Four hundred and thirty-four isolates of Salmonella enterica serotype Enteritidis were studied. They were grouped into five subsets defined by either the collection criteria or the parameter which formed the basis for subsequent analysis. Seventy-seven per cent harboured the serotype-specific plasmid (SSP). In 55% of the isolates this was the sole plasmid. Molecular variation in the SSP was detected in 17 (5%) of the isolates on the basis of restriction enzyme fragmentation pattern (REFP) analysis using Pst I and Sma I. The SSP variants were further characterized using additional restriction enzymes chosen to optimize the information content and analysed using a coefficient of similarity. A variant SSP designated pOG690 showed greater resemblance to the SSP of Salmonella enterica serotype Typhimurium than Enteritidis; 89% and 68% respectively for Pst I and 79% and 55% respectively for Sma I. In respect of the Pst I data pOG690 shared at least 55 kb of DNA with the Typhimurium SSP and 37 kb with the SSP of Enteritidis. This variant was associated with poultry (duck, goose, chicken) and all isolates belonged to phage type 9b. Other variants were associated with phage types 4, 6, 6a, 9a, 11, 15 and 24. The epidemiological implications of these results are discussed.  相似文献

目的研究2011-2012年杭州市肠道沙门菌临床分离株的型别,了解本地菌株分子流行病学特征。方法对66株肠道沙门菌临床分离株进行血清分型和多位点序列分型(MLST)。对其中主要血清型:鼠伤寒、甲型副伤寒、萨雷甲尼和肠炎沙门菌菌株进行脉冲场凝胶电泳(PFGE)分型。结果分布于21个血清型的66株沙门菌分成26个ST型别。发现一株纽波特沙门菌为新型ST1690。菌株血清型与MLST型别数据库中所对应的血清型符合率为100.00%。9株甲型副伤寒沙门菌的PFGE带型完全一致(P7型),与先前杭州流行菌株有差异(P1-P6型)。6株肠炎沙门菌分成4个PFGE型,型间最小相似性为92.70%。13株鼠伤寒沙门菌分为11个PFGE型,型间最小相似性为71.70%。7株萨雷甲尼沙门菌分成4个PFGE型别,型间最小相似性为91.00%。结论近年杭州腹泻病人中流行的肠道沙门菌菌株主要血清型为鼠伤寒、甲型副伤寒、萨雷甲尼和肠炎等。甲型副伤寒沙门菌菌株在杭州出现了新PFGE型别。MLST数据可以对沙门菌血清学鉴定提供一定的帮助。  相似文献

The relative frequency of salmonella strains isolated from hospitalized and non-hospitalized patients in Southern Israel changed during the period, 1994-6. Salmonella enterica serotype Typhimurium definitive phage-type 104 (DT104) appeared in Israel in 1994 and became the most prevalent strain in 1996. An outbreak of enteritis due to Salmonella enterica serotype Agona occurred in Israel, in October 1994 and lasted for 4 months. The relative frequency of Salmonella enterica serotype Enteritidis remained almost constant during these years, with seasonal fluctuations only. The importance of the increase in the prevalence of Typhimurium DT104 has been the epidemic spread of a multiresistant strain of R-type ACT (A, ampicillin; C, chloramphenicol; T, tetracycline) belonging to this phage-type. Since 1995 the frequency of Typhimurium DT104 isolates that possess, in addition to the above R-type, a chromosomally encoded resistance to the quinolone drug, nalidixic acid, increased tenfold. In 1996, 27% of the Typhimurium DT104 isolates were of R-type ACTN. S. Enteritidis exhibited over 95% susceptibility to at least eight of the most commonly used antibiotic drugs, and none of the isolates was resistant to quinolone or fluoroquinoline.  相似文献

