The aim of this study was to evaluate efficacy of an isoxazole derivative RM11 to accelerate reconstitution of selected immune activities in cyclophosphamide (CP)-immunocompromised mice. We demonstrated that administration of fifteen 10 mug intraperitoneal doses of RM11, following a sublethal (200 mug/kg) dose of CP, significantly stimulated the number of antibody-forming cells (AFC) to sheep erythrocytes (SRBC) as determined 35 days after the CP treatment. Similarly, treatment of the CP-injected mice with 7 doses of RM11 significantly enhanced generation of delayed type hypersensitivity (DTH) to ovalbumin (OVA). Moreover, in that model, the treatment of mice with RM11 accelerated the process of myelopoiesis. RM11 also counteracted the suppressive action of methotrexate (MTX) in the in vitro model of the humoral immune response to SRBC. The phenotypic studies with fluorocytometer revealed that intraperitoneal 10 mug dose of RM11 significantly elevated the percentage of mature (CD3(+), CD4(+) and CD8(+)) T cells in the spleen and down-regulated the content of CD19(+) cells. We conclude that RM11 may be of potential therapeutic value in restoration of the immune status in patients undergoing chemotherapy.  相似文献

目的研究饥饿对小鼠非特异性免疫和特异性免疫功能的影响。方法根据小鼠进食量的75%、50%、25%、100%分成高、中、低剂量及空白对照组,根据小鼠进食量计算出100%进食组每日进食量约为0.4g/只,高、中、低剂量组分别为0.3g/只、0.2g/只、0.1g/只,连续喂养10d后,测定各组小鼠的体重、胸腺和脾脏重量,计算胸脾指数;检测中性粒细胞的吞噬率、T淋巴细胞Ea花环形成率。结果与对照组比较,高、中、低剂量组的小鼠的体重、胸腺及脾脏指数明显降低(P〈0.05)。结论饥饿能够影响小鼠非特异性免疫功能。  相似文献

目的:研究牛至油对小鼠免疫功能的影响。方法:采用ConA诱导的小鼠脾淋巴细胞转化实验(MTT法)、NK细胞活性测定、小鼠腹腔巨噬细胞吞噬鸡红细胞实验等方法考察牛至油对小鼠免疫功能的作用。结果:牛至油在促进NK细胞活性和单核-巨噬细胞方面的具有明显的作用,但在ConA诱导的小鼠脾淋巴细胞转化能力试验中并未表现出显著的差异。结论:牛至油确实具有增强机体免疫功能的作用。  相似文献

目的研究康姜牌八宝颗粒剂对正常小鼠免疫功能的影响。方法将小鼠随机分为两组比较疗效。治疗组口服给予不同剂量的八宝颗粒剂(分别为2000mg/(kg·bw)、4000mg/(kg·bw)和12000mg/(kg·bw)),对照组采用生理盐水灌服。观察八宝颗粒剂对小鼠刀豆球蛋白(ConA)诱导的脾淋巴细胞转化、抗体生成细胞(溶血空斑数)、NK细胞的作用。结果康美牌八宝颗粒剂可促进ConA诱导小鼠脾淋巴细胞的增殖.促进小鼠抗体生成细胞的形成,提高NK细胞的活性。结论康美牌八宝颗粒剂可明显增强小鼠免疫能力。  相似文献

目的:研究青蒿琥酯对小鼠免疫功能的影响. 方法:溶血素含量用分光光度法测定,血清IgG和C3含量用单向免疫扩散法测定,淋巴细胞转化率、巨噬细胞吞噬百分率和吞噬指数镜检计数.结果:青蒿琥酯im 75 mg kg~(-1) bidfor 5—7d能降低SRBC致敏小鼠血清溶血素和IgG的含量,抑制抗体生成,但增加疟鼠补体C3的含量.青蒿琥酯能促进PHA诱导的小鼠体内淋巴细胞转化,能提高DNFB所致的迟发型超敏反应,并减少腹腔巨噬细胞的吞噬百分率和吞噬指数.结论:青蒿琥酯对体液免疫有抑制作用,但对细胞免疫有促进作用.  相似文献

