目的 探讨银屑病患者角质形成细胞(KC)对T淋巴细胞CD25、CD69表达的影响。方法 分离10例银屑病患者KC,密度梯度离心法分离单一核细胞(PBMC);流式细胞仪检测混合培养后T细胞活化标志CD25、CD69的表达。结果 银屑病患者皮损KC作用的自体外周血T细胞CD25、CD69表达水平分别与非皮损KC作用组及自体T细胞自然增殖组相比显著增高,银屑病非皮损KC+自体T细胞共培养组与自体T细胞自然增殖组相比,差异无统计学意义。银屑病皮损、非皮损KC作用的正常人T细胞CD25、CD69表达均显著高于正常人外周血T细胞自然增殖组,银屑病皮损KC+正常人T细胞共培养组与非皮损组相比,差异无统计学意义。结论 银屑病患者局部存在慢性炎症反应,可能是由于皮损KC免疫表型发生改变从而作为自身抗原,启动自身免疫反应。  相似文献

Abstract: Background: T cells have been shown to be highly relevant in psoriasis. CD26 is a novel T‐cell activation marker involved in various T‐cell functions, e.g. (i) co‐stimulation, (ii) migration and (iii) T‐cell memory response. In particular, CD26bright peripheral blood T cells have been shown to be altered in several autoimmune diseases. Objective: To characterize CD26‐expression of T‐cell subsets in psoriatic patients compared to healthy subjects. Methods: Peripheral blood was obtained from 15 untreated patients with severe psoriasis and from nine healthy subjects. The presence of specific CD26‐related T‐cell subsets was assessed by flow cytometry. Results: The CD26bright expression of CD8+ lymphocytes revealed a statistically significant (P<0.05) decrease in psoriatic patients. The majority of CD4+CD26bright cells are CD45RO+, whereas the minority of CD8+CD26bright cells are CD45RO+ in both groups. Conclusions: The present study demonstrates that the CD8CD26bright T‐cell population is markedly decreased in peripheral blood of psoriatic patients. Moreover, CD26 expression did not show a restriction to memory T cells. As CD26 is of relevance for T‐cell functions, future investigations should focus on elucidating these functions in psoriasis. It is attractive to speculate that the reduction in the CD8CD26bright subpopulation may represent a biomarker for recompartimentalization of activated T cells and a reduced CD8CD26bright count may correlate with increased responsiveness to T‐cell targeted treatments.  相似文献

The aim of this study was to investigate the characteristic cytokine pattern of patients with chronic idiopathic urticaria. Using flow cytometry, we examined the frequency of IL4, IL-10, IL-13 and IFN-gamma producing CD4+ and CD8+ T cells in the peripheral blood mononuclear cells at a single cell level. In patients with chronic idiopathic urticaria, the frequency of IL-10 producing CD4+ and CD8 + T cells was significantly higher than that of control subjects, while the frequency of IFN-y producing helper and cytotoxic T cells was significantly lower. The proportion of IL-4 producing CD4 + T cells from patients with urticaria was significantly lower. The ratio of IL-4 producing CD8 + T cells and the proportion of IL-13 producing CD4 + and CD8 + T lymphocytes did not show any significant difference between patients and controls. In our study, we could observe neither a dominant Th1 nor a dominant Th2 type cytokine pattern. We found a significant elevation in the intracellular IL-10 level which may be the cause of the down-regulated Th1 and Tc1 and partly Th2 lymphocyte functions.  相似文献

BACKGROUND: The usually protracted and indolent course of cutaneous T-cell lymphoma (CTCL) is consistent with an accumulation of lymphocytes rather than being a true proliferative disorder, perhaps as the result of defective lymphocyte apoptosis. Fas (CD95) is the main signalling membrane molecule involved in postactivation T-lymphocyte apoptosis. OBJECTIVES: To evaluate expression of Fas on circulating CD4+ lymphocytes in patients with CTCL. METHODS: Fas expression on peripheral blood CD4+ T cells in 16 patients with mycosis fungoides (patch and infiltrated plaque stages) and in four patients with Sézary syndrome was compared with that in 25 matched patients with lymphocyte-mediated cutaneous benign inflammatory disorders and in 15 subjects without inflammatory cutaneous diseases. RESULTS: Fas expression on peripheral CD4+ lymphocytes was significantly lower in patients with CTCL compared with subjects with benign inflammatory cutaneous disorders and with healthy donors. CONCLUSIONS: This pattern supports the hypothesis that a defect in T-cell apoptosis may play a part in the pathophysiology of CTCL, perhaps through abnormalities of the Fas/Fas ligand system. Alternatively, this decrease could be the result of the presence of the soluble Fas ligand molecule in the sera of patients with CTCL.  相似文献

