Two experiments were carried out in which rats were offered diets containing different amounts and types of dietary fibre, i.e. commercial stock diet and three semi-purified diets containing no fibre, 200 g wheat bran or 200 g pectin/kg. Dietary inclusion of fibre, and especially pectin, stimulated large bowel fermentation, as indicated by caecal hypertrophy and reduced caecal pH. After 3 weeks, mucosal:serosal zinc transfer and Zn accumulation by tissue were measured using the everted-gut-sac technique. In Expt 2, incubations were carried out in the presence and absence of 0.25 mM-ouabain to assess the importance of transfer by Na+,K+-ATPase-dependent mechanisms, and some observations on glucose transport were also made. Ouabain reduced rates of transfer of both Zn and glucose and also tissue Zn accumulation. There were no significant differences in rates of Zn transfer by everted sacs from duodenal, ileal and colonic sites, but accumulation of Zn by tissue was a more important fate than transfer across the serosal surface, and accumulation by duodenal tissue was approximately twice as great as by other tissues. Mucosal:serosal transfer of glucose by ileal tissue was much more sensitive to ouabain than was Zn transfer. Previous diet appeared to alter the capacity of the intestinal tissue to transfer Zn, with the highest rates of transfer being by colonic tissue from pectin-fed rats.  相似文献

One way in which regular consumption of dietary fibre could be related to the eventual development of coronary and other atherosclerotic disease is via the plasma·total or low-density lipoprotein cholesterol. Short-term human experiments under a variety of conditions have shown that wheat fibre does not affect plasma total cholesterol. However pectin and guar gum lower plasma cholesterol in man. Other viscous types of ‘dietary fibre’ and vegetable foods rich in pectin would repay similar investigation. There is clearly a dissociation between the effects of different types of dietary fibre on plasma lipids and on colonic function.  相似文献

There is considerable loss of DM during wet heat treatment of vegetables, leading to an increase in dietary fibre. Correction for the loss of DM indicates that the effects on total dietary fibre are minor. There is, however, depolymerization of the dietary fibre polysaccharides. The degradation is related to the severity of the heat treatment. Souring, freezing and mild microwave treatment have no effects. The viscosity is in general related to the extent of polymerisation. Microwave treatment has different effects on various cultivars of green beans, and the addition of salt (NaCl and CaCl2) to the boiling water changes the physico-chemical properties of soluble fibre in carrots, depending on the cation. The higher viscosity of the soluble fibre in raw carrots may partly explain the lower glucose and hormonal responses observed in healthy subjects when compared with blanched and microwave-cooked carrots. In studies on rats the amount of butyric acid in the distal colon has been shown to be higher with dietary components containing high amounts of resistant starch. Further, the fermentability is lower and the butyric acid concentration higher with composite foods than with the corresponding purified fibre fractions. In human studies the faecal concentration of butyric acid has been shown to increase in patients with ulcerative colitis when [beta-glucan-enriched oat bran (20 g fibre) is added to the diet for 12 weeks. The present study was undertaken to explore the effect of apple pulp on weight and composition of faeces. This material is rich in dietary fibre (DF; 620 g dry matter/kg) and contains appreciable amounts of polyphenols. Recent reports indicate that both condensed tannins (CT) and soluble polyphenols form cross-links with protein and inhibit digestive enzymes, affecting the protein digestibility, and may produce a stimulation of endogenous nitrogen excretion. Two groups of male Wistar rats were fed on either a control diet free of DF or a diet containing 100 g apple pulp DF/kg during 7 d after a 4 d adaptation period. Body-weight and food intake were monitored daily and faeces and urine were collected once daily. DF, water content and polyphenolic compounds were measured in faeces, and N content in both faeces and urine. Faecal weight increased in the fibre group by 280 and 240% when compared with wet and dry faecal weights of animals fed on the fibre-free diet. Soluble dietary fibre (SDF) excreted in faeces was 10.9% of the SDF ingested, which suggested a low resistance to fermentation of this fraction. Of the insoluble DF, 43% of the ingested fibre was fermented. Polyphenols were degraded in the intestinal tract. Of the ingested CT, 68.6% was recovered in faeces, while the soluble polyphenols were extensively degraded (85.7% of that ingested). On the other hand, a higher faecal N excretion was observed for the fibre-fed group, suggesting a decrease in the digestibility of the dietary protein and lower apparent digestibility of the dietary protein and lower apparent digestibility and N balance indices.  相似文献

