OBJECTIVE

To investigate the effect of brain functional recovery decoction (BFRD) on expression of vascular endothelial growth factor (VEGF) and angiopoietin-1 (Ang-1) protein in rats with cerebral ischemia reperfusion injury, and to explore the mechanism of action of BFRD.

METHODS

Using the suture-occlusion method, a Wistar rat model of focal cerebral ischemia reperfusion was established. The rats were randomly divided into treatment group, model group, and sham operation group. The treatment group was administered BFRD. In situ hybridization was used to detect VEGF mRNA expression. Immunohistochemistry was used to observe expression of Ang-1 protein.

RESULTS

VEGF mRNA expression was greater in the model group compared with the sham operation group (P < 0.05); Ang-1 protein expression was more obvious in the treatment group than the model group (P< 0.05).

CONCLUSION

BFRD promoted VEGF mRNA and Ang-1 protein expression in the brains of rats with cerebral ischemia, suggesting increased angiogenesis.  相似文献

Objective

The current study aims to investigate the effect of Hemagglutinating virus of Japan envelope (HVJ-E) on induction of apoptosis and autophagy in human prostate cancer PC3 cells, and the underlying mechanisms.

Methods

PC3 cells were treated with HVJ-E at various multiplicity of infection (MOI), and the generated reactive oxygen species (ROS), cell viability, apoptosis, and autophagy were detected, respectively. Next, the role of ROS played in the regulation of HVJ-E-induced apoptosis and autuphagy in PC3 cells were analysed. In the end, the relationship between HVJ-E-induced apoptosis and autuophagy was investigated by using rapamycin and chloroquine.

Results

Flow cytometry assay revealed that HVJ-E treatment induced dose-dependent apoptosis and that the JNK and p38 MAPK signaling pathways were involved in HVJ-E-induced apoptosis in PC3 cells. In addition, HVJ-E was able to induce autophagy in PC3 cells via the class III PI3K/beclin-1 pathway. The data also implyed that HVJ-E-triggered autophagy and apoptosis were ROS dependent. When ROS was blocked with N-acetylcysteine (NAC), HVJ-E-induced LC3-II conversion and apoptosis were reversed. Interestingly, HVJ-E-induced apoptosis was significantly increased by an inducer of autophagy, rapamycin pretreatment, both in vitro and in vivo.

Conclusion

HVJ-E exerts anticancer effects via autophagic cell death in prostate cancer cells.  相似文献

OBJECTIVE

To observe the effects of Fuxin decoction in rats with heart failure induced by doxorubicin injections.

METHODS

We employed a doxorubicin-induced heart failure model to investigate the effect of Fuxin decoction. Rats were orally administered with Fuxin decoction from 10 to 40 mg/kg. Organization structure changes were assessed by histology, circulating levels of oxidative stress-related biomarkers and inflammatory cytokines were detected via enzyme-linked immunosorbent assay.

RESULTS

Administration of Fuxin decoction improved the growth retardation in doxorubicin hydrochloride injection rats. Cardiac involvement was alleviated by Fuxin decoction, as well as changes of congestion systemic circulation in this rat models. Furthermore, Fuxin decoction regulates immune responses and suppresses oxidation reaction in doxorubicin hydrochloride-induced rats.

CONCLUSION

Fuxin decoction might be an interesting candidate for congestive heart failure.  相似文献

Objective

To examine whether a combinative treatment with curcumin enhances the effects of the epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) gefitinib on cell proliferation, clonogenic capacity and apoptosis in the drug-resistant lung cancer cell line NCI-H1975, and further investigate the molecular mechanisms involved.

Methods

NCI-H1975 cells were treated with curcumin and gefitinib alone or in combination, and cell proliferation, clonogenic capacity and apoptosis were examined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, clone forming experiments, and flow cytometry, respectively, while p38, extracellular regulated protein kinase (ERK)1/2, and protein kinase B (AKT) phosphorylation were examined using Western blotting.

