Objective:To explore the effects of IL-10 on acute leukemic immune evasion. Methods: Plasma concentrations of IL-10 were measured by ELISA in 56 first-visit acute leukemic patients. And expressions of IL-10 on leukemic cells in 30 patients were measured by indirect immunofluorescence technique.Results: Compared with those in control group,IL-10 concentrations increased significantly in first-visit acute leukemic patients. And there was a slight but not significant decrease of IL-10 in patients with acute lymphocytic leukemia(ALL) compared with those with acute non lymphocytic leukemic(ANLL). After intensive chemotherapy,there was a significant decrease of IL-10 in completely remitted (CR) patients ,especially in those with ANLL,but there was still a significant increase compared with those in control group.The positive rate of cells giving out yellow-green bright fluorescence on membranes was 10%-80% ;there were 18 patients expressing IL-10(18/30,60%) positively:among them 11 with ANLL(11/19,58%) and 7 with ALL(7/11,64 %) respectively while that of peripheral mononucleate cells in control group was 13%.Compared with that in control group,there was a significant increase of positive rate in ANLL and ALL but with no significant difference between ANLL and ALL. Conclusion: Probably as one of important mechanisms of acute leukemic immune evasion,IL-10 secreted by leukemic cells,contributing to the immunosuppressive state at the tumor site ,increase significantly in acute leukemic patients.  相似文献

目的 探讨急性白血病患者反复输注血小板的治疗效果。方法 对71例急性白血病反复输血及佃血小板的患者,采用淋巴细胞毒试验检测,研究这组患者的血小板校正增加值与之间的关系。结果 血小板输注有效的急性淋巴细胞白血病及急性非淋巴细胞白血病检测LCT阳性率分别为7.6%及4.2%;血小板输注无效的ALL及ANLL的LCT阳性率分别为71.4%及48.8%。  相似文献

The staining results of terminal deoxynucleotidyl transferase (TdT) in leukemic cells of peripheral blood or bone marrow in 55 patients showed that 22 (40%) patients were found positive for TdT staining: 17 patients with acute lymphoblastic leukemia (ALL), 2 acute non-lymphocytic leukemia (ANLL) and 3 hybrid acute leukemia (HAL). The positive rate of ALL (17/21) was significantly higher than that of ANLL (2/21, P < 0.005). The leukemic cells of HLA-DR+CD19+TdT- were demonstrated to express some of myeloid restricted antigens (HI98, HIM 4, HIM 5) (3/3), which suggested that these cases were leukemias of myeloid origin. None of the 8 patients with M 0/M 1 ANLL was positively stained for the enzyme. Our results showed that TdT detection is of great value in the diagnosis and differentiation of both poorly differentiated and hybrid acute leukemia.  相似文献

急性白血病细胞凋亡抑制基因Livin表达及临床意义   总被引:2,自引:0,他引:2  
目的 观察 Livin 基因在急性白血病(AL)病人白血病细胞中的表达及其临床意义.方法 采用半定量逆转录聚合酶链反应(RT-PCR)方法检测41例不同时期AL病人白血病细胞Livin mRNA的表达水平,以10例健康人作为正常对照.结果 AL病人白血病细胞Livin mRNA 阳性表达率为53.7%.其中急性淋巴细胞白血病(ALL)和急性非淋巴细胞白血病(ANLL)病人Livin mRNA 阳性表达率分别为53.8%、53.6%,二者比较差异无显著性,但均高于正常对照组(10.0%)(P=0.035、0.017).初治和复发AL病人白血病细胞Livin mRNA 阳性表达率分别为52.4%、85.7%,明显高于对照组(P=0.025、0.004).缓解组病人Livin mRNA阳性表达率为15.4%,明显低于初治组和复发组(P=0.004、0.030),而与对照组相比差异无统计学意义.白细胞计数≥50×109/L的初治白血病病人Livin mRNA的阳性表达率(77.8%,7/9)明显高于白细胞计数<50×109/L的初治病人(25.0%,3/12)(P=0.022),其表达水平也明显高于后者(T=6.50,P<0.05).结论 Livin mRNA过度表达参与了AL的发病,可作为AL的诊断、复发及预后判断的指标之一.Livin mRNA表达在AL细胞的增殖分化和(或) 抗凋亡过程中发挥了重要作用,可能是导致AL细胞对化疗药物不敏感的原因之一,可作为治疗的一个潜在靶点.  相似文献

