The effect of safflower phospholipid (SP) on plasma and liver lipids in rats fed a hypercholesterolemic diet was examined. Triglyceride mixture (SPO) of safflower oil and palm oil (8:2) containing almost comparable amounts of linoleic acid to safflower phospholipid was used as a control diet. Similarly, the effect of paste safflower phospholipid (PSP) which contains approximately 45% of neutral lipid was also compared to safflower oil (SO). Concentrations of total cholesterol in plasma and liver of rats fed the SP diet were markedly decreased in comparison with those of the other diets, but a slight reduction of total cholesterol in plasma and liver was observed in rats fed PSP diet. SP and PSP induced a reduction in the plasma level of low density lipoprotein (LDL) cholesterol as well as an increase in the level of high density lipoprotein (HDL) cholesterol. The activity of plasma lecithin-cholesterol acyltransferase (LCAT) was greatly increased in rats fed SP diet. These results suggest that the safflower phospholipids suppress the elevation of plasma and liver cholesterol and that this effect may depend on the phospholipid content in dietary lipid. 相似文献

The effect of dietary safflower phospholipid (Saf-PL) on the postprandial changes of steroids in the small intestinal and cecal contents was examined in rats fed a hypercholesterolemic diet. The triglyceride mixture (SP-Oil) containing a comparable amount of linoleic acid to Saf-PL was used as a reference fat source. Saf-PL suppressed the elevation of plasma cholesterol levels at all times after meal intake, when compared to SP-Oil. The reduction of plasma cholesterol in rats fed the Saf-PL diet was exclusively observed both in chylomicron plus very low density lipoprotein (VLDL) and low density lipoprotein (LDL) fractions. The rate of gastric emptying was not modified by the Saf-PL diet. The level of neutral steroids in the small intestinal contents was almost comparable in both groups, but in the cecal contents and feces it was significantly higher in rats fed the Saf-PL diet. On the other hand, the level of acidic steroids in the small intestinal contents tended to be higher in rats fed the Saf-PL diet than in those fed the SP-Oil diet, whereas in the cecal contents and feces it was comparable in the two diets. These results suggest that Saf-PL causes the accumulation of neutral steroids in the cecum due to the rapid transit through the small intestine. 相似文献

Recent data concerning the effect of soybean 7S globulin subunits on the upregulation of LDL receptors in Hep G2 cells identified the alpha' subunit as the candidate responsible for this biological effect. In vivo evaluation of this subunit on cholesterol homeostasis was hampered by the lack of suitable amounts of alpha' chain. A novel separation procedure allowed us to investigate the effects of alpha' subunit administration on plasma cholesterol and triglyceride levels, as well as on the activity of liver beta-VLDL receptors of rats fed a hypercholesterolemic (HC) diet. Rats were divided into 9 groups fed the following diets for 28 d: standard diet; HC diet; HC diets + 5, 10, and 20 mg/(kg body weight. d) of alpha' subunit; HC diets + 50, 100, and 200 mg/(kg body weight. d) of soybean 7S globulin; HC diet + 200 mg/(kg body weight. d) clofibrate. The highest dose of the alpha' subunit decreased plasma cholesterol and triglycerides 36 and 34%, respectively, in rats fed the HC diet; 10-fold amounts clofibrate reduced plasma cholesterol and triglycerides 38 and 41%. The activity of liver beta-VLDL receptors of rats fed the HC diet with the highest dose of the alpha' subunit had a 96% increase in binding compared with the HC diet group, thus restoring the receptor activity to that of rats fed the standard diet. These results represent the first in vivo evidence of both the plasma lipid-lowering properties and the upregulation of liver beta-VLDL receptors induced by the soybean alpha' subunit. 相似文献