Results

Compared with the effects of either agent alone, the combination of curcumin and gefitinib had a stronger suppressive effect on proliferation and the clonogenic capacity (P < 0.05), and showed an increased ability to promote apoptosis (P < 0.05) and reduce p38, ERK1/2, and AKT phosphorylation (P < 0.05).

Conclusion

Co-treatment of curcumin and gefitinib significantly improves the ability of gefitinib to inhibit cell proliferation, suppress the clonogenic capacity and enhance apoptosis in NCI-H1975 cells, and these effects are possibly mediated via a decrease in phosphorylation of proteins in downstream pathways of the epidermal growth factor receptor.  相似文献

Objective

To evaluate the effects of baicalin in human gastric cancer cells, including apoptosis-inducing effects, and to investigate its underlying mechanisms of action.

Methods

Cell proliferation and apoptosis assays were performed to investigate the anti-proliferation effects of baicalin in human gastric cancer BGC-823 and MGC-803 cells. Real time-quantitative polymerase chain reaction and Western blotting analysis were performed to elucidate the molecular mechanisms underlying the anti-tumor properties of baicalin.

Results

In BGC-823 and MGC-803 gastric cancer cells treated with 80, 120, and 160 µmol/L baicalin for 48 h, a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay showed that baicalin significantly inhibited cell proliferation in a dose-dependent manner, while flow cytometric analysis demonstrated that baicalin could induce apoptosis, also in a dose-dependent manner. Moreover, baicalin up-regulated the expression of caspase-3, caspase-9, and B cell lymphoma (Bcl-2)-associated X protein and down-regulated the expression of Bcl-2 at both the mRNA and protein level.

Conclusion

Baicalin has potential as a therapeutic agent for gastric cancer by inducing apoptosis in cancer cells.  相似文献

Objective

To investigate the effect of Sharbat-e-Deenar (SD) on acetaminophen (APAP)-induced hepatotoxicity in rat model.

Methods

Albino rats were treated with SD at the doses of 1, 2 and 4 mL/kg, p.o. against hepatotoxicity after APAP (2 g/kg, p.o. once only) intoxication. The blood, tissue biochemical parameters and histopathological observation were performed. The

Results

APAP exposure in rats significantly increased the level of biochemical parameters such as aspartate aminotransaminase, alanine aminotransaminase, lactate dehydrogenase, serum alkaline phosphatase, bilirubin, urea and creatinine into blood circulation which were reversed towards normal by SD therapy at all doses. The tissue biochemical parameters such as lipid peroxidation, reduced glutathione, adenosine tri-phosphatase and glucose-6-phosphatase were significantly restored after SD treatment against hepatotoxity. Histological analysis confirmed that SD-treated rats significantly alleviated of liver damage and reduced lesions caused by APAP intoxication. The biochemical changes are in good correlation with the histopathological observations.

Conclusion

On the basis of these results, it can be concluded that SD exerts hepatoprotective activity against APAP-induced liver injury.  相似文献

Objective

To investigate the effects of Tangnaikang (TNK), a mixture of five herbal plant extracts, on inflammation-mediated insulin resistance and β-cell apoptosis in SHR.Cg-Leprcp/NDmcr (SHR-cp) rats.

Methods

Seven-week-old SHR-cp rats were randomly divided into a control (CON) group and a TNK (3.24 g/kg) group. Wistar-Kyoto rats at the same age were used as the normal control group. After 7 weeks of continuous intragastric administration of TNK, the glucose metabolic status and insulin sensitivity of the rats were evaluated by assessing fasting serum glucose (FBG), the oral glucose tolerance test (OGTT), fasting serum insulin (FINS), and the insulin sensitivity index (ISI). Serum tumor necrosis factor-Δ (TNF-Δ), interleukin-6 (IL-6), C-reactive protein (CRP) and adiponectin were measured via enzyme-linked immunosorbent assays. Macrophage infiltration and apoptosis in adipose tissues were detected through F4/80 immunohistochemistry and the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. Islet morphology and β-cell apoptosis were investigated using double immunofluorescence staining and the TUNEL assay. The expression of cytokine genes in adipose tissue, the liver, and the pancreas was detected in real-time polymerase chain reaction assays. The expression and phosphorylation levels of insulin signaling-, inflammation-, and β-cell survival-related proteins in the liver and pancreas of SHR-cp rats were detected by western blotting.

