目的:研究RECK,MMP-9和TGF-β1在胃癌及正常胃组织中的表达,探讨其在胃癌的发生、发展、浸润和转移中的作用.方法:选择临床及病理资料齐全的存档胃腺癌蜡块标本54例,另取正常胃黏膜标本15例作对照.采用第二代通用型二步法监测系统(PV-9000)免疫组化方法检测RECK,MMP-9及TGF-β1在胃癌及正常胃组织中的表达.结果:RECK在胃癌组织中低表达(51.9%),并随肿瘤浸润深度的加深、淋巴转移的产生、远隔转移的发生、临床分期的提高、肿瘤病理分化程度的降低而降低(P<0.05).MMP-9在胃癌组织中高表达(75.9%),并随肿瘤浸润深度的加深、淋巴转移的产生、临床分期的提高、肿瘤病理分化程度的降低而增高(P<0.05).TGF-β1在胃癌组织中高表达(77.8%),并随淋巴转移的产生、临床分期的提高、肿瘤病理分化程度的降低而增高(P<0.05).胃癌组织中RECK与MMP-9、TGF-β1的表达呈负相关(r=-0.618,P<0.001;r=-0.620,P<0.001),MMP-9与TGF-β1的表达呈正相关(r=0.716,P<0.001).结论:胃癌组织中RECK低表达,MMP-9,TGF-β1高表达.RECK,MMP-9及TGF-β1可以作为评估胃癌发生浸润转移的重要指标.  相似文献

目的:探讨胃癌中MMP-9和TIMP-1蛋白表达与ZHX2蛋白表达的关系,并分析其与胃癌患者临床病理特征的相关性及其临床意义.方法:应用免疫组织化学Envision二步法检测62例胃癌组织及癌旁正常胃组织中MMP-9、TIMP-1及ZHX2蛋白的表达.用SPSS13.0统计软件分析MMP-9和TIMP-1蛋白表达与ZHX2蛋白表达的相关性,并分析其与患者的临床病理特征的关系.结果:MMP-9蛋白在62例胃癌组织及癌旁正常胃组织中阳性表达率分别为66.13%(41/62)和29.03%(18/62)(P<0.01),TIMP-1蛋白在胃癌组织及癌旁正常胃组织中阳性表达率分别为41.94%(26/62)和74.19%(46/62)(P<0.01).MMP-9蛋白表达在ZHX2阳性和阴性表达组中的阳性率分别为:84.44%(38/45)和17.65%(3/17)(P<0.05),与患者的年龄、性别、肿瘤大小、发生部位、大体类型、分化程度、淋巴结转移、远处转移、TNM分期均无关(P>0.05),而与肿瘤的浸润深度有关(P<0.05);TIMP-1蛋白表达在ZHX2阳性和阴性组中阳性率分别为:28.89%(13/45)和76.47%(13/17)(P<0.05),与患者的年龄、性别、肿瘤大小、发生部位、大体类型、分化程度、浸润深度、淋巴结转移、远处转移、TNM分期均无关(P>0.05).结论:MMP-9高表达可以作为评估胃癌恶性生物学行为及预后的指标,MMP-9/TIMP-1表达失衡与ZHX2表达相关,MMP-9和TIMP-1可为临床选择化疗药物提供参考依据.  相似文献

目的研究信号转导与转录活化因子(STAT3)和增殖核抗原(PCNA)在胃癌中的表达情况及其在胃癌发生发展中的作用。方法采用免疫组化二步法检测40例胃癌组织和40例正常组织中STAT3与PCNA的表达。结果胃癌组织中STAT3的阳性表达率为65.0%,明显高于正常组织的27.5%。PCNA的阳性表达率为57.6%,明显高于正常组织的25.0%。STAT3的表达与临床分期、淋巴结转移和分化程度相关(P0.05),与患者年龄、性别无关(P0.05),并且与PCNA的表达呈正相关。结论 STAT3的异常活化可促进细胞的过度增殖,而PCNA则反映了癌细胞的增殖状态。二者结合检测对胃癌的临床诊断及治疗有一定的价值。  相似文献

