Summary Dopamine (DA) stimulates the cAMP-generating system in the male rat hypothalamus only to a very low extent (25% above control). Diethylstilbestrol (DES), a synthetic estrogen, was found to be extremely potent (a 4- and 16-fold stimulation at 20 M and 100 M, respectively). Addition of either one to an incubation medium containing varying concentrations of the other resulted in a synergistic response. The potentiation by 20 M DES of the effect elicited by 100 M DA was the most remarkable, namely, a 3-fold stimulation of the combined response. A 4- and 7.5-fold stimulation of cAMP accumulation was observed when adenosine (100 M) or adenosine (100 M)+DA (100 M) were present in the incubation medium. Theophylline (0.5 mM), an adenosine antagonist, could effectively reduce this effect, as did adenosine deaminase (10 g/ml). Clomiphene (50 M), an estrogen antagonist, exhibited a marked decrease in DES+DA-elicited cAMP formation. Pimozide (40 M) had the ability to significantly block the stimulatory effects of DES and DA.  相似文献

Summary The correlations between the relaxing effect of papaverine derivatives, inhibition of low Km-phosphodiesterase (cAMP-PDE=EC 3.1.4.17) activity and cyclic 3,5-AMP (cAMP) levels in isolated rabbit ileum were investigated. There was a strong correlation between the relaxing effect, inhibition of PDE activity and cAMP content for eupaverine, ethylpapaverine and papaverine. Eupaverine was the most effective relaxing agent (I50=7.5 M) and the most potent inhibitor of PDE activity (Ki=0.6 M), followed by ethylpapaverine (I50=10 M); Ki=0.8 M) and papaverine (I50=20 M; Ki=2 M). In contrast, there was a strong relaxing effect (I50=6 M) but only slight inhibition of PDE activity (Ki=350 M) by tetrahydropapaveroline (THP). The adenylate cyclase stimulating effect of THP which was shown by others is most likely the reason for comparatively higher cAMP levels, which were found to be elevated about seven times over basal levels of 0.35 nmoles/g wet weight, and effective relaxation. Relaxation could be induced by exogenously added cAMP (I50=45 M) and dibutyryl-cAMP (I50=450 M). Our results support the assumption that smooth muscle relaxation in rabbit ileum is mediated by cAMP. 1.Acyclovir [9-(2-hydroxyethoxymethyl)guanine] interfered with embryonic development in vitro when assessed with the whole-embryo culture technique. The no-observed-effect level was at 10 M acyclovir; 2. Minor impairment of embryonic development (retarded development of ear anlagen) was observed in vitro at 25 M acyclovir in the culture medium. At high concentrations (100 or 200 M) development of the ear anlagen was largely inhibited. At concentrations of 50 M acyclovir or higher, additional disturbances of embryonic differentiation in vitro became obvious, resulting in gross structural abnormalities, especially of the brain (telencephalon); 3. Histological examinations confirmed and extended these observations: at 100 M acyclovir alterations of the neuroepithelium of the ventricles were pronounced, the telencephalon had developed poorly or was almost completely absent, and necroses were seen in the ear anlagen, the maxillar branch and within the somites; 4. In a limb bud culture (mouse embryos, starting with day 11 of gestation) acyclovir interfered with the differentiation of cartilaginous bone anlagen at concentrations of 200 M and more in the culture medium. A concentration of 100 M induced no significant effect. Thus, this organ culture system is less sensitive to the action of acyclovir when compared with whole-embryo culture; 5. The influence exerted by monoamines on acetylcholine release was studied in electrically stimulated slices of guinea pig nucleus basalis magnocellularis (nbM) prelabelled with 3H-choline (3H-Ch).Noradrenaline, 30 M, and clonidine, 1 M, reduced the evoked 3H-Ch efflux by about 50%, but phenylephrine, 100 M. did not; idazoxan, 0.1 M. but not prazosin, 1 M, antagonized these effects. pointing to the involvement of alpha2 receptors. Apomorphine, 1 or 30 M. reduced 3H-Ch efflux from nbM slices as well. The effect was shared by quinpirole, 1 or 10 M, but not by 2,3,4,5-tetrahydro-7,8-dihydroxy-1-phenyl-1H-3-benz-azepine (SKF 38393). 