Objective To investigate the effects of hypervolemic hemodilution (HH) with hypertonic saline plus hetastarch solution 40 injectio on hepatic ischemia-reperfusion (I/R) injury in rats. Methods Thirty male Wistar rats weighing 300-350 g were randomly divided into 3 groups ( n = 10 each): group I sham operation (group S); group II I/R and group Ⅲ HH. Partial liver ischemia was produced by clamping hepatic portal vein and left arteria hepatica for 30 min with atraumatic mini-clamp, followed by 2 h of reperfusion in I/R group and HH group. In HH group the animals were infused hypertonic saline plus hetastarch solution 40 injectio 10 ml/kg through vena caudalis over 30 min and then hepatic I/R was performed IS min after the infusion.The animals were killed at 2 h of reperfusion. The left liver was removed and blood sample was taken from inferior caval vein for determination of (1) serum alanine amino transferase (ALT) and aspartate amino transferase (AST) activities; (2) superoxide dismutase ( SOD) activity and malondialdehyde ( MDA) content in the liver; ( 3 ) microscopic examination. Results The serum ALT and AST activities and MDA content in the liver were significantly higher, SOD activity in the liver significantly lower after hepatic I/R and pathological changes in the liver severer in group I/R and HH than in group S. The serum ALT and AST activities and MDA content in the liver were significantly lower, SOD activity in the liver significantly higher after hepatic I/R and pathological changes in the liver milder in group HH than in group I/R. Conclusion Hypervolemic hemodilution with hypertonic saline plus hetastarch solution 40 injectio can ameliorate hepatic I/R injury by decreasing oxygen free radical production in rats.  相似文献

目的 了解雌激素对大鼠肝移植缺血再灌注损伤是否具有保护作用及其可能的作用机制.方法 将大鼠分为三组:雄性组、雌性组以及给予外源性雌激素的雄性组,分别应用两袖套法实施肝移植手术.术后6 h麻醉后处死,取血及肝组织标本.观察外周血丙氨酸氨基转移酶(ALT)和谷氨酸氨基转移酶(AST)以及一氧化氮(NO)水平,免疫组化染色分析内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS)在各组肝组织中的表达.结果 与雄性组和雌性组比较给予外源雌激素的雄性大鼠血清ALT水平和AST水平显著降低(分别为561.69 U/L比730.78 U/L和678.82 U/L,726.44 U/L比914.21 U/L和861.86 U/L);血清NO水平明显升高,平均为63.54 μmol/L;肝组织eNOS表达显著增加,iNOS表达显著降低.结论 雌激素对于大鼠肝移植缺血再灌注损伤具有保护作用,这种保护作用一部分是通过调节eNOS和iNOS表达、提高NO水平实现的.
Abstract:
Objective To investigate the protective effects of 17β-estradiol on ischemia reperfusion injury in rat liver transplantation. Methods The rats were divided into three groups: male to male group(MG), female to female group (FG), and male to male group which were given 17β-estradiol 4000 μg/kg 24 hours before liver transplantation intraperitoneally (M + EG). Then transplantation was performed. At 6 hours after portal vein reperfusion, blood samples were obtained to determine the levels of alanine aminotransferase(ALT), aspartate aminotransferase (AST), and nitric oxide(NO). The expression of endothelial nitric oxide synthase (eNOS) and inducible nitric oxide synthase (iNOS) in liver were observed by immunohistochemistry. Results ALT and AST levels in the M+EG group were lower than those in MG and FG (561. 69 U/L vs 730. 78 U/L and 678. 82 U/L; 726. 44 U/L vs 914. 21 U/L and 861. 86 U/L). The NO level (63. 54 μmol/L) was much higher than those in the MG and FG groups. The expression of eNOS in the M+EG group was higher than those in MG and FG, while the expression of iNOS in M+EG were lower than those in MG. and FG. Conclusion 17β-estradiol can attenuate ischemia reperfusion injury in rat liver transplantation by improving the balance of eNOS and iNOS.  相似文献