目的 探讨Survivin和Ki-67在胃癌组织中的表达及其与临床病理特征的关系。方法 采用免疫组织化学技术(S-P)法检测各期胃癌共50例及12例浅表性胃炎组织中Survivin和Ki-67的表达水平,并结合病理特征进行分析。结果 胃癌组织中Survivin阳性表达率为74%,显著高于浅表性胃炎组(P<0.001)。Survivin过表达与肿瘤浸润深度、淋巴结及远处转移、临床分期相关(P<0.05),而与胃癌组织分化程度无关(P>0.05)。Ki-67抗原阳性表达率在胃癌组织中为52%,显著高于浅表性胃炎组(P<0.01),并与胃癌组织分化程度、肿瘤浸润深度、淋巴结及远处转移、临床分期相关(P<0.05)。两种蛋白表达有关联(P<0.01)。结论 Survivin和Ki-67在胃癌组织中过表达,二者过表达与胃癌的发生、发展密切相关,可作为预后不良的指标。  相似文献

目的探讨低氧诱导因子(HIF)-1α在胃癌组织及转移淋巴结中的表达情况及在中老年胃癌早期发生、发展、转移及预后中的作用及意义。方法对临床未进行化疗的60例胃癌患者切除的胃癌组织及转移淋巴结进行苏木素-伊红(HE)染色确定标本的组织分化程度,采用免疫组织化学方法检测HIF-1α在胃癌组织及转移淋巴结中的表达情况。结果 HIF-1α在胃癌组织中的阳性率为71.67%(43/60),转移淋巴结中的阳性率为86.7%(52/60),而正常胃组织仅有2列表达弱阳性,正常淋巴结中均未有阳性率表达,有显著差异(P<0.05),其中高分化阳性率为55%(11/20),中分化为65%(13/20),低分化为95.0%(19/20),差异有统计学意义(P<0.05)。结论 HIF-1α在胃癌中呈高表达,与胃癌新生血管形成、癌组织浸润、转移有密不可分的关系。  相似文献

目的观察胃癌组织中NPRL2 mRNA的表达变化,并探讨其临床意义。方法采用逆转录聚合酶链反应(RT-PCR)法检测60例胃腺癌组织及其癌旁正常组织中NPRL2 mRNA的表达。结果 NPRL2 mRNA在胃癌及癌旁正常组织中均有表达,但胃癌组织中NPRL2 mRNA的表达显著降低(IOD比值为1.099±0.285),与癌旁正常组织(IOD比值为1.582±0.305)相比P<0.05。NPRL2 mRNA的表达与肿瘤分化程度、有无淋巴结转移、肿瘤浸润深度、临床分期及患者的年龄、性别无显著相关(P均>0.05)。NPRL2 mRNA在不同分化程度、不同临床分期、有无淋巴结转移及不同年龄、性别患者的胃癌组织中的表达无显著差异(P均>0.05)。结论胃癌组织中NPRL2mRNA表达降低,其在胃癌的发生、发展过程中可能发挥重要作用。  相似文献

目的探讨胃癌组织肿瘤坏死因子受体相关因子3(TRAF3)、肿瘤坏死因子受体相关因子6(TRAF6)阳性表达与肿瘤浸润、转移的关系。方法选择胃癌组织及配对癌旁正常组织标本各87份。制作组织芯片,采用免疫组化微阵列技术检测TRAF3、TRAF6表达,分析TRAF3、TRAF6阳性表达与胃癌浸润、转移的关系。结果胃癌组织TRAF3阳性表达率明显低于癌旁正常组织(P<0.01),TRAF6阳性表达高于癌旁正常组织(P<0.05)。TRAF3阳性表达与胃癌组织分化程度、TNM分期、淋巴结转移有关(P均<0.05),TRAF6阳性表达与胃癌组织分化程度、TNM分期有关(P均<0.05)。结论胃癌组织TRAF3阳性表达下调、TRAF6阳性表达上调,其表达变化可能参与胃癌的浸润及转移。  相似文献