According to the data collected at the Rome-based National Reference Centre for Enterobacteria, 266,547 Salmonella strains isolated from human sources (226,513) and from non human sources(40,034) were characterised in Italy during the period 1973–1995. The serotype of all isolates, and the antibiotype and phage type of selected isolates were determined. Human-source isolations grew from 4372 in 1973 to 12,310 in 1995; non-human source isolations, from 339 in 1973 to 3459 in 1995. Salm.Typhimurium ranked first in the list of the most common serotypes isolated from both sources in the period 1973–1988 except in the years 1975 and 1976 when it was overtaken by Salm. Wien. Since 1983 Salm. Enteritidis has been among the top ten isolates from animals, and ranked first in the list of isolates from humans in 1988 and from non human sources in 1991. During the last years the number of multidrug-resistant strains, mostly belonging to phage types 104 and 193 of Salm. Typhimurium has been rising. Salmonella strains have also been isolated from numerous extraintestinal infections, almost exclusively caused by Salm. Enteritidis and Typhimurium.  相似文献

Between 1998 and 2003, 5,161 isolates (3,182 human) of Salmonella enterica were received by the National Salmonella Reference Laboratory of Ireland. Serotyping, antimicrobial susceptibility testing and phage typing were performed by standard methods. The number of isolates of S. enterica serovar Typhimurium decreased from 579 (80%) in 1998 to 208 (19%) in 2003, while S. enterica serovar Enteritidis increased from 59 (8%) in 1998 to 219 (20%) in 2003. Definitive (DT) phage types 104 and DT104b accounted for a declining proportion of all Salmonella Typhimurium isolates (from n = 523 [90%] in 1998 to 126 [60%] in 2003). Numbers of Salmonella Enteritidis phage type 4 declined from 50 (85%) in 1998 to 59 (27%) in 2003. Twenty-eight isolates of typhoidal Salmonella were received with a history of recent travel in 17 cases. Resistance to multiple (four or more) antimicrobial agents was related to serotype and, where applicable, phage type, and was common in Salmonella Typhimurium. Salmonella Typhimurium predominated among isolates from cattle and pigs (n = 213 [58%]), while Salmonella Livingstone (n = 327) and S. Kentucky (n = 227) were predominant in isolates from poultry (total n = 554 [43%]). This paper discusses trends, and their implications, in Irish salmonella isolates since the establishment of the Reference Laboratory.  相似文献

The pigmy hog (Sus salvanius) is the smallest and the rarest wild suid in the world. This species is on the verge of extinction and the World Conservation Union has rated it among the most endangered of all mammals. This paper reports the investigation into an outbreak of salmonellosis among captive pigmy hogs at the Research and Breeding Centre of the pigmy hog conservation programme, Guwahati, Assam, India. Of 75 pigmy hogs (38 males and 37 females) maintained at the Centre, seven (9.3%) died within five days. The causative organism associated with the outbreak was identified as Salmonella Typhimurium (syn. S. enterica serovar Typhimurium). All the isolates of S. Typhimurium belonged to phage type DT193. The isolates harboured multiple plasmids. Five isolates harboured four (65.0 MDa, 4.2 MDa, 3.0 MDa, 1.3 MDa), while two isolates carried three plasmids (65.0 MDa, 4.2 MDa, 3.0 MDa). All strains showed resistance to amikacin, ampicillin, streptomycin and sulfamerazin; five strains were resistant to oxytetracycline and trimethoprim. All the strains were sensitive to chloramphenicol, ciprofloxacin, enrofloxacin and gentamicin. All seven isolates of S. Typhimurium were found to harbour stn, sopB and pefA genes. However, none of them was found to carry sefC and sopE genes.  相似文献

目的研究肠炎沙门菌菌株间的分子特征,建立了分子流行病学研究方法——多位点基因序列分型技术(MLST),并对食品中的肠炎沙门菌分离株进行分型研究。方法选择肠炎沙门菌的6个管家基因thrA、purE、sucA、aroC、hemD、dnaN及一个特异性的DNA标记SdfΙ作为目标基因。对上述基因分别进行PCR扩增及产物测序,用sequencer2.0软件对测序结果进行分析、比对核苷酸的差异。同时,将肠炎沙门菌株与GenBank中鼠伤寒沙门菌LT2相应的基因序列进行比较。结果在肠炎沙门菌标准菌株50041和18株食品分离株之间,6个基因PCR产物范围内的近60000个核苷酸序列完全相同,未观察到同一血清型内的基因突变。而与LT2相比,基因产物发生了1到6个点突变。在对SdfΙ的核苷酸序列比较研究中发现,肠炎沙门菌标准菌株与食品中分离株SdfΙ的核苷酸序列与GenBank中该序列的碱基对比,发生了C182T的点突变。即MLST技术揭示了在同一血清型内各菌株管家基因碱基序列的高度保守。结论MLST方法适用于不同血清型沙门菌株间的分型研究,而不适于同一血清型的菌株分型。  相似文献

