目的 按照ISO15189认可的要求,对UF-4000尿液有形成份分析仪进行性能评价,同时建立一种同一型号仪器的比对方案,以应用于临床。方法 对UF-4000的五项主要参数(RBC、WBC、EC、CAST、BACT)进行批内精密度、批间精密度、准确度、携带污染率、临床可报告范围、参考区间验证,与同机型检测结果的一致性比对。结果 UF-4000的批内精密度、批间精密度、准确度、携带污染率、临床可报告范围、参考区间验证等验证结果及UF-4000同机型检测结果的一致性比对结果均符合要求。结论 Sysmex公司生产的UF-4000尿液有形成分分析仪性能良好,能够满足临床需求。仪器间比对结果通过能满足同一机型组成流水线时检验结果的一致性要求,可明显提升工作效率。  相似文献

目的 根据ISO15189认可的要求和临床的实际需求,对HISCL-5000全自动高敏免疫分析仪进行性能验证,从而保证临床样本检验的准确性.方法 对HISCL-5000高敏免疫分析仪分别进行本底计数、批内精密度、批间精密度、线性验证、符合率(比对)、准确性、检出限的验证.结果 HISCL-5000高敏免疫分析仪检测传染病HBsAg、HBsAb、HBeAg、HCV-Ab、TP-Ab和HIVAg+Ab低浓度批内精密度CV分别为1.84%、3.82%、2.00%、1.96%、8.10%、1.45%;高浓度批内精密度CV分别为4.15%、1.60%、1.74%、1.52%、3.67%、1.57%,HBeAb低浓度批内精密度CV为5.23%,HBeAb高浓度批内精密度CV为2.83%.低浓度和高浓度样本批间精密度CV分别为HBsAg 1.96%、2.06%,HBsAb 3.64%、4.53%,HBeAg 3.29%、2.62%,HCV-Ab 2.30%、2.40%,TP-Ab 5.70%、8.17%,HIVAg+Ab 4.53%、2.53%,高浓度HBeAb 6.73%,低浓度HBeAb 1.40%.HBsAg和HBsAb线性验证R2值均>0.95,符合验证要求.两台仪器HBsAg和HBsAb定量检测结果 符合率Kappa≥0.75,表示两台仪器的结果 高度一致.HBsAg定量检测准确度偏差<10%,符合率大于90%以上,传染八项检出限验证结果 全部合格,仪器符合性能验证要求.结论 HISCL-5000全自动高敏免疫分析仪检测传染八项结果 准确性较高,可靠性较好,检验速率高效,随时满足临床需求,临床应用价值较高.  相似文献

目的 制备高浓度白细胞样本验证Sysmex XN-1000(B3)血细胞分析仪检测白细胞的性能.方法 从30份临床抗凝样本中分离白细胞,按照国家卫生行业标准WS/T 406-2012文件的要求对Sysmex XN-1000(B3)血细胞分析仪检测白细胞的性能进行验证.结果 任意混合多份临床样本分离白细胞可获得高浓度白细胞样本;Sysmex XN-1000(B3)血细胞分析仪检测白细胞的本底计数为0,批内精密度的CV为1.39%,低、中浓度水平质控品的日间精密度的CV分别为2.0%和1.9%,正确度偏倚为1.46%,在0~ 145.63×109/L检测范围内呈线性关系,携带污染率为0.06%,准确度的相对偏差范围为0.44%~ 5.74%.结果 任意混合多份临床样本,通过分离、富集可获得高浓度白细胞样本,用于Sysmex XN-1000(B3)血细胞分析仪检测白细胞的性能验证.  相似文献

