OBJECTIVE

To assess the antioxidant capacity of aqueous extracts of buds and flowers of Lonicera japonica Thunb. (BLJ and FLJ) using in vitro assays.

METHODS

We assessed the in vitro antioxidant activities of aqueous extracts of BLJ and FLJ and compared with that of classical antioxidants vitamin C and butylated hydroxytoluene, using several well-established methods including the 1,1-diphenyl-2-picryl-hydrazyl assay, 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) assay, reducing power assay, fluorescence recovery after photo-bleaching assay, β-carotene bleaching assay, ferric thiocyanate assay, and thiobarbituric acid method.

RESULTS

The aqueous extracts of both BLJ and FLJ had similarly potent antioxidant capacity. There were no significance differences between BLJ and FLJ in all the assays.

CONCLUSION

The aqueous extracts of both BLJ and FLJ have antioxidant activity with comparable efficacy. These findings suggest that both BLJ and FLJ may have the potential as natural antioxidants.  相似文献

Objective

To evaluate the anticancer, antioxidant and antimicrobial activities along with total phenolic and flavonoids contents extracted from Lespedeza bicolor indigenous to Khyber Pukhtoonkhwa, Pakistan.

Methods

Anticancer activity against human lung carcinoma (LU-1) and human prostate carcinoma (LnCap), antimicrobial against Aspergillus fumigates, Aspergillus niger, Fusarium solani, Mucor sp Klebsiella pneumonia and Enterococcus and antioxidant activity on 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) and Hydrogen peroxide radicals scavenging activity were assessed in methanol extract of Lespedeza bicolor.

Results

Lespedeza bicolor aerial parts and root extract showed IC50 value of 12.5 and 50 μg/mL against LU-1 whereas, ≤ 12.5 and 12 μg/mL were calculated against LnCap cell line. Minimum inhibition concentration (MIC) value of 20–35 μg/mL has been observed against Aspergillus fumigates, Aspergillus niger, Fusarium solani and Mucor sp in comparision with 1–2.5 μg/mL of Terbinafine used as a standard fungicide. MIC value of 20 and 35 μg/mL of Lespedeza bicolor aerial parts and root extract against bacterial pathogen Klebsiella pneumonia and 20–50 μg/mL against Enterococcus has been measured. DPPH radical scavenging activity of Lespedeza bicolor with IC50 values of ≤ 50 μg/mL and ≤ 200 μg/mL was observed whereas, hydrogen peroxide scavenging activity with IC50 values of ≤ 25 μg/mL for aerial parts and ≤ 50 μg/mL for the root extract of Lespedeza bicolor has been shown.

Conclusion

These data suggested that the methanolic extract of Lespedeza bicolor could be potential candidates for natural antioxidants and anticancer.  相似文献

OBJECTIVE

To assess the phytochemical contents and antioxidant activities of crude extracts from selected Tulbaghia species.

METHODS

Standard methods were used for preliminary phytochemical analysis. The total phenolic acid contents of the plant extracts were determined using the Folin-Ciocalteu method, and the total flavonoid contents were determined using the aluminum chloride colorimetric method. 1,1-Diphenyl-2-picrylhydrazyl and 2,2’-Azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) assays were used to evaluate the antioxidant activities.

RESULTS

Phytochemical screening showed flavonoids, glycosides, tannins, terpenoids, saponins, and steroids were present in the Tulbaghia species. The total phenolic acid and flavonoid contents varied in the different plant extracts, ranging from 4.50 to 11.10 mg of gallic acid equivalents per gram of fresh material and 3.04 to 9.65 mg of quercetin equivalents per gram, respectively. The IC50 values determined for Tulbaghia alliacea and Tulbaghia violacea based on 1,1-diphenyl-2-picrylhydrazyl (0.06 and 0.08 mg/mL, respectively) and 2,2’-Azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (0.06 and 0.03 mg/mL, respectively) were low and showed they had potential antioxidant activities.

CONCLUSION

Our results suggest that individual compounds from Tulbaghia species should be isolated for analysis of their antioxidant activity because some compounds may work best when pure.  相似文献

Objective

To evaluate the in vivo antiplasmodial activity and the oral acute toxicity of the Bombax buonopozense root bark aqueous extract.