1. Forty-eight male rats were fed sequentially for 14 d periods on diets containing different fibre contents. 2. One of the high-fibre diets was a commercial pelleted diet. The other was a low-fibre diet supplemented with 200 g wheat bran/kg. 3. At the end of each feeding period eight rats were killed. Liver microsomal cholesterol 7 alpha-hydroxylase (EC 1.14.1.-) activity and bile acid content of small intestine and colon were determined. 4. The different diets did not significantly alter the total intestinal bile acids, but affected the distribution and qualitative pattern in the colon and small intestine. 5. On the high-fibre diets deoxycholate, and hyodeoxycholate tended to be increased. 6. On the low-fibre diets the alpha, beta- and omega-muricholic acids tended to be increased. 7. Liver microsomal cholesterol 7 alpha-hydroxylase activity was lower in rats on the low-fibre and bran-supplemented low-fibre diets compared with that in rats fed on the commercial pelleted diet.  相似文献

Aims : This study aimed to compare the effect of advice to reduce both dietary fat and sugar, with the effect of advice to reduce dietary fat only, on dietary intake, body weight and body composition in normal to moderately overweight Scottish men. Methods : Subjects were randomly assigned to a control Group 0 (no advice), Group 1 (advice to reduce fat and sugar) or Group 2 (advice to reduce fat only). Dietary intakes, body weight, BMI, % body fat and waist:hip ratio were measured throughout the 6-month study period. Results : Both groups 1 and 2 significantly decreased percentage energy from fat after 6 months. Group 1 decreased percentage energy from added sugar and non-milk extrinsic sugar (NMES) from baseline to 6 weeks but there was no difference from baseline at 6 months. A small decrease in percentage body fat was observed in Group 1 (0.8%) without weight loss, whereas a larger decrease in percentage body fat was observed in Group 2 (1.4%) with a weight loss of 1.2 kg at 6 months. Conclusion : There may be no advantage in advising individuals to reduce sugar intakes in terms of promoting a 'healthy diet' or with respect to maintaining a healthy body weight status.  相似文献

The objective of this study was to determine the chemical composition of 28 white and coloured grain amaranth (Amaranthus spp.) genotypes. Neutral detergent fibre (NDF) concentration was greater while strach concentration was lower for coloured seeds genotypes than white seeds genotypes. Total dietary fibre followed a similar trend to that observed for NDF. Total tannin concentrations ranged between 20.7 and 0 g/kg with total and hydrolysed tannin concentrations being higher for white than for coloured seeds genotypes. Coloured seeds genotypes contained higher Mg and Ca concentrations than white seeds genotypes. However, seed colour had no influence on K, Na and P concentrations. Copper and Fe were the most variable micro-minerals in the evaluated genotypes with no significant effect of seed colour on the concentration of either mineral.  相似文献

In this study the effect of calcium absorption on some physical properties and composition of rat femurs was evaluated, comparing rats fed with raw whole corn (RC), tortillas made from extruded masa with 0.25% lime content (TEWL) and without lime (TE), and nixtamal tortillas (NT). The diets were formulated to contain the same amount of protein, oil, fiber, vitamins and minerals other than calcium. In all diets 0.20% calcium was added. At the end of the trials, the femurs were extracted, weighed and measured for ash, calcium and phosphorus content, some physical dimensions, and the crystallinity percentage. The femurs of rats fed with TEWL and NT were heavier, thicker, longer and had higher calcium content. On the other hand, the force required to break the femur of rats fed on ETWL and NT was 1.25 kg greater than that required to break the femurs of rats fed with RC. Higher crystallinity percentage values were observed in the femurs of the rats fed with NT (37.66%) and TEWL (36.98%) as compared to a 30.31% value obtained with the RC.  相似文献