Taurine supplementation has been shown to have an effect on lowering blood lipids in ovariectomized (OVX) rats. It therefore seemed desirable to find out whether the beneficial effect of taurine on OVX rats fed calcium-deficient diet could also be reproduced. Forty female Sprague-Dawley rats were divided into two groups. One group was OVX and the other group received a sham operation (Sham). Each rat group was further divided into the control diet and the taurine supplemented (2.0 g/100 g diet) diet group. All rats were fed on calcium-deficient diet and deionized water ad libitum for 6 weeks. Plasma and liver lipids were determined by using commercial kits. LDL-cholesterol concentrations were estimated with the equation of Friedewald et al. (1972). There were no significant differences in body weight gain and food intake between the control and taurine group within Sham and OVX groups, but body weight gain, food intake, and food efficiency ratio was higher in the OVX group. Concentrations of plasma total cholesterol, triglyceride, LDL-cholesterol were significantly lower in the taurine fed group of OVX rats fed Ca deficient diet, while HDL-cholesterol concentration was increased in the taurine fed group. Therefore, in this study, we examined whether taurine also prevented hypercholesterolemia induced by ovarian hormone deficiency in ovariectomized rats when they were fed a calcium-deficient diet. These results indicate that taurine may have some beneficial effects on hypercholesterolemia and hypertriglyceridemia in OVX rats fed calcium-deficient diet. 相似文献

The effects of dietary fats , consisting of different fatty acids, on body fat accumulation and uncoupling protein (UCP) in interscapular brown adipose tissue were studied in rats. Metabolisable energy in experimental diets based on safflower oil, soybean oil or beef tallow was measured strictly (experiment 1). Male Wistar rats were then meal-fed an isoenergetic diet for 8 weeks (experiment 2). Each group of rats showed the same weight gain during the 8-week experimental period. Carcass fat content was greater in rats fed the beef tallow diet than in those fed the with the safflower or soybean oil diets, whereas the weight of abdominal adipose tissue was the same for all three dietary groups. Gene expression of UCP1 and the UCP content of the interscapular brown adipose tissue was lower in the beef tallow diet group than in the other dietary groups. A negative correlation was observed between carcass fat content and n-6 unsaturated fatty acid content in dietary fats. These results suggest that the greater body fat accumulation in rats fed the beef tallow diet results from lower expression of UCP1 mRNA and lower UCP content in brown adipose tissue. n-6 Polyunsaturated fatty acids may be the most effective fatty acids in limiting body fat. 相似文献

The effects of diets containing fats and oils or fatty acids on the lipid metabolism were investigated in male rats of the Wistar strain fed hypercholesterolemic diets, especially focusing our attention on the correlation between dietary oleic acid (OLE) contents and the levels of plasma and liver total cholesterol (T-CHOL) or the fatty acid profiles in plasma and liver CHOL-ester. In the rats fed the free (FR)-type fatty acids, the concentrations of plasma and liver T-CHOL were high and the amounts of neutral steroids excreted into the feces were low when compared with those of rats given the triacylglycerol (TG)-type fatty acids, showing that TG-type fatty acids suppress the intestinal CHOL absorption more than the FR-type fatty acids do. The concentrations of plasma T-CHOL were highest in rats fed the oleic acid (OLE)-rich diets, followed in order by rats supplied with the palmitic acid (PAL)-rich diets, the hydrogenated coconut oil (HCO) diet, and the linoleic acid (LIN)-rich diets; the lowest was in rats given tristearin (TSTE) and linseed oil (LIS) diets. A positive correlation was obtained between the dietary OLE contents and the levels of plasma and liver T-CHOL or OLE in the plasma and liver CHOL-ester, and an inverse correlation between dietary OLE contents and the amounts of excreted neutral steroids. These results suggest that the dietary OLE contents regulate the levels of plasma and liver T-CHOL in CHOL-loaded rats. 相似文献

The effects of feeding olive and safflower oils on lipid and fatty acid composition of mammary tumors, plasma lipids and lipoproteins, and the nuclear magnetic resonance (NMR) spectra of plasma were investigated in rats. 7-12-Dimethylbenz[a]anthracene (DMBA)- and placebo-intubated male Sprague-Dawley rats were fed 20% fat diets containing 18:2n-6 (wt/wt) from either high-linoleic safflower oil (SL, 14.6% 18:2n-6), high oleic safflower oil (SO, 3.4% 18:2n-6), olive oil (OO, 1.1% 18:2n-6), or olive oil supplemented with 18:2n-6 (OL, 3.4% 18:2n-6) for 16 weeks. Our result indicated that tumor composition of 18:1n-9 and 18:2n-6 reflected the diet, but tumor neutral lipid (NL) was more reflective of diet than was tumor phospholipid (PL). The 20:4n-6 content of tumor PL was constant in all of the dietary groups despite threefold higher levels of 18:2n-6 in tumor PL from animals fed SL than from those fed SO, OO, or OL diets. This suggests a possible feedback inhibition of delta 6-desaturase by the higher content of 18:2n-6 associated with SL feeding No diet effects were obtained for tumor total lipid, NL, PL, cholesterol, and triglyceride contents. Plasma lipoprotein changes were more reflective of diet than tumorigenesis except for apolipoprotein-E, which was lower, and for very low-density lipoprotein cholesterol and low-density lipo protein, which were higher in tumor-bearing rats. Plasma NMR analysis indicated no difference in the average line widths of the methyl and methylene resonances for tumor-bearing and nontumor-bearing rats fed any of the diets. 相似文献