Methods

The in vivo antiplasmodial activity of the root bark aqueous extract of Bombax buonopozense against early and established rodent malaria infections in chloroquine sensitive Plasmodium berghei strain in mice was investigated, and oral acute toxicity of the aqueous root bark extract of Bombax buonopozense was also evaluated in mice.

Results

The findings of this study revealed significant (P < 0.05) and dose dependent decrease in parasitaemia in the parasitized groups treated with varying doses of the extract (50–200 mg/kg p.o.) in both suppressive and curative tests. There was also significant decrease in parasitaemia density in the chloroquine treated group. The aqueous extract was found no toxicity in mice and the oral LD50 was determined to be greater than 5000 mg/kg.

Conclusion

Bombax buonopozense root bark aqueous extract possesses potent antiplasmodial activity and may therefore, serve as potential sources of new antimalarial agents.  相似文献

OBJECTIVE

To evaluate the anti-platelet aggregation effects of extracts from 31 Traditional Chinese Medicines (TCM) with the property of activating blood and resolving stasis in terms of TCM theory.

METHODS

The 31 TCMs extracts were prepared using water, 90% ethanol and ethyl acetate., and the effects on anti-platelet aggregation were tested on a platelet aggregation analyzer in vitro with adenosine 5'-diphosphate, bovine thrombin and arachidonic acid (AA) as aggregation inducers, respectively. Aspirin was the positive control.

RESULTS

Lots of the tested TCMs had inhibitory effects with concentration-dependent manner on platelet aggregations induced by various agonists. Especially, some of the TCMs such as Chuanxiong (Rhizoma Chuanxiong), Yanhusuo (Rhizoma Corydalis Yanhusuo) and Danshen (Radix Salviae Miltiorrhizae) showed good anti-platelet aggregation effect similar or higher than that in positive control group.

CONCLUSION

The study provided scientific references that several TCMs such as Chuanxiong (Rhizoma Chuanxiong), Yanhusuo (Rhizoma Corydalis Yanhusuo) and Danshen (Radix Salviae Miltiorrhizae), possess the property of anti-platelet aggregation.  相似文献

Objective

To investigate saponins and various solvent extracts from Atriplex laciniata (A. Laciniata) against human parasites and various pests.

Methods

The samples from A. laciniata used in the activities were crude saponins (Al.SpF) and solvent samples including methanolic extract (Al.MeF), ethyl acetate (Al.EaF), choloroform (Al.CfF), n-hexane (Al.HxF) and water residual (Al.WtF). Anthelmintic potentials of the samples were analyzed against Pheretima posthuma (earthworms) and Ascaridia galli (round worms) using contact toxicity method. Insecticidal activities were performed against Heterotermes indicola (termite), Monomorium pharaonis (Pharaoh ant), Tribolium castaneum (flour beetle) and Rhyzopertha dominica (grain borer) using standard protocols.

Results

In anthelmintic assay, Al.CfF and Al.SpF were most effective against P. posthuma and A. galli with average death times of 25.62 and 29.65 min respectively. Likewise the anthelmintic assay, Al.SpF and Al.CfF were most effective against H. indicola causing 90.36% and 73.24% lethality respectively. Furthermore, in anti-Pharaoh activity Al.SpF, Al.WtF, Al.CfF, Al.MeF and Al.CfF exhibited highest activity with LD50 of 78, 220, 260, 330 and > 800 mg/mL respectively. Al.SpF and Al.CfF were highly effective against R. dominica causing 80.11% and 71.30% lethality respectively. Al.SpF was found most active against T. castaneum.

Conclusion

Our findings suggest that the Al.SpF, Al.CfF and Al.WtF extracted from A.laciniata L. may be the best options for the isolation of anthelmintic and bio-insecticidal compounds.  相似文献

OBJECTIVE

To evaluate the anti-tumor activity of ethyl acetate fraction (EFA), extracted with ethanol from the root of “Dai-Bai-Jie” in A549 cancer cells and its underlying mechanism.

