In the myocardium the inhibitory guanine nucleotide-binding regulatory proteins (Gi proteins) mediate negative chronotropic and negative inotropic effects by activation of K+ channels and inhibition of adenylyl cyclase. The concept of a uniform inhibitory action of Gi proteins on myocardial cellular activity has been questioned by the recent observations of adenosine-induced activation of the Na+/Ca2+ exchange and a carbachol-induced inhibition of the Na+/K+-ATPase activity in cardiac sarcolemmal membranes. The aim of the present study, therefore, was to reinvestigate the putative regulation of Na+/Ca2+ exchange and Na+/K+-ATPase activity in purified canine sarcolemmal membranes. These membranes were enriched in adenosine A1 (Maximum number of receptors, B max 0.033 pmol/mg) and muscarinic M2 (B max 2.9 pmol/mg) receptors and contained Gi2 and Gi3, two Gi protein isoforms, and Go, another pertussis toxin-sensitive G protein, as detected with specific antibodies. The adenosine A1-selective agonist, (–)-N 6-(2-phenylisopropyl)-adenosine, and the muscarinic agonist, carbachol, both inhibited isoprenaline-stimulated adenylyl cyclase activity by 25% and 35% respectively, and the stable GTP analogue 5-guanylylimidodiphosphate inhibited forskolin-stimulated adenylyl cyclase activity by 35% in these membranes. The characteristics of Na+/Ca2+ exchange and Na+/K+-ATPase activity as well as those of the ouabain-sensitive, K+-activated 4-nitrophenylphosphatase, an ATP-independent, partial reaction of the Na+/K+-ATPase, were in agreement with published data with regard to specific activity, time course of activity and substrate dependency. However, none of these activities were influenced by adenosine, (–)-N 6-(2-phenylisopropyl)-adenosine, carbachol, or stable GTP analogs, suggesting that Na+/Ca2+ exchange and Na+/K+-ATPase are not regulated by Gi proteins in canine cardiac sarcolemmal membranes.  相似文献

目的 研究四肽FMRFa对大鼠单个心室肌细胞Na+/Ca2+交换的作用。方法 用膜片钳全细胞记录法测定成年大鼠心室肌细胞Na+/Ca2+交换电流(INa+/Ca2+)和其他离子通道电流。结果 FMRFa对大鼠心室肌细胞INa+/Ca2+呈浓度依赖性抑制,100μmol·L-1浓度时抑制内向和外向INa+/Ca2+密度分别达60.1%和56.5%,对内向电流及外向电流的IC50分别为20μmol·L-1和34μmol·L-1。FMRFa5μmol·L-1抑制INa+/Ca2+内向和外向电流密度分别为38.7%和34.9%,但FMRFa5μmol·L-1及20μmol·L-1对L型钙电流、钠电流、瞬时外向电流和内向整流钾电流均无显著抑制作用。结论 FMRFa对大鼠心室肌细胞是一个特异性Na+/Ca2+交换抑制剂。  相似文献

Persistent neuroinflammation and microglial activation play an integral role in the pathogenesis of many neurological disorders. We investigated the role of voltage-gated sodium channels (VGSC) and Na+/H+ exchangers (NHE) in the activation of immortalized microglial cells (BV-2) after lipopolysaccharide (LPS) exposure. LPS (10 and 100 ng/ml) caused a dose- and time-dependent accumulation of intracellular sodium [(Na+)i] in BV-2 cells. Pre-treatment of cells with the VGSC antagonist tetrodotoxin (TTX, 1 μM) abolished short-term Na+ influx, but was unable to prevent the accumulation of (Na+)i observed at 6 and 24 h after LPS exposure. The NHE inhibitor cariporide (1 μM) significantly reduced accumulation of (Na+)i 6 and 24 h after LPS exposure. Furthermore, LPS increased the mRNA expression and protein level of NHE-1 in a dose- and time-dependent manner, which was significantly reduced after co-treatment with TTX and/or cariporide. LPS increased production of TNF-α, ROS, and H2O2 and expression of gp91phox, an active subunit of NADPH oxidase, in a dose- and time-dependent manner, which was significantly reduced by TTX or TTX + cariporide. Collectively, these data demonstrate a closely-linked temporal relationship between VGSC and NHE-1 in regulating function in activated microglia, which may provide avenues for therapeutic interventions aimed at reducing neuroinflammation.  相似文献

