The anti-cancer effect of PSP purified products, PSP-A, PSP-B, PSP-C and crude product PSP-Cr was compared on four human tumor cell lines in vitro. It was found that the inhibition rate of cell proliferation of PSP-A was higher than that of PSP-Cr (P<0. 05). On SPC cells, the inhibition rate of PSP-A at a dosage of 1000μg/ml was 62. 7%, being the highest as compared with those on the other three cell lines. Morphological changes were seen in all the four cell lines, especially in SPC cells after PSP-A treatment.  相似文献

Bufalin is one of the major active components of Chan Su, a traditional Chinese medicine. Studies at home and abroad have confirmed that bufalin can inhibit cell proliferation significantly in many human leukemia and solid cancer cell lines by inhibiting endothelial hyperplasia and angiogenesis, and inducing cell differentiation and apoptosis. With the deepening of study,it is showed that bufalin has extensive anticancer activities and it has potential clinical value with very low drug concentration.  相似文献

Background We have constructed anti-HER2 immunoliposomes containing magnetite nanoparticles (HML) that generate heat in an alternating magnetic field (AMF). The effective targeting and cytocidal abilities of HML have been achieved using cell culture models. This study aimed to investigate feasibility of this modality for breast cancer treatment using tumor-bearing mouse models. Material and methods The subcutaneous cancer nodules of BT474 (high HER2 expression) or SKOV3 (low HER2 expression) cells in nude mice were employed as models. HMLs were injected into these cancer nodules and were then exposed to an AMF for 30 min twice at 24 h intervals. Accumulation of magnetite and tumor growth rates were examined. Histological findings of the thermal effect were also examined. Results HMLs accumulated in only BT474 tumors. The tumor temperature increased to 45°C whereas the body temperature remained at around 38°C. Tumor regression was observed in the hyperthermic group and was sustained for 10 weeks after hyperthermia. Conclusion These results suggest that hyperthermia using HML is an effective and specific therapy for breast cancer overexpressing HER2. This therapy may provide an alternative way to treat recurrent cancer refractory to other modalities.  相似文献

目的:探讨RNA干扰(RNAi)对结肠癌SW480细胞人端粒酶逆转录酶(hTERT)基因的抑制效应。方法:化学合成靶向于hTERT基因的4个小分子干扰RNA(siRNA)序列,用阳离子脂质体为载体转染SW480细胞,分为siRNA1~4、siRNA阴性对照和空白对照6组。通过RT-PCR和蛋白质印迹法分别检测结肠癌细胞转染后细胞hTERT mRNA和蛋白水平,并用MTS法检测细胞生长增殖。结果:siRNA1~4组hTERT mRNA相对表达量分别为0.54±0.18、0.56±0.19、0.46±0.10和0.84±0.18,端粒酶相对活性分别为0.57±0.18、0.56±0.19、0.09±0.08和0.70±0.19,siRNA1~4组hTERT蛋白相对表达量分别为0.41±0.18、0.45±0.17、0.08±0.07和0.82±0.16,其中siR-NA1~3组的hTERT mRNA表达量、端粒酶活性和蛋白表达量与空白对照组的差异有统计学意义,P<0.05。SW480细胞在转染后48h时的细胞增长平均抑制率分别为42.14%、39.12%、48.05%和35.85%。提示不同siRNA序列对SW4...  相似文献

癌症治疗诱发的代谢综合征(cancer treatment induced metabolic syndrome,CTIMetS)是一种非常普遍且有害的疾病。代谢综合征包括中心性肥胖、血脂异常、高血压和胰岛素抵抗,是多种代谢紊乱集于一身的综合征。癌症幸存者与一般人群相比有着更高的心血管事件风险。目前,癌症治疗引起代谢综合征的分子机制尚不清楚。然而,人们发现可以通过肿瘤治疗类型的不同对其病理生理学进行分类。虽然CTIMetS的病因学可能与一般人群中的代谢综合征不同,但是有效的治疗和干预方案却可能是相似的。本文综述了不同癌症治疗方式后CTIMetS发展的可能机制,为预防CTIMetS的发生发展提供参考策略。  相似文献

目的 研究抗瘤新药PTS(抑瘤仙 )体外对肺癌细胞、耐药肺癌细胞及对正常支气管上皮细胞的作用 ,为其临床应用提供理论依据。方法 以大细胞肺癌株NCI H 460、耐药大细胞肺癌株H 460 /cDDP及正常支气管上皮细胞株 16HBE为实验对象 ,采用 2 4h活细胞跟踪法、MTT法及台盼蓝拒染记录细胞生长曲线法 ,进行PTS的体外抗瘤作用研究 ,同时以DDP(顺铂 )为阳性对照 ,PBS(平衡盐液 )为阴性对照。结果 PTS对受到直接损伤的肺癌细胞及耐药肺癌细胞均有杀伤作用 ,具有抗瘤作用和一定的抗耐药性。与DDP比较 ,PTS有更强的抗瘤作用 ,对于H 460细胞在第 48h 5 0 0mmol的DDP杀伤率为 62 .1% ,1/2 0 0的PTS为 81.3 % ;而PTS对于正常人支气管上皮 16HBE细胞的影响较小 (DDP为 3 9.5 % ,PTS为 2 1.6% ) ,具有较高的选择性。PTS应用后立即起效并达高峰 ,作用持续时间短。结论 PTS体外抗瘤作用明显 ,并具有选择性和一定的抗耐药性  相似文献