METHODS

“Dai-Bai-Jie” was extracted with 95% ethanol-aqueous (DBJ-1), 50% ethanol-aqueous (DBJ-2), and water (DBJ-3) by reflux method. 95% ethanol-aqueous extract was separated byethyl acetate (EFA) and n-butyl alcohol (DBJ-5), consecutively. The SRB method was used to evaluate the cytotoxic activity. Annexin V-FITC staining was applied to observe the apoptosis and analyze the cell cycle activated by EFA in A549 tumor cell. Western blot was used to detect the apoptosis/related proteins expressions. A549 tumor cellsbearing nude mice model was employed to measure the tumor volume, mice weight, and tumor inhibition ratio in order to verify the antitumor activity in vivo.

RESULTS

DBJ-1 and EFA showed better cytotoxic activity on A549 tumor cells with IC50 25 and 3.5 µg/mL, respectively. EFA can exhibit the proliferation, arrest cell cycle at G0/G1 phase, and induce apoptosis in A549 tumor cells in vitro. The mechanisms of apoptosis induced by EFA may be associated with decreasing Bcl-2 protein expression and increasing p53, Bax, Caspase-3, and Caspase-8 proteins expression. EFA also possessed significant anti-tumor efficacy in nude mice, and little toxicity was observed in the host.

CONCLUSION

EAF could induce A549 tumor cells apoptosis and G0/G1 cell cycle arrest. A549 tumor cells apoptosis induced by EAF may be associated with the decrease in the ratio of Bcl-2 and Bax mRNA levels, and increase in the expression of p53, Caspase-3, and Caspase-8 proteins.  相似文献

Objective This research aimed to evaluate the protective effects of bioactive compounds such as phenolic acids, flavonoids, and tannins present in four species extracted with methanol. Methods The total phenolic content of the methanolic extracts was measured spectrophotometrically. The effect of the extracts on cell viability in U266 cells was measured. The effects of extracts on free radical scavenging were assessed by the DPPH test and FRAP assay. Antibacterial effects of the natural products in this report were investigated by using the disc diffusion method. Results Our results clearly demonstrated that the methanolic extracts were characterized by a high amount of phenolic compounds. It has been speculated that ME-TA and ME-TAl exhibit a significant(P < 0.05) and dose-dependent antiradical potential. The exposure of cells to high doses of extracts almost completely suppressed cell growth in vitro. ME-TA and ME-TAl showed significant cytotoxic effects at a concentration of 100 μg/mL in the U266 cell line. ME-TAl and ME-CF inhibited the growth of B. subtilis and S. aureus, respectively, to the same extent as 10 μg/μL of chloramphenicol at a concentration of 1 mg/mL. Conclusion Overall, these results suggest that plants used in traditional medicine have a novel application as free radical scavengers, bacterial inhibitors and tumor suppressors.  相似文献

OBJECTIVE

To pool data on ethnobotanical medicine for the treatment of inflammatory disorders in Pakistan.

METHODS

We reviewed 237 research publications based on data from the six provinces in Pakistan (Punjab = 85, Khyber Pakhtunkhwa = 65, Sindh = 15, Balochistan = 8, Gilgit Baltistan = 22, Azad Jammu and Kashmir = 42) published until June 2015 in various journals. This was achieved using seven online databases: ScienceDirect, Google, Google Scholar, PubMed, Wiley Online Library, SpringerLink, and MEDLINE. Data were analyzed from different perspectives.

RESULTS

People from Pakistan made use of 371 plant species belonging to 263 genera and 99 families for the treatment of inflammatory disorders. Plants from the Asteraceae family were used most often. Herbs were the dominant growth form. Leaves were the plant parts used most often. Decoctions were the main preparation method. Nine plant species were used most frequently in the dwellers of most regions of Pakistan. A total of 111 plants were shown experimentally to have neither anti-arthritic nor anti-inflammatory activities, and 148 plant species were threatened. Eighty-four species had commercial importance. Twelve plant species were imported, and 25 plant species were exported, from Pakistan.

CONCLUSION

This review provides baseline data for plant species in Pakistan that have potential anti-inflammatory/anti-arthritic activities.  相似文献

OBJECTIVE

To investigate the inhibitory effect of Sumu (Lignum Sappan) plus Fuzi (Radix Aconiti Lateralis Praeparata) (SF) on the growth and metastasis of Lewis lung carcinoma.

METHODS

A lung carcinoma model was established by subcutaneously inoculating Lewis lung carcinoma cells into C57BL/6 mice. The mice were randomly divided into four groups of 13 mice (control, low-dose, moderate-dose, and high-dose), and gavaged once-daily for 21 consecutive days. The rates of tumor inhibition, metastasis, and metastasis inhibition were observed. The differential expressions of sP-selectin and vascular endothelial growth factor C (VEGFC) were compared between the treatment groups and the control group.

