Background: Hypotrophic newborns demonstrate a reduced blood platelet count and therefore may have haemostasis disorders. The antigen density of CD62P on the surface of platelets may indicate the activation of blood platelets.Materials and methods: We have studied 31 hypotrophic newborns, and have divided these into two groups: weighing less than the 5th centile and between the 5th and 10th centiles. The antigen density of CD62P was calculated according to the procedure recommended by DAKO QIFIKIT.Results: Hypotrophic newborns exhibited a median 22?000 of CD62P per platelet. There is a distinct gender difference (female median 29?768, male median 19?044 of CD62P per platelet, p?=?0.001). Newborns below the 5th centile demonstrated a median 30?000 of CD62P per platelet, whereas between the 5th and 10th centiles – a median 18?700. A negative correlation (R?=??0.53, p?=?0.002) was found between the antigen density of CD62P and birth weight.Conclusion: Hypotrophy affects the expression of CD62P. There is a negative correlation between the antigen density of CD62P and birth weight. 相似文献

目的:研究血小板膜CD62P表达率在冠状动脉介入治疗(PCI)术前和术后的变化。方法:检测60例急性心肌梗塞患者行PCI术前、术后即刻和术后7d的血小板膜CD62P表达率。结果:血小板膜CD62P表达率术前为(17.83±7.67)%、术后即刻降为(13.82±4.85)%.术后7d再降为(10.28±3.24)%.术前与术后即刻的CD62P表达率及术后即刻与术后7d的CD62P表达率均有显著差异(P均〈0.05)。结论:PCI术后血小板膜CD62P表达率较术前明显下降,重建闭塞冠脉血运和抗血小板治疗能减轻心血管事件中的炎症反应。 相似文献

Hemostasis in preterm newborns is characterized by low reserve functional capacity with special reference to the presence of such risk factors as asphyxia or infection. Platelets play vitally important role in hemostasis. Expression of CD62P is a marker of stimulated or activated blood platelets. The study involved a group of 16 preterm newborns, five girls and 11 boys. DAKO QIFIKIT was applied to calculate the number of these antigens. The mean CD 62P expression was found to be 23,792 per platelet. Correlation was found between antigen density and gestational age r = 0.954, p = 0.01. Evident deficit of P-selectin on the surface of platelets in preterm newborns may be at least in part responsible for platelet dysfunction, with special reference to interaction between circulating leukocytes and combating infection. 相似文献

目的 探讨血小板α颗粒膜糖蛋白CD62P和溶酶体膜糖蛋白CD63的表达与急性脑梗死的关系.方法 用流式细胞仪和单克隆抗体标记法检测61例急性脑梗死患者CD62P和CD63水平,并与59例正常对照组比较.结果 急性脑梗死患者CD62P和CD63的表达明显高于正常对照组(6.26%±1.68%比2.32%±1.14%和6.14%±1.89%比2.88%±1.15%,P<0.01).结论 急性脑梗死患者血栓的形成与血小板的活化密切相关,且活化血小板的检测可以作为脑梗死早期诊断一项较特异的指标. 相似文献

Measurements have been made of the antigen density of P-selectin (CD 62P) on the surface of platelets of healthy term newborns in relation to sex and in different umbilical vessels. The effect of thrombin or ADP on the density of CD 62P has been determined. The study was performed in 18 healthy term newborns, 10 girls and eight boys. Dako QIFIKIT was applied to calculate the number of these antigens. No differences were observed in the number of antigens of CD 62P on newborn platelets between the umbilical vein and artery in both sexes. On the other hand, boys demonstrated 31,245 antigens/plt, girls 26,397/plt (P = 0.045). A statistically significant increase in the expression of CD 62P was also observed following activation of thrombin in both sexes and in the umbilical vein and artery. ADP as a weaker agonist demonstrated less changes. A high number of the CD 62P antigens on the platelets of newborns may be interpreted as an effect of increased thrombinogenesis during birth. It can be suggested that this process is more pronounced in boys than girls. 相似文献