In rats, the effects of a 4-week supplementation of a fibre-free elemental diet with 100 or 200 g Plantago ovata seeds/kg was compared with that of the husks and wheat bran. The seeds increased faecal fresh weight up to 100%, faecal dry weight up to 50% and faecal water content up to 50%. The husks, at the high concentration only, were more effective and wheat bran less effective. Length and weight of the small intestine were not greatly affected by the seeds, but both variables increased significantly in the large intestine. The husks had more pronounced effects, especially in the small intestine, and wheat bran almost no effect. Faecal bacterial mass as estimated from the 2,6-diaminopimelic acid output was increased to the greatest extent by the seed-containing diet and by the high concentration of husks, but to a lesser extent by wheat bran. Faecal and caecal protein content was enhanced by the seeds and wheat bran, but to a lesser extent by the husks. Total acetate in caecal contents or faeces was highest on the seeds and husks diet and not elevated by wheat bran. Total faecal bile acid excretion was stimulated and beta-glucuronidase (EC 3.2.1.31) activity reduced by both Plantago ovata preparations, but not by wheat bran. Mucosal digestive enzyme activities were inhibited to different degrees by all dietary fibres in the jejunum, and sometimes activated in the ileum. These results suggest that Plantago ovata seeds are a partly-fermentable dietary fibre supplement which increases stool bulk; metabolic and mucosa-protective effects are also probable.  相似文献

One hundred nineteen vegetarian preschool children whose parents volunteered were studied in their homes. Length, weight, triceps and subscapular skinfolds, and head circumference were measured. Information on extensiveness of avoidances of animal food groups, parental dietary group, birth weight, parental heights, and the child's status with respect to breast feeding at the time of measurement was obtained by parental report. All infants under six months old were breast fed. More of the children's measurements were below the Harvard 50th percentiles for length and weight than would have been expected after six months of age, but not until then. Vegetarian children were also leaner (triceps and subscapular skinfolds) after six months of age than would be expected from Tanner-Whitehouse standards. Head circumferences were similar to standards and within normal limits at all ages. An association between smallness, lightness, and leanness was found among the vegetarians, with these characteristics being more pronounced among the older, non-breast fed, macrobiotic children on diets with limited variety in terms of animal food. While smallness and low weight-for-length were associated with leanness, largeness and high weight-for-length did not imply obesity in these subjects.  相似文献

Almond skins and blanch water are underutilized by-products of the almond processing industry. Nevertheless, they contain exploitable components that may contribute to the health benefits associated with almond consumption. We have compared natural almond skin powder (NS) prepared by a novel freeze-thawing method with blanched almond skin powder (BS). Microstructural studies were carried out, and we analyzed both types of almond skin for phenolic compounds (by HPLC), lipids (by solvent extraction), proteins (by micro-Kjeldahl), and fibre content (by the enzymatic-gravimetric AOAC method). Antioxidant activity (by measuring the reduction of the 2,2-diphenyl-1-picrylhydrazyl radical) was also monitored. We identified a combination of flavonols, flavan-3-ols, hydroxybenzoic acids and flavanones in NS, BS and in industrially obtained blanch water (BW). As expected, the total phenolic content was higher in NS compared to BW and BS, although the latter showed high antioxidant properties. Almond skins had high fibre content as well as significant amounts of lipid; both of these components may be relevant to fermentation in the large intestine. In addition, the processing of almond skins and blanch water clearly has economic potential for lowering the environmental impact of waste fill and pollution.  相似文献