目的 评价缺血后处理对大鼠肝缺血再灌注时肝细胞线粒体膜通透性转换和膜电位(△Ψm)的影响.方法 成年健康雄性SD大鼠40只,体重220~260 g,采用随机数字表法,将其随机分为5组(n=8):假手术组(S组)、苍术苷+假手术组(A+S组)、缺血再灌注组(IR组)、缺血后处理组(IPO组)和苍术苷+缺血后处理组(A+IPO组).采用阻断肝中叶和左叶60 min,恢复血流灌注6 h的方法 建立大鼠肝缺血再灌注模型.S组和A+S组仅游离肝门,不阻断血管;A+S组关腹前静脉注射苍术苷5 mg/kg;IR组制备肝缺血再灌注模型;IPO组于再灌注前行缺血后处理,再灌注1 min,缺血1 min,反复3次;A+IPO组于再灌注前静脉注射苍术苷5 mg/kg.于缺血前即刻和再灌注6 h时,采集左颈静脉血样,测定血清ALT和AST的活性.再灌注6 h时处死大鼠,取肝左叶组织,观察超微结构和细胞凋亡情况,计算凋亡指数,测定细胞色素c(Cyt c)的表达水平、△Ψm和线粒体通透性转换孔(MPTP)活性.结果 与S组比较,A+S组时血清ALT和AST的活性、凋亡指数、Cyt c表达、△Ψm和MPTP活性差异无统计学意义(P>0.05),IR组、IPO组和A+IPO组再灌注6 h时血清ALT和AST的活性、凋亡指数升高,Cyt c表达上调,△Ψm降低,MPTP活性升高(P<0.05);与IR组比较,IPO组血清ALT和AST的活性、凋亡指数降低,Cyt c表达下调,△Ψm升高,MPTP活性降低(P<0.05),肝组织病理学损伤减轻,A+IPO组各指标差异无统计学意义(P>0.05);与IPO组比较,A+IPO组血清ALT和AST的活性、凋亡指数升高,Cyt c表达上调,△Ψm降低,MPTP活性升高(P<0.05),肝组织病理学损伤加重.结论 缺血后处理可抑制肝细胞线粒体膜通透性转换,减少线粒体△Ψm的耗散,从而减轻大鼠肝缺血再灌注损伤.
Abstract:
Objective To investigate the effects of ischemic postconditioning on mitochondrial permeability transition and mitochondrial transmembrane potential(△Ψm)following hepatic ischemia-reperfusion(I/R)in rats.Methods Forty male SD rats weighing 220-260 g were randomly divided into 5 groups with 8 animals in each group:sham operation group(group S);atractyloside+sham operation group(group A+S);I/R group;ischemic postconditioning group(group IPO)and atractyloside+ischemic postconditioning group(group A+IPO).The animals were anesthetized with intramuscular injection of atropine 0.05 mg/kg.Hepatic I/R was produced by occlusion of hepatic blood flow for 60 min followed by 6 h reperfusion.In group A+S,atractyloside 5 mg/kg was injected intravenously before abdomen Was closed.In group IPO,the animals were subjected to 3 cycles of 1 min reperfusion interspersed with 1 min hepatic isehemia at the end of 60 min hepatic ischemia.In group A+IPO,atractyloside 5 mg/kg was injected intravenously before reperfusion. Venous blood samples were collected for determination of serum ALT and AST activities immediately before ischemia and at 6 h of reperfusion. The animals were then sacrificed.Their livers were removed for microscopic examination, detection of apoptosis and determination of cytochrome c (Cyt c) expression, △Ψm and mitochonerial permeability transition pore (MPTP)activity. Apoptosis index (AI) was calculated. Results There was no significant difference in serum ALT and AST activities, AI, Cyt c expression, △Ψm and MPTP activity between S and A + S groups (P>0.05). Compared with group S, serum ALT and AST activities and AI were significantly increased, Cyt c expression was up-regulated, △Ψm was decreased and MPTP activity was increased in groups I/R, IPO and A+IPO(P<0.05).Compared with group I/R, serum ALT and AST activities and AI were significantly decreased,Cyt c expression was down-regulated, △Ψm was increased and MPTP activity was decreased in group IPO(P<0.05), while no significant change was found in group A+IPO(P>0.05).Compared with group IPO,serum ALT and AST activities and AI were significantly increased, Cyt c expression was up-regulated, △Ψm was decreased and MPTP activity was increased in group A + IPO(P< 0.05).Microscopic examination showed that hepatic injury was reduced in group IPO compared with group I/R, while aggravated in group A+ IPO compared with group IPO. Conclusion Ischemic postconditioning can protect liver from I/R injury by attenuating the I/R-induced increase in MPTP opening and decrease in △Ψm in rats.  相似文献