目的:研究血清CD62P在急性心肌梗塞(AMI)患者行经皮冠状动脉介入术(PCI)前、后的表达变化。方法:60例AMI患者行PCI干预治疗,分别检测患者术前,术后即刻和术后7d的血清CD62P分子的表达,并对其结果进行比较分析。结果:PCI术后即刻AMI患者血清CD62P分子表达较术前显著降低[(42.68±2.35)ng/ml:(45.82±3.66)ng/ml,P0.05],术后7d血清CD62P分子表达[(38.82±3.14)ng/ml]较术后即刻又有显著降低(P0.05)。结论:急性心肌梗塞患者行经皮冠状动脉介入术后血清CD62P表达较术前明显下降,说明及时开通闭塞冠脉和有效的抗血小板治疗能减轻急性冠脉综合征的炎症反应。 相似文献

The effects of varying concentrations of platelet-activating factor (PAF), arachidonic acid (AA) and collagen on the expression of the platelet activation markers CD63 and CD62P were assessed in 10 normal subjects using flow cytometry. CD63 expression was significantly greater than CD62P expression, with PAF (80 nM) inducing mean maximum CD63 expression of 32.9 ± 6.4% and mean maximum CD62P expression of 5.5 ± 1.8%. AA (1 mM) induced maximum CD63 expression of 37.7 ± 7% and maximum CD62P expression of 9.3 ± 1%. Collagen (2-80 pg/ml) induced minimal expression but 800 pg/ml induced mean CD63 expression of 33.1 ± 4.1% and mean CD62P expression of 6.1 ± 0.8%. Greater CD63 and CD62P expression were induced by phorbol myristate acetate (1.6 pM, 70.9 ± 11% and 69.4 ± 9.9%, respectively) and thrombin (0.1 U/ml, 70.7 ± 9.3% and 73.5 ± 5.4%, respectively). With PAF and collagen only one platelet population was detected whereas with 1 mM AA two populations were observed. These results indicate that expression of platelet adhesion receptors depends on the nature and concentration of agonist and that subpopulations of platelets may exist. Importantly, PAF concentrations inducing moderate CD63 and CD62P expression did not induce platelet aggregation, suggesting that platelets can be activated independently of aggregation. 相似文献

目的 探讨2型糖尿病合并冠心病患者血小板表面CD62P和CD40L的表达水平及其和血清高敏C反应蛋白的关系.方法 选择单纯冠心病患者34例、单纯2型糖尿病患者33例、2型糖尿病合并冠心病患者30例及对照者30例,采用流式细胞术分别检测血小板表面CD62P和CD40L的表达水平,并与血清高敏C反应蛋白作相关分析.结果 2型糖尿病合并冠心病组血小板CD62P、CD40L和高敏C反应蛋白水平较对照组、单纯冠心病组及单纯2型糖尿病组显著升高(P<0.01),血小板CD62P和CD40L与血清高敏C反应蛋白水平呈正相关(r分别为0.343和0.495,P均<0.05).结论 2型糖尿病合并冠心病患者的血小板表面CD62P和CD40L表达水平明显升高,并与血清高敏C反应蛋白明显正相关.血小板表面CD62P和CD40L的表达水平可能是预示糖尿病患者合并冠状动脉粥样硬化的重要指标. 相似文献