The effects on body fat accumulation of long-term feeding of high fat diets of differing fatty acid composition were studied in rats. The rats were meal-fed isoenergetic diets based on safflower oil or beef tallow for 4 mo. Each diet was freshly prepared every day throughout the experimental period. Oxygen consumption and carbon dioxide production for 6 h after meals were measured between the 50th and 54th d of the experimental period. Oxygen consumption for 3 h after meals was significantly greater in the safflower oil diet group than in the beef tallow diet group, indicating greater diet-induced thermogenesis in the former group. From the assessment of respiratory quotient, the fat oxidation rate was also higher in the former. After the experimental period (4 mo), body fat accumulation was significantly less in the rats fed safflower oil. This difference was, at least in part, ascribed to increased diet-induced thermogenesis and fat oxidation. Serum triacylglycerol level was markedly lower in the rats fed safflower oil than in those fed beef tallow. The lipoprotein lipase activities in heart and soleus muscle after meals appeared to be higher in the former than in the latter. These results suggest that the consumption of the safflower oil diet increased lipoprotein lipase activity in heart and skeletal muscle, resulting in the elevation of fat oxidation rate and the depression of serum triacylglycerol level. 相似文献

Six polyphenolic compounds were isolated from ethylacetate extract secondary to 80% ethanol extraction of defatted safflower seeds. They were categorized into three types: lignans, flavones and serotonin derivatives. Female Sprague-Dawley rats weighing 163.4 +/- 6.3 g were ovariectomized (Ovx) and fed either ethylacetate extract at a level of 1% (w/w) or three types of safflower polyphenolic compounds at a level of 200 mg/kg in a diet containing 0.5% (w/w) cholesterol for four wk. The sham and Ovx control groups were fed the same diet without safflower components. Plasma GOT and GPT levels did not differ among the six experimental groups. The plasma levels of total cholesterol were reduced in the four safflower groups by 20-30% as compared to the Ovx control. The plasma level of HDL-cholesterol was higher in the Ovx+ethylacetate extract group or appeared to be in the three Ovx+safflower polyphenolic groups than in the Ovx control. The level of plasma triglyceride was also significantly lower in the Ovx+lignan group than in the Ovx control. The liver level of cholesterol was significantly reduced in the Ovx+ethylacetate extract group. Fecal excretion of cholesterol increased by the safflower lignans and flavones, whereas that of bile acid was not significantly changed by the safflower polyphenols. Matairesinol and acacetin isolated from safflower seeds reduced the cholesterol content in cultured HepG2 cells at a concentration of 0.01-0.1 microM and all three safflower polyphenolics decreased triglyceride content at the concentration of 0.1 microM. These results suggest that safflower polyphenols have the effect of improving blood lipid status via increasing HDL-cholesterol formation and cholesterol excretion without significant uterotropic action in estrogen-deficient animals. 相似文献

The present study compared the effects of dietary diacylglycerol (DG) and triacylglycerol (TG) oil on lipid metabolism in rats fed a 5% fat (AIN-76) diet for 6 weeks. The plasma and hepatic lipids, hepatic cholesterol-regulating enzyme activity, and hepatic and adipose tissue fatty acid metabolism enzyme activities were determined. Among plasma lipids, triglyceride, free fatty acid, and phospholipid concentrations were significantly lower in the DG group than in the TG group. A lower plasma TG level was accompanied by an increase in adipocyte lipoprotein lipase activity. The hepatic triglyceride level was significantly (P < .001) lowered in the DG group, which was attributable to an increased fatty acid oxidation enzyme (beta-oxidation) activity and a reduced fatty acid synthesis enzyme (glucose-6-phosphate dehydrogenase) activity. The plasma cholesterol concentration was significantly lower in the DG group and was accompanied by a lower hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activity. The DG oil used in this study was beneficial for enhancing lipid metabolism with apparent hypolipidemic effects. 相似文献