The effects of taurine on serum cholesterol levels and hepatic cholesterol 7alpha-hydroxylase activity (CYP7A1) were studied in rats fed cholestyramine or high amounts of sodium cholate in order to alter the intestinal pool of bile acids. Rats were fed a diet supplemented with 1% cholesterol and 0.25% sodium cholate (high cholesterol, control; C), and C supplemented with 4% cholestyramine (CH) or 0.75% sodium cholate (BA) for 14 d. Taurine groups were fed the diet supplemented with 3% taurine (CT, CHT and BAT). Compared to rats fed C and BA diets, serum cholesterol levels were significantly reduced in rats fed CT and BAT diets, but a significant reduction of serum cholesterol by taurine feeding was not observed in the CHT group as compared to the CH group. An increase in hepatic CYP7A1 activity due to taurine intake was observed in the CT and BAT groups. However, the simultaneous administration of cholestyramine and taurine (CHT group) did not increase hepatic CYP7A1 activity compared the intake of cholestyramine only (CH group). A significant increase in fecal bile acid excretion due to taurine intake was found only in rats fed the CT diet. In conclusion, it is suggested that taurine facilitates hepatic CYP7A1 activity regardless of the enlarged intestinal pool of bile acids due to increased intake of exogenous bile acid, and then reduces the serum cholesterol concentration.  相似文献

Feeding mineral-deficient diets enhances absorptive efficiency as an attempt of the body to compensate for the lack of an essential nutrient. Under certain circumstances, it does not succeed; and nutritional deficiency is produced. Our hypothesis was that inulin-type fructans (ITF), which are known to affect mineral absorption, could increase Ca and Fe bioavailability in Ca- and Fe-deficient rats. Male Wistar rats (n = 48, 4 weeks old) were assigned to 1 of 8 groups derived from 2 × 2 × 2 factorial design with 2 levels of added Fe (0 and 35 mg/kg), Ca (0 and 5 g/kg), and ITF (0 and 100 g/kg) for 33 days. The Fe status (hemoglobin, serum Fe, total Fe-binding capacity, transferrin saturation, liver minerals) was evaluated. Tibia minerals (Ca, Mg, and Zn), bone strength, and histomorphometry were determined. In nondeficient rats, ITF supplementation did not affect Fe status or organ minerals, with the exception of tibia Mg. Moreover, ITF improved bone resilience and led to a reduction in eroded surface per body surface and number of osteoclasts per area. In Ca-deficient rats, ITF increased liver (Fe and Zn) and tibia (Zn) mineral levels but impaired tibia Mg, yield load, and resilience. In conclusion, ITF worsened the tibia Mg levels and elastic properties when supplemented in Ca-deficient diets. In contrast, although bone Ca was not affected in nondeficient rats under the present experimental conditions, bone quality improved, as demonstrated by a moderate reduction in femur osteoclast resorption and significant increases in tibia Mg content and elasticity.  相似文献

ObjectivesTruly ketogenic rodent diets are low in carbohydrates but also low in protein. The aim of this study was to differentiate effects of ketosis, low carbohydrate (LC) and/or low-protein intake on energy and nitrogen metabolism.MethodsWe studied the nitrogen balance of rats fed LC diets with varying protein contents: LC diets consisted of 75/10, 65/20 and 55/30 percent of fat to protein (dry matter), respectively, and were iso-energetically pair-fed to a control (chow) diet to 12-wk-old male Wistar rats (n = 6 per diet).Previous studies demonstrated only LC75/10 was truly ketogenic. Food, fecal, and urine samples, as well as carcasses were collected and analyzed for heat of combustion and nitrogen (Kjeldahl method). Blood samples were analyzed for plasma protein, albumin, and triacylglycerol.ResultsAll LC groups displayed less body weight gain, and the degree of reduction was inversely related to digestible crude protein intake (daily weight gain compared with chow: LC75/10: −50%; LC55/30: −20%). Nitrogen excretion by urine was related to digestible protein intake (chow: 0.23 ± 0.02 g nitrogen/d; LC75/10: 0.05 ± 0.01 g nitrogen/d). Renal energy excretion was closely associated with intake of digestible crude protein (r = 0.697) and renal nitrogen excretion (r = 0.769). Energy-to-nitrogen ratio in urine was nearly doubled with LC75/10 compared with all other groups. Total body protein was highest with chow and lowest with LC75/10. Rats fed with LC75/10 displayed features of protein deficiency (reduced growth and nitrogen balance, hypoproteinemia, depletion of body protein, and increased body and liver fat), whereas the effects with the non-ketogenic diets LC65/20 and LC55/30 were less pronounced.ConclusionThese results suggest that truly ketogenic LC diets in growing rats are LC diets that are also deficient in protein for growth.  相似文献