目的:探讨CD4+CD25+T细胞在特应性皮炎发病中的可能作用.方法:用流式细胞仪对40例AD患儿和45例健康儿童外周血CD4+CD25+、CD4+、CD25琳巴细胞亚群等进行检测,并结合临床资料进行相关分析.结果:AD儿童组CD4+CD25+/CD4+与对照组相比明显升高(P=0.018,P<0.05),婴幼儿组无明显差异(P=0.15,P>0.05),两组CD4+、CD25+细胞相比、轻中重组两两相比、局限型与泛发型相比,其差异均无统计学意义(均大于0.05).结论:CD4+CD25+调节性T细胞可能在特应性皮炎的发病中起一定的作用,但对病情的判断无重要意义.  相似文献

Flow cytometry was used to detect the levels of peripheral CD4+ and CD4+CD25+ T lymphocytes in these samples. Results Compared with the controls, the percentage of CD4+CD25+ regulatory T lymphoeytes in peripheral lymphocytes was significantly lower in patients with progressive vitiligo than those in patients with stable vitiligo and normal human con-trois [(2.43±0.30)% vs (3.49±0.39)% and (3.34±0.24)%, both P <0.05], but no significant difference was found between patients with stable vitiligo and normal human controls (P>0.05). A significantly nega-tive correlation was observed between the percentage of CD4+CD25+ regulatory T lymphocytes and lesion area in patients with progressive vitiligo (r = -0.48, P <0.05), but not in patients with stable vitiligo (P >0.05). There was no significant correlation between the course of disease and the percentage of peripheral CD4+CD25+ regulatory T lymphocytes in patients with progressive vitiligo or stable vitiligo (both P > 0.05). 目的 检测不同病期白癜风患者外周血CD4+CD25+调节性T细胞水平,探讨其与白癜风发病的关系.方法 白癜风患者34例,进展期19例,稳定期15例.通过流式细胞仪对不同病期白癜风患者外周血CD4+、CD4+CD25+T细胞水平进行检测,并与20例正常人比较.结果 进展期患者外周血中CD4+CD25+调节性T细胞占外周血淋巴细胞的表达率低于正常对照组(P<0.05);稳定期患者与正常对照组比,差异无统计学意义(P>0.05);进展期患者低于稳定期患者,差异有统计学意义(P<0.05).进展期患者CD4+CD25+调节性T细胞占外周血淋巴细胞表达率与皮损面积呈负相关(P<0.05),稳定期则无相关性(P>0.05).进展期与稳定期患者CD4+CD25+调节性T细胞占外周血淋巴细胞水平与病程均无明显相关性(P>0.05).结论 白癜风患者外周血中存在异常比例的cD4+CD25+调节性T细胞,可能与白癜风的发病有关.  相似文献

目的:监测梅毒患者外周血CD4~+、CD8~+调节性T细胞的表达水平,考察其在临床的参考价值。方法:将2014年12月至2015年9月,我院收治并确诊的32例梅毒患者作为研究对象。其中,Ⅰ期、Ⅱ期、潜伏期梅毒患者分别有11例、15例和6例。采用细胞免疫芯片检测(淋巴细胞CD4~+、CD8~+绝对数检测)技术检测并计算患者外周血中的调节性T细胞CD4~+、CD8~+表达水平及CD4~+、CD8~+的比值。并与同期就诊我科的30例非梅毒患者的相应指标数据进行比较。结果:与非梅毒患者比较,梅毒患者外周血中CD4~+、CD8~+细胞绝对值计数均显著下降(P0.05);CD4~+、CD8~+比值显著低于非梅毒患者组(P0.05)。在Ⅰ期、Ⅱ期、潜伏期梅毒患者间CD4~+、CD8~+绝对值计数均值水平的差异有统计学意义(P0.05)。结论:在梅毒患者外周血中CD4~+、CD8~+表达水平及CD4~+、CD8~+比值均显著降低,而在梅毒感染的不同疾病发展时期CD4~+、CD8~+表达水平也有着显著的差异。  相似文献

目的 检测尖锐湿疣患者外周血CD8+T细胞白介素-2、白介素-12、干扰素-γ、白介素-4的表达水平,探讨其在尖锐湿疣发病中的作用.方法 应用流式细胞仪检测60例尖锐湿疣患者和20例正常人对照者外周血CD8+T细胞白介素-2、白介素-12、干扰素-γ、白介素-4的表达水平.结果 尖锐湿疣患者外周血CD8+T细胞白介素-2、白介素-12、干扰素-γ阳性细胞百分比均显著下降(P<0.01);白介素-4+CD8+T细胞百分比与正常人对照组比较,差异无统计学意义(P>0.05);干扰素-γ+/白介素-4+T细胞比值显著低于正常人对照组(P<0.01).结论 尖锐湿疣患者Tc1细胞水平低下,Tc1/Tc2显著低于正常人对照.  相似文献