目的 观察大豆异黄酮 (SI)对进食高脂饲料的去卵巢大鼠脂质和肝组织过氧化状态的影响。方法 50只SD大鼠随机分为 5组 ,分别接受假手术 +基础饲料 ;去卵巢 +高脂饲料 ;去卵巢 +高脂饲料 +SI 3个剂量组 2 0 ,60 ,180mg/ (kg·bw·d)处理 ,实验期 8周。 结果 与模型组比较 ,SI可显著抑制高脂饲料所致的去卵巢大鼠肝脏和血清甘油三酯水平升高 (P <0 .0 5) ,但对肝脏总胆固醇变化无影响 (P >0 0 5)。与假手术组和模型组比较 ,SI降低肝脏MDA含量均有显著性差异 (P <0 0 1)。结论 SI对去卵巢大鼠肝脏高甘油三酯有降低作用 ,并能改善高脂饮食所致的肝脏过氧化状态异常 ,减轻对机体的过氧化损伤 相似文献

Diets low in carotenoids have been associated with greater risks of developing certain cancers. Diets high in fats, especially unsaturated fats, have also been associated with increased risks of developing cancer. This study was undertaken to evaluate the effect of fat type on the beta-carotene status in a rat model. Rats were fed diets containing 2 g/kg beta-carotene or placebo and either 10% lard or corn oil for 13-16 weeks. Plasma vitamin A was unaffected by the dietary regimes. Liver vitamin A levels were significantly increased in the beta-carotene groups and were greater in the lard-fed animals. No detectable level of plasma beta-carotene was found in the rats fed placebo diets. In animals given dietary beta-carotene, plasma beta-carotene levels were significantly reduced in the group fed corn oil compared to the group fed lard. Liver beta-carotene levels were higher in the groups fed corn oil. The results suggest that the type of dietary fat can significantly affect the circulating and tissue levels of beta-carotene. 相似文献

The effect of dietary phosphatidylethanolamine (PE) and phosphatidylcholine (PC) added to cholesterol-free semipurified diet on serum lipoprotein and hepatic and fecal lipids was compared to the effect on rats fed soybean oil (controls). The dietary PE, but not PC, caused a decrease in serum cholesterol, phospholipid, apolipoprotein A-I (apoA-I) and apoE and an increase in high molecular weight apoB. The simultaneous addition of PC and ethanolamine also decreased serum apoA-I and cholesterol. The distribution patterns of phospholipid subclasses in the liver and fatty acid composition of hepatic and plasma phospholipids were also altered by dietary PE. Both PE and PC increased to a similar extent the excretion of fecal neutral steroids and hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activity compared to the controls. The present study, therefore, demonstrated for the first time that the PE in the soybean phospholipid preparation is responsible for the alterations of profiles of serum lipids and apoproteins in rats. 相似文献

目的 观察复合膳食纤维(DFC)对高脂血症大鼠肝脏结构及功能的长期影响。方法 健康成年SD大鼠84只,雌雄各半。经高脂饲料诱导形成高脂血症模型后,按体重及血胆固醇水平均衡的原则分为7组,其中2组分别以基础饲料及小麦纤维作为正常和高脂对照组,其余5组给不同剂量的DFC,实验期为3个月。通过生化分析及形态观察的方法,研究DFC对大鼠肝脏结构与功能的影响。结果(1)实验第60d时,DFC含量最高的G组(64%DFC)的血浆ALT活性显高于正常和高脂对照组,其余各DFC组的血浆ALT活性、各DFC组AST活性与正常及高脂对照组比较均无显性差异;(2)实验第90d时,较高剂量的3组(分别含DFCl6%,32%和64%)的血浆ALT、AST活性显高于正常对照组,其中最高剂量组的血浆ALT活性显高于高脂对照组,而较低剂量的2组(DFC含量分别为4%,8%)的血浆ALT、AST活性在实验各个阶段与对照组比较均无显性差异;(3)各DFC组的血浆ALT及AST活性在实验第60d及90d时均随DFC含量的增加而增高,呈现明显的剂量—反应关系;(4)各剂量DFC组肝细胞脂肪变性不明显,但呈现不同程度的气球样变,较高剂量组(32%)肝细胞可见到嗜酸性变。结论 长期摄入高剂量DFC会导致高脂血症大鼠肝脏形态结构和功能的改变,因此实际应用时应该以低剂量为宜。 相似文献