10 M, and was antagonized by sulpiride, 1 M, but not by R-(+)-8-chloro-2,3,4,5-tetra-hydro-3-methyl-5-phenyl-1H-3-benzazepin-7-ol (SCH 23390). 1 M, suggesting the involvement of the D2 receptor subtype.5-hydroxytryptamine (5-HT) 0.3–30 M, and alphamethyl-5-HT, 10 M, significantly increased 3H-Ch efflux from nbM slices; the 5-HT2 antagonist ritanserin, 1 M. prevented this response. 2-methyl-5-HT, 1–30 M, inhibited the evoked 3H-Ch efflux and its effect was prevented by the 5-HT3 antagonist 1H,3,5H-tropan-3-yl-3,5-dichlorobenzoate (MDL 72222). 1 M.These findings indicate that i) catecholamines inhibit nbM neurons through alpha2 and D2 receptors and that ii) a complex serotonergic modulation of cholinergic function exists in the nbM, involving the activation of various receptor subtypes. which can mediate opposite responses. This study investigated the toxic effects of the insecticides lindane and chlorpyrifos, the herbicide diuron, the organometallic antifoulant tributyltin (TBT), and the surfactant sodium dodecyl sulfate (SDS) on the early life stages of Paracentrotus lividus (Echinodermata, Euechinoidea), Ciona intestinalis (Chordata, Ascidiacea), Maja squinado and Palaemon serratus (Arthropoda, Crustacea) in laboratory acute toxicity tests. The assays studied embryogenesis success from fertilized egg to normal larvae in P. lividus (48 h incubation at 20 °C) and C. intestinalis (24 h incubation at 20 °C), and larval mortality at 24 and 48 h in M. squinado and P. serratus. For P. lividus, the median effective concentrations (EC50) reducing percentages of normal larvae by 50% were: 350 g l–1 for chlorpyrifos, 5500 g l–1 for diuron, 4277 g l–1 for SDS, and 0.309 g l–1 for TBT. For C. intestinalis, the EC50 values affecting embryogenesis success were 5666 g l–1 for chlorpyrifos, 24,397 g l–1 for diuron, 4412 g l–1 for lindane, 5145 g l–1 for SDS, and 7.1 g l–1 for TBT. The median lethal concentrations (LC50) for M. squinado larval survival were 0.84 g l–1 (24 h) and 0.79 g l–1 (48 h) for chlorpyrifos, 2.23 g l–1 (24 h) and 2.18 g l–1 (48 h) for lindane, and 687 g l–1 (48 h) for SDS. For P. serratus the LC50 values obtained were 0.35 g l–1 (24 h) and 0.22 g l–1 (48 h) for chlorpyrifos, 3011 g l–1 (24 h) and 3044 g l–1 (48 h) for diuron, 5.20 g l–1 (24 h) and 5.59 g l–1 (48 h) for lindane, and 22.30 g l–1 (24 h) and 17.52 g l–1 (48 h) for TBT. Decapod larvae, as expected, were markedly more sensitive to the insecticides than sea urchins and ascidians, and SDS was the least toxic compound tested for these organisms. Lowest observed effect concentrations (LOEC) of TBT for sea urchin and ascidian embryos, chlorpyrifos and lindane for crustacean larvae, and SDS, were similar to those found in many coastal areas indicating that there would be a risk to invertebrate embryos and larvae from exposure in the field to these pollutants.  相似文献

Summary (1)We have studied the ability of some regulatory peptides to induce a mitogenic (incorporation of tritiated thymidine) response in human peripheral blood mononuclear cells (PBMC) and to modify the response produced by phytohaemagglutinin (PHA), a well known PBMC mitogen. (2) Human calcitonin gene-related peptide (hCGRP), human or salmon calcitonin (hCT, sCT), neurokinin A (NKA) and neurokinin (4–10) (up to 1 M for each peptide) did not produce per se any significant PBMC stimulation. (3) hCGRP (0.1 nM –1 M) produced a concentration dependent enhancement of the response to a submaximal concentration of PHA (1 g/ml). On the other hand, hCGRP decreased the mitogenic response to a maximal concentration of PHA (25 g/ml). (4) Neither hCT nor sCT (0.1 nM–1 M) had a significant influence on the response to PHA (1–25 g/ml). (5) Both NKA and NKA (4–10) produced a concentration-dependent (1 fM –10 pM) enhancement of the response to 1 g/ml PHA, while these compounds had no effect on the response to 25 g/ml PHA. (6) These findings suggest a potent modulatory action of CGRP and NKA, two peptides present in sensory and other nerves, on immune function which is possibly mediated via C2 receptors for CGRP and NK-2 tachykinin receptors, respectively.  相似文献