In order to investigate the relationship between dietary amino acids and protein, and activities of intestinal sucrase [EC 3.2.1.26] and leucine aminopeptidase [EC 3.4.11.1, LAPase] in rats, the effect of supplementation of amino acids into a protein-free diet and a low casein diet containing sucrose as the carbohydrate source on these enzyme activities was studied. The segmental weights of the small intestine and its mucosa of rats fed the protein-free diet supplemented with L-methionine or with L-methionine and L-threonine at 0.1 or 0.2% levels were significantly higher than those of rats fed the protein-free diet or one supplemented with L-glutamic acid, but there was no difference in the segmental activities of the sucrase and LAPase among rats fed these diets. On the other hand, the supplementation of methionine or methionine plus threonine to the 5% or 10% casein diet produced remarkable increases in the segmental weights of the small intestine and its mucosa as well as in the segmental activities of the sucrase and LAPase. There was no difference between the segmental sucrase activity of rats fed the 10% casein diet supplemented with 0.2% methionine ad libitum and that of rats fed this diet under restricted feeding conditions, although the segmental LAPase activity was affected by the amount of food consumed.  相似文献

To test a possible influence of dietary fibre on intestinal sugar uptake, rates of absorption of 10 mM-D-[U-14C] glucose and 10 mM-D-[U-14C] xylose were measured in either jejunum or (distal) caecum, by in vivo lumen perfusion, in immature female fowls preconditioned to a standard diet containing (g/kg) either 0, 100, 200 or 400 added dried grass, 200 powdered cellulose, or 200 grass with a polysaccharidase enzyme supplement. When birds were killed after perfusion, dimensions of (unperfused) parts of their alimentary tracts were determined, and recoveries of 14C radioactivity in some body tissues were compared with measured activities absorbed. On average, absorption of glucose was 1.9 and 1.2 times faster than xylose in jejunum and caecum respectively, although these differences varied with dietary treatment and order of perfusion. Increasing grass in the diet caused significant changes in xylose absorption rate in both jejunum and caecum, but only when it was perfused before glucose. With any one sugar and intestinal segment, mean rates of absorption were correlated positively with corresponding mean rates of fluid loss from perfusate. Although their influence on sugar absorption was not well defined, the dietary fibre treatments had more pronounced effects on gross dimensions of parts of the alimentary tract and, hence, potentially on total rates of absorption. Compared with the basal diet, addition of 100, 200 or 400 g grass/kg or 200 g cellulose/kg caused significant increases in small intestine length while 200 g grass/kg with supplementary enzyme did not, and combined caecal length increased with the 400 g grass/kg and the supplementary enzyme treatments. Absorbed 14C activity was recovered in plasma after jejunal perfusions but not caecal ones, whereas it was recovered in liver and in the flushed perfused segment after both types of perfusion. Since there was overlap in absorption rates between jejunum and caecum, this result suggests that the liver may be able to distinguish and treat differently compounds absorbed in the two regions.  相似文献