Results

TNK (3.24 g/kg) treatment significantly decreased body weight, FBG and FINS; improved impaired glucose tolerance; increased the ISI; reduced serum levels of TNF-Δ, CRP and IL-6; and increased serum adiponectin. The mRNA expression of inflammatory markers was markedly reduced in the liver, pancreas, and adipose tissue. F4/80- and TUNEL-positive cells in adipose tissues were decreased, as was the number of TUNEL-positive β-cells. The phosphorylation of c-Jun N-terminal kinase and that of insulin receptor substrate-1 at serines 307 and 1101 was significantly decreased. In the pancreas, the expression of nuclear factor kappa-light chain-enhancer of activated B cells-p65 was significantly decreased, and phosphoinositide 3-kinase and IRS-2 were significantly increased.

Conclusion

TNK was able to improve insulin resistance and β-cell apoptosis in SHR-cp rats, which might be associated with its ability to relieve the overall and local metabolic inflammatory responses observed in SHR-cp rats.  相似文献

OBJECTIVE

To evaluate the anti-platelet aggregation effects of extracts from 31 Traditional Chinese Medicines (TCM) with the property of activating blood and resolving stasis in terms of TCM theory.

METHODS

The 31 TCMs extracts were prepared using water, 90% ethanol and ethyl acetate., and the effects on anti-platelet aggregation were tested on a platelet aggregation analyzer in vitro with adenosine 5'-diphosphate, bovine thrombin and arachidonic acid (AA) as aggregation inducers, respectively. Aspirin was the positive control.

RESULTS

Lots of the tested TCMs had inhibitory effects with concentration-dependent manner on platelet aggregations induced by various agonists. Especially, some of the TCMs such as Chuanxiong (Rhizoma Chuanxiong), Yanhusuo (Rhizoma Corydalis Yanhusuo) and Danshen (Radix Salviae Miltiorrhizae) showed good anti-platelet aggregation effect similar or higher than that in positive control group.

CONCLUSION

The study provided scientific references that several TCMs such as Chuanxiong (Rhizoma Chuanxiong), Yanhusuo (Rhizoma Corydalis Yanhusuo) and Danshen (Radix Salviae Miltiorrhizae), possess the property of anti-platelet aggregation.  相似文献

OBJECTIVE

To investigate the effect of acupuncture at acupoint of Shenshu (BL 23) in ovariectomized rats and sham-operated rats.

METHODS

Female Sprague-Dawley rats were divided into the following four groups (n = 12 each): sham operation only (Sham), sham operation with acupuncture (Sham + ACP), ovariectomy only (OVX), and ovariectomy with acupuncture (OVX + ACP). Operations were performed at the 9th week of age. Acupuncture of Shenshu (BL 23) was started at the 11th week of age and conducted 3 times per week until the 26th week of age.

RESULTS

Among ovariectomized rats, the acupuncture-treated rats had significantly lower body weights and cross-linked N-telopeptide of type 1 collagen levels in the urine after 3 weeks of acupuncture treatment, higher bone mineral density of the lumbar vertebra after 7 weeks, and smaller trabecular separation of the sixth lumbar vertebra, measured by micro-computed tomography, after 17 weeks than control rats. These effects were not observed in sham-operated rats. Blood concentrations of estradiol were significantly higher in the OVX + ACP group than in the untreated group. Acupuncture treatment increased plasma testosterone levels in sham-operated rats, while this increase was not observed in OVX rats.

CONCLUSION

Acupuncture of Shenshu (BL 23) prevents bone loss and structural changes by suppressing bone resorption.  相似文献

OBJECTIVE

To investigate effect of icariin on apoptosis in hippocampal neurons cultured in high glucose, and the possible mechanism behind the action.