目的研究核因子-κB(NF-κB)和β-catenin在胃癌及癌前病变中的表达及与胃癌临床病理特征的关系。方法应用免疫组化检测72例胃癌组织及12例正常胃组织中NF-κBp65与β-catenin表达。结果β-catenin和NF-κBp65在胃癌组织中的阳性表达率明显高于正常胃组织(P<0.05),二者在胃癌组织中的阳性表达率为65.3%和61.1%。β-catenin的表达与肿瘤病理分化程度、有无淋巴结转移、病理分型、浸润深度密切相关(P<0.05)。NF-κBp65的表达与肿瘤大小、有无淋巴结转移、病理分型、肿瘤浸润深度密切相关(P<0.05)。结论β-catenin和核因子-κB在胃癌组织表达有相关性,提示其与胃癌的发生、发展和转移有关,二者可作为判断胃癌预后的指标和化疗的靶向。  相似文献

目的研究再生基因蛋白(Reg)Ⅳ、骨桥蛋白(OPN)及表皮生长因子受体(EGFR)在胃腺癌中的表达及与其临床意义。方法应用S-P免疫组化技术检测65例胃腺癌及其相应的癌旁正常组织中RegⅣ、OPN、EGFR蛋白的表达。结果 65例胃腺癌组织中RegⅣ、OPN、EGFR蛋白的阳性表达率53.8%(35/65),63.1%(41/65),56.9%(37/65)均高于癌旁正常组织12.3%(8/65)、4.6%(3/65)、18.5%(12/65)(P<0.05)。RegⅣ蛋白的表达与胃癌分化程度相关有统计学意义(P<0.05),OPN蛋白的表达与浸润深度、肿瘤分化程度、临床分期相关有统计学意义(P<0.05),EGFR蛋白的表达与胃腺癌的浸润深度、淋巴结转移、临床分期相关有统计学意义(P<0.05)。RegⅣ、OPN与EGFR蛋白在胃腺癌组织中的表达呈正相关关系(P<0.05),RegⅣ与OPN蛋白在胃腺癌组织中的表达呈正相关关系(P<0.05)。结论 RegⅣ、OPN、EGFR蛋白均在胃癌组织中表达呈正相关,并在胃癌的发生发展发挥重要作用。  相似文献

目的讨论整合素α2β1和CD44v4在胃癌组织中的表达及意义。方法采用免疫组化SP法检测85例胃癌标本和50例癌旁正常胃组织中整合素α2β1和CD44v4的表达情况,分析二者的表达与胃癌生物学行为以及它们之间的相关性。结果①整合素α2β1与CD44v4在胃癌标本中的阳性表达率均高于癌旁正常组织(P<0.05)。②整合素α2β1和CD44v4在胃癌中的表达与胃癌的分化程度、TNM分期、淋巴结转移和浸润深度有关(P<0.05)。③整合素α2β1和CD44v4在胃癌中的表达呈正相关(r=0.517,P<0.05)。结论整合素α2β1和CD44v4在胃癌中高表达且在胃癌的浸润、转移过程中起正协同作用,并且与胃癌的分化程度、TNM分期、淋巴结转移和浸润深度密切相关,与患者的年龄、性别以及病理组织学类型无关。  相似文献

AIM: To investigate the expression of COX-2 proteins in gastric mucosal lesions and to assess the relationship between COX-2 expression and type, pathologic stage, differentiation, or lymph node metastasis in gastric cancer and the relationship between COX-2 expression and H pylori infection in gastric mucosal lesions. METHODS: Thirty patients with gastric carcinoma underwent surgical resection. Samples were taken from tumor site and paracancerous tissues, and ABC immunohistochemical staining was used to detect the expression of COX-2 proteins. H pylori was determined by rapid urea test combined with pathological stating/14C urea breath test. RESULTS: The positive rate and staining intensity of mutant COX-2 gene expression in gastric cancer were significantly higher than those in paracancerous tissues (66.7% vs 26.7%) (P<0.01, P<0.001). There was a significant correlation between COX-2 and pathologic stage or lymph node metastasis type of gastric carcinoma (76.0% vs 20.0%, 79.2% vs 16.7%) (P<0.05). No correlation was found between COX-2 expression and type or grade of differentiation (P>0.05). COX-2 expression of intestinal metaplasia (IM) or dysplasia (DYS) with positive H pylori was significantly higher than that with negative H pylori (50.6% vs 18.1%, 60.0% vs 33.3%) (P<0.05). CONCLUSION: COX-2 overexpression was found in a large proportion of gastric cancer tissues compared with matched non-cancerous tissues and was significantly associated with advanced tumor stage and lymph node metastasis. Overexpression of COX-2 plays an important role in tumor progression of gastric cancer. COX-2 may also play a role in the early development/promotion of gastric carcinoma and is associated with H pylori infection.  相似文献