Background and Purpose

N-arachidonoyl glycine (NAGly) is a lipoamino acid with vasorelaxant properties. We aimed to explore the mechanisms of NAGly''s action on unstimulated and agonist-stimulated endothelial cells.

Experimental Approach

The effects of NAGly on endothelial electrical signalling were studied in combination with vascular reactivity.

Key Results

In EA.hy926 cells, the sustained hyperpolarization to histamine was inhibited by the non-selective Na+/Ca2+ exchanger (NCX) inhibitor bepridil and by an inhibitor of reversed mode NCX, KB-R7943. In cells dialysed with Cs+-based Na+-containing solution, the outwardly rectifying current with typical characteristics of NCX was augmented following histamine exposure, further increased upon external Na+ withdrawal and inhibited by bepridil. NAGly (0.3–30 μM) suppressed NCX currents in a URB597- and guanosine 5′-O-(2-thiodiphosphate) (GDPβS)-insensitive manner, [Ca2+]i elevation evoked by Na+ removal and the hyperpolarization to histamine. In rat aorta, NAGly opposed the endothelial hyperpolarization and relaxation response to ACh. In unstimulated EA.hy926 cells, NAGly potentiated the whole-cell current attributable to large-conductance Ca2+-activated K+ (BKCa) channels in a GDPβS-insensitive, paxilline-sensitive manner and produced a sustained hyperpolarization. In cell-free inside-out patches, NAGly stimulated single BKCa channel activity.

Conclusion and Implications

Our data showed that NCX is a Ca2+ entry pathway in endothelial cells and that NAGly is a potent G-protein-independent modulator of endothelial electrical signalling and has a dual effect on endothelial electrical responses. In agonist pre-stimulated cells, NAGly opposes hyperpolarization and relaxation via inhibition of NCX-mediated Ca2+ entry, while in unstimulated cells, it promotes hyperpolarization via receptor-independent activation of BKCa channels.  相似文献

Four catechins, epigallocatechin-3-gallate, epigallocatechin, epicatechin-3-gallate, and epicatechin, inhibited activity of the Na+,K+-ATPase. The two galloyl-type catechins were more potent inhibitors, with IC50 values of about 1 μM, than were the other two catechins. Inhibition by epigallocatechin-3-gallate was noncompetitive with respect to ATP. Epigallocatechin-3-gallate reduced the affinity of vanadate, shifted the equilibrium of E1P and E2P toward E1P, and reduced the rate of the E1P to E2P transition. Epigallocatechin-3-gallate potently inhibited membrane-embedded P-type ATPases (gastric H+,K+-ATPase and sarcoplasmic reticulum Ca2+-ATPase) as well as the Na+,K+-ATPase, whereas soluble ATPases (bacterial F1-ATPase and myosin ATPase) were weakly inhibited. Solubilization of the Na+,K+-ATPase with a nonionic detergent reduced sensitivity to epigallocatechin-3-gallate with an elevation of IC50 to 10 μM. These results suggest that epigallocatechin-3-gallate exerts its inhibitory effect through interaction with plasma membrane phospholipid.  相似文献