目的 评价罗氏Cobas e601及雅培Architect i2000两种化学发光免疫分析系统检测抗-HCV的分析性能和一致性.方法 按照CLSI推荐的方法对两台分析仪检测抗-HCV的精密度、检出限、阴阳性符合率等性能进行验证试验;两台仪器分别检测120例有反应性血清样本及30例无反应性样本,用Kappa检验评价检测结果的一致性.结果 两台仪器的精密度良好;检出限均为0.3NCU;阴阳性符合率为100%;两个分析系统检测抗-HCV总体符合率为70.67%,无反应性、e601 S/CO≥15.0、i2000 S/CO≥6.0样本结果的符合率为100%,e601 S/CO为1.01 ~5.0和i2000 S/CO为1.01 ~2.0样本结果的符合率为0和44.4%:经统计分析κ=0.35在Kappa系数区段中一致性程度属于弱(κ=0.35,P<0.05).结论 两个分析系统检测抗-HCV的分析性能均良好,适用于临床标本的常规检测,对于两台仪器检测结果不一致的样本,尤其是e601 S/CO 1 ~ 15及i2000 S/CO 1~6的样本,建议进行补充实验(RIBA,NAT)来明确诊断.  相似文献

目的分析不同血细胞分析仪检测结果的可比性。方法以希森美康XN-1000型血细胞分析仪、配套试剂、原装配套校准品、质控品作为参比系统;迈瑞BC-5600型血细胞分析仪、配套试剂、原装配套校准品、质控品作为待评系统。依据行标WS/T406-2012要求收集血液样本40份,参考美国临床、实验室标准化协会(CLSI)EP9-A2文件,分别应用两台血细胞分析仪测定血小板计数(PLT)、红细胞压积(Hct)、血红蛋白(HGB)、红细胞计数(RBC)、白细胞计数(WBC),并予以分析。结果PLT、Hct、HGB、RBC、WBC相对偏差符合要求比例,分别为98.00%、95.25%、98.00%、96.00%、100.00%,五个测定参数相对偏差符合比例,均≥80%,临床评估可接受;线性回归分析可知,两台仪器具有良好相关性,差异无统计学意义(P0.05)。结论在使用多台仪器检测时,要加强实验仪器的质量控制,定期对不同的检测仪器进行校准和比对,以确保检测结果的准确性和一致性,从而更好的服务临床。  相似文献

目的 免疫胶体金法用于快速检测受试者手指全血的促甲状腺素(TSH),可以定量分析且快速简便.本研究拟对免疫胶体金法定量检测TSH的精密度性能及检测仪器间是否存在差异进行实验研究.方法 设计7个不同浓度的促甲状腺激素(TSH)质控品和人血清样本,每天对各样本重复测定4次,持续5天,计算重复性(批内CV)和中间精密度(CV).选用33份不同浓度的TSH血清标本,分别用检测仪器1和检测仪器2进行判读分析,比较两者结果的相关性.结果 质控品和病人血清TSH浓度为1.20、2.51、3.46、5.02、7.63、10.03、11.08 μIU/mL,对应的批内CV分别为30.00%、17.84%、16.50%、15.00%、13.92%、10.62%、9.62%,中间精密度CV分别为29.23%、20.19%、18.15%、16.60%、15.34%、10.42%、8.76%.二台检测仪器检测TSH值相关性r为0.968,无统计学差异.结论 免疫胶体金法定量检测TSH在低浓度TSH水平的精密度变异较大,但在中高浓度TSH尤其>10μIU/mL情况下,检测精密性能良好.不同检测仪器间检测结果也无明显差异.  相似文献

目的验证上海透景公司生产的化学发光法血清鳞状上皮细胞癌相关抗原(SCC)定量测定试剂盒在非配套免疫检测系统中的性能,并初步评价其临床应用价值。方法参考CLSI EP15-A2及EP9-A2方案,在雅培全自动免疫分析仪i2000SR上验证上海透景化学发光法SCC定量测定试剂盒,主要评价试剂盒的精密度、正确度、线性范围和生物参考区间。采用上海透景公司提供的多项肿瘤标志物质控品进行精密度的评价;同时,收集雅培化学发光法SCC检测结果均匀分布在测量范围内的冻存血清标本进行比对试验和线性范围的评价。另外,选取120例表观健康体检人员的新鲜血清标本进行生物参考区间的评价。将所得的结果与厂家提供的数值进行比较,判断其是否一致。结果测定多项肿瘤标志物质控品,血清SCC低值和高值质控品实验室内CV分别为5.37%和4.02%,均小于厂家提供的数值(≤10%),精密度验证通过。40例标本进行比对试验时,透景与雅培试剂盒SCC检测结果相关性良好,说明二者之间具有一致性。线性范围验证时,血清SCC的线性范围为0.1~70.0 ng/mL,决定系数R^2大于0.995。120例表观健康体检人员的血清SCC检测结果均在厂家提供的参考区间范围之内。结论上海透景化学发光法血清SCC定量测定试剂盒在非配套免疫检测系统(雅培全自动免疫分析仪i2000SR)中具有良好的精密度,与雅培化学发光法SCC试剂盒的检测结果和线性范围一致,可初步满足临床需要。  相似文献