METHODS

Hippocampus was obtained from newborn 24 h Sprague-Dawley rats and then primarily cultured. Then hippocampal neurons were divided into normal control group, high glucose group, icariin group, icariin + protein kinase B (Akt) inhibitor group and Akt agonist group. After each group was cultured in different conditioned medium for 72 h, we detected the apoptosis of neurons with flow cytometry, and the expression of Akt, p-Akt, B-cell lymphoma-2 (Bcl-2) and Bcl-2 associated X protein (Bax) with western blot.

RESULTS

Compared with the normal control group, the apoptosis rate of hippocampal neurons increased significantly (P < 0.01), and p-Akt/Akt and Bcl-2/Bax decreased significantly (P < 0.01) in high glucose group. Compared with the high glucose group, the apoptosis rate of hippocampal neurons decreased significantly (P < 0.01), and p-Akt/Akt and Bcl-2/Bax increased significantly (P < 0.05) in icariin group and Akt agonist group. Compared with the icariin + Akt inhibitor group, the apoptosis rate of hippocampal neurons decreased significantly (P < 0.01), and p-Akt/Akt and Bcl-2/Bax increased significantly (P < 0.01) in icariin group.

CONCLUSION

Icariin could reduce the apoptosis of neurons cultured in high glucose, which may be achieved by increasing the phosphorylation of Akt protein in Akt signal pathway, then increasing the expression of Bcl-2 and inhibiting the expression of Bax.  相似文献

Objective

The aim of this study is identify the intervention mechanism of the effects of electro-acupuncture on the expression of Ang/Tie-2 mRNA and protein in rats with acute cerebral infarction induced by middle cerebral artery occlusion (MCAO).

Methods

Altogether 120 Wistar rats were subjected to MCAO by inserting a nylon filament, and then divided into 3 groups: control group, injured group and electro-acupuncture group. The injured and electro-acupuncture groups were further divided into the following 7 subgroups according to the time after MCAO: 3, 6, 12, 24 h, 3, 7 and 12 day, with 8 rats in each subgroup. The electro-acupuncture group was given electro-acupuncture treatment at Shuigou (GV 26) instantly after operation. The rats were killed at different time points according to their groups, and then the expression levels of Ang/Tie-2 mRNA and protein were detected using Real-Time PCR and immunohistochemical staining.

Results

The mRNA and protein expression levels of Ang/Tie-2 in the electro-acupuncture group were significant higher than that in the injured group.

Conclusion

The results suggested that electro-acupuncture could significantly regulate the expression of Ang/Tie-2 mRNA and protein in the rats with acute cerebral infarction induced by MCAO, and enhance angiogenesis after ischemic penumbra.  相似文献

OBJECTIVE

To investigate a possible mechanism for protective effects of a decoction of the Qinggan Lishui formula (QF) on retinal ganglion cells (RGCs) in a rat model of microbead-induced chronic intraocular hypertension (COH).

METHODS

The COH model was generated by injecting microbeads (superparamagnetic iron oxide) into the anterior chamber of rat eyes. QF was given by intragastric administration (gavage) once daily at a dose of 6.2 g/kg until day 28, following microbead injection. Cholera toxin B subunit (CTB) retrograde labeling and immunohistochemistry were used to evaluate changes in the number of RGCs in the retina. Terminal dUTP nick end labeling (TUNEL) staining was used to assess apoptotic changes in RGCs.

RESULTS

Microbead injection induced a steady increase in intraocular pressure (IOP) of rats. Elevated IOP resulted in a progressive reduction in the number of CTB-labeled RGCs, 2-4 weeks after microbead injection. QF administration may moderately reduce IOP in the rat COH model and attenuate reduction of the number of CTB-labeled RGCs in COH rats. Furthermore, elevated IOP resulted in a progressive increase in the number of TUNEL-positive RGCs, 2–4 weeks after microbead injection, suggestive of an increase in the extent of RGC apoptosis. There was a significant reduction in the number of TUNEL-positive signals in QF-treated COH retinas, compared with untreated COH retinas.