Summary (1) Longitudinal muscle strips from the human small intestine (jejunum/ileum) responded to electrical field stimulation (1–50 Hz) with frequency-related primary contractions which were largely atropine- (3 M) sensitive. When the tone was raised by addition of galanin (0.3 – 1 M), prostaglandin (PG) E2 (1–10 M) or neurokinin A (NKA, 0.1 M), a frequency-related relaxation was evident which was potentiated by atropine. All the responses to field stimulation were abolished by tetrodotoxin (1 M), thus indicating their neural origin. (2) The atropine-sensitive primary contraction to field stimulation was virtually abolished by omega conotoxin fraction GVIA (CTX, 0.1–0.3 M) while the relaxations were CTX-resistant. The field stimulation-induced relaxations, which were observed in the presence of atropine and guanethidine (3 M), were also unaffected by apamin (0.1 M). (3) NKA and substance P (SP) produced a concentration- (1 nM–1 M for both peptides) related contraction, NKA being about 53 times more potent than SP. [Pro9]SP sulphone and [MePhe7]-NKB, selective agonists of the NK-1 and NK-3 receptor, respectively, were barely effective. On the other hand, [\Ala8]NKA(4–10), a selective NK-2 receptor agonist, had a potent contractile activity, similar to that of NKA. (4) Galanin (1 nM–1M) produced an atropine- and tetrodotoxin-resistant concentration-related contraction of longitudinal muscle of human isolated small intestine. The response to galanin did not show any sign of fading and was particularly suitable to study the evoked relaxations. (5) Calcitonin gene-related peptide (CGRP) (10–100 nM) consistently inhibited the nerve-mediated contractions of strips from the ileum while the effect on the jejunum was less pronounced. Vasoactive intestinal polypeptide (VIP, 0.1–1 M) inhibited nerve-mediated contractions both in the ileum and the jejunum. (6) These experiments indicate that both cholinergic excitatory and non-adrenergic non-cholinergic nerves affect motility of the longitudinal muscle of the human small intestine. Furthermore, several neuropeptides produce potent motor effects, the contractile response to tachykinins being apparently mediated by activation of NK-2 receptors.  相似文献

Mouse embryos at the preimplantation stage were used in vitro for assessing the risk exerted by arsenic on early stages of embryonal development. We studied the effects of sodium arsenite on morphological development, proliferation, and micronucleus formation.A concentration of 100 M sodium arsenite immediately killed the embryos, and 1 M almost completely inhibited formation of blastocysts. Micronuclei were induced starting from 0.7 M arsenite. Concentrations below 0.1 M (0.1 M corresponds to 7.5 ppm arsenic) did not show significant effects with respect to any of the endpoints examined.Arsenic did not influence radiation risk under various experimental conditions; all the effects after combining moderately toxic doses of sodium arsenite and X-rays corresponded to the effect obtained by the addition of the single effects.  相似文献

We have investigated the ability of human CGRP (CGRP) to inhibit the electrically-evoked myogenic contractions of the guinea-pig ureter, in comparison with the K channel opener, cromakalim, and the adenylate cyclase activator, forskolin. CGRP (0.1 nM -0.1 M) produced a concentration-dependent inhibition of the evoked contractions; its action was prevented by the CGRP receptor antagonist, CGRP(8–37) (1 M), while it was unaffected by the nitric oxide (NO) synthase inhibitor, L-nitroarginine (30 M). The effect of CGRP was antagonized in a noncompetitive manner (depression of E max, no change in EC50) by glibenclamide (1–10 M), a blocker of ATP-sensitive potassium channels (KATP). A substantial fraction of the inhibitory effect of CGRP was glibenclamide-resistant, however. Glibenclamide also blocked the inhibitory action of cromakalim (0.1–10 M) without affecting the inhibition produced by forskolin (0.1–30 M). When tested in a low-K medium (extracellular K reduced from 5.9 to 1.2 mM), the inhibitory effects of CGRP, cromakalim and forskolin were enhanced. The inhibitory effect of forskolin was partly antagonized by glibenclamide when tested in a low-K medium.CGRP (0.1 M), cromakalim (3 M) and forskolin (10 M) inhibited the contractile response to KCl (80 M), which is characterized