We examined the intracellular mechanisms for endothelin-1-induced positive and negative inotropic components that coexist in the mouse ventricular myocardium using isolated ventricular tissue and myocytes from 4-week-old mice. In the presence of SEA0400, a specific inhibitor of the Na+–Ca2+ exchanger, endothelin-1 produced positive inotropy. Endothelin-1, when applied to cardiomyocytes in the presence of SEA0400, did not change the peak amplitude of the Ca2+ transient but increased intracellular pH and Ca2+ sensitivity of contractile proteins. On the other hand, in the presence of dimethylamiloride (DMA), a specific inhibitor of the Na+–H+ exchanger, endothelin-1 produced negative inotropy. In cardiomyocytes, in the presence of DMA, endothelin-1 produced a decrease in peak amplitude of the Ca2+ transient. In the presence of both DMA and SEA0400, endothelin-1 produced neither positive nor negative inotropy. Positive inotropy was blocked by BQ-123 and negative inotropy by BQ-788. These results suggested that endothelin-1-induced positive inotropy is mediated by ETA receptors, activation of the Na+–H+ exchanger and an increase in intracellular pH and Ca2+ sensitivity and that the negative inotropy is mediated by ETB receptors, activation of the Na+–Ca2+ exchanger and decrease in Ca2+ transient amplitude.  相似文献

N,N-dimethyl-D-erythro-sphingosine (DMS), an N-methyl derivative of sphingosine, is an inhibitor of protein kinase C (PKC) and sphingosine kinase (SK). In previous reports, DMS-induced intracellular Ca2+ increase concentration ([Ca2+]i) was studied in T lymphocytes, monocytes, astrocytes and neuronal cells. In the present study, we studied DMS-induced increase of [Ca2+]i in HCT116 human colon cancer cells. We found that the DMS-induced increase of [Ca2+]i in colon cancer cells is composed of Ca2+ release from intracellular Ca2+ stores and subsequent Ca2+ influx. The Ca2+ release is not related to modulation of inositol 1,4,5-trisphosphate (IP3) receptor or ryanodine receptor. On the other hand, the Ca2+ influx is mediated largely through Ca2+ channels sensitive to verapamil, nifedipine, Ga3+, and La3+. Furthermore, we found that the response is inhibited by bepridil and Ni2+, specific inhibitors of Na+-Ca2+-exchanger, suggesting involvement of Na+-Ca2+ exchanger in the DMS-induced [Ca2+]i increase in colon cancer cells. Summary To clarify the effects of hypoxia on stimulus-release coupling, we have examined the effects of hypoxia on nicotine-induced catecholamine (noradrenaline and adrenaline) release from, and 22Na+ influx, 45Ca2+ influx and cytosolic free Ca2+ concentration ([Ca2+]i) in, cultured bovine adrenal chromaffin cells. Experiments were carried out in media pre-equilibrated with 21% O2/79% N2 (control) or with 0% O2/100% N2 (hypoxia). Cells were stimulated with either nicotine (activating nicotinic acetylcholine (ACh) receptors) or a high K+ concentration (55 mmol/1 KCI; directly activating voltage-dependent Ca2+ channels). Hypoxia reduced both nicotine- and high K+-induced catecholamine releases from the cells, but the reduction of the former (to about 30% of the control value) was more pronounced than that of the latter (to about 40% of the control value). Nicotine-induced 22Na+ influx, which is considered to reflect the function of nicotinic ACh receptors, was inhibited by hypoxia. Both nicotine- and high K+-induced 45Ca2+ influx into the cells were reduced by hypoxia, but the reduction of the former was more pronounced than that of the latter. Nicotine- and high K+-induced increases in [Ca2+]i were reduced by hypoxia to about 30% and 40% of the control values, respectively. These results suggest that hypoxia reduces cation influxes (Na+ and Ca2+) through both the ligand-gated cation channels of the nicotinic ACh receptor and the voltage-dependent Ca2+ channels. The effects of ouabain, an inhibitor of the plasmalemmal Na+/K+-ATPase activity, were examined in human isolated bronchus. Ouabain produced concentration-dependent contraction with –logEC50=7.16±0.11 and maximal effect of 67±4% of the response to acetylcholine (1 mM). Ouabain (10 M)-induced contraction was epithelium-independent and was not depressed by inhibitors of cyclooxygenase and lipoxygenase, antagonists of muscarinic, histamine H1-receptors and -adrenoceptors, or neuronal Na+ channel blockade. The inhibition of ouabain contraction in tissues bathed in K+-free medium, and the inhibition by ouabain of the K+-induced relaxation confirm that the contractile action of ouabain is mediated by inhibition of Na+/K+-ATPase. Furthermore, depolarization (16.4±0.9 mV) was observed in human isolated bronchus by intracellular microelectrode recording. Ouabain (10 M)-induced contractions were abolished by a Ca2+-free solution but not by blockers of L-type Ca2+ channels. In human cultured bronchial smooth muscle cells, ouabain (10 M) produced a sustained increase in [Ca2+]i (116±26 nM) abolished in Ca2+-free medium. Incubation with a Na+-free medium or amiloride (0.1 mM) markedly inhibited the spasmogenic effect of ouabain thus suggesting the role of Na+/Ca2+ exchange in ouabain contraction while selective inhibitors of Na+/H+-antiport, Na+/K+/Cl-antiport, or protein kinase C had no effect. Ouabain (10 M) failed to increase inositol phosphate accumulation in human bronchus. Ouabain (10 M) did not alter bronchial responsiveness to acetylcholine or histamine but inhibited the relaxant effects of isoprenaline, forskolin, levcromakalim, or sodium nitroprusside. These results indicate that ouabain acts directly to produce contraction of human airway smooth muscle that depends on extracellular Ca2+ entry unrelated to L-type channels and involving the Na+/Ca2+-antiporter.  相似文献