Expression and significance of CD44s, CD44v6, and nm23 mRNA in human cancer   总被引:18,自引:0,他引:18  
AIM: To investigate the relationship between the expression levels of nm23 mRNA, CD44s, and CD44v6,and oncogenesis, development and metastasis of human gastric adenocarcinoma, colorectal adenocarcinoma,intraductal carcinoma of breast, and lung cancer.METHODS: Using tissue microarray by immuhistochemical (IHC) staining and in situ hybri-dization (ISH), we examined the expression levels of nm23mRNA, CD44s, and CD44v6 in 62 specimens of human gastric adenocarcinoma and 62 specimens of colorectal adenocarcinoma; the expression of CD44s and CD44v6in 120 specimens of intraductal carcinoma of breast and 20 specimens of normal breast tissue; the expression of nm23 mRNA in 72 specimens of human lung cancer and 23 specimens of normal tissue adjacent to cancer.RESULTS: The expression of nm23 mRNA in the tissues of gastric and colorectal adenocarcinoma was not significantly different from that in the normal tissues adjacent to cancer (P>0.05), and was not associated with the invasion of tumor and the pathology grade of adenocarcinoma (P>0.05). However, the expression of nm23 mRNA was correlated negatively to the lymph node metastasis of gastric and colorectal adenocarcinoma (r = -0.49, P<0.01; r = -4.93, P<0.01). The expression of CD44s in the tissues of gastric and colorectal adenocarcinoma was significantly different from that in the normal tissues adjacent to cancer (P<0.05;P<0.01). CD44v6 was expressed in the tissues of gastric and colorectal adenocarcinoma only, the expression of CD44v6 was significantly associated with the lymph node metastasis, invasion and pathological grade of the tumor (r = 0.47, P<0.01; r = 5.04, P<0.01). CD44sand CD44v6 were expressed in intraductal carcinoma of breast, the expression of CD44s and CD44v6 was significantly associated with lymph node metastases and invasion (P<0.01). However, neither of them was expressed in the normal breast tissue. In addition, the expression of CD44v6 was closely related to the degree of cell differentiation of intraductal carcinoma of breast (x2= 5.68, P<0.05). The expressional level of nm23mRNA was closely related to the degree of cell differentiation (P<0.05) and lymph node metastasis (P<0.01), but the expression of nm23 gene was not related to sex, age, and type of histological classification (P>0.05).CONCLUSION: Patients with overexpression of CD44s and CD44v6 and low expression of nm23 mRNA have a higher lymph node metastatic rate and invasion. In addition, overexpression of CD44v6 is closely related to the degree of cell differentiation. Detection of the three genes is able to provide a reliable index to evaluate the invasion and metastasis of tumor cells.  相似文献