by a distinct phasic and tonic component: cromakalim selectively inhibited the phasic response to KCl with CGRP and forskolin inhibited both components. The inhibitory effect of CGRP on the phasic contraction to KCl was partly glibenclamide (1 M) sensitive, while that on the tonic contraction was glibenclamide-resistant. The inhibitory action of forskolin on both components of the response to KCl was unchanged by glibenclamide. The inhibitory effect of cromakalim on the phasic response was prevented by glibenclamide.In sucrose gap electrophysiological experiments, CGRP (0.3 M) produced hyperpolarization of the membrane, increased the delay between stimulus application and onset of the action potential, shortened the action potential duration and eventually suppressed both action potential and phasic contractions in 4 out 8 cases tested. The membrane hyperpolarization and the ability of CGRP to suppress the evoked action potential were completely prevented by glibenclamide (1 M), but an inhibitory effect on the action potential duration and contractility persisted in the presence of glibenclamide.We conclude that activation of glibenclamide-sensitive K channels mediates part of the smooth muscle relaxant action of CGRP, and accounts for the ability of CGRP to suppress phasic contractions evoked through electromechanical coupling. However, a glibenclamide-resistant component also exists in the smooth muscle relaxant activity of CGRP. The possible involvement of cAMP accumulation as an intermediate link between CGRP receptor occupation and activation of K channels by CGRP remains speculative. Correspondence to: C. A. This study in normal male subjects further investigates the effects of dopaminergic-antidopaminergic interactions as manifested by the prolactin response to dopamine and neuroleptic drugs. Incremental doses of dopamine hydrochloride (4 g/min, 15 g/min, 60 g/min, 300 g/min) were infused fused at a constant rate over 90–120 min after a fixed dose of a neuroleptic drug (sufficient for about half of the maximal prolactin response) had been given IV. A dose of dopamine in the order of 15–60 g/min appeared to match the los of endogenous dopaminergic inhibition due to the antidopaminergic effect of the neuroleptic drug. The lactotrophic cells of the pituitary gland are suggested to serve as a model in man for the study of some basic neurohormonal mechanisms.  相似文献

Summary Cyclic adenosine-3,5-monophosphate (cAMP) phosphodiesterases (PDE) were partially purified from human and rat gastric mucosa. Drugs known to affect the cyclic nucleotide system and/or gastric secretion were tested for effects on the PDE-activities from both species.In rat gastric mucosa PDE-activity can be detected in the 100 000×g sediment (K m =8.3 M; V max=3.2 nmoles cAMP hydrolyzed/mg protein x min) and the cytoplasma (K m =5.6 M; V max=2.6 nmoles cAMP hydrolyzed/mg protein x min).The most effective inhibitors of the particle-bound activity are papaverine (K i =4 M, non-competitive) and 3-isobutyl-1-methylxanthine (K i=14 M, competitive). There was only a modest competitive inhibition by theophylline (K i =495 M). PDE-activity in the cytoplasma was inhibited competitively by these three drugs (papaverine: K i =6.5 M; 3-isobutyl-1-methylxanthine: K i =37 M; theophylline: K i =152 M.In human gastric mucosa PDE-activity can be detected in the particular fraction (K m =23.9 M; V max=1.2 nmoles cAMP hydrolyzed/mg protein x min), and the soluble fraction (K m =12.1 M; V max=2.4 nmoles cAMP hydrolyzed/mg protein x min).PDE-activity in the 100 000×g sediment was inhibited by papaverine (K i =5.6 M, non-competitive), 3-isobutyl-1-methylxanthine (K i =16 M, non-competitive), theophylline (K i =165 M, non-competitive), and N6-2-O-dibutyryl-cAMP (K i =746 M, competitive).Inhibition in the 100 000×g supernatant was noncompetitive with 3-isobutyl-1-methylxanthine (K i =7.1 M and papaverine (K i =8.5 M), but competitive with N6-2-O-dibutyryl-cAMP (K i =170 M), and theophylline (K i =225 M). This study indicates that PDE-activities of the two species are qualitatively similar, but quantitative differences exist.  相似文献