以Fura-2/AM为细胞内钙离子的荧光指示剂,用AR-CM-MIC阳离子测定系统,直接测定了体外培养的新生大鼠神经细胞内游离钙([Ca2+]i)值,并观察了小檗碱(Ber)的影响。结果表明,Ber对神经细胞静息[Ca2+]i无明显影响,Ber1~100μmol·L-1能剂量依赖地抑制去甲肾上腺素和H2O2引起的[Ca2+]i升高,其IC50分别为39.9和17.9μmol·L-1。高剂量Ber(10~100μmol·L-1)能抑制高K+引起的[Ca2+]i升高。姐果提示,Ber对去甲肾上腺素,高K+及H2O2引起的[Ca2+]i升高的抑制作用可能是其抗脑缺血作用机制之一。  相似文献

Ca2+ ions are essential to myonecrosis, a serious complication of snake envenomation, and heparin seems to counteract this effect. We investigated the effect of local injection of Bothrops jararacussu venom in mouse fast-twitch extensor digitorum longus (EDL) muscle, without or with heparin, on functional/molecular alterations of two central proteins involved in intracellular Ca2+ homeostasis, sarco(endo)plasmic reticulum Ca2+-ATPase (SERCA) and Na+/K+-ATPase. EDL-specific SERCA1 isoform expression dropped significantly just after venom administration (up to 60% compared to control EDL values at days 1 and 3; p < 0.05) while SERCA2 and Na+/K+-ATPase α1 isoform expression increased at the same time (3-6- and 2-3-fold, respectively; p < 0.05). Although not significant, Na+/K+-ATPase α2 isoform followed the same trend. Except for SERCA2, all proteins reached basal levels at the 7th day. Intravenous heparin treatment did not affect these profiles. Ca2+-ATPase activity was also decreased during the first days after venom injection, but here heparin was effective to reinstate activity to control levels within 3 days. We also showed that B. jararacussu venom directly inhibited Ca2+-ATPase activity in a concentration-dependent manner. Our results indicate that EDL SERCA and Na+/K+-ATPase are importantly affected by B. jararacussu venom and heparin has protective effect on activity but not on protein expression.  相似文献

BACKGROUND AND PURPOSE

Pulmonary transepithelial Na+ transport is reduced by hypoxia, but in the airway the regulatory mechanisms remain unclear. We investigated the role of AMPK and ROS in the hypoxic regulation of apical amiloride-sensitive Na+ channels and basolateral Na+K+ ATPase activity.