CONCLUSION

QF decoction may provide a protective effect for RGCs in COH retinas by reducing RGC loss; these effects may be mediated by inhibition of RGC apoptosis.  相似文献

OBJECTIVE

To evaluate the application of Traditional Chinese Medicine (TCM) Four-diagnostic Auxiliary Apparatus in disease diagnosis.

METHODS

The liver cancer patients and healthy controls were recruited from Shanghai Integrated Chinese and Western Medicine Hospital and Beijing University of Traditional Chinese Medicine, respectively. Then, the included subjects were diagnosed by the Four-diagnostic auxiliary apparatus.

RESULTS

Thirty liver cancer patients and 30 paired healthy controls were enrolled in this study. Based on the apparatus, the pulse wave velocity was significantly higher in patients compared with controls (P < 0.05). The number of patients with purple tongue and ecchymosis were more than controls (P < 0.05). The number of patients (10%) with yellow tongue coating were higher than the controls (0%). Patients were inclined to be with water type and fire type constitution.

CONCLUSION

TCM Four-diagnostic auxiliary apparatus can be applied in clinical diagnosis of body constitution and health status of subjects. It promotes the accuracy and speed for disease diagnosis and TCM standardization.  相似文献

OBJECTIVE

To assess the medication adherence reporting in clinical trials the field of Traditional Chinese Medicine (TCM) and the impact factors of medication adherence.

METHODS

Reviewed and evaluated were all randomized clinical trials in the field of TCM in treatment of type 2 diabetes mellitus published in Chinese journals in 2012, in terms of their medication adherence, adherence measurement, and impacted factors of adherence.

RESULTS

Finally 124 studies were included. None studies reported the medication adherence. The factors impacting medication adherence couldn't be analyzed due to none reporting adherence.

CONCLUSION

Medication adherence reporting was poor in clinical trials in TCM research. Establishing standards for adherence assessment and reporting may be one of the important steps to improve the quality of clinical studies.  相似文献

Objective

Our aim was to explore whether heat stress protein (HSP) 9 preferentially expresses under heat stress and affects the expression of other heat stress proteins as well as to explore the effect of HSPB9 overexpression and knockdown on apoptosis in DF-1.

Methods

We used gene cloning to construct an overexpression vector of the target gene, and synthesized the target gene interference fragment to transfect the chicken fibroblast cell line. Gene and protein expression, as well as apoptosis, were detected by RT-qPCR, Western blot, and flow cytometry.

Results

Chicken DF-1 cells showed an early state of apoptosis in the early stages of HSPB9 overexpression. In the later stages, as HSPB9 expression increased, the cells showed inhibition of apoptosis. When the cells were under heat stress, HSPB9 expression was much higher and earlier than the expression of HSPB1 and HSPA2. In addition, high expression of HSPB9 had a negative effect on HSPB1 and HSPA2 expression. This negative feedback decreased the percentage of early stages of apoptotic cells and promoted cell survival.

Conclusion

HSPB9 expression, although rapid, is detrimental to cell survival early during its overexpression. In heat stress, HSPB9 overexpression, while inhibiting the expression of HSPA2 and HSPB1, is beneficial to cell survival.  相似文献

Objective

To evaluate the in vivo antiplasmodial activity and the oral acute toxicity of the Bombax buonopozense root bark aqueous extract.

Methods

The in vivo antiplasmodial activity of the root bark aqueous extract of Bombax buonopozense against early and established rodent malaria infections in chloroquine sensitive Plasmodium berghei strain in mice was investigated, and oral acute toxicity of the aqueous root bark extract of Bombax buonopozense was also evaluated in mice.

Results

The findings of this study revealed significant (P < 0.05) and dose dependent decrease in parasitaemia in the parasitized groups treated with varying doses of the extract (50–200 mg/kg p.o.) in both suppressive and curative tests. There was also significant decrease in parasitaemia density in the chloroquine treated group. The aqueous extract was found no toxicity in mice and the oral LD50 was determined to be greater than 5000 mg/kg.

Conclusion

Bombax buonopozense root bark aqueous extract possesses potent antiplasmodial activity and may therefore, serve as potential sources of new antimalarial agents.  相似文献