目的探讨血小板源性CD40配体(CD40L)、血小板P选择素(CD62P)在哮喘患者外周血中的表达及其临床意义。方法体外分离哮喘患者的血小板,用流式细胞仪测定二磷酸腺苷(ADP)诱导前后血小板源性CD40L、CD62P的表达,并与健康人比较。同步测定所有受试者的肺功能,分析与CD40L、CD62P表达的关系。结果①哮喘急性发作组(n=37)、哮喘缓解组(n=22)、健康对照组(n=13)血小板源性CD40L的阳性表达率分别为:(20.1±7.3)%、(5.2±3.1)%、(4.1±2.2)%,CD62P分别为(46.0±11.1)%、(12.6±4.8)%(、10.0±2.7)%;ADP诱导20分钟后CD40L的阳性表达率分别为:(19.7±7.6)%(、20.1±5.6)%(、25.4±12.8)%,CD62P分别为(45.5±11.4)%、(45.5±10.5)%(、43.2±15.4)%。与健康对照组、哮喘缓解组相比,哮喘急性发作组CD40L、CD62P的表达水平显著升高(P<0.05),而ADP诱导后其差异无统计学意义(P>0.05)。②健康对照组血小板源性CD40L、CD62P的表达在ADP诱导20min时达到最高水平,且CD40L、CD62P表达水平呈正相关(P<0.01)。③哮喘急性发作组FVC占预计值百分比(FVC%Pred)、FEV1占预计值百分比(FEV1%Pred)、FEV1占FVC百分比(FEV1%)较对照组显著降低(P<0.05),哮喘患者血小板源性CD40L和CD62P的表达水平与其肺功能成负相关(P<0.05)。结论哮喘急性发作患者存在外周血血小板源性CD40L、CD62P的表达增高,表达水平与其肺功能成负相关,血小板源性CD40L在哮喘的发病中起着一定的作用。 相似文献

目的 探讨不同剂量腺苷与AMP579对冠心病(CHD )患者血小板表面CD62P表达的影响.方法 选择经临床症状、心电图改变及心肌酶学确诊的CHD患者30例及正常健康者15名,采集肘静脉血,分别给予不同剂量的腺苷与AMP579,用双抗体夹心酶联免疫吸附法( ELISA)检测患者及正常人用药前后血小板CD62P表达值.结果 CHD患者的血小板CD62P的表达升高,与正常人比较差异有统计学意义(P<0.05);腺苷和AMP579均能降低CHD患者血小板CD62P的表达,分别与用药前比较差异有统计学意义(P<0.05),但同一药物在450 μmol/L、1 000 μmol/L、3 000 μmol/L 3种浓度下对CD62P表达的影响无统计学意义(P>0.05).结论 CHD的发生可能与CD62P表达升高有关;腺苷和AMP579可通过降低CD62P表达而降低血小板与内皮下基质的黏附,从而起到防止血栓的作用. 相似文献

目的探讨糖尿病合并急性冠状动脉综合征患者血小板表面血小板膜糖蛋白Ⅱb/Ⅲa纤维蛋白原受体(PAC-1)和P选择素(CD62P)的表达水平及其与血浆同型半胱氨酸(Hcy)的关系。方法选择单纯急性冠状动脉综合征患者40例、糖尿病合并急性冠状动脉综合征患者24例及对照组30例,采用全血流式细胞术结合三色荧光技术检测血小板表面PAC-1和CD62P的表达水平,并与血浆Hcy作相关性分析。结果糖尿病合并急性冠状动脉综合征患者的血小板PAC-1、CD62P和血浆Hcy水平较对照组明显升高(P0.05),同时血小板CD62P表达水平较单纯急性冠状动脉综合征患者明显升高(P0.05);血小板PAC-1、CD62P与血浆Hcy水平呈正相关(r分别为0.441和0.408,均P0.05)。结论糖尿病合并急性冠状动脉综合征患者血小板表面PAC-1、CD62P表达水平明显升高,并与血浆Hcy呈明显正相关。血小板表面PAC-1和CD62P的表达水平可能对预示糖尿病患者发生血栓事件具有重要的意义。 相似文献