EXPERIMENTAL APPROACH

H441 human airway epithelial cells were used to examine the effects of hypoxia on Na+ transport, AMP : ATP ratio and AMPK activity. Lentiviral constructs were used to modify cellular AMPK abundance and activity; pharmacological agents were used to modify cellular ROS.

KEY RESULTS

AMPK was activated by exposure to 3% or 0.2% O2 for 60 min in cells grown in submerged culture or when fluid (0.1 mL·cm−2) was added to the apical surface of cells grown at the air–liquid interface. Only 0.2% O2 activated AMPK in cells grown at the air–liquid interface. AMPK activation was associated with elevation of cellular AMP : ATP ratio and activity of the upstream kinase LKB1. Hypoxia inhibited basolateral ouabain-sensitive Isc (Iouabain) and apical amiloride-sensitive Na+ conductance (GNa+). Modification of AMPK activity prevented the effect of hypoxia on Iouabain (Na+K+ ATPase) but not apical GNa+. Scavenging of superoxide and inhibition of NADPH oxidase prevented the effect of hypoxia on apical GNa+ (epithelial Na+ channels).

CONCLUSIONS AND IMPLICATIONS

Hypoxia activates AMPK-dependent and -independent pathways in airway epithelial cells. Importantly, these pathways differentially regulate apical Na+ channels and basolateral Na+K+ ATPase activity to decrease transepithelial Na+ transport. Luminal fluid potentiated the effect of hypoxia and activated AMPK, which could have important consequences in lung disease conditions.  相似文献

Thromboxane A2 is a novel endogenous secretagogue of Cl secretion in the distal colon. Here, we examined if the Cl secretion caused by platelet-activating factor (PAF; 1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine) is mediated by thromboxane A2 production using isolated mucosae of the rat colon. Furosemide (100 μM) and 5-nitro-2-(3-phenylpropylamino)-benzoate (NPPB; 300 μM) completely inhibited PAF (10 μM)-induced increase in short-circuit current (Isc) across the mucosa, indicating that PAF caused a Cl secretion in the rat colon. A selective thromboxane A2 receptor antagonist (sodium(E)-11-[2-(5,6-dimethyl-1-benzimidazolyl)-ethylidene]-6,11-dihydrobenz[b,e]oxepine-2-carboxylate monohydrate; KW-3635), and a selective thromboxane synthase inhibitor (sodium 4-[α-hydroxy-5-(1-imidazolyl)-2-methylbenzyl]-3,5-dimethylbenzoate dihydrate; Y-20811) inhibited the PAF-induced Cl current in a concentration-dependent manner. The IC50 values of KW-3635 and Y-20811 were 2.1 and 0.5 μM, respectively. 30 μM KW-3635 and 1 μM Y-20811 inhibited the PAF response by 92% and 83%, respectively. These inhibitors did not affect the prostaglandin E2-induced increase in Isc. A 5-lipoxygenase-activating protein inhibitor (3-[1-(p-chlorobenzyl)-5-(isopropyl)-3-t-butylthioindol-2-yl]-2,2-dimethyl-propanoic acid sodium; MK-886) (5 μM) did not affect the PAF-induced Cl current. The present study suggests that the PAF-induced Cl secretion in the rat colonic mucosa is mainly mediated by a release of thromboxane A2.  相似文献