We explored the dual influence of the patient's age and the infecting serotype on the blood invasiveness patterns of non-Typhi Salmonella enterica (NTS). Blood invasiveness ratio (BIR) was calculated as the ratio between the number of blood and blood + stool isolates. Analysis of 14,951 NTS isolates showed that the BIR increased drastically above the age of 60 years, reaching levels 3.5-7 times higher compared to age group < 2 years. Different patterns of age-related invasiveness were observed for the five most common NTS serotypes (Enteritidis, Typhimurium, Virchow, Hadar, Infantis). Among children < 2 years, the BIR was highest for serotype Virchow and lowest for serotype Hadar, while in persons > or = 60 years it was highest for serotypes Enteritidis and lowest for serotype Infantis. The tendency of NTS serotypes to invade the bloodstream was significantly influenced by the patient's age, however the impact of age differed for various NTS serotypes.  相似文献

Salmonella surveillance: a global survey of public health serotyping   总被引:2,自引:0,他引:2  
To better understand the global epidemiology of salmonellosis and the national surveillance programmes used for salmonella infections in humans, we conducted a global survey of the 191 WHO Member States. We gathered information on the total number of salmonella isolates serotyped, and the 15 most commonly isolated serotypes from humans in 1990 and 1995. Of the 104 countries that responded, 76 (73.1%) conducted public health surveillance for salmonella and 69 of these (90.8%) conducted serotyping as part of the surveillance. Fifty-nine countries (56.7% of those responding) provided information about the most commonly isolated serotypes in 1995. Three serotypes, Enteritidis, Typhimurium and Typhi accounted for 76.1% of all isolates reported in 1995. One of these three was the most common serotype identified in 93.2% of countries reporting data for that year. In 1995, Enteritidis was the most frequently isolated serotype in 35 countries, followed by Typhi (12 countries) and Typhimurium (8 countries). The global pandemic of Salmonella Enteritidis continued to expand. The mean national proportion of all salmonella isolates that were Enteritidis increased globally from 25.6% in 1990 to 36.3% in 1995. Serotyping is a frequently used component of a public health response to the global challenge of salmonellosis. Support for serotyping as part of national salmonella surveillance, and for rapid international communication of the results via a new WHO electronic website will help target future prevention strategies.  相似文献

The present study was conducted to investigate serotype distribution, antimicrobial resistance patterns, carriage of class 1 integron, and clonality of Salmonella strains isolated from patients aged 0-12 years in Tehran, Iran, during 2007-2008. A total of 139 Salmonella isolates were studied. Salmonella serotypes Enteritidis, Infantis, and Typhimurium included 84.9% of isolates, Enteritidis accounting for 41.7%. The most prevalent resistances were to doxycycline (64.7%), nalidixic acid (61.2%), tetracycline (51.8%), and streptomycin (42.8%). Fifty-three (38.1%) isolates contained class 1 integron. Eight different gene cassettes were identified, aadA1 being the most frequently encountered. Pulsed-field gel electrophoresis showed that integron-positive Salmonella strains belonging to serotypes Infantis, Enteritidis, and Typhimurium were attributed to two, three, and five different pulsotypes, respectively. The findings indicated that the distribution and drug resistance pattern of most prevalent Salmonella serotypes were broadly similar to that reported globally from human isolates. Presence of class 1 integrons was common among Salmonella serotypes in Tehran, Iran. Concurrent clonal expansion and horizontal transmission events seem to contribute to increase in drug resistance prevalence among Salmonella serotypes.  相似文献