目的 探讨血小板源性CD40配体(CD40L)、血小板P选择素(CD62P)在肺癌患者外周血中的表达及其临床意义。方法 体外分离肺癌患者的血小板,采用流式细胞仪测定二磷酸腺苷(ADP)诱导前后血小板源性CD40L、CD62P的表达水平,并与健康人作比较。结果 ①肺癌组、健康对照组血小板源性CD40L的阳性表达率分别为(13.6±8.2)%、(3.9±1.7)%;CD62P分别为(62.6±20.5)%、(12.5±3.2)%;ADP诱导20min后CD40L的阳性表达率分别为(23.9±11.9)%、(26.4±9.4)%;CD62P分别为(74.9±16.6)%、(41.4±15.2)%。与健康对照组相比,肺癌组CD40L、CD62P的表达水平显著升高(P〈0.01);而ADP诱导后,肺癌组CD62P表达水平明显高于对照组(P〈0.01),而CD40L与对照组相比差别无统计学意义(P〉0.05)。②ADP诱导前后,不同病理类型肺癌患者血小板源性CD40L、CD62P表达水平的差别均无统计学意义(P均〉0.05)。③ADP诱导前后,不同期别肺癌患者血小板源性CD40L、CD62P表达水平的差别有统计学意义(P均〈0.01)。有远处转移患者血小板源性CD40L、CD62P表达水平明显高于无远处转移患者(P〈0.05)。④肺癌患者血小板源性CD40L、CD62P表达水平在ADP诱导20min时达到最高水平,且CD40L、CD62P表达水平呈正相关(r=0.803,P〈0.01)。结论 肺癌患者存在外周血血小板源性CD40L、CD62P表达水平的增高,且随病情的进展CD40L、CD62P的表达增高,因此检测血小板源性CD40L、CD62P表达水平对研究肺癌的临床分期、预后判断有一定的参考价值。 相似文献

Human vasoactive intestinal peptide (VIP) and epoprostenol (prostacyclin) have vasodilatative effects in the pulmonary circulation. Both VIP and epoprostenol are successfully used to treat pulmonary hypertension in humans and experimental animal models. The positive effects of epoprostenol on the course of this disease are achieved through vasodilatation and inhibitory effects on platelet activity. Since VIP also binds specifically to platelets, we compared the in vitro effects of VIP and epoprostenol on platelet P-Selectin (CD62P) expression and primary haemostasis. Anti-aggregative effects of VIP (10?6?mol and 10?8?mol) and epoprostenol (50, 5 and 0.5?ng/ml) on platelets were determined by agonist-induced CD62P expression and in vitro bleeding time (PFA-100? system). Blood from healthy individuals was either incubated with epoprostenol, VIP or saline control and was analysed by whole blood flow cytometry and the PFA-100?. Prior to flow cytometric analysis, the platelets were stimulated with either arachidonic acid (AA) or adenosine diphosphate (ADP). Whole blood flow cytometry analysis showed that epoprostenol inhibited dose-dependently agonist-induced CD62P expression, whereas VIP did not inhibit CD62P expression. PFA analysis revealed substantial closure time prolongation by epoprostenol and again no effects of VIP. These results indicate that VIP, in contrast to epoprostenol, has no effect on platelet CD62P expression and primary haemostasis. 相似文献

Effect of tumor stage and nephrectomy on CD62P expression and sP-selectin concentration in renal cancer 总被引:4,自引:0,他引:4
The CD62P receptor and its soluble form sP-selectin is a marker of platelet (PLT) activation, and constitutes a ligand for CD24 antigen of neoplastic cells and tumor stroma components. The aim of the study was to evaluate the relationship dynamics of the percentage of CD62P+ platelets, the level of the receptor expression and the concentration of soluble form of sP-selectin in renal cancer. Examinations were performed before and after nephrectomy in patients with renal cancer (group A - 25, T2N0M0; group B - 27, T2N1-2M0) and in control group (C - 24 subjects). The two groups A and B showed an increased subpopulation of CD62P+ platelets (p<0.01) and elevated sP-selectin concentration (p<0.001) before and after nephrectomy. Although following nephrectomy sP-selectin concentration decreased markedly, it was still higher 3 months after the procedure compared to control group (p<0.05). Following nephrectomy, however, no statistically significant differences were found in the % of CD62P+ platelets and the receptor expression. Greater dynamics of changes before and after nephrectomy in the percentage of CD62P+ platelets (B1:B2 p<0.05) and the receptor expression (B1:B3 p<0.001) was observed in patients with local lymph node involvement (group B) while sP-selectin concentration was similar in both groups. Nephrectomy did not normalize intravascular activation of PLT and TNM had no significant effect on the expression of CD62P and concentration of sP-selectin. 相似文献