目的 探讨前列地尔对兔肝脏缺血再灌注损伤时,有效减少肝细胞凋亡的机制.方法 将健康新西兰兔36只随机分为3组:对照组、缺血再灌注组和前列地尔组,3组分别在再灌注60min和90min时,检测血清谷丙转氨酶(ALT)、谷草转氨酶 (AST)、乳酸脱氢酶(LDH)水平.取肝中叶检测兔诱导型一氧化氮合酶(iNOS)、髓过氧化物酶(MPO)以及bcl-2、bax和Caspase-3蛋白表达;并用原位缺口末端标记法(TUNEL)染色比较各组肝细胞凋亡.结果 与对照组比较,缺血再灌注组和前列地尔组在再灌注后ALT、AST、LDH 水平均大幅上升(P<0.05);但前列地尔组兔在再灌注60、90min时ALT、AST、LDH 水平明显低于缺血再灌注组(P<0.05);与对照组比较,缺血再灌注组肝细胞bcl-2、bax、Caspase-3的表达明显增强;前列地尔组与缺血再灌注组比较表达均减弱,但仍强于对照组.TUNEL 法显示前列地尔组、缺血再灌注组与对照组比较凋亡细胞数增多,前列地尔组与缺血再灌注组比较凋亡细胞数减少;与对照组比较,缺血再灌注组与前列地尔组iNOS 与MPO的活性明显增强,前列地尔组与缺血再灌注组比较,该两者活性明显减弱.结论 前列地尔在肝脏缺血再灌注损伤时能有效地保护肝功能,减少肝细胞的损伤,其作用机制可能是通过减少细胞脂质过氧化,从而降低bcl-2、bax、Caspase-3等凋亡基因的表达.

Methods Thirty-six rabbits were made the model of liver ischemia-reperfusion injury, and randourly divided into three groups:Control group, Ischemia-Reperfusion Injury group and Alprostadil intervention group. The alanine aminotransferase (ALT) , aspartate aminotransferase (AST) , lactate dehydrogenase (LDH) were determined; Each group inducible nitric oxide synthase (iNOS) , myeloperoxidase (MPO) and bcl-2, bax, Caspase-3 and apoptosis of the hepatocyte by TUNEL were assayed at 60 and 90 min after reperfusion. Results The ALT, AST, LDH concentration in plasma in Ischemia-Reperfusion Injury group and Alprostadil intervention group were increased obviously at 60, 90 min after reperfusion and it was significantly higher than that in the Control group in the same time point (P<0.05). And the ALT, AST, LDH concentration in plasma in Alprostadil intervention group was significantly lower than that in the in Ischemia-Reperfusion Injury group in the same time point (P<0.05). The level of bcl-2, bax, Caspase-3 in the liver tissue in the Control group was smaller, but the obvious increase of the expression those was found in the Ischemia-Reperfusion Injury group and Alprostadil intervention group. Compared with those in the Ischemia-Reperfusion Injury group group, the expression of bcl-2, bax, Caspase-3 in the Alprostadil intervention group werw obviously smaller (P<0.05). The contents of iNOS and MPO in liver tissue in the Ischemia-Reperfusion Injury group and Alprostadil intervention group were significantly higher than that in the Control group (P<0.05). Conclusion Alprostadil could be used to protect liver ischemia-reperfusion injury, it could decrease oxygen free radicals generation, inhibit neutrophils aggregating and activating in the liver, thereby inhibiting expression of bcl-2, bax, Caspase-3.  相似文献

目的 探讨干细胞相关基因Oct4与Wnt/β-catenin在人肝癌组织及肝癌细胞系中的相互作用.方法 PCR法检测Oct4、Wnt/β-catenin及其他相关基因在肝癌组织、癌旁组织及正常肝组织的表达差异.使用SiRNA-Oct4沉默人肝癌HepG2细胞Oct4的表达,实时荧光定量PCR法检测Wnt/β-catenin基因表达.检测RNAi后细胞迁移以及克隆形成能力.结果 肝癌患者的上述组织中,Oct4和β-catenin在癌内表达最高,癌旁次之,正常肝最低.使用SiRNA-Oct4沉默人肝癌HepG2细胞Oct4后,Oct4表达明显下调,β-catenin、Wnt10b表现为与Oct4表达呈正相关,TCF3表达与Oct4呈负相关.RNAi后HepG2细胞的迁移能力、克隆形成能力均下降.结论 Oct4在肝癌组织内高表达而在正常肝细胞中的表达极低.RNAi后HepG2细胞的迁移能力、克隆形成能力均下降可能与Wnt/β-catenin通路功能减弱有关.