目的对两种全自动血液分析仪检测结果进行比对分析,观察其检测结果的一致性。方法根据美国化学标准化委员会(NCCLS)标准文件EP9-A2的要求,用本科室的两台全自动血液分析仪对白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、血小板(PLT)、红细胞压积(HCT)、红细胞平均体积(MCV)六个项目进行比对实验,验证两台仪器的一致性。结果两台仪器的精密度与相对偏差均在允许范围内,两台仪器的检测值用配对检验,差异无统计学意义(P>0.05),线性回归方程符合均要求。结论两台仪器检测结果具有较好的准确性和可比性,能够满足临床需要。  相似文献

Over 200 schizophrenic patients belonging to three major and interrelated pedigree complexes have been investigated over the past 30 years in a North Swedish geographically isolated population, presently numbering about 6,000. An intensive investigation of a number of biochemical correlates and genetic markers in a few selected families belonging to one of the major pedigrees has indicated new strategies for the current research program.
Schizophrenia, as defined operationally, is significantly associated with decreased activities of two enzymes (1) blood platelet monoamine oxidase, (2) plasma dopamine-β-hydroxylase, and (3) with the genetic marker Gc2 (group specific antigen). Both enzymes are subject to genetic variation. A positive score for linkage between schizophrenia and low plasma DBH activity has been calculated, but, so far, available data are insufficient for discrimination between linkage and partial contribution of genetically controlled low plasma DBH to the pathogenesis of the disease. Alternatively, both mechanisms could be involved.
As a model for continued research, schizophrenia is explained as based on a double dominant-recessive genotype (Aabb), representing a vulnerability which in about 50 % of cases develops into clinical schizophrenia. It is suggested that the dominant mutation (A) operates on or affects MAO activity, and that the recessive genotype (bb) is instrumental in low variates of DBH activity and very likely such variates within the normal range of physiological variation. Moreover, it is suggested that the combined effects of MAO- and DBH-reduced efficiency on the metabolism of e.g. dopamine could be an essential pathogenic mechanism for the schizophrenic illness which is segregating in this population.  相似文献

HLA-A,-B,-C,-DRB1 and -DQB1 alleles have been studied in Chimila Amerindians from Sabana de San Angel (North Colombian Coast) by using high resolution molecular typing. A frequent extended haplotype was found:HLA-A*24:02-B*51:10-C*15:02-BRB1*04:07-DQB1*03:02 (28.7%) which has also been described in Amerinndian Mayos Mexican population (Mexico, California Gulf, Pacific Ocean). Other haplotypes had already been found in Amerindians from Mexico (Pacific and Atlantic Coast), Peru (highlands and Amazon Basin), Bolivia and North USA. A geographic pattern according to HLA allele or haplotype frequencies is lacking in Amerindians, as already known. Also, five new extended haplotypes were found in Chimila Amerindians. Their HLA-A*24:02 high frequencies characteristic is shared with aboriginal populations of Taiwan; also, HLA-C*01:02 high frequencies are found in New Zealand Maoris, New Caledonians and Kimberly Aborigines from Australia. Finally, this study may show a model of evolutionary factors acting and rising one HLA allele frequency (-A*24:02), but not in others that belong to the same or different HLA loci.  相似文献