This study investigates the distribution of pulsed-field gel electrophoresis (PFGE) profiles within Salmonella enterica serotype Enteritidis phage type (PT) 4 and S. Typhimurium definitive phage type (DT) 104, from cases of human infection in nine European countries from 2000 to 2004. Isolates were subtyped using standardized methods and gel images submitted by each participating country to the coordinating centre (Health Protection Agency Centre for Infections, London, UK), where they were entered into a central database, developed within BioNumerics software, and designated using an agreed nomenclature. S. Enteritidis PT4 (n=3637) was differentiated into 38 different profiles. Simpson's index of diversity (D) of profiles ranged from 0.2 to 0.4. Profile SENTXB.0001 represented at least 80% of all profiles in each country. S. Typhimurium DT104 (n=1202) was differentiated into 28 different profile types. Simpson's D was at least 0.6 in all countries except in Austria and Italy. In both these countries over 74% of S. Typhimurium DT104 profiles were STYMXB.0013. Profile STYMXB.0061, was predominant in Denmark, Spain, Finland and England and Wales where it represented between 36% and 45% of profiles. Profile STYMXB.0001 represented nearly half of all profiles in Scotland and 23% in England and Wales. PFGE is proving useful for further discrimination within S. Enteritidis PT4 and S. Typhimurium DT104. Ascertainment of international outbreaks involving common serotypes and phage types may be increased by the timely pooling of PFGE profiles within a central database readily accessible to all participating countries.  相似文献

The plasmid pOG670, a 54 kb, conjugative plasmid that specifies resistance to ampicillin and kanamycin and belonging to the incompatibility group X (IncX), was transferred into 10 isolates of Salmonella enterica serotype Enteritidis belonging to 10 different phage types (PT1, 2, 3, 4, 8, 9, 9b, 10, 11 and 13). Acquisition of the plasmid by these strains did not result in the loss of any resident plasmids but resulted in phage type conversion in 8 of the 10 strains (PT1, 2, 4, 8, 9, 9b, 10 and 11). The observed changes in phage type were found to result from the loss of sensitivity to 3 of the 10 typing phages used (phages 3, 5 and 7). Where the conversion resulted in a change to a defined phage type, both the new and original PTs belonged to the same, previously described, evolutionary lines. Enteritidis PTs 1, 4 and 8, commonly associated with poultry world-wide, were converted to PTs 21, 6 and 13a respectively. The results indicate a different route for phage type conversion Enteritidis from others reported in the literature and, although IncX plasmids are not normally present in PT8 or PT13a, may suggest a possible mechanism/link connecting these phage types.  相似文献

Salmonellae of non-typhoidal serovars are the most important pathogens involved in foodborne diseases in humans all over the world. The incidence rates of two major Salmonella serovars, i.e. S. enterica serovar Enteritidis (S. Enteritidis) and S. enterica serovar Typhimurium (S. Typhimurium), in the Slovak Republic in 2000-2003 are given. Over the period studied, 829 S. Enteritidis strains and 258 S. Typhimurium strains isolated from patients with salmonellosis were investigated in the National Reference Centre for Salmonella Phage Typing. The S. Enteritidis strains were differentiated into 16 phage types, with phage type 8 being dominant since found in 73.6%, 53.8%, 62.8% and 45.6% of strains in 2000, 2001, 2002 and 2003, respectively. The following most frequent phage types were 4 and 13a. New phage types, i.e. 15, 5, 25 and 14b, were identified from salmonellosis outbreaks in 2003. The S. Typhimurium strains were also differentiated into 16 phage types with phage type DT104 strains being prevalent and showing an increase from 7.4% in 2000 to 44.6% in 2003; 54.2% of them were resistant and of R type ACSSut. The second most frequent phage type in 2000-2001 was 2b in 2003 DT41. The frequency of the other phage types was not epidemiologically significant.  相似文献