The release of glutamate from striatal synaptosomes induced by depolarisation with 4-aminopyridine (4-AP) was studied by a method based on the fluorescent properties of the NAPDH formed by the metabolism of the neurotransmitter by glutamate dehydrogenase.Ca2+-dependent, depolarisation-induced glutamate release was inhibited in a concentration-dependent manner by the selective histamine H3 agonist immepip. Best-fit estimates were: maximum inhibition 60±10% and IC50 68±10 nM. The effect of 300 nM immepip on depolarisation-evoked glutamate release was reversed by the selective H3 antagonist thioperamide in a concentration-dependent manner (EC50 23 nM, Ki 4 nM).In fura-2-loaded synaptosomes, the increase in the intracellular concentration of Ca2+ ([Ca2+]i) evoked by 4-AP-induced depolarisation (resting level 167±14 nM; Δ[Ca2+]i 88±15 nM) was modestly, but significantly reduced (29±5% inhibition) by 300 nM immepip. The action of the H3 agonist on depolarisation-induced changes in [Ca2+]i was reversed by 100 nM thioperamide.Taken together, our results indicate that histamine modulates the release of glutamate from corticostriatal nerve terminals. Inhibition of depolarisation-induced Ca2+ entry through voltage-dependent Ca2+ channels appears to account for the effect of H3 receptor activation on neurotransmitter release. Modulation of glutamatergic transmission in rat striatum may have important consequences for the function of basal ganglia and therefore for the control of motor behaviour.  相似文献

Summary The role of inhibition of Na+/Ca2+ exchange in the positive inotropic, negative chronotropic and vasorelaxant responses to amiloride and some of its analogues was investigated in isolated cardiovascular tissues from female Wistar rats. The compounds tested were amiloride, 5-(N-ethyl-N-isopropyl)amiloride (EIPA, a potent inhibitor of Na+/H+ exchange), phenamil and 2,4-dimethylbenzamil (DMB), both potent Na+ channel inhibitors with activity against Na+/Ca2+ exchange, and 5-(N-4-chlorobenzyl)-2,4-dimethylbenzamil (CBDMB), a potent inhibitor of Na+/Ca2+ exchange with reduced activity against Na+ channels compared with its parent compound DMB.Phenamil, DMB and CBDMB increased the force of contraction of right ventricular papillary muscles with similar potencies (-log EC50 values: 4.77 ± 0.06, 5.09 ± 0.09, 4.97 ± 0.17 respectively), while amiloride and EIPA gave small negative inotropic responses. All compounds gave negative chronotropic responses at similar concentrations to those which exerted inotropic effects. Inhibition of KCl contraction of endothelium-free aortic rings was observed with all compounds tested. Phenamil, DMB and CBDMB but not amiloride or EIPA showed a shift to the left of the concentration-response curves in the presence of intact endothelium.These results provide further evidence for positive inotropic and endothelium-dependent vasorelaxant effects of amiloride analogues mediated by inhibition of Na+/Ca2+ exchange. The influence of reducing external Na+ concentration ([Na+]ex) upon vascular smooth muscle contractility was investigated using the rat isolated aorta. NaCl from the physiological saline solution (PSS) was replaced with either choline-Cl, sucrose, or LiCl to give the following [Na+]ex (mM): 115, 85, 55, and 25 (115NaPSS to 25NaPSS). Small reductions in [Na+]ex (115NaPSS) induced a biphasic contraction, comparable in amplitude with the control one induced by phenylephrine 10–6 M. Elimination of the endogenous catecholamine participation using either phentolamine 10–5 M or guanethidine 3.10–6 M similarly reduces these contractions to 25% (sucrose replacement). A similar relaxing effect was obtained with D600 10–5 M, an antagonist of the voltage operated Ca2+ channels (25–30% residual tension for all the substitutes). Large reductions in [Na+]ex (25NaPSS) induced contractions comparable in amplitude and shape, but less sensitive to phentolamine and guanethidine (residual tension 65–75 %, sucrose replacement) and insensitive to D600 (all the substitutes). The Na+/K+ ATPase inhibitor ouabain (10–4 M) elicited slowly developing contractions, the amplitude being 115% of the phenylephrine 10–6 M control.Phenylephrine further contracted the 115NaPSS precontracted preparations, but was significantly less effective in 25NaPSS, although the precontraction levels were similar for the same substitute used. The amplitude of the superimposed phenylephrine contractions exhibited [Na+]ex dependence. Phenylephrine 10–6 M failed to further contract the ouabain 10–4 M precontracted rings.We conclude that relatively small reductions in [Na+]ex are able to induce contractions of rat aorta primarily through release of endogenous catecholamines, probably through neural Na+/Ca2+ exchange. Larger reductions in [Na+]ex appear to cause contraction through muscular Na+/Ca2+ exchange.  相似文献