目的:探讨测定血清白细胞介素-8(IL-8)在恶性血液病诊断及其疗效评价中的意义.方法:57例恶性血液病患者分为急淋(ALL)、急非淋(ANLL)、慢粒(CML)、霍奇金病(HD)和非霍奇金淋巴瘤(NHL)组,分别测定血清IL-8并与正常对照组比较;其中CML组尚进行了治疗前后的比较.结果:各组的血清IL-8水平均高于正常对照组(P<0.01);NHL组的血清IL-8水平高于HD组(P<0.01)和其余各疾病组(P<0.05),且经治疗后较治疗前降低(P<0.05);HD组的血清IL-8水平低于ALL、ANLL和CML组(P<0.05).结论:测定血清IL-8水平可作为恶性血液病的诊断指标,亦可作为区分HD和NHL的参考指标,在CML的病程中观察血清IL-8的动态变化可能有助于对病情变化以及对治疗反应的判断.  相似文献

目的:探讨原代白血病细胞survivin基因表达与化疗药物诱导凋亡的敏感性关系。方法:RT—PCR方法检测初发ANLL细胞suntivin mRNA的表达。DNA片段原位末端标记(TUNEL)方法检测不同化疗药物体外诱导的白血病细胞凋亡。结果:原代ANLL细胞与HHT、AcLa、Ara—C孵育15h后,survivin mRNA阳性表达的ANLL细胞凋亡率低于阴性表达者。联用HHT Ara—C或AcLa Ara—C体外处理原代ANLL细胞15h后.survivin mRNA阳性表达的ANLL细胞凋亡率亦低于阴性表达者。且survivin mRNA阴性的ANLL患者骨髓缓解(BMR)率高于survivin mRNA阳性者((2/2)对(3/11))。结论:sunrvivin基因阳性表达的ANLL细胞对化疗药物诱导凋亡不敏感。  相似文献

用 R T P C R 检测了28 例急性白血病患者 W T1 表达水平。结果显示:19 例 W T1 表达阳性,阳性率679 % ;其中16 例 A N L L 中有11 例表达阳性,12 例 A L L 有8 例表达阳性。表明 W T1 在急性白血病中有高度表达,可作为一种新的检测标记物,在白血病疗效观察、判定预后及 M R D 研究中有重要意义。  相似文献

The purpose of the study was to investigate the apoptosis of leukemic cells induced by Boswellia Carterii Birdw(BCB). The target leukemia cell line HL60 and bone marrow leukemic cells from 30 acute non-lymphocytic leukemic(ANLL) patients (3 M1 11 M2a 10 M3 1 M4a 5 M5b) were studied. Apoptosis was detected by morphological observation, DNA electrophoresis, percentage of DNA fragmentation test and flow cytometric cell cycle analysis. It is concluded that BCB can induce apoptosis in ANLL cells and HL60 cells.  相似文献

用RT-PCR检测了28例急性白血病患者WT1表达水显示:19例WT1表达一,阳性率67.9%;其中16例ANLL中有11例表达阳性,12例ALL有8例表达阳性。表明WT1在急性白血病中有高度表达,可作为一种新的检测标记物,在白血病疗效观察、判定预后及MRD研究中有重要意义。  相似文献

目的:研究人端粒重复序列结合因子(TRF1)在30例急性白血病细胞中的表达,了解TRF1表达与急性白血病分型的关系,以及与端粒酶活性的关系.方法:用荧光定量RT-PCR定量检测30例急性白血病细胞中TRF1 mRNA表达情况,同时检测hTERT mRNA的表达,并分析两者之间的相关性.结果:TRF1在急性非淋巴细胞性白血病(ANLL)细胞中的表达0.0126(0.0127~0.0546)明显低于正常人骨髓单个核细胞中的表达0.0457(0.00839~0.262)(P<0.001),但在急性淋巴细胞性白血病(ALL)细胞中的表达0.0745(1.92×10-6~0.193)与正常人骨髓单个核细胞比较差异无显著性,且在ANLL细胞中的表达明显低于ALL细胞中的表达(P=0.001).TRF1和hTERT在急性白血病细胞和正常人骨髓单个核细胞中的表达无显著相关性(r=-0.173,P=0.207).结论:TRF1 mRNA在ANLL细胞中表达明显降低,而在ALL细胞中表达无显著改变,提示ANLL细胞中TRF1可能通过调控端粒长度对端粒酶起间接负调控作用.  相似文献

研究急性非淋巴细胞白血病细胞中原癌基因c-myc的表达及临床意义。方法采用蛋白质免疫印渍和单克隆抗体免疫细胞化学染色方法检测30例ANLL细胞中原癌基因c-myc的表达。结果19例表达阳性,阳性率为63%。ANLL中白血病细胞的阳性细胞百分率为29%-96%,c-myc在不同病人的白血病细胞中表达水平不一样;c-myc的表达和患者骨髓中白血病细胞百分率显著相关,和患者年龄,性别,外周血白细胞计数。  相似文献