RESULTS

The tumor weights differed significantly between the treatment groups and the control group (P < 0.05). Administration of SF in the moderate-dosage and low-dose groups significantly inhibited the expression of sP-selectin and VEGFC (both P < 0.05), suggesting anti-tumor activity.

CONCLUSION

SF can inhibit the growth and metastasis of Lewis lung carcinoma.  相似文献

Objective

To investigate the effect of Sharbat-e-Deenar (SD) on acetaminophen (APAP)-induced hepatotoxicity in rat model.

Methods

Albino rats were treated with SD at the doses of 1, 2 and 4 mL/kg, p.o. against hepatotoxicity after APAP (2 g/kg, p.o. once only) intoxication. The blood, tissue biochemical parameters and histopathological observation were performed. The

Results

APAP exposure in rats significantly increased the level of biochemical parameters such as aspartate aminotransaminase, alanine aminotransaminase, lactate dehydrogenase, serum alkaline phosphatase, bilirubin, urea and creatinine into blood circulation which were reversed towards normal by SD therapy at all doses. The tissue biochemical parameters such as lipid peroxidation, reduced glutathione, adenosine tri-phosphatase and glucose-6-phosphatase were significantly restored after SD treatment against hepatotoxity. Histological analysis confirmed that SD-treated rats significantly alleviated of liver damage and reduced lesions caused by APAP intoxication. The biochemical changes are in good correlation with the histopathological observations.

Conclusion

On the basis of these results, it can be concluded that SD exerts hepatoprotective activity against APAP-induced liver injury.  相似文献

OBJECTIVE

To investigate the effect of brain functional recovery decoction (BFRD) on expression of vascular endothelial growth factor (VEGF) and angiopoietin-1 (Ang-1) protein in rats with cerebral ischemia reperfusion injury, and to explore the mechanism of action of BFRD.

METHODS

Using the suture-occlusion method, a Wistar rat model of focal cerebral ischemia reperfusion was established. The rats were randomly divided into treatment group, model group, and sham operation group. The treatment group was administered BFRD. In situ hybridization was used to detect VEGF mRNA expression. Immunohistochemistry was used to observe expression of Ang-1 protein.

RESULTS

VEGF mRNA expression was greater in the model group compared with the sham operation group (P < 0.05); Ang-1 protein expression was more obvious in the treatment group than the model group (P< 0.05).

CONCLUSION

BFRD promoted VEGF mRNA and Ang-1 protein expression in the brains of rats with cerebral ischemia, suggesting increased angiogenesis.  相似文献

Objective

To investigate the neural differentiation capacity of water extraction of velvet antler.

Methods

Velvet antler (Cervus Nippon Temminck) polypeptide (VAP) was used to differentiate neural stem cells (NSCs) towards neurons in the study. Firstly, we obtain the polypeptides of VAP by water extraction. Secondly, we observed the morphology, assayed the factors in the media by enzyme-linked immunosorbent assay, and detected the special neural molecules by immunofluorescence staining. NSCs were cultured on the cell climbing film. After neuronal differentiation, differentiated NSCs were mounted for immunocytochemistry with immunofluorescence technique.

Results

The differentiating cells look like neuron, some special factors, such as Glial cell line-derived neurotrophic factor, nerve growth factor, in the media can be detected while differentiated neuron -like cells can express the special neural molecules.

Conclusion

Differentiation of NSCs towards neurons can be induced by velvet antler polypeptide.  相似文献

Objective

To evaluate the effects of baicalin in human gastric cancer cells, including apoptosis-inducing effects, and to investigate its underlying mechanisms of action.

Methods

Cell proliferation and apoptosis assays were performed to investigate the anti-proliferation effects of baicalin in human gastric cancer BGC-823 and MGC-803 cells. Real time-quantitative polymerase chain reaction and Western blotting analysis were performed to elucidate the molecular mechanisms underlying the anti-tumor properties of baicalin.