AIM: To investigate the expression and significance of G3BP and RhoC proteins in esophageal squamous carcinoma (ESC).
METHODS: The expression of G3BP and Rhoc proteins in 80 cases of ESC was detected by immunohistochemistry. The relationship was studied between the expression of the two proteins and tumor size, differentiation degree, TNM stage, lymph node metastasis and prognosis of ESC.
RESULTS: The positive expression rate of G3BP in ESC was 71.25%; and the rate in the lymph node metastasis group was significantly higher than that in the nonlymph node metastasis group (Z = -2.283, P = 0.022), but no relations were found between G3BP expression and tumor size, differentiation degree and TNM stage (P 〉 0.05). The group with G3BP positive expression had shorter survival time than the group with G3BP negative expression (P = 0.000). The positive expression rate of RhoC in ESC was 66.25%; and the rate in the lymph node metastasis group was significantly higher than that in the non-lymph node metastasis group (Z = -2.115, P 〈 0.05), but no relations were found between RhoC expression and tumor size, differentiation degree and TNM stage (P 〉 0.05). The RhoC positive expression group had a shorter survival time than the RhoC negative expression group (P 〈 0.001. The expression of G3BP protein correlated positively with the expression of RhoC in ESC tissues (rs = 0.656, P 〈 0.001).
CONCLUSION: The expression of G3BP and RhoC protein is closely related to the lymph node metastasis and survival in ESC patients. G3BP and RhoC proteins can be considered as predictors of prognosis in ESC patients.  相似文献

目的 探讨信号转导和转录激活因子5(STAT5)和WW结构域氧化还原酶(WWOX)在非小细胞肺癌(NSCLC)中的表达及相关性研究.方法 用免疫组化SP法检测40例手术切除的NSCLC组织和20例正常肺组织中STAT5和WWOX的蛋白表达.结果 STAT5蛋白在NSCLC中的表达明显高于正常肺组织,而WWOX在NSCLC中的表达明显低于正常肺组织(P<0.01);STAT5表达水平与NSCLC组织学类型有关,在腺癌组织中的表达明显高于鳞癌(P<0.01),WWOX的表达与淋巴结转移相关,有淋巴结转移的表达低于无淋巴结结转移(P<0.01);STAT5蛋白和WWOX蛋白在NSCLC组织中的表达呈负相关(P<0.05).结论 在NSCLC中存在STAT5的过度表达以及WWOX的失表达,STAT5表达水平与NSCLC组织学类型有关,而WWOX的表达与淋巴结转移相关.提示在NSCLC的发生机制中,WWOX可能通过抑制STAT5系统,促进细胞凋亡,起到抑癌作用.  相似文献

AIM: To evaluate the relationship between the expression of cell adhesion molecules (CAMs) and the biological behavior of gastric carcinoma. METHODS: Expression of syndecan-1, E-cadherin and integrin β3 were evaluated by immunohistochemical study in a total of 118 gastric carcinomas and 20 nontumor gastric mucosas. RESULTS: The expressions of syndecan-1 and E-cadherin were significantly lower in gastric carcinoma compared to non-tumor gastric mucosa, and the low expression rates were positively correlated to the tumor invasion depth, vessel invasion, lymph node metastasis and distant metastasis (P 〈 0.01 in all cases). However, the expression of integrin β3 was significantly higher in gastric carcinoma compared to non-tumor gastric mucosa, and the high expression rates were positively correlated to the tumor invasion depth, vessel invasion, lymph node metastasis and distant metastasis (P 〈 0.01 in all cases). In addition, the three protein expressions were correlated to the tumor growth pattern (P 〈 0.01, P 〈 0.01, and P 〈 0.05 respectively), but not correlated to tumor differentiation (P 〉 0.05, P 〉 0.05 and P 〉 0.05 respectively). Positive correlation was observed between the expressions of syndecan-1 and E-cadherin, but they which were negatively correlated to the expression of integrin β3 (P 〈 0.01 in all cases). Univariate analysis demonstrated that the mean survival time and 5-year survival rate were lower in the cases with low expressions of syndecan-1 and E-cadherin and high expression of integrin β3 (P 〈 0.01, in all cases). COX multivariate analysis showed that the expression level of syndecan-1 could be an independent prognostic index of gastric carcinoma (P 〈 0.01), whereas E-cadherin and integrin β3 could not be independent indexes (P 〉 0.05, P 〉 0.05 respectively).CONCLUSION: The low expression of syndecan-1 and E-cadherin and the high expression of integrin β3 are significantly correlated with the invasion and metastasis o  相似文献