PURPOSE: Bio-reduction/activation of anti-cancer drug beta-lapachone (beta-lap) is mediated by NAD(P)H: Quinone oxidoreductase (NQO1). We investigated the feasibility of using mild temperature hyperthermia to increase the anti-cancer effect of beta-lap by up-regulating NQO1 expression. METHODS: NQO1 expression in FSaII fibrosarcoma of C3H mice and A549 human lung cancer cells was evaluated with Western blot analysis and immunostaining of cells at different times after water-bath heating. Clonogenic cell survival method was used to determine the sensitivity of cells to heating, beta-lap, and in combination. The growth of FSaII tumors in the right hind legs of C3H mice was studied after heating the tumors at 42 degrees C for 1 h with water bath, an i.p. injection of beta-lap to host mice or an i.p. injection of beta-lap 24 h after heating the tumors. RESULTS: Heating at 42 degrees C for 1 h significantly increased the expression of NQO1 in the cancer cells with a maximum increase occurring 8-24 h after heating. The sensitivity of cancer cells to beta-lap treatment progressively increased until 24 h after heating most likely due to the increase in NQO1 expression. Heating the FSaII tumors at 42 degrees C for 1 h and treating the host mice with an i.p. injection of 50 mg/kg beta-lap 24 h after the tumor heating was far more effective than heating alone or beta-lap treatment alone to suppress the tumor growth. CONCLUSION: Mild temperature heat shock elevates the NQO1 expression in cancer cells, which in turn markedly increases the sensitivity of the cells to the bioreductive drug beta-lap in vitro and in vivo.  相似文献

目的:总结国内外关于TRAIL的抗肿瘤机制及应用的研究进展.方法:应用MEDLINE及CNKI期刊全文数据库系统,以"TRAIL、机制和治疗"等为关键词,检索2000-2010年的相关文献.纳入标准:1)TRAIL的结构及受体;2)TRAIL的作用机制;3)TRAIL的应用及临床试验.根据纳入标准,纳入分析23篇文献.结果:TRAIL在TNF家族中最显著的特征是对肿瘤细胞和转化细胞具有选择性的凋亡诱导作用,而对正常的组织和细胞不具有明显的毒性反应,并且与其他试剂联合应用具有协同抗肿瘤作用.结论:相对于化疗药物的毒副反应,TRAIL的独特优势为其在肿瘤治疗中奠定了良好基础,可以为目前的肿瘤治疗提供更有效,更安全的新方法.  相似文献

Tropomyosin-related kinase A (TrkA) is emerging as an important player in carcinogenic progression. TrkA overexpression, which is associated with cell growth, proliferation, survival, and invasion, has been observed in pancreatic cancer. We therefore synthesized HS-345, a novel TrkA inhibitor, and evaluated its anti-cancer effect and underlying mechanism of action in pancreatic cancer. In this study, HS-345 effectively inhibited the growth and proliferation in three pancreatic cancer cell lines (PANC-1, MIA PaCa-2, and BxPC-3). Activation of the TrkA/Akt signal cascade was also inhibited by HS-345 treatment in a dose-dependent manner. The pro-apoptotic effect of HS-345 was evidenced by increased levels of cleaved caspase-3 and cleaved PARP, and decrease of Bcl/Bax expression via mitochondria membrane potential, as well as elevated numbers of TUNEL-positive apoptotic cells. HS-345 was additionally found to exert anti-angiogenic effect by decreasing the expression of HIF-1α and VEGF, major factors of angiogenesis, which were also demonstrated by the suppression of tube formation and migration of VEGF-treated human umbilical vein endothelial cells along with inhibition of blood vessel formation by HS-345 in a Matrigel plug assay with mice. Results of our investigation show that HS-345 inhibited the TrkA/Akt signaling pathway resulting in cell growth/angiogenesis inhibition and apoptosis induction. Based on our data, we suggest that HS-345 is a potential candidate for treating pancreatic cancer.  相似文献

[目的]在社区中培训一支具有一定专业水平的肿瘤防治队伍,以整体提高社区常见肿瘤的早诊、早治率。[方法]对不同层次的人员分别制订针对性强,切实可行的培训计划,并采取多种形式的方法进行培训。[结果]通过培训前后的效果评估。表明受训人员的肿瘤防治知识均有不同程度的提高。[结论]在社区中有计划地开展肿瘤专业培训,对普及肿瘤防治知识,提高社区肿瘤防治人员的素质很有必要。  相似文献