The present study aimed to demonstrate that interactions of cations, hydrogen peroxide (H2O2) and the Na+-Ca2+exchanger stimulate Ca2+ release and oscillations of cytosolic Ca2+ [Ca2+]i in non-transfected Chinese Hamster Ovary (CHO) C1 cells and in transfected CHO (CK1.4) cells that contained an expression vector coding the Na+-Ca2+ exchanger sequence. 2. The [45Ca2+] uptake assay, fura-2 fluorescence imaging and 22 and 23 factorial orthogonal statistics provide comparative, direct, efficient, quantitative and transient methods to delineate the effects of such interactions on Ca2+ influx, Ca2+release and [Ca2+]i in C1 and CK1.4 cells. 3. In contrast to the control of either Na+-, Ca2+- or H2O2-free or CI cells, an elevated [45Ca2+] uptake was induced by Ca2+, Na+ and H2O2 individually and in combination, intracellular Ca2+ release was activated by H2O2 and by combinations of either H2O2 and Na+, H2O2 and the Na+-Ca2+ exchanger, Na+ and the Na+-Ca2+ exchanger or by H2O2, Na+ and the Na+-Ca2+ exchanger and a rise in [Ca2+]i was triggered by H2O2, Na+ and a combination of Na+ and the Na+-Ca2+exchanger. 4. These results indicate that interactions between H2O2, Na+ and the Na+-Ca2+ exchanger stimulate intracellular Ca2+mobilization via Ca2+-induced Ca2+ release mechanisms, ATP-activated G-protein coupled P2y-purinoceptor-sensitive pathways, Na+-Ca2+ exchanger-mediated Ca2+ influx and cation-π interaction (a strong non-covalent force between the cation and the π face of an aromatic structure in the transmembrane protein). 5. The present findings provide important clues for understanding Ca2+ signal transduction mechanisms from the plasma membrane to the endoplasmic reticulum.  相似文献

BACKGROUND AND PURPOSE

The aim of this study was to clarify the mechanisms by which hydrogen sulphide (H2S) affects ion secretion across rat distal colonic epithelium.

EXPERIMENTAL APPROACH

Changes in short-circuit current induced by the H2S-donor, sodium hydrosulphide (NaHS; 10 mmol·L−1), were measured in Ussing chambers after permeabilization of the apical membrane with nystatin. Cytosolic Ca2+ concentration ([Ca2+]i) and Ca2+ in intracellular stores were measured with fluorescent dyes. Changes in mitochondrial membrane potential were estimated with rhodamine 123.

KEY RESULTS

NaHS had a biphasic effect on overall currents across the basolateral membrane: an initial inhibition followed by a secondary stimulation. Both a scilliroside-sensitive action on the Na+-K+-ATPase and modulation of glibenclamide-sensitive and tetrapentylammonium-sensitive (i.e. ATP-sensitive and Ca2+-dependent) basolateral K+ channels were involved in this action. Experiments with rhodamine 123 revealed that NaHS induced a hyperpolarization of the mitochondrial membrane. NaHS evoked a biphasic change in [Ca2+]i, an initial decrease followed by a secondary increase, known to be mediated by the release of stored Ca2+. Initial falls in [Ca2+]i were not mediated by a sequestration of Ca2+ in intracellular Ca2+ storing organelles, as the Mag-Fura-2 signal was unaffected by NaHS. Falls in [Ca2+]i were inhibited by 2′,4′-dichlorobenzamil, an inhibitor of the Na+-Ca2+-exchanger, and attenuated in Na+-free buffer, suggesting a transient stimulation of Ca2+ outflow by this transporter, directly demonstrated by Mn2+ quenching experiments.