用扫描电镜(SEM)研究了26例急性白血病细胞表面形态特征,同时结合光镜形态学(FAB)、免疫学和细胞遗传学分型(MIC),结果发现,SEM可更准确地区分急性淋巴细胞白血病(ALL)和急性非淋巴细胞白血病(ANLL)。在ANLL中,白血病粒细胞以表面横嵴为特征,白血病单核细胞以基底宽的膜性皱起为特征,SEM观察结果与MIC分型符合率为90.5%。在ALL病例中,SEM区分其免疫亚型有一定局限性。ANLL的疗效与SEM下白血病细胞的分化程度可能相关。  相似文献

目的检测c-kit R在急性白血病(acute leukemia,AL)的表达并进一步分析ANLL中c-kit R表达与染色体异常核型的关系及其意义.方法采用流式细胞术(floW cytometry,FCM)分别检测并比较92例急性淋巴细胞性白血病(ALL)和81例ANLL初诊患者骨髓单个核细胞(BMMNC)跨膜c-kit R表达的阳性率及阳性水平,并设立20例正常人骨髓标本作为阴性对照组;采用R带显带方法对c-kit R阳性表达的ANLL进行染色体核型分析.结果ALL患者的c-kit R表达的阳性率及阳性水平与正常人相比差异无显著性(P>O.05).ANLL患者的c-kit R表达率及阳性水平则均明显高于正常人和ALL患者(P值分别<0.01).ANLL中c-kit R阳性表达者异常核型的百分率为74.1%,明显高于c-kit R阴性组者(26.1%)(P<O.05).异常核型的类型有5q-、-17、 21、del 3p、t(8;21)、6p 、t(1;8)、复杂易位、t(15;17)、t(1;12)、-7、四倍体、20q-等十余种类型.结论c-kit R可作为髓系免疫学标记,用以协助ANLL的诊断.c-kit R阳性者伴有细胞遗传学异常,但是,c-kit R阳性与染色体异常核型的类型之间不存在特殊对应关系.c-kit原癌基因不是ANLL发病机制的唯一要素.  相似文献

目的 体外诱导培养正常儿童与B系急性淋巴细胞白血病(B-ALL)儿童的树突状细胞(DC),比较二者的生物学特性.方法 分离10例B-ALL初诊患儿(ALL组)和10例正常儿童(对照组)的外周血单个核细胞,以rhGM-CSF(20 ng/mL)、rhIL-4(10 ng/mL)及TNF-α(10 ng/mL)联合培养8 d,显微镜下观察细胞形态,流式细胞仪检测细胞免疫表型(CD11c、CD80、CD83、CD86),ELISA检测培养上清中IL-12的浓度,混合淋巴细胞反应(MLR)检测抗原递呈功能,电化学法检测上清中葡萄糖浓度.结果 ALL组细胞未表现出DC的典型形态,CD11c、CD80、CD83、CD86表达均较对照组细胞低(P< 0.05),分泌IL-12的能力弱于对照组细胞(P<0.05),刺激T淋巴细胞增殖的能力弱于对照组细胞,ALL组细胞培养上清中的葡萄糖浓度高于对照组(P<0.05).结论 B-ALL来源的DC成熟度异常,功能减弱,葡萄糖代谢异常可能是其成熟异常的原因之一.  相似文献

Summary The expression of three heat shock proteins (HSPs)-HSP90α, HSP70, HSP27 in cells obtained from 22 patients with leukemia, K562 erythroleukemia cell line, and normal blood cells was observed by means of RNA dot blot analysis. The results showed that the expression of the HSP27 gene was enhanced in 4 cases of acute lymphoid leukemia (ALL), 7 cases of acute nonlymphoid leukemia (ANLL) and 2 cases of myelodysplastic syndrome. (MDS) as compared with that of the normal blood Cells, yet there was no significant difference in the HSP27 expression between the ALL and ANLL cells. The expression of HSP70 in all the 5 ALL and ANLL patients was much lower than that of the normal subjects, except 1 case of ALL and 1 case of MDS, in which the expression was obviously enhanced. All the cases including 11 ANLL, 5 ALL and 1 MDS had higher HSP90α expression than the normal subjects. The enhanced expression pf HSP90α in leukemia cells may be associated with the active and indefinite proliferation of leukemia cells. Our results also suggest that the high expression of the HSP27 gene may not be confined to a specific type of acute leukemia.  相似文献

