Objective: To evaluate the therapeutic effects and mechanisms of Qidan granule in blemycinA5-induced pulmonary interstitial fibrosis (PIF)in rats. Methods: PIF models were established by blemycinA5-induced in rats. They were treated by Qidan granule and Hydrocortisone respectively. The pathological changes and collagen protein disposition were observed, and the expression of TGF-β, TNF-α proteins were measured by immunohistochemical technique . Results: The pulmonary alveolitis and fibrosis were alleviated remarkably in Qidan granule group compared with those in the model control group and hydrocortisone group (P <0. 01). The expression of TGF-β and TNF-α protein were higher in Qidan granule group than those in normal group , and were significantly less than those in the model control group and in hydrocortisone group (P < 0. 01). Conclusion: Qidan granule would ameliorate the pulmonary alveolitis and fibrosis. TGF-β and TNF-α might play an important role in the development of alveolitis and fibro  相似文献

目的 探讨表皮生长因子 (EGF)对作为皮肤组织工程种子细胞之一的真皮成纤维细胞中 cyclin D1和 CDK- 4表达的影响 ,以从细胞周期探讨 EGF促进真皮成纤维细胞生长的机制 .方法 用含 10 0 m L· L- 1 胎牛血清的 DMEM,加入5 0 mg· L- 1的 EGF培养 SD大鼠的真皮成纤维细胞 ,通过MTT检测、流式细胞仪分析观察细胞的生长状态 ,免疫组化检测 cyclin D1和 CDK- 4的表达 ,结合流式细胞仪分析观察细胞的周期变化 .结果  MTT检测、流式细胞仪结果都显示 5 0mg· L- 1 的 EGF能显著促进真皮成纤维细胞的生长增殖 ,免疫组化结果显示 ,二者一致 .结论  5 0 mg· L- 1 的 EGF对真皮成纤维细胞的生长有极大的促进作用 ,促进了细胞周期蛋白 cyclin D1和 CDK- 4的表达 ,使细胞 G1期变短 .  相似文献

Summary: In order to study the role of annexin Ⅱ , a recombinant expression vector, pZeoSV2 (+)ANN Ⅱ , containing the annexin Ⅱ cDNA, was developed. The 1.1-kb-length annexin ⅡeDNA was inserted into a expression vector, PZeoSV(+) and transfected into HL-60 cells which had low baseline expression of Ann- I. pZeoSV(+) ANN I was analyzed by restriction mapping and the Ann-I sequence identified. The ability of the transfected cells, non-transfected and mock-transfected cells to stimulate t-PA-depend plasminogen activation was compared. The results showed that HL-60 with pZeoSV (+)ANN I transfection could significantly increase the plasminogen activation (8. 9± 1.2 U) in vitro with the difference being significant as compared with non-transfected (1.5±0. 4 U) and mock-transfected cells (4.2±0. 9 U), respectively. Antiannexin Ⅱ oligonucleotides significantly inhibited the binding ability of t-PA and plasminogen to annexin Ⅱ , and obviously reduced the plasminogen activation in vitro. The above findings showed human umbilical vein endothelial cells (HUVECs) treated with sense or missense oligonucleotides indicated no significant change in binding of t-PA and PLG. Treatment of HUVECs with antian nexin Ⅱ oligonucleotides could significantly reduce the plasminogen activation by 2.4± 0. 3 U as compared with sense oligonucleotide group in binding of t-PA and PLG. These results, therefore,suggest that Ann- Ⅱ can bind plasminogen and participate in the stimulation of t-PA-dependent ac tivation of plasminogen, and that interference with Ann- Ⅱ mRNA by antisense oligonucleotidemay be a new strategy for the therapy of bleeding in patients with hyperfibrinolysis.  相似文献

The toxic effect of fertilizer Diammonium phosphate resulted in alterations of 5‘-Nucleotidase activity of tissues-liver,kidney and muscles of fish C.batrachus at varying intervals and exposures.Alterations in 5‘-Nucleotidase activity of body organs gave an idea of the toxicity caused by the fertilizer.Thus the enzyme 5‘-Nucleotidase can be used to monitor the pollution in aquatic ecosystem.  相似文献