Mice were injected with ethanol (2.5 g/kg body wt., i.p.) 1-3 times daily for 17 days or 3 times daily for 49 days. The primary and secondary antibody titers to sheep red blood cells were determined. In addition, microhematocrits, white blood cell counts, white blood cell differential counts, and organ-to-body-weight ratios were determined. Despite continuous large doses of ethanol, the ethanol-treated mice responded as well as saline-treated controls in all parameters tested. The results of this study are discussed in the context of other studies on the effects of ethanol on the immune system.  相似文献

目的:研究地黄低聚糖(RGOS)对快速老化模型小鼠(SAMP8)造血功能的影响。方法:采用造血祖细胞体外克隆培养等实验血液学方法。结果:RGOS10,20mg·kg-1可使SAMP8小鼠的CFU-S、CFU-GM、CFU-E和BFU-E明显增多,并可使外周血中降低的WBC数明显增加。提示RGOS可刺激SAMP8小鼠造血干细胞、祖细胞的增殖分化。集落刺激活性实验显示RGOS可使小鼠体内的集落刺激因子产生明显增多。小鼠骨髓组织学研究结果也进一步证实了RGOS增强SAMP8小鼠造血功能的作用。结论:RGOS可以增强SAMP8小鼠的造血功能。  相似文献

Safrole, a component of piper betle inflorescence, is a documented rodent hepatocarcinogen and inhibits bactericidal activity and the release of superoxide anion (O(2-)) by polymorphonuclear leukocytes (PMNs). In the present study, we investigated the effects of safrole on immune responses, including natural killer (NK) cell cytotoxicity, phagocytic activity and population distribution of leukocytes from normal BALB/c mice. The cells population (cell surface markers) and phagocytosis by macrophages and monocytes from the peripheral blood mononuclear cells (PBMCs) were determined, and NK cell cytotoxicity from splenocytes of mice after oral treatment with safrole was performed using flow cytometric assay. Results indicated that safrole did not affect the weights of body, spleen and liver when compared with the normal mice group. Safrole also promoted the levels of CD11b (monocytes) and Mac-3 (macrophages) that might be the reason for promoting the activity of phagocytosis. However, safrole reduced the cell population such as CD3 (T cells) and CD19 (B cells) of safrole-treated normal mice by oral administration. Furthermore, safrole elevated the uptake of Escherichia coli-labelled fluorescein isothiocyanate (FITC) by macrophages from blood and significantly stimulated the NK cell cytotoxicity in normal mice in vivo. In conclusions, alterations of the cell population (the increase in monocytes and macrophages, respectively) in safrole-treated normal BALB/c mice might indirectly influence the immune responses in vivo.  相似文献

It was found in vitro that Tremella polysaccharides (TP) (50, 100, 150 and 200 micrograms/ml) augmented lymphocyte proliferation induced by Con A and did not antagonize the suppressive effect of hydrocortisone on lymphocyte proliferation. In vivo TP promoted the plaque-forming cell (PFC) response to SRBC in mice. TP 50 and 100 mg/kg ip for 5 d produced 77.6% and 81.8% increases in PFC response respectively. At the doses of 150 and 200 micrograms/ml, TP decreased the interleukin 2 (IL-2) activities in the supernatant of culture media of mouse spleen cells. TP (50 micrograms/ml) enhanced the lymphocyte proliferation induced by Con A and increased the PFC response to SRBC by 47.1% in 14-month-old mice.  相似文献