A two-step real-time SYBR Green I multiplex polymerase chain reaction (PCR) assay with melting curve analysis was developed for rapid detection of 19 Salmonella serotypes frequently encountered in humans, animals, and animal-associated meat products within the European Union. The first-step single-tube reaction (Multiplex PCR I), consisting of five primer pairs, classified an initial test panel of eight Salmonella serotypes into five groups on the basis of characteristic amplicon melting temperatures produced by each strain. Following designation into groups, two subsequent triplex reactions (Multiplex PCR II-G1 and II-G3) allowed for further identification of five Salmonella serotypes by their melting peak temperatures. Primers for serotype differentiation were designed to target the genes encoding either phase 1 and 2 flagellar antigens fliC and fljB or unique serotype-specific loci. In addition, the assay simultaneously screened for the presence of the ampicilin-amoxicillin, chloramphenicol-florfenicol, streptomycin-spectinomycin, sulfanomides, and tetracycline (ACSSuT)-type multidrug resistance pattern, indicated by the floR gene, and for the Salmonella virulence plasmid encoded by the svp operon in Salmonella serotype Typhimurium. The established multiplex assays were successfully tested on 97 isolates, comprising 37 distinct Salmonella serotypes and 12 non-Salmonella strains. The two-step assay correctly detected 19 of 37 Salmonella serotypes and all non-Salmonella strains produced negative results. Of the 19 serotypes detected in the assays, 7 serotypes, including Salmonella serotypes Ohio, Goldcoast, Livingstone, Kedougou, Enteritidis, Kentucky, ACSSuT-type Salmonella serotype Typhimurium DT104 and DT104b, as well as non-ACSSuT-type Salmonella serotype Typhimurium strains, were definitively identified. The developed multiplex real-time SYBR Green I PCR assay represents a more rapid and reliable method for identification of large numbers of Salmonella serotypes prevalent throughout the European Union than assays using phenotypic serotyping methods.  相似文献

Foodborne infections are an important public health problem in China. In 2008, we conducted surveillance for laboratory-confirmed nontyphoidal Salmonella to monitor trends for this infection in China and to build capacity for rapid detection and response to foodborne outbreaks. Salmonella isolates from patients with diarrhea were sent from hospitals to local public health laboratories for confirmation, serotyping, and antimicrobial susceptibility testing. A total of 126 hospitals in 44 cities and counties from 8 provinces provided isolates and epidemiologic data for analysis. Of 23,140 stool specimens submitted to clinical laboratories, 662 (3%) grew Salmonella enterica. Salmonella were most commonly detected between April and October. The median age of infected patients was 27 years; 34% of infections occurred in patients <5 years old. Of the 662 isolates, we found 73 serotypes, of which serotype Enteritidis (31%) and serotype Typhimurium (26%) were the most common. The prevalence of resistance was high for clinically important antimicrobial agents, including ampicillin (41%) and ciprofloxacin (6%). More than 60% of isolates, including 35% of all Typhimurium, were resistant to three or more antimicrobial agents. In this first multiprovince surveillance report of laboratory-confirmed Salmonella infections in China, we found that Enteritidis and Typhimurium are the most common serotypes and that efforts to reduce antimicrobial resistance among Salmonella in China are needed. Although no outbreaks were detected using this system, efforts to improve this system's capacity to do so are underway.  相似文献

A 4-year retrospective study was undertaken at the Kilifi District Hospital on the coast of Kenya to determine the antibiotic-susceptibility patterns and genotypes of non-typhi Salmonella (NTS) isolates from children. Overall, during the period 1994-97, positive cultures were obtained from 543 (14%) of 3885 blood samples, 364 (30%) of 1210 stool samples and 143 (11%) of 1283 cerebrospinal fluid (CSF) samples. NTS were isolated from 151 (27.8%), 72 (19.8%), and 11 (7.7%) of these positive cultures, respectively. The total 234 NTS isolates were serotyped: the most frequent were Salmonella enterica serotype Enteritidis (41%) and S. enterica serotype Typhimurium (38%). Antibiotic sensitivity testing was done using ampicillin (amp), chloramphenicol (chl), gentamicin (cn), co-trimoxazole (s-t), cefuroxime (cxm), ciprofloxacin (cip), cefotaxime (ctx), amoxicillin-clavulanic acid 20 micrograms-10 micrograms (amc), and tobramycin (tob). Of the 234 isolates, 43 were sensitive to all antibiotics tested and 133 were multiple drug resistant (MDR). The most common resistance type seen was amp, cn, cxm, s-t, ctx, amc, tob (36/234). Our results indicate a high proportion of MDR amongst the isolates from Kilifi. We conclude that 2 major serotypes of salmonella, i.e., S. enterica serotype Typhimurium and S. enterica serotype Enteritidis, of micro-epidemic nature that have been previously unrecognized in Kilifi are responsible for infection in Kilifi district on the coast of Kenya and that over half (56.8%) of total NTS isolates are MDR.  相似文献