Although the effects of dietary fibre in a normal diet on intestinal function are well known, the effects of supplementing enteral diet with fibre on intestinal function are not so clear. The aims of the present study were two fold: firstly to compare intestinal function during ingestion of a self selected diet and fibre free polymeric enteral diet and secondly to investigate the effect of adding the fibre source, soy polysaccharide to the same polymeric diet, on bowel function. Six healthy subjects were randomly assigned to 3 treatment periods (7 days) of a self selected diet (SSD), 2 l polymeric enteral diet (ED), or 2 l of the same polymeric enteral diet supplemented with 20 g/l of soy polysaccharide fibre (SPED). Parameters measured were bowel frequency/day, mean daily wet stool weights and whole gut transit time. Highest stool wet weights (g/24 h +/- SEM) were obtained from subjects consuming a SSD (180.3 +/- 30.6). This was significantly higher (p < 0.02) than those consuming an ED (81.9 +/- 14.5) but not a SPED (123 +/- 22.3). Whole gut transit (h +/- SEM) on a SSD (47 +/- 5.9) was significantly (p < 0.05) quicker than on an ED (73 +/- 4.9), but not significantly different from those ingesting a SPED (54 +/- 5.5). Bowel frequency/day (+/- SEM) in subjects consuming a SPED (1.0 +/- 0.1) was significantly (p < 0.02) higher than in subjects ingesting an ED (0.8 +/- 0.1) but was not significantly different when compared to a SSD (1.0 +/- 0.2). We conclude that ingestion of an ED results in significantly longer whole gut transit time, significantly decreased daily stool wet weights and decreased bowel frequency when compared to a SSD. The ingestion of 30 g of soy polysaccharide in a polymeric enteral diet not only tends to normalise whole gut transit time and daily stool wet weights, but also significantly increases bowel frequency when compared with values seen during the consumption of a fibre free polymeric enteral diet.  相似文献

1. Five pigs initially of 40-50 kg live weight were fitted with simple gastric cannulas which permitted complete evacuation and sampling of gastric digesta once daily. 2. The effects of addition of four types of dietary fibre (wheat bran (WB; 40 g/kg), sodium carboxymethylcellulose (CMC; 40 g/kg), high-methoxy citrus pectin (Pe; 40 g/kg) and granulated guar gum (G; 40 g/kg] on gastric emptying of a semi-purified diet during 4 h following a meal were measured. 3. Each of the test diets and the control diet (C) were given to each pig for 1 week using a 5 x 5 Latin-square arrangement. Digesta were collected before and 0.5, 1, 2 or 4 h after feeding on the last 5 d of each week. 4. The mean gastric pH was not significantly affected by diet except 2 h after feeding (CMC higher than C) and 4 h (Pe, G and CMC higher than C). 5. Compared with diet C, the rate of gastric emptying of digesta was significantly slower for diet G, 1, 2 and 4 h after feeding, and 2 and 4 h after feeding for diet CMC. 6. The rates of gastric emptying of digesta components were not significantly reduced by dietary fibre except for dry matter (DM) (diet CMC 2 h and diet B 4 h after feeding), total nitrogen (TN) (diet G 2 h after feeding) and total glucose (diet Pe 2 h after feeding). 7. There were no significant effects of diet on trichloroacetic-acid-soluble N:TN. 8. When gastric emptying was expressed in terms of half-time (T50) values, significant increases (compared with diet C) were found for digesta (diets G and CMC), DM (diet WB) and TN (diet G). 9. The apparent viscosity of the gastric digesta was significantly higher when diets Pe, G, and CMC were given than diets C or WB. Diets Pe and CMC were very viscous in the meal before ingestion, but diet G was not; its high viscosity developed after it had reached the stomach. 10. It is concluded that although those types of dietary fibre which increased meal or gastric viscosity reduced the rate of gastric emptying of digesta, this effect was confined to the liquid phase, because DM, total glucose and TN emptying were largely unaffected. The hypothesis that a reduced rate of gastric emptying may be an important determinant of the decreased rates of glucose absorption observed when such sources of dietary fibre are eaten is not supported by the results presented.  相似文献