Abstract:
Objective To investigate the expression of Oct4 in liver cancer, and the interrelation of the Oct4 and Wnt/β-catenin genes in hepatocellular carcinoma( HCC) cell line HepG2. Methods RTPCR technique was used to detect the expression of Oct4 and β-Catenin in HCC specimens; RNAi was used to knock-down the expression of Oct4 in HepG2, and the change of Wnt/β-catenin related genes were detected by Real time-PCR. Results In HCC specimens, the expression of Oct4 and β-Catenin in tumor and cirrhotic liver tissues were stronger than normal liver tissues. In SiRNA Oct4 HepG2 cells, the expression of Oct4 was downregulated, and β-catenin as well as Wnt10b were in a positive correlation with Oct4, TCF3 was in negative correlation with Oct4. Clone formation and move ability of the HepG2 were downregulated. Conclusions The expression of Oct4 was higher in tumor tissues than in normal liver tissues. Silencing Oct4 by SiRNA-0ct4 in HepG2 resulted in decreased ability of clone formation and cell movement.  相似文献

目的 观察大鼠部分肝缺血再灌注损伤后切除对残肝再生的影响.方法 将75只健康雄性SD大鼠随机分为5组:肝脏左叶和中叶(约占全肝70%)切除组(Control组)、肝脏左叶和中叶缺血10min再灌注30min后切除组(I10R30组)、类推得到I60R30组、I90R30组、I90R60组.术后6、12、24h等时间点,测定再生肝重量(RLW);自动生化分析仪检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)含量;酶联免疫吸附试验(ELISA)检测血清肿瘤坏死因子(TNF)-α含量;通过免疫组织化学法检测残肝增殖细胞核抗原(Ki-67)表达.结果 术后12h,I60R30、I90R30和I90R60组RLW值分别为(1.80±0.03)%、(1.82±0.10)%、(1.87±0.05)%;Ki-67值分别为(58.35±2.18)%、(59.73±3.06)%、(62.65±2.24)%,均明显高于对照组(P<0.05).缺血再灌注干预各组ALT和AST明显高于对照组(P<0.05).术后6h和12h,I60R30、I90R30和I90R60组TNF-α明显高于对照组(P<0.05).结论 大鼠即将被切除的肝脏先缺血再灌注后切除,对残肝再生具有促进作用;诱导产生的TNF-α表达量增多是促进肝再生的原因之一.
Abstract:
Objective To investigate the effects of ischemia reperfusion injury before partial hepatectomy on liver regeneration in rats. Methods Seventy-five male healthy SD rats were randomly classified into 5 groups: group control, in which rats were only subjected to 70% hepatectomy; group I10R30, 70% liver hepatectomy after 10 min of ischemia and 30 min of reperfusion in the resected liver; By analogy, group I60R30, group I90R30 and group I90R60 were constructed. At 6th, 12th and 24th h after operation, RLW was determined; serum alanine aminotransferase (ALT) and aspartate transaminase (AST) activities were measured by using autoanalyzer; the levels of serum tumor necrosis factor (TNF)-α were determined by ELISA and the expression level of Ki-67 was detected by using immunohistochemical methods in the residual liver tissues. Results At 12th h after partial hepatectomy, the rate of RLW in group I60R30, group I90R30 and group I90R60 was (1.80±0.03)%, (1.82±0.10)%, (1.87±0.05)% respectively; the rate of Ki-67 was (58.35±2.18)%, (59.73±3.06)%, (62.65±2.24)% respectively, which was significantly higher than that in the group control (P<0.05). The levels of ALT and AST in rats with ischemia reperfusion injury were higher than in the group control (P<0.05). At 6th h and 12th h after operation, the expression levels of TNF-α in groups I60R30, I90R30 and I90R60 were significantly higher than those in the group control (P<0.05). Conclusion Ischemia reperfusion injury in the resected liver before partial hepatectomy could improve liver regeneration of the remnant liver in rats. 【Abstract】〓Objective〓To summarize the experience in treatment of the obstructive jaundice by preoperative percutaneous puncture of liver bile duct drainage. Methods〓Eight-six patients with obstructive jaundice were included and divided into two groups according to the operation procedure, 44 cases were given preoperative percutaneous transhepatic cholangial drainage(observation group) and then,.underwent internal or external drainage or radical operation,.42 cases were served as control group without preoperative percutaneous transhepatic cholangial drainage..Intraoperative blood loss, operative time,..hospital stay and complications were compared between two groups. The preoperative and postoperative liver function tests,such as total bilirubin(TB),.serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) were evaluated and compared in two groups. Results〓The operation time,.blood loss,.length of hospital stay and complication rates between observation group were lower than that in control group (P<0.05). The levels of TB, ALT, AST in 14 days after operation reduced in two groups, but were significantly decreased in observation group,.as compared to control group (P<0.05). Conclusion〓For patients with obstructive jaundice, preoperative percutaneous transhepatic cholangial drainage would shorten the length of hospital stay,.reduce complications occurrence,.and improve liver function.  相似文献