目的 考察三七多糖(PPN)对小鼠免疫功能的影响.方法 分别以133.5、267.0、534.0 mg·kg-1·d-1的PPN多糖经口灌胃给予小鼠30 d,于试验的第30天测定小鼠的细胞免疫、体液免疫、单核巨噬细胞、NK细胞的活性.结果 PPN各剂量组的淋巴细胞转化能力均高于阴性对照组,且高剂量组与阴性对照组的差异具有显著性(P <0.01);PPN各剂量组的左右耳片重量差值均高于阴性对照组,且高、中剂量与阴性对照的差异具有显著性(P<0.01、P<0.05);PPN剂量组的抗体生成细胞数均高于阴性对照组,且高、中剂量与阴性对照的差异具有显著性(P<0.01、P<0.05);PPN各剂量组的抗体积数均高于阴性对照组,且高剂量与阴性对照的差异具有显著性(P <0.05);PPN各剂量组的腹腔巨噬细胞对鸡红细胞的吞噬率及吞噬指数均高于阴性对照组,高剂量的吞噬率与阴性对照组的差异有显著性(P<0.05).结论 PPN可提高小鼠的细胞免疫和体液免疫功能.  相似文献

目的 考察绿原酸对小鼠细胞免疫和体液免疫的影响.方法 通过DNCB致小鼠耳肿胀的迟发超敏反应(DTH)、小鼠脏器巨噬细胞清除异物的功能、对5%鸡红细胞的吞噬功能的影响以及对环磷酰胺致小鼠溶血素低下的影响实验,研究绿原酸对机体细胞免疫和体液免疫的影响.结果 绿原酸可明显增强DTH的耳肿胀度,增强巨噬细胞吞噬功能,提高机体血清溶血素的含量.结论 绿原酸可明显增强机体细胞免疫和体液免疫的功能.  相似文献

Treatment of mice with the direct-acting alkylating agent methyl methanesulfonate produced marked suppression of the humoral response to sheep erythrocytes and suppression of T cell responses to foreign antigens. These effects occurred without loss of spleen, thymus or body weight and in the absence of peripheral blood, splenic or bone marrow cytotoxicity. In comparison, exposure to urethan decreased spleen weights, number and viability of spleen cells, and numbers of circulating lymphocytes. Significant suppression of T cell mitogen responsiveness was observed at all dose levels of urethan. Thymus weights, proliferative responses to the B cell mitogen lipopolysaccharide and delayed hypersensitivity responses were decreased at the highest urethan dose. Cyclophosphamide treatment significantly depressed thymic weight, lymphoproliferative responses of T and B cells, antibody production and delayed hypersensitivity responses. These results suggest differential sensitivity in components of the host defense system to weak carcinogens.  相似文献

The potential effects that the thiocarbamate herbicide Ordram has on the immune system of mice was evaluated following 12 days of acute dosing by oral gavage. Dosages of Ordram ranging from 20 to 320 mg/kg/day had no consistent significant effects on a variety of immune parameters investigated. The immune parameters measured were the following: body and lymphoid organ weights; splenic natural killer (NK) cell activity; lymphoproliferative responses to B and T lymphocyte mitogens and allogeneic spleen cells in a one-way mixed lymphocyte reaction; and delayed-type hypersensitivity and antibody responses to sheep red blood cells (SRBC). The effects that the immunosuppressant cyclophosphamide has on these immune parameters was also examined. The results indicate that Ordram does not appear to affect key parameters of the immune system of mice under the conditions of exposure employed.  相似文献

Cyclophosphamide-treated mice are proposed as a model to assay immunomodulation by antimicrobial agents in immunocompromised animals. Cyclophosphamide-treated BALB/c mice developed temporary leukopenia, myelopenia and spleen atrophy that was followed by splenomegaly. Cells from both atrophic and hypertrophic spleens exhibited impaired responses to mitogens and suppressed the mitogen-driven proliferation of normal splenocytes. This experimental model was applied to the study of immunomodulation by ceftriaxone. Ceftriaxone did not worsen the cyclophosphamide-damaged immunity mechanisms. On the contrary, cyclophosphamide-induced suppression of lymphoproliferation in response to concanavalin A was attenuated by ceftriaxone.  相似文献