The aim of this study was to determine the involvement of the central cholinergic system in the rise in blood pressure evoked by the thromboxane A2 (TxA2) analog, U-46619, given centrally. Intracerebroventricular (i.c.v.) injections of U-46619 (0.5, 1.0 and 2.0 g) caused dose- and time-related increases in blood pressure and decreased heart rate in awake rats. U-46619 (1 g; i.c.v.) also produced an approximately 65% increase in posterior hypothalamic extracellular acetylcholine and choline levels. Pretreatment with SQ-29548 (8 g; i.c.v.), selective TxA2 receptor antagonist, completely inhibited both the cardiovascular responses and the increase in acetylcholine and choline levels to subsequent injection of U-46619 (1 g; i.c.v.). Atropine (10 g; i.c.v.), nonselective muscarinic receptor antagonist, pretreatment did not affect the cardiovascular responses observed after U-46619 (1 g; i.c.v.). Pretreatment with the nonselective nicotinic receptor antagonist, mecamylamine (50 g; i.c.v.) attenuated the pressor effect of U-46619 (1 g; i.c.v.). Higher doses of mecamylamine (75 and 100 g; i.c.v.) pretreatments did not change the magnitude of the blockade of pressor response to U-46619; however, they abolished the bradycardic effect of U-46619 dose-dependently. Interestingly, pretreatment of rats with methyllycaconitine (10 g; i.c.v.) or -bungarotoxin (10 g; i.c.v.), selective antagonists of 7 subtype of nicotinic acetylcholine receptors (7nAChRs), partially abolished the pressor response to i.c.v. injection of U-46619 (1 g). Similar to the mecamylamine data, the use of higher doses of methyllycaconitine (25 and 50 g; i.c.v.) produced the same magnitude of blockade that was observed after the 10 g methyllycaconitine pretreatment, but it completely abolished the bradycardic effect of U-46619 (1 g; i.c.v.) at the dose of 25 g. The present results show that central administration of U-46619 produces pressor and bradycardic effect and increase in hypothalamic acetylcholine and choline levels by activating central TxA2 receptors. The activation of central nicotinic receptors, predominantly 7nAChRs, partially mediates the cardiovascular responses to i.c.v. injection of U-46619.  相似文献

Twelve patients suffering from partially reversible chronic obstructive pulmonary disease (COPD) took past in a single blind, randomised, 4-way cross-over trial to determine the optimal dose and duration of action of the anticholinergic agent oxitropium bromide (OTB) inhaled as a nebulised solution.Single doses of 500, 1000, 1500 and 2000 g nebulised OTB were compared during a 6 hour-observation period. Lung function test results indicated that 500 and 100 g OTB only induced slight bronchodilatation, whereas 1500 and 2000 g OTB produced a significantly greater increase in mean FEV1 compared to 500 g. There was a trend for 2000 g to be superior to 1000 g, but 2000 g and 1500 g were not significantly different. Significant bronchodilatation (>15% rise in FEV1 from baseline) persisted for 6 h after 1500 g. A significant decrease in airway resistance (Raw) was observed following inhalation of 2000 g. The mean decrease in Raw was 33% after 30 min, 20% after 4 h and 12% after 6 h.In this trial, 2000 g OTB administered by an ultrasonic nebuliser was the optimal dose, but a satisfactory result was also obtained with 1500 g.  相似文献

The effects of the putative selective P2X purinoceptor agonist, ,-methylene-l-adenosine 5-triphosphate (me-l-ATP), were determined at rat neuronal and smooth muscle P2X purinoceptors.Me-l-ATP had no effect on the extracellularly recorded membrane potential of the rat isolated vagus nerve preparation at concentrations up to 300 M. In contrast, the archetypal P2X purinoceptor agonist, , methylene ATP (meATP;1–100 M), produced concentration-related depolarisation responses with a mean EC50 value of 10.8 M. The depolarising effects of meATP were not attenuated by me-l-ATP (100 M). In voltage clamp experiments on single nodose ganglion neurones, ATP (100 M), but not me-l.