目的 探讨人脐带间充质干细胞在中度脂肪肝大鼠肝脏缺血再灌注损伤的保护作用.方法 通过高脂高胆同醇饮食建立SD大鼠中度脂肪肝模型.60只大鼠分为人脐带间充质干细胞处理组(MSC组)和对照组,每组各30只大鼠.MSC组缺血再灌注术前1 d和术后第3天分别从尾静脉注射含2×106人脐带间充质干细胞的缓冲液1.5ml.对照组则在相同时间点注射PBS缓冲液1.5ml.术后第1、4和7天(每个时间点10只),检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、肝组织中超氧化物歧化酶活性(SOD)、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)的变化.肝组织病理切片HE染色观察肝组织损伤和白细胞浸润情况.结果 术后第1、4天,与对照组相比,MSC组血清ALT、AST、TNF-α和肝组织MDA明显降低,两组比较差异有统计学意义(P<0.05).术后第7天,两组ALT、AST和TNF-α比较,差异无统计学意义(P>0.05).而SOD在术后各时间点两组间的比较差异均无统计学意义.肝组织病理检杳则发现MSC组肝细胞坏死和白细胞浸润明显较对照组减轻.结论 人脐带间充质干细胞输注能有效减轻大鼠脂肪肝缺血再灌注损伤.
Abstract:
Objective To investigate the protective effect of human umbilical cord mesenchymal stem cells (huc-MSC) on the ischemia-reperfusion injury (IRI) of moderately fatty liver in rats. Methods The rat fatty liver model was established by fat-rich diet feeding. 60 female SD rats were randomly divided into MSC-treated and control groups (n = 30). MSC-treated group was infused with MSC (2 x 106 cells resuspended in 1.5 ml of sterile phosphate-buffered saline solution, PBS) by intra-venous injection through the tail vein. The first dose was administered on day 1 before IRI, followed by another dose on day 3 postIRI. The control group was injected with sterile PBS alone at the same intervals. Blood and liver samples were collected at day 1,4 and 7 (10 rats at each time point) post-IRI respectively to test enzyme activities,biochemical and histological changes. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), superoxide dismutase (SOD), malondialdehyde (MDA) and tumor necrosis factor α (TNF-α) in liver tissue were measured. The pathological changes of liver tissue and apoptosis of hepatocytes were also assessed. Results Compared to the control group, the levels of ALT, AST, TNF-α, and MDA declined in the MSC-treated group on day 1 and day 4 post-IRI ( P < 0. 05 ), while no difference was observed on day 7 post-IRI ( P > 0. 05). There was no difference of the level of SOD between the two groups on day 1,4 and 7 post-IRI (P >0. 05). Pathological examination revealed inflammatory injury in the MSC-treated group was alleviated compared with that in the control group. Conclusion huc-MSC effects protection on the IRI of fatty liver in rats.  相似文献

Objective To investigate the expression of MCP-1 and TGF-β1 in early stage of ob-structive jaundice and explore its relation to liver correlated injury indexes in rats. Methods Fifty male Wistar rats were randomized into the control group, sham-operated group and biliary obstruction group. On day 10, serum ALT and BIL-T levels were determined in inferior caval blood and hepatic MDA was estimated in liver homogenates. Meanwhile, serum TGF-β1 level was measured by ELISA and the MCP-1 infiltration determined with immunohistochemistry. Results Serum ALT and BIL-T values were elevated. Meanwhile, the liver MDA content was increased. The positive expression of MCP-1 was augmented and serum TGF-β1 was over-expressed in the early stage after obstructive jaun-dice in rats. The hepatic MCP-1 expression had a positive correlation with the elevated ALT, BIL-T and MDA levels. However, the serum TGF-β1 content only had a positive correlation with ALT and BIL-T. Conclution Hepatic MCP-1 and serum TGF-β1 expression in the early stage after biliary tract obstruction is related to liver injury and extrahepatic cholestasis. Meanwhile, the hepatic MCP-1 ex-pression is correlated with hepatic oxidation stress. They have an important significance in liver injury in rats during the early period of obstructive jaundice.  相似文献