OBJECTIVE: To obtain insight into the molecular mechanism of acute leukemia in elderly patients in relation to the clinical features of the disease. METHODS: DNA specimens were obtained from 34 elderly patients with acute nonlymphoblastic leukemia (ANLL), 9 elderly patients with acute lymphoblastic leukemia (ALL) and 10 healthy blood donors. Polymerase chain reaction (PCR) coupled with single-strand conformation polymorphism (SSCP) was performed to detect T cell receptor (TCR) V gamma I-J gamma gene rearrangement. RESULTS: In the 34 elderly ANLL patients, 47.1% (16/34) were found with TCRV gamma I-J gamma gene rearrangement, showing a higher rate of the rearrangement than in younger ANLL patients (P<0.025). TCRV gamma I-J gamma gene rearrangement was identified in 7 (77.8%) of the 9 elderly patients with ALL, and 3 of the identified cases had oligoclonal/ subclonal rearrangement, suggesting a higher rate of oligoclonal/subclonal rearrangement in elderly ALL patients than in younger ALL patients (P<0.01). None of the blood donors was found with TCRV gamma I-J gamma gene rearrangement. The complete remission rate was lower in the elderly ANLL patient with TCRV gamma I-J gamma gene rearrangement than in those without (P<0.05). CONCLUSION: The detection of TCRV gamma I-J gamma gene rearrangement can be of value in assessing the therapeutic effect and making prognostic prediction in elderly patients with acute leukemia.  相似文献

目的探讨细胞间粘附分子-3(inter cellular adhesion molecule-3,ICAM-3)在急性白血病(acute leukemia,AL)中的表达及其在临床疗效、髓外浸润中的意义。方法采用骨髓涂片免疫组织化学法检测了33例急性非淋巴细胞白血病(acute nonlymphoblstic leukemia,ANLL)、21例急性淋巴细胞白血病(acute lymphoblastic leukemia,ALL)患者治疗前后及10例正常对照骨髓单个核细胞ICAM 3的表达率。结果化疗前AL组ICAM-3表达率均显著高于正常对照组,ANLL组与ALL组比较ICAM-3表达率差异无统计学意义(P>0.05),且T-ALL组与B-ALL组ICAM-3表达率差异无统计学意义(P>0.05);ICAM 3的表达与患者发病年龄、性别、就诊时血红蛋白(Hb)、血小板(BPC)等因素无关,与骨髓原始细胞比例呈正相关(r=0.467,P<0.01);外周血白细胞(WBC)≥100×109患者ICAM 3的表达率比WBC<100×109组高(P<0.05);所有AL患者经过一个疗程化疗后ANLL、ALL未缓解组的ICAM-3表达率明显高于缓解组(P<0.01),但缓解组与对照组之间ICAM-3表达率差异无统计学意义(P>0.05);组织浸润组骨髓AL细胞ICAM-3表达率高于无组织浸润组(P<0.01)。结论ICAM-3在AL中的表达明显高于正常对照组,其表达水平的高低可作为AL观察临床疗效及判断预后的指标之一。同时,AL的组织浸润机制可能与细胞间粘附分子表达升高有关。  相似文献

目的 :探讨急性非淋巴细胞白血病 (ANLL)细胞survivinmRNA表达及与疗效关系。方法 :应用RT -PCR方法检测 2 8例初发ANLL患者和 6例正常对照骨髓单个核细胞survivinmRNA的表达。结果 :ANLL患者survivinmRNA表达阳性率 75 .0 % (2 1/ 2 8)明显高于正常对照的 2 / 6 (χ2 =3.919,P <0 .0 5 ) ;survivinmRNA表达阴性的ANLL患者接受 1个疗程化疗后骨髓缓解 (BMR)率高于阳性者 (83.3%vs 2 5 .0 % ,P =0 .0 2 3)。结论 :survivin基因在ANLL细胞高表达 ,可能是ANLL患者对化疗药物不敏感的原因之一  相似文献

目的 探讨增殖细胞核抗原 (PCNA)及原癌基因Bcl- 2在儿童急性白血病 (AL)骨髓细胞的表达及临床意义。方法 应用S -P免疫组化法检测 41例儿童AL骨髓标本PC NA和Bcl- 2。结果 急性淋巴细胞白血病 (ALL)组及急性非淋巴细胞白血病 (ANLL)组PC NA表达率与对照组比较均有显著差异 (P <0 .0 1,P<0 .0 5 ) ,ALL标危组显著高于高危组 (P <0 .0 5 ) ;ALL及ANLL组的Bcl- 2表达率高于对照组 (P <0 .0 1) ,标危组显著低于高危组 (P <0 .0 5 )。PCNA高表达者缓解率显著高于低表达者 (P <0 .0 1) ;Bcl- 2阳性表达者 3年无病生存率显著低于阴性表达者 (P <0 .0 1)。结论 PCNA、Bcl- 2基因在儿童AL中存在表达异常 ,二者均参与发病过程 ,但其作用机制可能是独立的 ,联合检测有助于判断预后。  相似文献