-ATP (1–300 M), evoked rapid ( < 20 ms onset) inward currents when applied using a concentration-clamp method. In receptor binding studies to rat brain membranes, me-d-ATP and meATP competed with high affinity for [3H]Lx meATP binding sites, with mean pIC50 values of 7.7 and 8.3, respectively. However, me-l-ATP possessed low affinity for these sites and competed only at concentrations in excess of 10 M (mean pIC50 value 4.1).In prostatic segments of the rat vas deferens, me-l-ATP (1–100 M) and meATP (0.3–100 M) each produced concentration-related contractile responses with mean EC50 values of 17.1 and 3.6 M, respectively. Me-l-ATP (1–10 M) evoked fast inward currents in freshly dispersed vas deferens smooth muscle cells, indicative of an action at ligand-gated ion channels. Binding sites in vas deferens membranes labelled using 1 nM [3H]meATP exhibited high affinity for me-l-ATP, meATP and me-d-ATP with mean PIC50 values of 7.7, 8.4 and 7.3, respectively.These results indicate that me-l-ATP exhibits neither agonist nor antagonist properties at P2X purinoceptors on rat vagal neurones and possesses only very low affinity for [3H]meATP binding sites in rat brain. In contrast, me-l-ATP is a potent, high affinity agonist at smooth muscle P2X purinoceptors of the rat vas deferens. This selective agonist action of me-l-ATP suggests that P2X purinoceptors in smooth muscle and neurones are different and represent distinct P2X purinoceptor subtypes.  相似文献

Summary Effects of dynorphin-(1–13), Leu5-enkephalin,d-Ala2,d-Leu5-enkephalin (DADLE), and for comparison bremazocine, on plasma noradrenaline concentration and mean arterial pressure (MAP) were studied in pithed rabbits. In the first series of experiments, the sympathetic outflow was stimulated electrically via the pithing rod at 2 Hz twice for 3 min each (S1, S2). Drugs were administered before S2. Bremazocine 10 g/kg+2g/kg/h and 100 g/kg+20 g/kg/h, dynorphin 1 and 3 g/kg/min, Leu5-enkephalin 100 g/kg/min and DADLE 10 and 30 g/kg/min all diminished the electrically-evoked increase in plasma noradrenaline and MAP. The effects were antagonized by naloxone. In the second series, an infusion of noradrenaline (2 g/kg/min) was given twice for 3 min each (N1, N2). Drugs were administered before N2. Bremazocine 100 g/kg+20 g/kg/h slightly enhanced the pressor effect of exogenous noradrenaline, whereas dynorphin 3 g/kg/min, Leu5-enkephalin 100 g/kg/min and DADLE 30 g/kg/min caused no significant change. In the third series, the sympathetic outflow was stimulated continuously at 2 Hz, and the interaction of dynorphin and DADLE was studied. Dynorphin 1 g/kg/min and DADLE 10g/kg/min initially decreased MAP to a similar extent. The effect of DADLE faded with time. When, during continuous infusion of DADLE 10 g/kg/min, and after return of MAP to the pre-DADLE level, dynorphin 1 g/kg/min or DADLE 10 g/kg/min was infused additionally, the effect of dynorphin was unchanged, whereas that of DADLE was almost abolished. We conclude that the opioid peptides as well as bremazocine decrease action potential-evoked release of noradrenaline and, secondarily, blood pressure. They act at peripheral sites, presumably prejunctional opioid receptors at postganglionic sympathetic axons. Dynorphin on the one hand, and Leu5-enkephalin and DADLE on the other hand, appear to act at different receptors, dynorphin probably at a - and DADLE and Leu5-enkephalin at a -receptor.  相似文献

The adenosine analogue N6-cyclopentyladenosine (CPA), acting via postjunctional A1 receptors, has been shown to enhance contractions of the rat vas deferens induced by adenosine 5-triphosphate (ATP), the sympathetic cotransmitter in this tissue. The aim of the present study was to examine the ability of CPA to enhance contractions induced by other contractile agents. CPA (0.01–0.3 M) enhanced contractions induced by exogenous ATP (10 M), 5-hydroxytryptamine (5-HT) (3 M), tyramine (10 M), 2-methyl-5-hydroxytryptamine (2-Me-5-HT) (10 M) and KCl (35 mM) and this enhancement was blocked by an A1-selective concentration (3 nM) of 1,3-dipropyl-8-cyclopentylxanthine (DPCPX). CPA failed to enhance contractions induced by exogenous noradrenaline (NA) (1 M or 10 M), bradykinin (0.1 M), phenylephrine (3 M) or carbachol (10 M).The contractions induced by ATP (10 M), 5-HT (3 M), 2-Me-5-HT (10 M) and KCl (35 mM) were unaffected by tetrodotoxin (1 M) as well as by desensitisation of the P2x-purinoceptors with the ATP analogue adenosine 5-(, -methylene) triphosphonate. The contractions induced by tyramine (10 