CONCLUSIONS AND IMPLICATIONS

ATP-sensitive and Ca2+-dependent basolateral K+ conductances, the basolateral Na+-K+-pump as well as Ca2+ transporters were involved in the action of H2S in regulating colonic ion secretion.  相似文献

The human embryonic kidney (HEK) 293 cell line is widely used in cell biology research. Although HEK293 cells have been meticulously studied, our knowledge about endogenous G protein-coupled receptors (GPCR) in these cells is incomplete. While studying the effects of bradykinin (BK), a potent growth factor for renal cells, we unexpectedly discovered that BK activates extracellular signal-regulated protein kinase 1 and 2 (ERK) in HEK293 cells. Thus, we hypothesized that HEK293 cells possess endogenous BK receptors. RT-PCR demonstrated the presence of mRNAs for BK B1 and BK B2 receptors in HEK293 cells. Western blotting with BK B1 and BK B2 receptor antibodies confirmed this result at the protein level. To establish that BK receptors are functional, we employed fluorescent measurements of intracellular Ca2+, measured changes in extracellular acidification rate (ECAR) as a reflection of the Na+/H+ exchange (NHE) with a Cytosensor™ microphysiometer, and assessed ERK activation by Western blotting with a phospho-specific ERK antibody. Exposure of HEK293 cells to BK produced a concentration-dependent rise in intracellular Ca2+ (EC50 = 36.5 ± 8.0 × 10−9 M), a rapid increase in tyrosine phosphorylation of ERK (EC50 = 9.8 ± 0.4 × 10−9 M), and elevation in ECAR by ∼20%. All of these signals were blocked by HOE-140 (B2 receptor antagonist) but not by des-Arg10-HOE-140 (B1 receptor antagonist). We conclude that HEK293 cells express endogenous functional BK B2 receptors, which couple to the mobilization of intracellular Ca2+, increases in ECAR and increases in ERK phosphorylation.  相似文献

Aim:

To investigate the reverse mode function of Na+/Ca2+ exchangers NCX1.1 and NCX1.5 expressed in CHO cells as well as their modulations by PKC and PKA.

Methods:

CHO-K1 cells were transfected with pcDNA3.1 (+) plasmid carrying cDNA of rat cardiac NCX1.1 and brain NCX1.5. The expression of NCX1.1 and NCX1.5 was examined using Western blot analysis. The intracellular Ca2+ level ([Ca2+]i) was measured using Ca2+ imaging. Whole-cell NCX currents were recorded using patch-clamp technique. Reverse mode NCX activity was elicited by perfusion with Na+-free medium. Ca2+ paradox was induced by Ca2+-free EBSS medium, followed by Ca2+-containing solution (1.8 or 3.8 mmol/L CaCl2).

Results:

The protein levels of NCX1.1 and NCX1.5 expressed in CHO cells had no significant difference. The reverse modes of NCX1.1 and NCX1.5 in CHO cells exhibited a transient increase of [Ca2+]i, which was followed by a Ca2+ level plateau at higher external Ca2+ concentrations. In contrast, the wild type CHO cells showed a steady increase of [Ca2+]i at higher external Ca2+ concentrations. The PKC activator PMA (0.3-10 μmol/L) and PKA activator 8-Br-cAMP (10-100 μmol/L) significantly enhanced the reverse mode activity of NCX1.1 and NCX1.5 in CHO cells. NCX1.1 was 2.4-fold more sensitive to PKC activation than NCX1.5, whereas the sensitivity of the two NCX isoforms to PKA activation had no difference. Both PKC- and PKA-enhanced NCX reverse mode activities in CHO cells were suppressed by NCX inhibitor KB-R7943 (30 μmol/L).

Conclusion:

Both NCX1.1 and NCX1.5 are functional in regulating and maintaining stable [Ca2+]i in CHO cells and differentially regulated by PKA and PKC. The two NCX isoforms might be useful drug targets for heart and brain protection.  相似文献