Starting with the integument, we see many organs are contractile sacs or multiples thereof, which tubes or bags constitute the major part of the entire body. Recognition of this basic unit and its characteristics sheds new light, individually and collectively, on many disorders previously considered unrelated. Muscular tears and perforations develop in the walls of these chambers, being no way peculiar to those organs, wherein, hydrochloric acid occurs. So, it is not necessary to explain the absence of excessive acid from patients who exhibit holes in the gastric, uterine, aortic, duodenal, rectal, pulmonary, retina, and other walls. Muscle, not acid is the great common factor relating idiopathic disorders in the gastrointestinal tract to each other and to similar diseases in other systems. When the units are linked together, the lesions tend to appear as arthropathies, i.e. at the joints. Rephrasing common-place observations, frees us from conventional, conceptual cul-de-sacs. An observation is only as good as its interpretation, so all possibilities must be considered, otherwise, we will remain blinded by our misconceptions.  相似文献

Zusammenfassung Der Einfluß von verschiedenen Nahrungsmitteln auf Methoden zur Bestimmung von Adrenalin (AD), Noradrenalin (NA), Vanillinmandelsäure (VMS), Metanephrinen (MN), Homovanillinsäure (HVS) und 5-Hydroxyindolessigsäure (5-HIE) im 24 h-Harn zur Diagnose des Phäochromozytoms bzw. Karzinoid-Syndroms wurde untersucht. Die in die Untersuchung einbezogenen Nahrungsmittel waren: Tee, Kaffee, Mandeln, Ananas, Käse, Walnüsse, Vanillepudding, Bananen, Tomaten und Milchschokolade. Außerdem wurde der Einfluß des Zigarettenrauchens auf die Bestimmung von AD, NA, VMS und MN untersucht.Walnüsse führten zu einer starken Erhöhung der 5-HIE-Ausscheidung. Bananen erhöhten die Ausscheidung von AD, NA, VMS, MN und 5-HIE. Kaffee und Ananas bewirkten eine geringe Zunahme der MN-Werte. Rauchen von 20–30 Zigaretten/Tag beeinflußte keine der vier Variablen.Wenn die beschriebenen Methoden benutzt werden, sollte lediglich auf den Verzehr von Bananen und Walnüssen vor und während der Harnsammelperioden verzichtet werden, da die oberen Normgrenzen im Harn überschritten werden könnten. Ein Verzicht auf Kaffee und Ananas in normalen Mengen ist nicht erforderlich. Es besteht kein Anlaß, weiterhin die bisherigen umfangreichen Restriktionen der übrigen Nahrungsmittel beizubehalten.  相似文献

A series of surface-functionalized poly(ether ether ketone) (PEEK) films has been prepared by selective wet-chemistry; they are hydroxylated polymer (PEEK-OH) obtained by reduction, aminated polymer (PEEK-[]-NH2) prepared by coupling a diisocyanate reagent to PEEKOH (PEEK-[]-NCO) followed by hydrolysis, and carboxylated and aminocarboxylated polymers (PEEK-[]-GABA and PEEK-Lysine) resulting from the coupling of aminoacids to PEEK-[]-NCO. The aminated and carboxylated substrata promoted the adhesion and growth of CaCo2 cells in the presence of serum. Fibronectin (FN), an extra-cellular matrix protein, has been covalently fixed and/or adsorbed on various PEEK substrata, in the presence or not of a polymeric surfactant (Pluronic F68). The performances of the FN-grafted substrata (PEEK-[]-FN(1) and PEEK-[]-FN(2)) were significantly higher than those of reference substrata simply coated with FN (PEEK-OH(+FN)(1) and (2), PEEK-[]-NH2(+FN)(1) and (2)), considering the adhesion and spreading of CaCo2 cells in the absence of serum. Moreover, the stability of the adherent cells on the FN-adsorbed substrata dramatically depended on the experimental conditions applied during the PEEK coating with FN.  相似文献

Homer's epic tale of the 20-year return of Odysseus from the Trojan War is investigated with particular reference to Jung's theory of individuation. Odysseus' meetings with 'the anima' in the form of goddesses, sirens and female monsters and his visit to Hades demonstrate the confrontation and humanization of aspects of the archetypal level of the psyche, central to Jung's theories of psychic growth and development. Jung's important ideas of the psychoid level and the transcendent function are explored and linked both to his investigations into medieval alchemy and with findings from contemporary neuroscience. The importance of Jung's constructive method of the interpretation of dreams and myths is shown to be central.  相似文献