M) and 2-Me-5-HT (10 M) were blocked by prazosin (100 nM) or by imipramine (1 M). Ketanserin (10 nM) antagonised the response to 5-HT giving a dose-ratio of 12.9 corresponding to an apparent pA2 of 9.1.In conclusion, the A1-mediated effect was clearly selective for certain contractile agents and not due to a non-specific increase in contractility of the tissue. CPA enhanced contractions induced by both ATP and indirect sympathomimetics which release endogenous NA, and this enhancement of the two sympathetic cotransmitters may have a functional significance, and demonstrates the complexity of the neuromodulatory effects of adenosine in the rat vas deferens.  相似文献

We aimed at studying the mechanism(s) of the inhibitory effect exerted by calcitonin gene-related peptide (CGRP) on the spontaneous activity of the guinea-pig isolated renal pelvis. In organ bath experiments, CGRP (1–100 nM) produced a concentration-dependent (EC50 8 nM) partial inhibition (Emax about 35% inhibition of motility index) of spontaneous contractions. The potassium (K) channel opener, cromakalim (3–10 M) promptly suppressed the spontaneous contractions in a glibenclamide- (10 M) sensitive manner. Glibenclamide (10 M) did not affect the inhibitory action of CGRP. The calcium (Ca) channel agonist, Bay K 8644 (1 M), markedly enhanced the spontaneous activity of the renal pelvis and reduced the inhibitory effect of CGRP. The protein kinase A inhibitors Rp-cAMPS (300 M), H8 (100 M) and H89 (10 M), and the blockers of intracellular Ca handling bysarcoplasmic reticulum, ryanodine (100 M) and thapsigargin (1 M) did not affect the response to CGRP. The response to CGRP was likewise unaffected by the nitric oxide synthase inhibitor, L-nitroarginine (30 M) and by the protein kinase G inhibitor, KT5823 (3 M). Furthermore, the inhibitory action of CGRP was not modified by lowering the extracellular concentration of K (from 5.9 to 1.2 mM) nor by increasing (from 2.5 to 3.75 mM) or decreasing (from 2.5 to 0.25 mM) the extracellular Ca concentration. Replacement of 80% glucose with 2-deoxyglucose (2-DOG) reduced the amplitude of spontaneous contractions, both in the absence and presence of 10 M glibenclamide. In the presence of 2-DOG, the inhibitory action of CGRP was enhanced at a similar extent, either in the absence or presence of glibenclamide. In sucrose gap, the effect of CGRP (0.1 M for 5 min) was separately analyzed in the proximal (close to the kidney) and distal (close to the ureter) regions of the renal pelvis. Both preparations discharged spontaneous (pacemaker) action potentials having different shape, duration and frequency. CGRP had no effect on pacemaker potentials in the proximal renal pelvis while producing about 30% reduction of the frequency of pacemaker potentials and motility index in the distal renal pelvis. Cromakalim (3 M) abolished pacemaker potentials in both regions of the renal pelvis.In conjunction with the results of previous studies in the guinea-pig ureter, the present findings document the existence of remarkable regional differences in the effector mechanisms initiated by CGRP receptor occupancy in the guinea-pig pyeloureteral tract. CGRP appears to be inherently unable to activate glibenclamide-sensitive K channels in the guinea-pig renal pelvis, a mechanism which is central for its ability to suppress latent pacemakers in the ureter. Within the renal pelvis, the sensitivity to the inhibitory effect of CGRP appears in the more distal region, from which an ureter-like action potential is recorded.  相似文献

The ATP-induced increase in tritium outflow from cultured chick sympathetic neurons prelabelled with [3H]-noradrenaline was investigated.Seven days-old dissociated cell cultures of embryonic paravertebral ganglia, loaded with [3H]-noradrenaline (0.05 M), were superfused in the presence of (+)-oxaprotiline and exposed to ATP, ATP-analogues, or 1,1-dimethyl-4-piperazinium (DMPP) for 2 min. ATP (3 LM-3 mM), 2-methylthio-ATP (3–100 M), as well as DMPP (10 and 100 M) induced a significant overflow of tritium. The EC50-value of ATP was 20 M. Both the ATP-induced and the DMPP-induced tritium overflow was Ca2+-dependent and sensitive to tetrodotoxin (0.3 M) and -conotoxin (0.1 M); in