关性,提示在肝细胞癌中,Caveolin-1的异常与Caspase-3细胞凋亡紊乱无关。  相似文献

AIM: To evaluate the relationship of expression of paxillin, syndecan-1 and EMMPRIN proteins with clinicopathological features in hepatocellular carcinoma (HCC). METHODS: Fifty-one patients who underwent HCC resection were recruited in the study. Paxillin, syndecan-1 and EMMPRIN proteins in HCC tissues were detected with immunohistochemical staining. RESULTS: Of 51 cases of HCC, 23 (45%) exhibited paxillin protein positive expression. Of 42 cases of adjacent non-tumor liver tissues, 24 (57%) exhibited positive expression. Positive paxillin protein expression was associated with low differentiation (r= 0.406, P= 0.004), with the presence of portal vein thrombosis (r = 0.325, P = 0.021), with extra-hepatic metastasis (r=0.346, P=0.014). Of 51 cases of HCC, 28 (55%) exhibited syndecan-1 protein positive expression. Of 42 cases of adjacent non-tumor liver tissues, 23 (55%) exhibited positive expression. Positive snydecan-1 protein expression was associated with well differentiation (r=0.491, P=0.001), with no extra-hepatic metastasis (r=0.346, P=0.014). Of 51 cases of HCC, 28 (55%) exhibited EMMPRIN protein positive expression. Of 42 cases of adjacent non-tumor liver tissues, 21 (50%) exhibited positive expression. Expression of EMMPRIN protein was not associated with serum AFP level, HBsAg status, presence of microsatellite nodule, tumor size, presence of cirrhosis and necrosis, differentiation, presence of portal vein thrombosis, extra-hepatic metastasis, disease-free survival and overall survival (P>0.05). Expression of paxillin protein was correlated conversely with the expression of syndecan-1 protein in HCC (r = -0.366, P = 0.010). CONCLUSION: Expression of paxillin and syndecan-1 proteins in HCC may affect its invasive and metastatic ability of the tumor. There may be a converse correlation between the expression of paxillin and syndecan-1 protein in HCC. Expression of EMMPRIN protein may be detected in HCC, but it may play little role in the invasion and metastasis of HCC.  相似文献

目的 探讨钠氢交换蛋白1(NHE1)mRNA在原发性肝细胞肝癌组织及癌旁组织的表达及意义。方法采用RT—PCR检测诊断为HCC患者34例的肝癌及癌旁组织中NHE1mRNA的表达情况。结果所有肝癌组织和癌旁组织NHE1均有表达,相对表达量分别为2.892±0.882和0.802±0.206,两者存在显著性差异(P〈0.001)。作为对照的21例肝组织NHE1的相对表达量为0.872±0.186。肝癌组织与对照肝组织比较存在显著性差异(P〈0.001);癌旁组织与对照肝组织比较差异无统计学意义(P〉0.10)。将从同一患者取材的肝癌组织和癌旁组织检测到的NHE1mRNA的表达量进行配对t检验,两者间存在显著性差异(P〈0.001)。结论NHE1mRNA在肝癌组织的表达增高,可能在肝癌的发生中起一定的作用。  相似文献

AIM: To investigate the expression of p57kip2 and its relationship with clinicopathology, PCNA and p53 in primary hepatocellular carcinoma (HCC). METHODS: Expression of p57kip2, PCNA and p53 in tumor tissues from 32 patients with HCC and 10 liver tissues of normal persons was detected with Elivision immunohistochemical technique. RESULTS: The p57kip2 protein positive-expression rate in HCC was 56.25%, lower than that in normal tissues (100%, P<0.05). The reduced expression of p57kip2 protein correlated significantly with moderate or low differentiation of tumor cells (P = 0.007 <0.05), high clinical stage (P= 0.041 <0.05) and poor prognosis (P= 0.036 <0.05), but did not correlate significantly with metastasis, tumor size, level of AFP and age (P>0.05). The PCNA positive-expression rate was 56.25%, which was correlated significantly with the expression of p57kip2 (P= 0.025<0.05). The p53 positive-expression rate was 46.88%, which was not correlated significantly with the expression of p57kip2 (P>0.05). CONCLUSION: There is a marked loss or absence of p57kip2 expression and high expression of PCNA in HCC, which are involved in carcinogenesis and development of HCC. The p57kip2 and p53 may induce apoptosis via different mechanisms.  相似文献

AIM: To evaluate the prognostic signif icance of HIF- 2α/EPAS1 expression in hepatocellular carcinoma (HCC). METHODS: Surgical specimens from 315 patients with HCC as well as 196 adjacent noncancerous lesions and 22 cases of normal liver tissue were investigated by immunohistochemistry (IHC) for HIF-2α/EPAS1 using a standard detection system. Correlations with clinicopathological factors, VEGF, microvessel density (MVD), and prognosis were analyzed. RESULTS: Immunoreactivity of HIF-2α/EPAS1 was positive in 69.5% of HCC, 55.6% of adjacent noncancerous tissue, and 0% of normal liver tissue. And it was significantly correlated with tumor grade, venous invasion, intrahepatic metastasis, necrosis, and capsule infiltration. Correlation analysis of HIF-2α/EPAS1 with angiogenic factor VEGF (P < 0.001), and MVD (P = 0.016) was also noted. HIF-2α/EPAS1 protein was less frequently expressed in low MVD cases, whereas a high rate of expression was noted in cases with both medium and high MVD (P = 0.042). By Kaplan-Meier analysis, strong HIF-2α/EPAS1 staining (> 50% of tumor cells) in HCC correlated with a shortened survival in patients (Cox's regression, P < 0.001, r = 3.699). CONCLUSION: We conclude that HIF-2α/EPAS1 expression may play an important role in tumor progression and prognosis of HCC. Assessment of HIF-2α/EPAS1 expression in HCC may be used as a diagnostic tool and possibly a target in the treatment of HCC.  相似文献