Salmonella is one of the most common foodborne pathogens in humans. Laboratory-based surveillance for non-typhoidal Salmonella infection was conducted in Guangdong Province, China to improve understanding about the disease burden and detection of dispersed outbreaks. Salmonella isolated from patients with diarrhea were sent from 16 sentinel hospitals to local public health laboratories for confirmation, serotyping, antimicrobial susceptibility testing, and pulsed-field gel electrophoresis (PFGE). PFGE patterns were analyzed to identify clusters representing potential outbreaks. Between September 2009 and October 2010, 352 (4%) Salmonella isolates were obtained from 9167 stool specimens. Salmonella enterica serotype Typhimurium (45%) and Salmonella enterica serotype Enteritidis (13%) were the most common serotypes, and multidrug resistance was high, especially in Salmonella Typhimurium isolates. PFGE patterns of obtained Salmonella isolates were found to be diverse, but a unique PFGE pattern comprising 53 Salmonella Typhimurium isolates were found to occur almost exclusively in infants. Epidemiologic studies are ongoing to determine whether a common exposure is the source of the Salmonella Typhimurium strain frequently isolated from infants.  相似文献

Salmonellae are a common cause of foodborne disease worldwide. The World Health Organization (WHO) supports international foodborne disease surveillance through WHO Global Salm-Surv and other activities. WHO Global Salm-Surv members annually report the 15 most frequently isolated Salmonella serotypes to a Web-based country databank. We describe the global distribution of reported Salmonella serotypes from human and nonhuman sources from 2000 to 2002. Among human isolates, S. Enteritidis was the most common serotype, accounting for 65% of all isolates. Among nonhuman isolates, although no serotype predominated, Salmonella enterica serovar Typhimurium was reported most frequently. Several serotypes were reported from only 1 region of the world. The WHO Global Salm-Surv country databank is a valuable public health resource; it is a publicly accessible, Web-based tool that can be used by health professionals to explore hypotheses related to the sources and distribution of salmonellae worldwide.  相似文献

目的分析7株侵袭性非伤寒沙门菌(iNTS)的血清型和耐药性。方法针对收集的7株iNTS菌株,开展血清学鉴定、药物敏感性实验以及全基因组测序,并对血清型、MLST型和耐药基因进行鉴定、注释和分析。结果7株iNTS菌株中,包含1株鼠伤寒血清型和2株Ⅰ4,[5],12:i:-(ST34型)、2株肠炎血清型、1株科瓦利斯血清型和1株未知血清型Ⅰ4,[5],12:d:-(ST279型),其中6株为单相菌,沙门菌二相鞭毛基因缺失或假基因化可能有助于增强沙门菌侵袭性。未发现替加环素、氨曲南、阿米卡星及头孢类、碳青霉烯类抗生素耐药株,存在1株八重耐药鼠伤寒沙门菌。耐药基因与耐药表型基本相符。结论本研究中的iNTS菌株存在八重耐药株,整体耐药水平尚低但不可忽视。  相似文献

An examination of salmonella isolates collected by the Scottish Agricultural College Veterinary Services Division from April 1994 to May 1995 was conducted to determine the extent to which Salmonella enterica serotype Typhimurium phage type 104 (DT104) occurred and to investigate the antimicrobial resistance patterns of isolates. Typhimurium DT104 was the predominant salmonella and was isolated from nine species of animal. All isolates of this phage type possessed resistance to at least one antimicrobial and 98% of the isolates were resistant to multiple antimicrobials with R-type ACTSp the predominant resistance pattern. Various other resistance patterns were identified and transferable resistance to the veterinary aminoglycoside antimicrobial apramycin was demonstrated in three strains. A retrospective study for gentamicin resistance in isolates from the Scottish Salmonella Reference Laboratory collection revealed a human isolate of Typhimurium DT104 resistant to gentamicin but sensitive to apramycin and a bovine isolate with apramycin and gentamicin resistance.  相似文献