addition, it was inhibited by the 2-adrenoceptor agonist 5-bromo-6-(2-imidazoline-2-ylamino)-quinoxaline (UK-14,304; 1 M). The effects of ATP and DMPP were not additive. The ATP-induced as well as the DMPP-induced overflow of tritium was diminished by the P2-purinoceptor antagonists suramin (300 M) and reactive blue 2 (3 M); in all 4 cases, the inhibition amouted to approximately 40%. The tritium overflow induced by ATP or DMPP was almost abolished by the nicotinic receptor antagonist mecamylamine (10 M) and markedly inhibited by hexamethonium (100 M). Neither ATP nor electrical stimulation caused an overflow of tritium from cultures loaded with [3H]-choline.The results suggest that ATP at molar concentrations induces noradrenaline release from cultured chick sympathetic neurons via an action on a subclass of the nicotinic cholinoceptor.  相似文献

The frequency of spontaneous action potentials of locus coeruleus (LC) neurons was recorded extracellularly in pontine slices of the rat brain. Ethanol (1–100 mM) elevated the firing rate in most neurons; this effect was concentration-dependent. (S)--amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA; 0.03–1 M), kainate ( 0.1–3 M), N-methyl-D-aspartate (NMDA; 1–30 M), substance P 0.01–1 M, nicotine 0.1–10 M and ,-methylene ATP ,-meATP; 0.3–30 M, all increased the firing. Application of ethanol g10–100 mM to the superfusion medium for 10 min, reproducibly and concentration-dependently inhibited the facilitatory effect of NMDA g10 M. However, the inhibitory effect of ethanol (100 mM) decreased during a 30-min superfusion period and after the washout of ethanol the sensitivity of LC neurons to NMDA (10 M) tended to overshoot above their initial level. Although NMDA was more potent in the absence than in the presence of external Mg2+, ethanol (100 mM) continued to depress the facilitatory effect of a low concentration of NMDA (EM) in a Mg2+-free medium. By contrast, in a medium containing normal Mg2+, ethanol (100 mM) failed to significantly interfere with the increase in firing rate induced by a high concentration of NMDA (30 M). The effects of kainate (0.5 M), AMPA (0.3 M) and nicotine (1 M) were also depressed by ethanol (100 M), while the effects of substance P (0.03 M) and ,-meATP (30 M) were not changed. In conclusion, ethanol selectively counteracts the opening of cationic channels caused by excitatory amino acid (EAA) receptor agonists and nicotinic acetylcholine receptor agonists. During a longer lasting incubation with ethanol, the inhibition of the NMDA-induced excitatory effect declines, indicating the development of tolerance. Correspondence to: P. TMB-8 has been characterized as an inhibitor of the release of Ca+ from intracellular pools. We have studied the modification of the pressor responses to selective l-adrenoceptor agonists (methoxamine and phenylephrine), and to selective 2-adrenoceptor agonists (B-HT 920 and B-HT 933) in pithed rats, produced by TMB-8. We have compared this modification with that produced by the calcium antagonist nifedipine. Nifedipine (100 g/kg, 300 g/kg, and 1000 g/kg) inhibited in a dose-dependent manner the pressor responses to the 1- and 2-adrenoceptor agonists, the dose-response curves to the 2-adrenoceptor agonists being shifted further to the right. TMB-8 at a dose of 3000 g/kg did not modify the pressor effects of the l-adrenoceptor agonists, and neither did it reinforce the inhibition of such responses produced by nifedipine. By contrast, TMB-8 pretreatment (0.03 g/kg, 0.3 g/kg, 3 g/kg, 30 g/kg, 300 g/kg and 3000 g/kg) inhibited the responses to both 2-adrenoceptor agonists, the inhibition being more pronounced with B-HT 920. A similar effect was obtained with 0.03 g/kg TMB-8 and 0.3 g/kg TMB-8, particularly in the case of B-HT 920. It was stronger with higher doses, but similar for all doses over 3 g/kg. The inhibition of the pressor responses mediated by the stimulation of 2-adrenoceptors by TMB-8 was less in rats treated with the Ca2+ entry promoter BAY K 8644 (300 g/kg), and could also be reduced by the continuous infusion of CaCl2 (0.25 g/min). These results suggest that in pithed rats TMB-8 may also behave as an inhibitor of the Ca+ influx into vascular smooth muscle.  相似文献