The effect of dietary supplementation of vitamin E on serum cholsterol, triglycerides, lipoproteins and lipoprotein composition was studied in rabbits fed a one per cent cholesterol diet for a period of twelve weeks. Vitamin E supplemented animals were found to maintain significantly lower concentrations of serum cholesterol, triglycerides and very low density lipoproteins (d less than 1.006). The difference in the serum lipid content was mainly in the very low density lipoproteins of d less than 1.006. The possibility of an increased clearance of chylomicron remnants and decreased inhibition of extrahepatic lipoprotein lipase in hypercholesterolemic rabbits as a result of vitamin E supplementation is discussed in the light of the results obtained. 相似文献

The alleviation mechanism of methionine toxicity by dietary glycine was investigated in weanling rats fed a high-methionine diet. When rats were fed a 10% casein diet containing 2% methionine, the activities of methionine adenosyltransferase, cystathionine beta-synthase, and cystathionine gamma-lyase, which participate in the methionine metabolism in the transsulfuration pathway, were significantly enhanced. But the addition of 2% glycine to the high methionine diet did not cause further increase in these enzyme activities; the activities of methionine adenosyltransferase and cystathionine beta-synthase were rather decreased while cystathionine gamma-lyase activity was not altered. Methionine transaminase activity was essentially insensitive to the dietary addition of methionine and glycine. In rats fed a high methionine diet, the hepatic methionine level was significantly increased with a concomitant decrease in the levels of glycine, serine, and threonine. The addition of glycine to the high methionine diet effectively suppressed the enhancement of the hepatic methionine level and almost completely restored the glycine level, but it only partially restored the serine level and further decreased the threonine level. From these results, it is suggested that the alleviating effect of dietary glycine on methionine toxicity is primarily elicited by the restoration of the hepatic glycine level rather than by an increase in hepatic enzyme activity. 相似文献

In order to evaluate a) the respective roles of adipose and muscle lipoprotein lipase (LPL) in the clearing of alimentary lipemia and b) the role of the resulting nonesterified fatty acids (NEFA) in controlling hepatic ketogenesis and liver triglyceride content, a number of parameters related to lipid metabolism were studied over the 24 hour period (the dark period being from 1930 to 0730 hours), in rats ad libitum fed either a low-fat (LF) or a high-fat (HF) diet containing respectively 1.1% and 41.5% lard. During spontaneous feeding (from 1500-1800 hours onwards), LPL activity in LF rats increased in adipose tissue and decreased in heart; in rats fed the HF diet for 3 weeks, the postprandial rise in adipose LPL was smaller and there was no decrease in heart LPL before 2100 hours. In HF rats, unlike the LF, feeding resulted in a large increase in circulating NEFA and total ketone concentrations, an increase in liver beta-hydroxybutyrate dehydrogenase activity, and a decrease in hepatic triglyceride content. The findings clearly indicate that in HF rats, muscle LPL controls the postprandial rise in plasma NEFA concentrations, which in turn appear to determine the extent of ketonemia and liver triglyceride changes. The possible control of these metabolic events by insulin is discussed. 相似文献

OBJECTIVE: This study investigated the effects of soymilk on lipid metabolism in Sprague-Dawley rats fed a cholesterol-enriched (0.3%) diet. METHODS: Thirty male Sprague-Dawley rats weighing 230.0 +/- 9.8 g were randomly assigned to one of three groups: control, S1 (containing 15% soymilk powder in the diet), and S2 (22.5%). After 8 wk, lipid profiles of the plasma, liver, and feces were determined. RESULTS: Body weight gain, daily food intake, and feeding efficiency showed no differences across groups (P > 0.05). The experimental groups had significantly lower plasma levels of cholesterol, triacylglycerol, and low-density lipoprotein cholesterol than the control group (P < 0.05) at weeks 4 and 8. However, total fecal excretion of neutral steroid did not significantly differ across groups (P > 0.05). CONCLUSION: Soymilk affects the metabolism of plasma cholesterol in Sprague-Dawley rats. 相似文献