Results

In BGC-823 and MGC-803 gastric cancer cells treated with 80, 120, and 160 µmol/L baicalin for 48 h, a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay showed that baicalin significantly inhibited cell proliferation in a dose-dependent manner, while flow cytometric analysis demonstrated that baicalin could induce apoptosis, also in a dose-dependent manner. Moreover, baicalin up-regulated the expression of caspase-3, caspase-9, and B cell lymphoma (Bcl-2)-associated X protein and down-regulated the expression of Bcl-2 at both the mRNA and protein level.

Conclusion

Baicalin has potential as a therapeutic agent for gastric cancer by inducing apoptosis in cancer cells.  相似文献

Objective

To evaluate the clinical efficacy of Chinese medicine formula HB01 on exudative age-related macular degeneration (AMD) patients who received HB01 for 6 months by analyzing relative indexes.

Methods

From July 2006 to August 2015, 139 eligible exudative AMD patients (183 eyes) were enrolled into this study and received aqueous extract from herbs of HB01 by oral twice a day (100 mL for once) for 6 months. The best corrected visual acuity, center macular thickness (CMT), hemorrhage area, fluorescein leakage area as the main outcomes were estimated and compared before and after treatment.

Results

After 3 and 6 months of treatment, visual acuity obtained a greater improvement (P < 0.01). After 1, 3 and 6 months of treatment, CMT obviously decreased (P < 0.05). After 3 and 6 months of treatment, the hemorrhage area and fluorescein leakage area also significantly narrowed (P < 0.01). During treatment, no significant adverse events relating to HB01 treatment were elucidated.

Conclusion

On the basis of these results, HB01 could improve visual acuity, and reduce hemorrhage and fluorescein leakage of patients with exudative AMD, which may be an effective alternative and supportive treatment for exudative AMD.  相似文献

Objective

To determine differences in cerebral activity evoked by acupuncture and conventional stroke treatment, and identify the treatment targets.

Methods

In total, 21 patients were randomly divided into two groups. Group A (11 patients) received both acupuncture and conventional treatment, while Group B (10 patients) received conventional treatment only. Resting-state functional magnetic resonance imaging (fMRI) was performed on each participant before and after treatment. Regional homogeneity (ReHo) analysis was performed to investigate the potential mechanism of acupuncture treatment by comparing differences in cerebral activity between treatments.

Results

Group A showed higher ReHo in the frontal lobe (BA6, BA46), supra-marginal gyrus (BA40), middle temporal gyrus (BA21), cerebellum, and insula. Group B showed higher ReHo in the frontal lobe (BA6) and parietal lobe (BA3, BA7).

Conclusion

Acupuncture and conventional treatment triggered relatively different clinical efficacy and brain responses. Acupuncture treatment more significantly improved the symptoms of stroke patients. More marked changes in sensory, emotional, and motor areas (including the frontal lobe, middle temporal gyrus, cerebellum, and insula) might reflect the specific acupuncture mechanism.  相似文献

OBJECTIVE

To investigate the antidepressant-like effect of active fraction of Polyrhachis vicina Roger (AFPR) in a rat depression model, and to elucidate the underlying mechanism.

METHODS

AFPR was extracted with ethanol followed by petroleum ether. Its antidepressant-like effect was investigated in mice by tail suspension test (TST), forced swimming test (FST) and open field test (OPT). A repeated dose of reserpine (0.5 mg/kg, daily for 14 d) was used to establish a rat depression model. Fluoxetine was used as positive control agent. The effect of AFPR on reserpine-induced ptosis, hypothermia and akinesia, the levels of monoamines and their metabolites, and the activity of monoamine oxidase (MAO) in hippocampus and prefrontal cortex were determined.

RESULTS

Administration of AFPR by gavage at 160 and 320 mg/kg significantly reduced the duration of immobility in the FST and TST, and did not affect locomotor activity in the OPT. In the reserpine-induced depression model, AFPR attenuated anhedonia, demonstrated by reversing hypothermia, akinesia and sucrose consumption. AFPR significantly increased the concentration of monoamines, including dopamine, serotonin, noradrenaline and acetylcholine.

CONCLUSION

AFPR normalized the metabolism rates of noradrenaline, serotonin and dopamine, and the activity of MAO, which were altered by chronic reserpine exposure. The findings suggest that modulation of the monoaminergic neurotransmitter system likely underlies the antidepressant-like effect of AFPR.  相似文献
